Effects of Calendula Officinalis L. Extract on the Activation of Murine Lymphocytes and Macrophages (금잔화 추출물이 생쥐의 임파구 및 대식세포의 활성에 미치는 영향)

  • The purpose of this research was to investigate the effects of Calendula officinalis extract (CE) on the activation of murine lymphocytes and macrophages. CE was administered p.o. once a day for 7 days at the concentration of 500 mg/kg. The administration of CE increased the viability of thymocytes, but decreased the viability of splenocytes. Also, CE increased the viability of thymocytes and splenocytes at the concentration of $5{\mu}g$/ml in vitro. The administration of CE did not affect the population of thymic $CD4^+$/$CD8^+$ cells and splenic $CD4^+$/$CD8^+$ cells. Furthermore, the administration of CE enhanced the phagocytic activity of peritoneal macrophages, but decreased the phagocytic activity in vitro. CE decreased the production of nitric oxide from peritoneal macrophages in vivo and in vitro system. These results suggest that CE enhance of cell viability by a direct influence on thymocytes and phagocytic activity by an indirect influence on peritoneal macrophages.

Effect of Lespedeza Cuneata G. Don on the Activity of Murine Immune Cells (야관문이 생쥐의 면역세포 활성에 미치는 영향)

  • The purpose of this research was to investigate the effects of 50% ethylalcohol extracts of Lespedeza cuneata G. Don (LE) on the activity of murine immune cells. LE was administered orally once a day for 7 days at the dose of 250 mg/kg. LE increased the cell viability of thymocytes, splenocytes and peritoneal macrophages in vitro and in vivo system, but decreased the cell viability of thymocytes and splenocytes in the presence of concanavalin A in vivo system. Also, the administration of LE was increased the production of ${\gamma}$-interferone, but did not affect the production of interleukin-2 and interleukin-4. Furthermore, LE decreased the phagocytic activity of peritoneal macrophages in vitro and in vivo system, but enhanced the production of nitric oxide. These results suggest that LE has a immuno-regulative action via stimulation of immune cells proliferation.

Effect of Ginseng Saponin on the Proliferation and Viability of Murine Thymocyte, in vitro (생쥐 흉선세포의 증식과 생존력에 미치는 인삼 사포닌의 영향)

  • Ginseng saponin had an effect on the proliferation and viability of cultured murine thymocytes. When the thymocytes were cultured in various concentrations of ginseng saponin, the number of thymocytes increased at $10^{-5}$% ginseng saponin but decreased at $10^{-5}$%. There was little change in the number of thymocytes when cultured in IL 2(Interleukin 2), a factor known for its influence on the proliferation and maturation of thymocytes. When the thymocytes were cultured in various concentrations of IL 2 with $10^{-5}$% ginseng saponin, the number of total cells increased at 1.5% or 3% IL 2 when cultured for 9 hours, or at 6% IL 2 for 12, 24, or 48 hours. But there was little change in the number of viable cells. In vitro, ginseng saponin had an effect on the activity of ADA(Adenosine Deaminase), an enzyme known to affect the production of IL 2. There was a 25% increase in the activity of ADA in the presence of $10^{-5}$% ginseng saponin.

  • Immunoregulative action of 70% ethyl alcohol fraction of Cervus nippon was investigated in vitro. The fraction enhanced the proliferation of thymocytes and the population of CD4$\^$+/CD8$\^$-/ single-positive cells in thymocytes. CN-E enhanced [Ca$\^$2+]$\sub$i/, the production of TNF-${\alpha}$ and IL-1 ${\beta}$, and phagocytosis in peritoneal macrophages. The fraction did not induce the proliferation of splenocytes and DNA fragmentation in thymocytes. These results indicate that 70% ethyl alcohol fraction of Cervus nippon regulates immunological action by stimulating murine thymocytes and macrophages.

Antitumor Activity of the Aqueous-alcoholic Extracts from Unripe Cotton Ball of Gossypium indicum

  • The present study investigated the antitumor activity of the aqueous-alcoholic extracts from unripe cotton balls of Gossypium indicum. An Exposure of murine B16 melanoma and L1210 lymphoma cells to the extracts resulted in their severe deaths in time- and concentration-dependent manners. Of the extracts, hydrophilic fractions were most efficacious for the antitumor activity and found to contain certain amounts of catechin and its derivatives. The hydrophilic extract fraction C36B2-8 had approximately 10 times more cytotoxic effects on B12 and L1210 cells than on isolated murine thymocytes. High concentrations (>150 $\mu$g/ml) of C 36B3-8 mainly induced necrotic cell death. At low concentrations (<100 $\mu$g/ml), however, C 36B3-8 induced not only necrosis but also apoptosis of the two tumor cell lines, which was proved by the TUNEL staining and DNA fragmentation techniques. The data indicate that certain ingredients of the cotton ball extract of G. indicum have an antitumor activity.

