Our previous study has suggested that muscarinic receptor present in the mouse oocytes, and Ca/sup 2+/ waves elicited by acetylcholine (ACh) are similar to those induced by sperm. A numerous study reported that ACh could cause early activation events in mouse oocytes overexpressing the Ml muscarinic receptor (Williams et al., 1992; Moore et al., 1993; Kim et al., 1998). Egg yolk is widely used to extract lecithin, which is utilized in the food and cosmetics industry. After lecithin is removed, the rest of egg yolk is generated as a by-product. Thus, it is necessary to properly utilize it. In this study, egg yolk protein extracts were produced using ethanol (EYE-E) and water (EYE-W). Their antioxidant and immunomodulatory effects were then evaluated. Antioxidant activities of EYE-E and EYE-W were determined using cellular antioxidant capacity (CAC) assay and comet assay. EYE-E and EYE-W showed significant (p<0.05) scavenging effects on intracellular reactive oxygen species (ROS) in a dose dependent manner. At a concentration of 50 ㎍/mL, EYE-W showed higher (p<0.05) antioxidant activity than EYE-E. EYE-E and EYE-W also exhibited protective effects against DNA damage caused by oxidative stress. After treatment with EYE-E and EYE-W, DNA damage level of 48.7% due to oxidative stress was decreased to 36.2% and 31.8% levels, respectively. In addition, EYE-E and EYE-W showed immunomodulatory effects by regulating Th1 cytokines (TNF-α and IL-2) and Th2 cytokines (IL-10 and IL-4) in Balb/c mouse splenocytes. These data suggest that EYE-E and EYE-W could be used as functional food ingredients with excellent antioxidant and immunomodulatory activities in the food industry.
Egg consumption has been limited to avoid cardiovascular diseases such as atherosclerosis and hyperlipidemia, because the yolk contains high levels of cholesterol. This study was conducted to evaluate the effect of the water-soluble component of egg-yolk on the growth efficiency, immune modulation, and changes in serum lipid levels in BALB/c mice. A total 5 wk old 120 BALB/c male mice were divided into 4 groups and were fed 0, 2, 10, and 20 mg/d water-soluble egg yolk extract (WSEYE) for 5 wk. Water-soluble egg yolk extract (WSEYE) uptake resulted in a significant reduction in daily weight gain and feed efficiency rate (FER). The mouse groups treated with 2 and 20 mg/d WSEYE showed a significant increase in populations of monocytes at the third wk and B-lymphocyte activity at the fifth wk. In addition, WSEYE uptake did not influence serum immunoglobulin E levels. In serum lipid-profile studies, treatment of WSEYE did not alter total cholesterol and low-density lipoprotein levels; however, blood triglyceride levels were significantly diminished in mice treated with 2 mg/d at the third wk (p<0.05), and the level of high-density lipoprotein was significantly increased in the mice group treated with 2 and 10 mg/d WSEYE after 5 wk (p<0.05). Taken together, the data demonstrate the beneficial effects of WSEYE in the diet on immune modulation and serum lipid profiles in mouse models; therefore, this study suggests that ingestion of water-soluble fraction of egg yolk might not be related to the increased risk of heart disease, but can be an excellent candidate for maintaining health.

Insulin-like Growth Factors (IGFs) and IGF-binding protein act as intra-ovarian regulators that modulate the proliferation and differentiation of the granulosa and theca cells. Moreover, the IGF system is involved in metabolism by modulating the synthesis and degradation of glycogen and protein in animals. However the effect of the IGF system on egg productivity or body growth in KNOC has not been studied in depth. Therefore, this study was performed to investigate differences of serum IGFs and binding protein expressions between two groups showing high and low egg production or body weight and to elucidate the relationship of IGFs with egg productivity and body growth. KNOCs were divided into high and low groups depending on their egg productivity or body growth, and sera were collected every 10 wk from 20 till 60 wk. Serum IGF-I and -II concentration were measured by RIA using human and mouse antiserum and chicken standards. IGFBP was detected by Western ligand blotting. IGF-I concentrations were significantly greater in the high egg production group compared with those in the low egg production group (30 wk, p<0.01; 20 and 40 wk, p<0.05). Also, differences in IGF-II amounts between the two groups were detected at 60 wk (p<0.05). But IGFBPs in the low egg production group were more intense than that in the high egg production group through the egg laying period. The correlation between IGF-I concentration and number of egg production is significantly positive (20 wk, r=0.2729: p<0.05; 40 wk, r=0.3500: p<0.01), while IGF-II shows no correlation with egg productivity. In male KNOC, IGF-I and -II concentrations in the high body weight group are lower than that in the low body weight group. Body weight also shows a negative correlation with the serum IGF-II concentration in male chickens (20 wk, r=-0.5901: p<0.01). Consequently, we suggest that IGFs and binding protein are (in)directly involved in the egg productivity and body growth in KNOC.
