This study was peformed to investigate the effects on skeletal muscle recovery with aqua-exercise; swimming to take the muscle endurance for 20 days on two group of white rats which were the low extremity atrophy group(control groups) by fixed for two weeks and aqua-exercise group(experimental groups) after it. The effects was observed with light and electron microcope to measure the morphological changes of muscle fibers. The results obtained were as follow. 1. Light microscope: In the case of control groups, quadriceps fibers had been irregular alignment, decreased muscle width and the irregular alignment nuclear appeared, as it is degenerative muscle fibers. In the case of experimental groups, the fibers had been regular alignment cells and fibers. The nucleus of muscle had been normal characterized by oval shape and fiber sarcomere clearly classified. 2. Electron microscope: In the case of control groups, there were the quadriceps which was Z-line streaming phenomenon induced at the sarcomere and cells nuclear separated from basal membrane. It was not only observed the sarcomere alignment irregularly and mitochondria damaged, but also vacuoles found. In the case of experimental groups, A band, I band, H band had been clearly appeared, classified at the myofibrils of quadriceps, and electronic dense M-line found in sarcomere. There were observed satellite cells and basal laminas that usually to be appeared at the time of mitochondrial development, skeletal muscle fiber regeneration or development. This results suggest that the aqua-exercise assisted to inhibit the degenerative morphological changes of skeletal muscle cells and help to recover from abnormal states. Especially, it is considered to effect on a normal structural formation.

Purpose : The purpose of this study was to determine the changes in ultrasound imaging of the lower limbs in patients with degenerative osteoarthritis who received a total knee replacement (TKR). Methods : The participants for the study were ten patients who were to receive a total knee replacement. Measurements were taken a total of three times: before receiving a total knee replacement, and one week and two weeks after receiving a total knee replacement. The vastus medialis, rectus femoris muscle, tibialis anterior and gastrocnemius muscle were measured using ultrasound imaging. Results : Muscle thickness of the vastus medialis, rectus femoris muscle, tibialis anterior and gastrocnemius muscle had no significant interaction over time whether surgery was performed or not. White area index (WAI) and density of vastus medialis, rectus femoris muscle, tibialis anterior and gastrocnemius muscle had no significant interaction over time whether surgery was performed or not. Conclusion : In conclusion, lower quality muscles were due to a decrease in muscle fiber and an increase in fat fiber. Therefore, initial physical therapy after total knee replacement should consider this point and be designed appropriately as a therapeutic approach for total knee replacement patients.

The aim of this experiment was to observe and analysis clinical effect of the 'Sexiang shuhuo Jing' on histological change for 14days after skeletal muscle injury in rats. The gastrocnemius muscles of rats were damaged by electromechanical and serial cryosections of the damaged muscle were prepared at 1, 5, 10, 14 days after injury. Muscle sample of the both control and 'Sexiang Shuhuo Jing' treated group were prepared for histological analysis by optical microscope and electron microscopy. 'Sexiang Shuhuo Jing' treatment group's skeletal muscle recovery was much more faster than control group. After 5 day's 'Sexiang Shuhuo Jing' treatment group's basically recovery normal structure of muscle fiber. After 14 day's control group's damaged muscle were basically recovery structure of muscle fiber but still has some factor of pathological impression but in Sexiang Shuhuo Jing treatment group's can't be found that.
Purpose: The purpose of this study was to examine the effects of daily exercise before steroid treatment on mass, the type I and II fiber cross-sectional area, and myofibrillar protein content of hindlimb muscles in a rat model. Method: Adult male Sprague-Dawley rats were randomly assigned to one of three groups: a control group(n=10) that had a normal saline injection for 7days, a steroid group(n=10) that had a steroid injection for 7days, and an exercise-steroid group(n=10) that ran on the treadmill for 7days before a steroid treatment. Body weight and food intake were measured every day. At 15 days all rats were anesthetized and the soleus, plantaris and gastrocnemius muscles were dissected. Result: The exercise-steroid group showed significant increases as compared with the steroid group in body weight, muscle weight of the soleus and gastrocnemius, type II muscle fiber cross-sectional area of plantaris, and myofibrillar protein content of the soleus, plantaris, and gastrocnemius. As compared with the control group, the steroid group showed significant decreases in body weight and diet intake, muscle weight, the type II fiber cross-sectional area and myofibrillar protein content of the soleus, plantaris, and gastrocnemius muscles. Conclusion: Daily exercise before steroid treatment attenuates hindlimb muscle atrophy, with type II muscle changes more apparent than type I muscle changes.
