Many infraspecific taxa within Lespedeza maximowiczii and hybrids with related species have been described, but taxonomic verification remains controversial. We examined the morphological traits of hybrids (L. chiisanensis and L. patentibicolor) and infraspecific taxa (var. tomentella, elongata, and tricolor) and analyzed their genetic structures using microsatellite loci. Flower and leaflet shapes in var. tomentella and elongata were within the range of variation of those in var. maximowiczii, and individuals in the two former varieties were grouped into var. maximowiczii. Lespedeza maximowiczii var. tricolor was similar to L. buergeri in terms of the structure and flower color, whereas the leaflet and bracteole shapes of var. tricolor were similar to those of var. maximowiczii. Based on the genetic structure (K = 3), var. tricolor had a mixed lineage with L. maximowiczii and L. buergeri. In addition, these formed a distinct lineage at K = 5. For two hybrids, the flower and leaflet structure in L. chiisanensis did not differ from those in L. maximowiczii, whereas the flowers of L. patentibicolor were within the range of variation of L. bicolor. In addition, L. chiisanensis and L. patentibicolor were assigned to L. maximowiczii and L. bicolor, respectively, based on the genetic structure. We treated var. tomentella and elongata as a forma, f. friebeana, because L. friebeana preceded var. tomentella, whereas var. tricolor was treated as a distinct species, L. tricolor. Lespedeza chiisanensis was recognized as a synonym of L. maximowiczii. Lespedeza patentibicolor was considered to be L. bicolor.
Molecular characterization of ten marine cyanobacterial isolates belonging to the order Oscillatoriales was carried out using the phycocyanin locus (cpcBA-IGS) and the 16S-23S internally transcribed spacer region. DNA sequences from the phycocyanin operon discriminated ten genotypes, which corresponded to seven morphotypes identified by traditional microscopic analysis. The cpcB coding region revealed 17% nucleotide variation, while cpcA exhibited 29% variation across the studied species. Phylogenetic analyses support the conclusion that the Phormidium and Leptolyngbya genera are not monophyletic. The nucleotide variations were heterogeneously distributed with no or minimal informative nucleotides. Our results suggest that the discriminatory power of the phycocyanin region varies across the cyanobacterial species and strains. The DNA sequence analysis of the 16S-23S internally transcribed spacer region also supports the polyphyletic nature of the studied oscillatorian cyanobacteria. This study demonstrated that morphologically very similar strains might differ genotypically. Thus, molecular approaches comprising different gene regions in combination with morphological criteria may provide better taxonomical resolution of the order Oscillatoriales.
Isolates of Trichoderma spp. collected from Pleurotus ostreatus and P. eryngii beds, which included loosened substrate compactness and development of green colour, were grouped into three species. The occurrence of different species of Trichoderma was as T. cf. virens(70.8%), T. longibrachiatum(16.7%) and T. harzianum(12.5%). The conidia of Trichoderma spp. were ellipsoidal, obovoid and phialides were bowling pins, lageniform and the length of phialides was $3.5{\sim}10.0{\times}1.3{\sim}3.3{\mu}m$. Phialides of T. cf. virens and T. harzianum were tending clustered, but it was solitary disposition in T. longibrachiatum. T. cf. virens was characterized by predominantly effuse conidiation, sparingly branched, and fertile to the apex and it was penicillate type. RAPD analysis could detect variability amongst three different species of Trichoderma using two newly designed URP-primers. However, intra-specific variation could not be detected in all the isolates except for rDNA sequence data classified Trichoderma isolates into three distinct groups representing three species. The profiles of rDNA sequences of isolates representing a species showed high similarity in T. cf. virens and T. harzianum. However, there was a variation in rDNA sequences of isolates representing T. longibrachiatum. The results of present study reveals that molecular techniques of RAPD and rDNA sequencing can greatly aid in classification based on morphology and precise identification of fast evolving species of Trichoderma.