  • Experimental studies were done to research the clinical effects of Juglandis Semen aqua-acupuncture and acupuncture(BL13, BL17, BL13ㆍ17 and free points) on the anti-allergic inflammation response. The following results have been obtained ; JSAA was increased in the lymphocyte proliferation in cultured murine thymocytes and splenocytes. The production of IL-1β was decreased in Juglandis Semen aqua-acupuncture and acupuncture(BL13, BL17 and BL13ㆍ17) group. The production of IL-4 was decreased in Juglandis Semen aqua-acupuncture and acupuncture(BL13, BL17 and BL13ㆍ17) group. The production of total IgG was decreased in Juglandis Semen aqua-acupuncture(BL13 and BL13ㆍ17) and acupuncture(BL13, BL17 and BL13ㆍ17) group. According to the above results, Semen aqua-acupuncture and acupuncture(BL13, BL17 and BL13ㆍ17) both enhances the specific immune response in T and B cells and depress the allergic inflammation reaction.

Effect of the Seeds of Coix Lachryma-Jobi L. var. Ma-yuen Stapf. on the Proliferation of L1210 cells, a Leukemic cell-lines (율무가 백혈병세포주인 L1210 세포의 증식에 미치는 영향)

  • The purpose of this research was to investigate the effects of Coix Lachryma-Jobi L. var. Ma-yuen Stapf. on the proliferation of L1210 cells and immune cells. The hexane fraction of Coix Lachryma-Jobi decreased the proliferation of L1210 cells and induced DNA fragmentation of L1210 cells. Also, the fraction decreased the cell viability of murine thymocytes and splenocytes, but increased the phagocytic activity of murine peritoneal macrophages. In addition, fatty acids and fatty acid methyl esters decreased the proliferation of L1210 cells and induced DNA fragmentation of L1210 cells. The main components of the fraction are fatty acid and their derivatives. These results indicate that Coix Lachryma-Jobi has a cytotoxicity on tumor cells and immune cells.

Comparative Study of Ginseng Radix and Insamhodo-tang on Immune Reaction (인삼과 인삼호도탕(人參胡挑湯)의 면역반응에 대한 비교 연구)

  • Immunological activities of Ginseng Radix water extract (GE) were compared with theirs of Insamhodo-tang water extract (IHT). GE and IHT (500 mg/kg) were administered p.o. twice a day for 5 days to C57BL/6 mice, respectively. IHT decreased the cell viability of murine thymocytes and splenocytes, but increased the population of $CD4^+$ cells in thymocytes, and the population of $Thy1^+$ cells and $CD4^+$ cells in splenocytes in contrast to GE. In addition, IHT decreased the phagocytic activity of peritoneal macrophages, but increased the production of nitric oxide from peritoneal macrophages in contrast to GE. These results suggest that IHT decrease a specific- and nonspecific immune reaction in comparison with GE.

  • The effects of genistein on murine thymocytes for inducing apoptotic cell death and phagocytic activity of peritoneal macrophage were studied in vitro. Addition of genistein (10 and 50$\mu$M) to cultured thymocytes from BALB/c mice definitely promoted DNA fragmentation. Also, cytofluorometric analysis of these cells demonstrated a reduction in mitochondrial transmembrane potential ($\Delta$Ψm). But, repeated administration of genistein (1 mg/mouse/day) to mice for 7 days did not cause any detectable DNA fragmentation. Genistein decreased lucigenin chemiluminescence and engulfment of fluorescein-conjugated E. coli particles in peritoneal macrophage. These results suggest that genistein induce an apoptosis of thymocyte via reduction in $\Delta$Ψm and decrease phagocytic activity of peritoneal macrophage in vitro.

  • The purpose of this research was to investigate the effects of Indongdeungjikolpitang water extract(IJTE) on the complication of diabetes. IJTE did not affect the level of blood glucose in alloxan- or streptozotocin-induced hyperglycemic mice, but inhibited the motility of gastrointestine. IJTE inhibited the writhing syndrome induced by acetic acid, the permeability of evans blue into peritoneal cavity induced by acetic acid, the paw edema induced by histamine, and the formation of cotton pellet granuloma. IJTE increased the cell viability of thymocytes and splenocytes. IJTE decreased the release of ${\gamma}-interferone$(${\gamma}-IFN$) and interleukin-2(IL-2), but did not affect the release of interleukin-4(IL-4) from murine thymocytes. IJTE increased the release of IL-4 and decreased the release of tumor necrosis $factor-{\alpha}$($TNF-{\alpha}$) and $interleukin-1{\beta}$($IL-1{\beta}$), but did not affect of ${\gamma}-IFN$ and IL-2 from murine splenocytes. IJTE decreased the release of $TNF-{\alpha}$ and $IL-1{\beta}$ from murine peritoneal macrophages. IJTE decreased the production of niric oxide(NO) from murine peritioneal macrophages and increased the phagocytic activity of murine peritoneal macrophages. These results suggest that IJTE has an anti-inflammatory action via the inhibition of $TNF-{\alpha}$, $IL-1{\beta}$ and NO production from immune cells.