Cancer is the main cause of death in developed countries. However, in underdeveloped countries infections and parasitic diseases are the main causes of death. There are raising scientific evidences indicating that parasitic infections induce antitumor activity against certain types of cancers. In this study, the effects of Toxoplasma gondii and Toxocara canis egg antigens in comparison with Bacillus Calmette Guerin (BCG) (known to have anticancer distinctive) on WEHI-164 fibosarcoma transplanted to BALB/c mice was investigated. Groups of 6 male BALB/c mice injected with T. gondii antigen, BCG, or T. canis egg antigen as case groups and alum alone as control groups. All mice were then challenged with WEHI-164 fibrosarcoma cells. The mice were examined for growth of the solid tumor and the tumor sizes were measured every other day up to 4wk. The mean tumor area in T. gondii, BCG, or alum alone injected mice in 4 different days of measurements was $25\;mm^2$, $23\;mm^2$, and $186\;mm^2$ respectively, Also the mean tumor area in T. canis injected mice in 4 different days was $25.5\;mm^2$ compared to the control group (alum treated) which was $155\;mm^2$. T. gondii parasites and T. canis egg antigens induced inhibition of the tumor growth in the fibrosarcoma mouse model. We need further study to clarify the mechanisms of anti-cancer effects.
dbcAMP에 의해 성숙이 억제된 생쥐난자의 수정능을 조사하기 위해 본 실험을 행하였다. dbcAMP로 일시 성숙이 억제되었던 난자를 배양액내에서 정자와 섞고 24시간 배양한 후 발생한 2세포기의 배아형성, 정자의 관입, 전액형성을 조사하여 2세포기로 배아발생이 진척된 것을 수정율의 기준으로 정하였다. dbcAMP를 처리하지 않은 난자는 약 53.3%의 수정율을 보였으며,dbcAMP가 함유되 배양액에서 배양한 후 기본 배양액에서 성숙시킨 난자들의 수정율은 dbcAMP의 처리시간에 비례하여 낮으나, 정자의 관입은 정상적으로 일어났다. 전자현미경적 관찰에 의하면 dbcAMP 처리는 난자의 미세구조에 어떠한 변화도 야기하지 않았다. 따라서 dbcAMP를 사전처리하여 성숙을 억제한 난자라할지라도 어느 정도의 수정능력을 보유하고 있다고 사료된다.
Polyclonal antibodies labeled with a tracer have been commonly used as secondary antibodies in immunochemical assays to quantify the concentration of antibody-antigen complexes. The majority of these antibodies conjugated with a tracer are commercially available, with the exception of few untouched targets. This study focused on the production and application of mouse anti-quail IgY as an intermediate antibody to link between quail egg yolk IgY and goat anti-mouse IgG-HRP as primary and secondary antibodies, respectively. Subsequently, the produced mouse anti-quail IgY was labeled with horseradish peroxidase (HRP) and its efficiency on enzyme linked immunosorbent assay (ELISA) was compared with that of commercial rabbit anti-chicken IgY-HRP. As an intermediate antibody, mouse anti-quail IgY was successfully produced with good affinity and sensitivity (1:10,000) to the primary and secondary antibodies. Subsequently, mouse anti-quail IgY was effectively conjugated with HRP enzyme, resulting in a secondary antibody with good sensitivity (1:10,000) to quail anti-V. parahaemolyticus and V. vulnificus IgY. The detection limit was $10^5$ CFU/ml for both V. parahaemolyticus and V. vulnificus. The efficiency of the produced conjugate to detect quail IgY on ELISA was comparable to that of the commercial rabbit anti-chicken IgY-HRP, and hence the produced and labeled mouse anti-quail IgY-HRP can be used as a secondary antibody to detect any antibody produced in quail.