The purpose of this study was to examine the effects of exercise on muscle weight and Type I and II fiber cross-sectional area of affected and unaffected hindlimb muscles in rats with neuropathic pain induced by unilateral peripheral nerve injury. Methods: Neuropathic pain was induced by ligation and cutting of the left L5 spinal nerve. Adult male Sprague-Dawley rats were randomly assigned to one of two groups: The Pain+Exercise (PE) group (n=21) and the Sham+Exercise (SE) group (n=20). All rats had 28 sessions of treadmill exercise at grade 10 for 30 minutes, twice/day at 10 m/min for 14 days. Body weight, food intake and activity were measured every day. At 15 days all rats were anesthetized and soleus, plantaris and gastrocnemius muscles were dissected. Muscle weight and Type I, II fiber cross-sectional area of the dissected muscles were measured. Results: The PE group showed significant increases (p<.05), as compared to the SE group for body weight and total diet intake, muscle weight of the unaffected soleus and plantaris, and in Type I and II fiber cross-sectional area of unaffected three muscles and affected plantaris. Conclusion: Exercise for 14 days attenuates unaffected soleus, plantaris and gastrocnemius muscle atrophy in neuropathic pain model.
Purpose: There was no controlled study on botox injection and neurectomy for treatments of muscle hypertrophy. Although many studies have shown the clinical effects of each treatment, it was not able to evaluate and compare the effects of each treatment because there was no comparison of the two treatments under the same experimental condition. Hence, the aim of this study is to com Methods: The study was carried out on 21 rabbits. 9 rabbits received botox injection(botox injection group), and neurectomy was performed to another 9 rabbits (neurectomy group). 3 rabbits did not receive any treatment(control group). To compare and analyze the effects of muscular atrophy, muscle was stained with NADH-TR, and the changes in size of the muscle fiber were examined. And the electromyography was examined. In each group, muscle fiber was stained and electromyography was performed 2, 3, and 6 months after injection or operation. Results: In histological test and electromyography, in the neurectomy group, the size of muscular fiber and amplitude of electromyography decreased until 2 months after neurectomy. And decreased results were maintained with the passage of time. It showed irreversible aspect. On the other hand, in the botox injection group, the decrease in the size of muscular fiber and amplitude of electromyography was observed until 2 months after injection. In 3 months after the injection, it was slowly getting back to original size and had almost recovered by 6 months after the injection. It showed reversible aspect. Conclusion: This study shows researches about clinical effect of botox injection and neurectomy coincide with the results of experiment under the same experimental condition.
Objective: The effects of maternal undernutrition during midgestation on muscle fiber histology, myosin heavy chain (MyHC) expression, methylation modification of myogenic factors, and the mammalian target of rapamycin (mTOR) signaling pathway in the skeletal muscles of prenatal and postnatal goats were examined. Methods: Twenty-four pregnant goats were assigned to a control (100% of the nutrients requirement, n = 12) or a restricted group (60% of the nutrients requirement, n = 12) between 45 and 100 days of gestation. Descendants were harvested at day 100 of gestation and at day 90 after birth to collect the femoris muscle tissue. Results: Maternal undernutrition increased (p<0.05) the fiber area of the vastus muscle in the fetuses and enhanced (p<0.01) the proportions of MyHCI and MyHCIIA fibers in offspring, while the proportion of MyHCIIX fibers was decreased (p<0.01). DNA methylation at the +530 cytosine-guanine dinucleotide (CpG) site of the myogenic factor 5 (MYF5) promoter in restricted fetuses was increased (p<0.05), but the methylation of the MYF5 gene at the +274,280 CpG site and of the myogenic differentiation (MYOD) gene at the +252 CpG site in restricted kids was reduced (p<0.05). mTOR protein signals were down-regulated (p<0.05) in the restricted offspring. Conclusion: Maternal undernutrition altered the muscle fiber type in offspring, but its relationship with methylation in the promoter regions of myogenic genes needs to be elucidated.
