Eleven accessions of Hedera helix, three accessions of Hedera rhombea, one accession of Fatshedera lizei, and one accession of Fatsia japonica were collected and their genetic diversity was measured by using 10 RAPD primers. Approximately ninety seven percentage of polymorphism was detected, because broad germplasm, three genus, was used. Total 97 bands were scored and a dendrogram was constructed by using an UPGMA method. Accessions belonging to Hedera helix tightly clustered in one group: eight accessions showed extremely narrow genetic differences and the other three accessions also showed significant similarity. Despite of their genetic similarity, they showed morphological variations. The morphological variation with a narrow genetic differences indicated that the ivy cultivars have been indeed developed from a mutation breeding program. Genetically most unrelated Fatsia japonica showed a genetic distance of 0.63 on the average between other species. An accession from Fatshedera lizei developed by crossing between two genus, Hedera helix and Fatsia japonica, was allocated together with accessions from Hedera rhombea.
In order to presume the relationships between two species of P. ternata and P. tripartia, and their populations of the Korean Pinellia, RAPD analysis was performed. The length of the amplified DNA fragments ranged from 300 to 2,500bp. Seventy scorable RAPD makers were found from the PCR reactions with 7 random oligoprimers and were analyzed by Nei-Li's genetic coefficient. Also, some regional groups instead of same taxa were clustered from the phenogram of UPGMA analysis and NJ tree. Populations within each species were clustered at low genetic distance, there had the closed relationship. According to the regional individuals, Pinellia ternata was showed the variation pattern of morphological (leaf shape and flower color) and cytological characters(somatic chromosome numbers). So we suggested to difference of characteristic variety based on variety of habitat. According to this study, new species (Pinellia sp.) was affiliated with Pinellia and had the closest relationship with Hallasan and Japan population. The RAPD data was very useful to define the genetic variation and to discuss the relationships among the intraspecific taxa and their populations of the Korean Pinellia.
It has been made clear that there exist a definite rhythm in the accumulative process of carbohydrates of soybeans, and that there is a great variation in nitrogen metabolism. In spite of earlier or later manuring, it is possible to make use of the above stated conclusion. But in the case of later manuring, there is so short a time of growth before harvest, resulting in the lack of normal growth and in a great effect upon harvest. In the general terms, the first half period of growth may be called that of nitrogen assimilation, and the late half, that of carbohydrate assimilation. It has delayed comparatively distinction between these periods whether manuring was done earlier or later. In other words, the lack of carbohydrates in the first half period of growth has been found to deter the process of protein synthesis. And the lack of protein in a body in the latter half period of growth has been found to weaken photosynthesis that should be active, resulting in the decrease of harvest. Consequently, lack of protein in the body during the period of pod adhesion definitely brought about lack of growth in the latter period of growth. It follows, therefore, that in order to increase harvest, it is necessary to manure comparatively in the early period, and that contents of various forms of nitrogen.
RAPD markers were analyzed in order to detect the genetic variation and diversity of the fifty-two melon lines. SDS extraction method produced more and purer DNA than CTAB method. RAPD reaction conditions were optimized as follows ; 10ng template DNA, 270nM primer, $200{\mu}M$ each of dATP, dCTP, dGTP and dTTP, $0.3{\mu}unit$ dynazyme and 10x buffer brought to $15{\mu}l$ final volume with distilled water. The adequate annealing temperature was $39^{\circ}C$ and forty cycles of amplification produced the best RAPD band patterns. Among a total of 123 bands from 12 random primers, 25 polymorphic bands(20%) were selected as reliable markers. The average number of polymorphic bands per primer was 2.1 among the 52 lines. Intragroup genetic relationship based on the marker difference was closer than intergroup genetic relationship. The 52 lines could be grouped into two major group (Korean landraces and melon lines) and then melon group subdivided into two subgroups (net melon lines and no-net melon). This result corresponded to morphological grouping. Eight RAPD markers separated the Korean landraces and melon groups and four RAPD markers separated net melon and no-net melon groups.
Allozyme study for open pollinated seeds of forty nine pitch pine families in an $F_1$-hybrid seed orchard demonstrated that allozyme variants in aspartate aminotransferase(AAT), glutamate dehydrogenase(GDH) and leucine aminopeptidase(LAP) systems are encoded by at Least eight loci; five for AAT, one for GDH and two fur LAP. Allozyme variations showed polymorphisms at seven of the eight loci, except GDH. Average number of alleles examined over six loci were 2.33 and 2.67 for maternal and progeny groups, respectively. Average heterozygosity and genetic diversity computed over six loci were, respectively, 0.235 and 5.409 for maternal tree group, 0.238 and 5.569 for progeny group. The proportion of $F_1$-hybrid seeds estimated by allozyme analysis was 0.77%. The estimated proportion of $F_1$ hybrid seeds by allozyme study is in good agreement with the value 0.73% estimated by morphological study for the proportion of pitch ${\times}$ Loblolly $F_1$ hybrid seedlings at a nursery. Indications for Wahlund effect, high levels of self-fertilization and for non-random matings in the $F_1$ hybrid seed orchard call for cautions in estimating allele frequency changes and mating probabilities for the parental and progeny groups.