The leopard danio, Danio rerio var. frankei is a spotted color morph of the zebrafish, Danio rerio caused by a pigment mutation. The structural differences of fertilized egg and egg envelope are poorly documented. To clarify this, we compared the fertilized egg morphology and ultrastructures of surface structures, the micropyle and the cross section of fertilized egg envelopes of zebrafish and leopard danio, variation species of zebrafish using a light and electron microscopes. Although the fertilized egg sizes were different, the external shapes of the fertilized eggs of two species couldn't be differentiated under the light microscope. The characteristics of fertilized eggs, such as a spherical shape, a non-adhesive quality and a large perivitelline space, were shown to be related to spawning habit. In ultrastructure of fertilized egg envelope, there is no morphological difference of micropyle between two species. By contrast, the ultrastructure and the numbers of knob-like structures and semihemisphere-like structures per unit area on the outer surface, and the number of lamellae of inner layer on the fertilized egg envelope section displayed definite species specificity. Collectively, our data indicate that the ultrastructure of fertilized egg envelope in the zebrafish could be differentiated by species variation.
This study was carried out to investigate the variations of dimension and exterior shape of vessel elements, morphology of spiral thickening and ray-vessel pit in korean diffuse-porous woods (56 species, 18 families), The tangential pore diameter and vessel element length was increased, whereas the pore number per unit area($1mm^2$) was decreased from pith outwards. The tangential pore diameter was decreased but the length of vessel element was not changed from earlywood to latewood within an annual ring. However, the dimensional variations of vessel element was not able to be recognized among relative positions in peripheral variation. The exterior shape of vessel element could be classified into four types; Type 1 is without tail, Type 2 with ligulate tail, Type 3 with broad taper tail and Type 4 with the very short length between perforations. The distribution frequency of Type 2 and 3 was relatively high in comparision with the others. According to the prominence, distribution position and branched shape, the spiral thickenings could be divided into five types. The spiral thickenings occurred 52% in the species observed. Thus it was doubtful to consider the simple presence of spiral thickening as diagnostic index in diffuse-porous woods. The morphology of ray-vessel pit could be grouped as reticulate, scalariform, oval, linear and coalescent type. Most of species examined showed oval and linear type.
Genetic and morphological variations of Rhinogobius giurinus were surveyed. Populations of R.giurinus were clearly divided into two forms (Form-A and Form-B). Starch gel variation of this species. Three loci (Aco, Mpi, aGpd) out of 27 showed fixed allelic differences between Form-A and Form-B and they are well differentiated from each other genetically (Rogers' S=0.871). These two forms, moreover, are found to be allopatric in distribution and morphologi cally different in body length and caudal fin color pattern. Therefore, they are considered as two distinct taxa of subspecific rank. In addition, the genetic relationships among 5 taxa within 4 species of the genus Rhinogobius were investigated. Three species of the Rhinogobius brunneus complex (R.sp. OR, R. sp. CB and R. sp. CO) are well differentiated from each other genetically and two taxa of R. giurinus are genetically divergent from three species of the Rhinosotius brunneus complex (average Nei's D=0.603, average Rogers'=0.534).
The development of the lobe pattern in the human dentition plays a part in the form and function of each individual teeth. In order to determin the morphological categories used to describe the occlusal surfaces of the maxillary and mandibular molars, the variation of the developmental grooves which separate each lobe in the molars was examined and analysed. The obtained conclusions were as follow. 1. Most of the maxillary first molars with more distinct and more developmental grooves than the other molars but in most cases of the third molar, a heart-shaped outline due to poorly developed or abscent distolingual cusp was most frequent and in this case the third molar had the 3 cusps separated by the central developmental groove and the buccal developmental groove. 2. In most cases, the mandibular first molar had the 5-cusp type that the groove patter resembles a Y, the second molar the 4-cusp type arranged in such a way that the buccal and lingual developmental grooves meet the central developmental groove at right angle on the occlusal surface and many instances of the mandibular third molars had the 5-cusp thpe with a+groove pattern which separatess the mesiolingual cusp from the distobuccal cusp and the 4-cusp type with a+groove pattern. 3. The maxillary and mandibular third molar were most variable in the developmental groove.
