HPLC를 이용한 봉약침의 주요 성분에 관한 연구 (A Study on Major Components of Bee Venom Using HPLC)

  • This study was designed to study on major components of various Bee Venom(Bee Venom by electrical stimulation in Korea; K-BV I, Bee Venom by Microwave stimulation in Korea; K-BV II, 0.5mg/ml, Fu Yu Pharmaceutical Factory, China; C-BV, 1mg/ml, Monmouth Pain Institute, Inc., U.S.A.; A-BV) using HPLC(High performance liquid chromatography). The results were summarized as follows : 1. HPLC method is useful for analysis of Bee Venom when solution rate is above 1:4000. 2. Analysis of Apamin using HPLC, the Retention time was 8.7min, and standard measurement curve was a function of y=4E+06x+21245. 3. Analysis of Melittin using HPLC, the Retention time was 29.0 min, and standard measurement curve was a function of y=4E+06x+23015. 4. Concentration of Melittin was about 297times than Apamin in K-BV I, and about 329times in K-BV II at same 1:500 solution rate, abnormally about 12 times in C-BV at 1:4000 solution rate. 5. Chinese Bee Venom using HPLC, the point from 5 to 7min(Retention time) showed a big extraordinary peak. These data from the study can be applied to establish the standard measurement of Bee Venom and prevent pure bee venom from mixing of another components. I think it is desirable to study more about safety of Bee Venom as time goes by.

현부리경탕가미방(玄附理經湯加味方)을 병행한 무월경 환자 치험2례(例) (Two cases of secondary amenorrhoea treated by Hyunburikyungtang gamibang)

  • Objectives : Secondary amenorrhoea is the absence of menses for three months in a woman with previously normal menstruation or nine months for women with a history of oligomenorrhoea. It can be caused by stress, extreme weight loss, and excessive exercise. The purpose of this study was to report the clinical effects of herbal medicine on secondary amenorrhoea.Methods : We employed oriental medical treatments; herbal - medication (Hyunburikyungtang gamibang), acupuncture and moxibustion. We treated the patients one or two times a month with oriental therapy method. They took medicine three times a day after a meal. During taking medicine, we let the patients avoid fatty food, flour based food and alcohol. We evaluated the status of the patient, on the basis of the state of menstration and F2 level of Yangdorak. Because we diagnosed the condition of patients with the pattern of liver depression and qi stagnation, so F2 level of Yangdorak was an important point of the diagnosis. Yangdorak machine was Tormeter Iw - zen at Towatech Co.,Ltd.Results : After taking treatment - several times acupuncture and moxibustion during some period and taking herbal-medicine, they had menstrain naturally without taking any hormone drug. Also the amount of menstration has gradually increased. The F2 level of Yangdorak returned to normal range. The feeling of cold on hands, feet and lower abdomen was much improved.Conclusions : Herbal medicine (Hyunburikyungtang gamibang) with oriental medical treatments, acupuncture and moxibustion was effective in the treatment of secondary amenorrhoea.

레이저침의 원리와 연구동향 분석 (Fundamentals and Research Trend of Laser Acupuncture)

  • Objectives : Being introduced in the 1980s, laser acupuncture studies have been steadily carried on such as its thermal effects on human body. However, the mechanisms of effect are not yet clearly explained. Generally, laser acupuncture could be favorable for pediatric patients or people under special conditions (hemophilia, needle-phobia, pathogen-carriers, etc.) because of lower risk of infection, bleeding, pain or fainting. But there are still problems for using laser acupuncture equivalently as the traditional metal needle. The goal of this study is to review the fundamentals, history and present trend of laser acupuncture study, and to suggest how to develop laser acupuncture as an oriental medical device. Method : We performed literature search using PubMed(http://www.ncbi.nlm.nih.gov/pubmed) and Scopus(http://www.scopus.com). Search conditions are laser acupuncture, laser needle, LLLT(as title or abstract) or Litscher G(as author). Main target studies are physical fundamentals, clinical trials, physiological results and device proposals. Results : Many research have shown the similarities of laser acupuncture and traditional metal acupuncture and have reported clinical and physiological effects of laser acupuncture. But, still laser acupuncture is not widely used in Korea due to high-cost of equipments and the lack of clear understanding of laser devices. Additionally, there are still challenges in standardization and device development. Conclusions : Frequencies, laser modes, waveguides, adaptor types, polarization and signal modulations are suggested as future study subjects on laser acupuncture. More various studies and efforts for device development have to be performed to provide oriental medical doctors with evidence based confidence about usage of laser acupuncture.