To establish the phylogenetic relationships of Dictyophora spp., rDNA-ITS regions of 11 strains of veiled lady mushroom collected from various countries were amplified and sequenced. It was observed that the 11 strains were divided into four groups based on PCR band patterns of each ITS region cleaved by eight different restriction enzymes in cleaved amplified polymorphic sequence analysis (CAPS). The phylogenic relationship of each group by cleaved amplified polymorphic sequence (CAPS) analysis matches well with previously reported morphological phylogeny, such as 5 strains of D. indusiata, 4 strains of D. echinovolvata, and a strain of Phallus rugulosus. Sequence analysis using the cluster V methods showed more detail classification than CAPS analysis. The 5.8S region showed two point nucleotide base exchanges from G to A according to four groups, and four groups were subdivided by sequence variation of ITS I and ITS II regions. But sequence variation of Phallus rugulosus was not showed in full ITS region. This study further delineates the taxonomic level at which ITS sequences, in comparison to ribosomal gene sequence, are most useful in systematics and other mushroom study.
In present paper, the morphological characters in perfect stage of Microsphaera polygoni(DC.) Sawada on Robinia pseudoacasia were investigated and the phylogenetic relationship between Gen. Erysiphe and Microsphaera was discussed with variation of appendages. The results are summarized as follows: 1. The perithecia of the powdery mildew fungus on Robinia pseudoacasia were rarely formed on the surface of the leaves just before difoliation in late autumn and their forming period was very short. 2. Powdery mildew fungus on R. pseudoacasia was identified as Microsphaera polygoni(DC.) Sawada in Korea. 3. Appendage of the fungus are both Erysiphe and Microsphaera types in shape, and plenty of intercalary types are intervened between both types. Number of perithecia bearing upper various appendages appears the normal distribution with both poles of typical Erysiphe and Microsphaera types 4. If Blumer's theory on the phylogenetic relationship of Erysiphaceae is right, variation of the appendages of the fungus might be evolved from Gen. Erysiphe to Gen. Microsphaera.
Diploid plants were obtained by anther culture of tetraploid poplar(Populus alba L. X P.glandutosa Uyeki). The effect 2,4D on callus formation from anther culture was greater than any other auxins tested. The highest average number of multiple shoots per callus was obtained when zeatin was used at levels of 6-8 ${\mu}$M. Regenerated shoots were excised and transferred to MS basal medium. Rooted plantlets were subsequently transferred to pots containing artificial soil mix. Finally 100 plane were transplanted in nursery located in forest Genetics Research Institute. for the 300 anther clones growing in greenhouse for 6 months after transplanting, 33% were slow-growing, 47% were rapid-growing and 20% had huge leaf size with rapid-growing characteristics. Chromosome study showed a narrow range of variation from diploid to tetraploid. DNA polymorphism studies using various RAPD markers revealed some extend of differences among the anther-clones in their band pattern.
7 populations of rare endemic Berchemia berchemiaefolia were analysed using multivariate analysis for 9 characteristics of leaf morphology. The aim of this study was to examine geographic variation to support a genetic resource conservation plan of B. berchemiaefolia in Korea. In the morphological characters, nine characters of leaf were 10.25 cm (blade length), 4.10 cm (maximum width), 2.52 (blade length/maximum width), 3.22 cm (upper 1/3 width), 3.42 cm (lower 1/3 width), 0.95 (upper 1/3 width/lower 1/3 width), 1.24 cm (petiole length), 8.91 (blade length/petiole length), 8.16ea (vein number), respectively. Nested anova showed that were statistically significant differences among populations as well as among individuals within populations in all 9 quantitative characters. In 7 of 9 characters, variance components among individuals within populations were higher than those among populations. Cluster analysis using complete linkage method showed two groups (Chungbuk and Gyeongbuk districts) to Euclidean distance 1.2. Among principal components, primary 3 principal components appeared to be major variables because of the loading contribution of 87.3%. The first contribution was blade length, blade length/maximum width and blade length/petiole length; the second one was maximum width, upper 1/3 width and lower 1/3 width; the third one was petiole length, respectively.
Asarum consists of low-growing herbs and is a genus in the Aristolochiaceae family with species found in the north temperate zones with most species in Asia. We evaluated the nine taxa with the trnL - trnT region of the chloroplast genome to estimate phenotypic relationships within genus Asarum in Korea. Alignment of the DNA sequences required the addition of numerous gaps. Sequence variation within the Asarum was mostly due to nucleotide inserts/deletions, although several indels and inserts were found. Another source of sequence divergence was length variation due to stretches of short repeats that occur at the trnL - trnT region in all the Asarum. A + T content for nine Korean species of genus Asarum ranged between 74.7% and 78.3%. These values were higher than those for the angiosperm alignments of the total trnL and trnT region (64.5~67.1%). Within genus Asarum, A. patens was strikingly different from the others in the three phylogenetic analyses (MP, ML, and NJ). However, some internal nodes were poorly supported. Within Korean Asarum, four species were unsolved portions. Possible reasons for the striking non-congruence between the previous morphological traits and the trnL - trnT based on phylogeny were discussed.