This paper proposes a method to improve speech recognition performance by extracting various pronunciations of the pseudo-morpheme unit from an eojeol unit corpus and generating a new recognition unit considering pronunciation variations. In the proposed method, we first align the pronunciation of the eojeol units and the pseudo-morpheme units, and then expand the pronunciation dictionary by extracting the new pronunciations of the pseudo-morpheme units at the pronunciation of the eojeol units. Then, we propose a new recognition unit that relies on pronunciation by tagging the obtained phoneme symbols according to the pseudo-morpheme units. The proposed units and their extended pronunciations are incorporated into the lexicon and language model of the speech recognizer. Experiments for performance evaluation are performed using the Korean speech recognizer with a trigram language model obtained by a 100 million pseudo-morpheme corpus and an acoustic model trained by a multi-genre broadcast speech data of 445 hours. The proposed method is shown to reduce the word error rate relatively by 13.8% in the news-genre evaluation data and by 4.5% in the total evaluation data.
Diatoms have been used in examining water quality and environmental change in freshwater systems. Here, we analyzed molecular profiling of seasonal diatoms in the Han River, Korea, using the hypervariable region of 18S V1-V3 rRNA and pyrosequencing. Physicochemical data, such as temperature, DO, pH, and nutrients showed the typical seasonal pattern in a temperate region. In addition, cell counts and chlorophyll-a, were recorded at high levels in spring compared to other seasons, due to the diatom bloom. Metagenomic analysis showed a seasonal variation in the phytoplankton community composition, with diatoms as the most frequently detected in spring (83.8%) and winter (69.7%). Overall, diatom genera such as Stephanodiscus, Navicula, Cyclotella, and Discostella were the most frequent in the samples. However, a large number of unknown Thalassiosirales diatoms were found in spring (35.5%) and winter (36.3%). Our molecular profiling revealed a high number of diatom taxa compared to morphological observation. This is the first study of diatoms in the Han River using molecular approaches, providing a valuable reference for future study on diatoms-basis environmental molecular monitoring and ecology.
Prunus sargentii complex of subgenus Cerasus is an Eastem Asiatic plant group that exhibits a broad range of morphological variation and includes P. takesimensis, P. yedosensis, P. verecunda, and P. sargentii. In this study, a morphological analysis was undertaken to determine whether the observed morphological variation was primarily attributable to morphological discontinuities among the taxa. P. sargentii, which distributed eastem area in Korea, northern area in Japan and far east Russia had umbel like inflorescence and additionally was characterized by sticky bud and leaf twigs, compared with P. serrulata complex. Also, P. verecunda in Korea and Japan was characterized by umbel like inflorescence and presence of hair in leaf, petiole and pedicel, and was treated as a variety of P. sargentii. Evidence obtained from multivariate morphometric analyses indicated that the entity of P. takesimensis formed a cohesive group somewhat distinct from P. sargenti.. Especially, P. takesimensis was characterized by relatively small flowers (26-32mm in diameter) and many flowers [(2)3-5] per umbel inflorescence, compared with P. sargentii (34-48mm and 2(3) per inflorescence) and should be recognized as an independent and endeImic taxon in Korea. Additionally, P. yedosensis, which was known to have umbel inflorescence (short peduncle type) with pubescent style based on the type specimen, was comprised of corymb inflorescence (long peduncle type) as well. The morphological differentiation between these two types of P. yedosensis was not considered sufficient to warrant recognition of specific status because of the putative hybrid origin, no distinctive geographical distribution pattern, and existence of various peduncle length on Island Jeju-do of Korea.