Alcalase hydrolysis of liquid egg white was used to produce 5-hydroxytryptophan (HTP) under various conditions and investigate the sleep-potentiating activity of liquid egg white hydrolysate (LEH) on pentobarbital-induced sleep. Alcalase hydrolysis yielded the highest content of 5-HTP ($13.50{\mu}g/mL$), while neutrase hydrolysis showed the lowest 5-HTP content ($5.23{\mu}g/mL$). The liquid egg white to water ratio (1:1) was optimal for the production of 5-HTP with high amino-nitrogen (A-N) content and degree of hydrolysis. The 5-HTP, amino-nitrogen, and degree of hydrolysis increased until 24 h of hydrolysis and slightly increased thereafter during hydrolysis with 2% and 5% enzyme addition. 5-HTP administration at doses of 6 and 9 mg/kg significantly increased sleep duration and decreased sleep latency time compared to that in the control (p<0.05). LEH (150 mg/mouse), which was equivalent to 5-HTP at 6 mg/kg, significantly decreased sleep latency time and increased sleep duration time compared to that in the control (p<0.05). Oral administration of LEH showed sleep-potentiating effects because of 5-HTP. The sleep-potentiating activity of LEH may have occurred through 5-HTP in our pentobarbital-induced sleep model. LEH may be a valuable alternative to sleep enhancement and may be used as a sleep-potentiating agent.
Objective & Methods : Experimental studies were done to research the clinical effects of Juglandis Semen aqua-acupuncture and acupuncture($BL_{13}{\cdot}BL_{17}$) on the anti-allergic response. anaphylaxis provoked by the compound48/80, delayed type hypersensitivity response to picryl chloride and SRBC and inflammation response to egg albumin. Results : The following results have been obtained; 1. Juglandis Semen aqua-acupuncture and acupuncture($BL_{13}{\cdot}BL_{17}$) group were increased the survival rate in compound 48/80 induced systemic anaphylactic reaction. 2. Picryl chloride induced contact dermatitis and delayed type hypersensitivity in SRBC challenged mouse were significantly decreased in Juglandis Semen aqua-acupuncture and acupuncture($BL_{13}{\cdot}BL_{17}$) group. 3. Inflammation response-WBC, CRP and Nitric Oxide in egg albumin induced allergic rat were significantly decreased in Juglandis Semen aqua-acupuncture and acupuncture($BL_{13}{\cdot}BL_{17}$) group. Conclusions : According to the above results, Semen aqua-acupuncture and acupuncture($BL_{13}{\cdot}BL_{17}$) both depress the allergy reaction.
만손주혈흠충 충란의 soluble eff antigen (SEA)을 mini-pump를 이용하여 마우스, 복강내에 지속적으로 주입시키면서 말초혈액내의 호산구수 변동, delayed hypersensitivity인 ear swelling test, 간조직병변, 효소 표식면역법을 이용한 혈청내 항체가를 경시적으로 관찰한 결과 다음 성적을 얻었다. 1. 말초혈액 호산구수는 SEA투여후 2~3주후에 최고에 달하였고 그후 서서히 감소하였다. 2. Ear swelling test 실시에서 SEA투여 2주후에 종창이 최고에 달하고 그후 서서히 감소하였다. 3. 간조직병변은 SEA투여후 4주에 육아종형성을 관찰하였고 그후 조직손상이 복귀하였다. 4. 현청에서의 항체가는 SEA투여후 4주후에 양성대조군보다 높았으며 시간이 경과함에 따라 항채가가 증가하였다.