Pseudorabies virus를 이용한 신장, 신수 및 경문에서 투사되는 중추신경계내 표지영역에 관한 연구 (Central Localization of the Neurons Projecting to the Kidney, UB23 and GB25 Using the Pseudorabies Virus)

  • Objective : To investigate the relation to the organs, shu points and mu points. The labeled common locations of the spinal cord and brain were observed following injection of pseudorabies virus(PRV) into the the kidney, UB23 and GB25. Methods : After survival times of 96 hours following injection of PRV, The fifteen rats were perfused, and their spinal cord and brain were frozen sectioned($30{\mu}m$). These sections were stained by PRV immunohistochemical staining method, and observed with light microscope. Results : In spinal cord, PRV labeled neurons projecting to the kidney, BL23 and GB25 were founded in cervical, thoracic, lumbar and sacral spinal segments. Dense labeled areas of cervical segments were overlap in lateral cervical n. and lamina III-V area. Thoracic segments were overlap in lateral spinal n., intermediolateral n. and lamina V-X areas. Lumbar segments were overlap in lamina I-V areas. Sacral segments were overlap in lamina IV, V and X areas. In brain, PRV labeled areas projecting to the kidney, UB23 and GB25 were overlap in the A1 noradrenalin cells/C1 adrenalin cells/caudoventrolateral reticular n./rostroventrolaterai n., raphe obscurus n,, raphe pallidus n., raphe magnus n., gigantocellular reticular n., locus coeruleus, subcoeruleus n., A5 cell group and paraventricular hypothalamic n.. Conclusions : This results suggest that PRV labeled overlap areas of projecting to the kidney may be correlated to shu and mu points related to the kidney. These morphological results provide that organs-shu(transport) and mu(alarm) points interrelationship may be related to the central autonomic pathways.

봉약침액(蜂藥鍼液)이 PLA2, COX-2, iNOS, AA 및 미치는 영향(影響)에 관(關)한 연구(硏究) (The Effects of Bee Venom on PLA2, COX-2, iNOS, AA and PG in RAW 264.7 Cells)

  • Objectives : The purpose of this study was to investigate the effect of Bee Venom on the lipopolysaccharide-induced expression phospholipase $A_2$, cyclooxygenase-2 and inducible nitrogen oxide synthase, and the generation of arachidonic acid, prostaglandin D2 and E2 in RAW 264.7 cells, a murine macrophage cell line. Methods : The expression of phospholipase $A_2$, cyclooxygenase and inducible nitrogen oxide synthase was determined by western blotting with corresponding antibodies, and the generation of arachidonic acid, prostaglandin $D_2$ and $E_2$ was assayed by ELISA method in RAW 264.7 cells. The non-toxic concentrations (0.1 to $5\;{\mu}g/ml$) of bee venom determined by MTT assay, were used in this study. Results : 1. Bee venom inhibited lipopolysaccharide-induced expression of phospholipase $A_2$ in a dose dependent manner after 48 hours treatment. 2. Bee venom inhibited lipopolysaccharide-induced expression of cyclooxygenase-2 in a dose dependent manner after 24 and 48 hours treatment. 3. Bee venom inhibited lipopolysaccharide-induced expression of inducible nitrogen oxidesynthase in a dose dependent manner after 48 hours treatment. 4. The generation of arachidonic acid, prostaglandin $D_2$ and $E_2$ was not much affected by the treatment of bee venom on the lipopolysaccharide-induced generation of arachidonic acid, prostaglandin $D_2$ and $E_2$ in RAW 264.7 cells.