To demonstrate the effect of physical irritancy(massages) on the skeletal muscles of immobilization ddY mice models induced by right side sciatic nerve neurectomy, the cross sectional histological profiles of the muscularis (M) gastrocnemius, M. tibialis cranialis and M. tibialis caudalis were observed after 28 days of treatment of physical irritancy with the changes of body weight thickness of hind limb and individual muscle weights. In addition, changes of demonstrated with diameter of individual muscle fiber and muscle fasciculata, and number of muscle fiber in each of three types of muscles located in the calf. The massages were used in this study as physical irritancy. The experimental groups were divided into five groups, 1) Sham-operated group(Sham), 2) Neurectomized but not physical irritated control group(Control), 3) Neurectomized and physical irritated at knee pint regions(T1), 4) Neurectomized and physical irritated at calf regions(T2), and 5) Neurectomized and physical irritated at achilles tendon regions(T3). The experimental animals were used 5 per groups. The changes of number or diameter of muscle fiber in each muscles were calculated using automated image analyzer. The results are as follow : Thickness(diameter) of muscle fiber of M. gastrocnemius, M. tibialis cranialis and M. tibialis cranialis in right side of hind limb were remarkedly decreased in Control, T1, T2 and T3 groups compared to that of Sham group. An experiment was conducted to determine the effects of caponization on the growth performance, breast and thigh muscles physical properties and fiber diameter of the Pectoralis major and Gastrocnemius pars externa in Taiwan country chicken cockerels. Caponized birds were surgically altered at 10 weeks of age. Birds were fed grower and finisher diets ad libitum during an eighteen-week experimental period. The results indicated that the live weight and feed intake in the capons were significantly (p<0.05) higher and the shank length was significantly (p<0.05) longer than in intact birds. There were no significant (p>0.05) differences in feed conversion and mortality between two treatments at 28 weeks of age. Compared with intact birds, the capons had greater (p<0.05) tenderness in the breast and thigh muscles. Cohesion of the breast muscle in the capons was significantly (p<0.05) better than in the intact birds, but the thigh muscles were not significantly (p>0.05) affected. No treatment differences (p>0.05) were associated with cooking loss, muscle chewiness, and elasticity. The capons had a significantly (p<0.05) smaller fiber diameter in the Pectoralis major, but were not significantly (p>0.05) different in the fiber diameter of the Gastrocnemius pars externa. It is concluded that castration did not depress growth compared with the intact birds, but did improve muscle tenderness. This difference was most pronounced in the thigh muscles.
Experiment was conducted to study the effect of sources of dietary fiber on the intestinal morphology of geese. Sixty white Roman geese of two-week-old were divided randomly into six groups and were fed with isoenergetic and isonitrogenous diets which contain alfalfa meal, barley hull, rice hull, purified cellulose, lignin, or pectin as the major dietary source of fiber. Different sources of dietary fiber significantly influenced the villi height and the crypt depth in the duodenum, and the villi height and the muscle layer thickness in the ileum (p < 0.05). The duodenal villus in the geese that fed diets with alfalfa meal, rice hull or pectin supplemented were significantly longest ($968.5{\mu}m$), whereas the lignin group was significantly shortest villus and deepest crypt depth (p < 0.05). The thicknesses of the ileal and caecal muscle layer were significantly thicker in the geese fed with cellulose supplemented diets than in those fed with the other treatment diets. The caecum of the barley bran fed geese possessed significantly longest villi and the most thick muscle layers (p < 0.05). From scanning electronic microscopic observation, the leafy and plate-like in the duodenal villi morphology of geese represented a more effective nutrient absorption in the small intestine. The morphology of ileal villi in geese was similar from herbivorous rabbit and from the African Green monkey.