아유르베다와 티베트 의학의 기본이론과 한의학과의 비교 고찰 (A Comparative Study on Traditional Korean Medicine and Ayurveda, Tibetan Medicine)

  • Background : There has been increased global interest in traditional medicine. Ayurveda and Korean medicines are most world-widely spread as Complementary and Alternative Medicine(CAM). Objectives : This review gives an overview of basic principles and commonalities of Ayurveda, Tibetan and Korean medicine. Method : I have summarized the results of the research into the following categories: basic theoies, history, physiology and pathogenesis. Result : 1. Ayurveda is the medical science that grasps individual constitution through Tridosha, a combination of five primary elements(space, wind, fire, water, and earth), and systematically explains physiological and pathological phenomena which reveal according to the constitution. 2. The theoretical basis forming the Tibetan Medicine is the substance of phlegm, bile, and wind each of which is divided into five kinds. These physiological substances can be transformed into pathological factors when certain environment is formed. 3. Ayurveda, Tibetan and Korean medicine, three main axes of the traditional medicine arouse western medical researchers' interest by their peculiar views of the disease and the system of their medical theories. And they are expected to render services to human health. Conclusions : We can expect the potential of the approaching between Ayurveda, Tibetan and Korean medicine.

만성 두통에 대한 뇌청혈해독탕의 임상적 효과 (The clinical effect of Noechongyolhaedok-tang on the chronic headache)

  • Objective : Chronic headache is the most common headache. The objective of this study is to find the clinical manifestation of chronic headache and has been carried out to investigate the effects of Noechongyolhaedok-tang on the chronic headache. Method : This observation was carried out on 73 patients with chronic headache. They were applied to Noechongyolhaedok-tang and the degree of improvement of chronic headache was evaluated by VAS. Results : 1. Distribution of study population by sex, age : male : female = 1 : 3, 30s-50s group = 81% Duration of onset : over 10years = 48% Reported site of chronic headache : the whole = 28% Clinical pain characteristics : be torn = 25% Frequency of headache : everyday = 72% Time of attack : irregular = 52% 2. In VAS scores, there were significant differences between before and after Noechongyolhaedok-tang treatment. There were no differences between before and after treatment in each sex and the duration of onset Conclusion : The effectiveness of Noechongyolhaedok-tang on the chronic headache was shown through VAS. Further study is needed about Noechongyolhaedok-tang.

봉약침액(蜂藥鍼液)이 Scopolamine으로 유발(誘發)된 기억(記憶) 장애(障碍)에 미치는 영향(影響) (The Effect of Bee Venom on Scopolamine Induced Memorial Impairment)

  • Alzheimer's disease (AD) is the most prevalent form of neurodegenerative disease associated with aging in the human population. This disease is characterized by the following 4 structural changes : Atrophy of the Cortex, Parasympathetic, and other neural cells, the existence of Neurofibrillary tangles (NFTs), and the accumulation of Senile plaques. NFTs and Senile plaques is known to be the index of this disease. Senile plaques disturbs the neutro transmission and depletes of Acetylcholine. So, Recovery of Acetylcholine is the primal objective for treating Alzheimer's disease. So, Inhibiting the activity of Acetylcholine Esterase (AChE), which causes the hydrolysus of acetylcholine into choline and acetate, can be seen as a key role for treating Alzheimer's disease. Increasing body of evidence has been demonstrated that Bee Venom Acupuncture (BV) could compete with complex protein involving in multiple step of $NF-_{\kappa}B$ activation and exert the anti -inflammatory potential of combined inhibition of the prostanoid and nitric oxide synthesis systems by inhibition of IKK and $NF-_{\kappa}B$. The effect of BV through behavioral tests after memory impairment induced by Scopolamine. We examined the improving effect of BV on the Scopolamine (1 mg/Kg, i.p.)-induced memorial impairment using passive avoidance response and water maze tests in the mice. BV (0.84, $1.67\;{\mu}g/ml$) reversed the Scopolamine-induced memorial impairment in dose dependent manner. This study therefore suggests that BV acupuncture method may be useful for prevention of development or progression of AD.

  • The purpose of this study was to investigate the effect of Bee Venom on the lipopolysaccharide(LPS), sodium nitroprusside(SNP), hydrogen peroxide($H_2O_2$)-induced expressions of cyclooxygenase-2(COX-2), p38, jun N-terminal Kinase(JNK) and extra-signal response kinase(ERK) in RAW 264.7 cells, a murine macrophage cell line. Method : The expressions of COX-2, p38, JNK and ERK were determined by western blotting with corresponding antibodies. Results : 1. The 0.5, 1 and $5\;{\mu}g/ml$ of bee venom inhibited significantly LPS and SNP-induced expression of COX-2 compared with control, respectively. The 0.5, 1 and $5\;{\mu}g/ml$ of bee venom inhibited insignificantly $H_2O_2$-induced expression of COX-2 compared with control, respectively. 2. The 0.5, 1 and $5\;{\mu}g/ml$ of bee venom inhibited significantly LPS, SNP and $H_2O_2$-induced expression of p38 compared with control, respectively. 3. The 1 and $5\;{\mu}g/ml$ of bee venom inhibited significantly SNP-induced expression of JNK compared with control, respectively. All of bee venom inhibited insignificantly LPS and $H_2O_2$-induced expression of JNK compared with control, respectively. 4. The $5\;{\mu}g/ml$ of bee venom inhibited significantly SNP-induced expression of ERK, the $0.5\;{\mu}g/ml$ of bee venom increased significantly $H_2O_2$-induced expression of ERK compared with control. The 0.5, 1 and $5\;{\mu}g/ml$ of bee venom inhibited insignificantly LPS-induced expression of ERK compared with control, respectively.

백작약약침(白芍藥藥鍼)이 백서(白鼠)의 위궤양(胃潰瘍)에 미치는 면역조직화학적(免疫組織化學的) 연구(硏究) (The immunohistochemical studies of Herbal drug-acupuncture of Radix paeoniae lactiflorae on Gastric ulcer induced by HCI-aspirin in Rats)

  • In order to study the immunohistochemical effects of herbal drug-acupuncture of Radix paeoniae lactiflorae on gastric ulcer induced by HCl-aspirin in rats. this experiment was done by herbal drug-acupuncture to Wisu($B_{21}$). Chung-Wan($CV_{12}$), Chok-Samni($S_{36}$) loci to measure histological features of ulcer lesion, the change of numbers of parietal cell, chief celI, gastrin and somatostatin-immunoreactive cell. The obtained results were as follows; 1. The ulcerative lesions of gastric mucosa were decreased to WiSU($B_{21}$) loci followed by Chung-Wan($CV_{12}$) and Chok-Samni($S_{36}$) loci compared to the control groups. 2. In the numbers of parietal cell, the most remarkable decrease was observed to Wisu($B_{21}$) loci followed by Chung-Wan($CV_{12}$) and Chok-Samni($S_{36}$) loci compared to the control groups. 3. In the numbers of chief cell, the most remarkable increase was observed to Wisu($B_{21}$) loci followed by Chung-Wan($CV_{12}$) and Chok-Samni($S_{36}$) loci compared to the control groups. 4. In the numbers of gastrin-immunoreactive cell, the most remarkable decrease was observed to Wiu($B_{21}$) loci followed by Chung-Wan($CV_{12}$) and Chok-Samni($S_{36}$) loci compared to the control groups. 5. In the numbers of somatostatin-immunoreactive cell, the most remarkable decrease was observed to Wisu($B_{21}$) loci followed by Chung-Wan($CV_{12}$) and Chok-Samni($S_{36}$) loci compared to the control groups.
