Modification of carbohydrate compositions of 31/36 kDa proteins of plerocercoids (sparganum) of Spirometra mansoni grown in different intermediate hosts

  • We purified specific 31/36 kDa antigenic molecules from sparganum in different intermediate hosts (snakes and mice) and analyzed their monosaccharide compositions. Compositional analysis showed that glucose and man nose concentrations were 2-3 fold higher in the 31/36 kDa molecule purified from snakes than those from mice. This result implies that antigenic glycoproteins of sparganum from snakes might be modified in mammalian sparganosis with respect to their carbohydrate composition.

Variation in Fucoidan Contents and Monosaccharide Compositions of Korean Undaria pinnatifida (Harvey) Suringar (Phaeophyta)

  • Three different forms of Undaria pinnatifida, the southern form (U. pinnatifida f. typica), the northern form (U. pinnatifida f. distans), and Samcheok form (recently cultivated strain), were examined for the contents and compositions of fucoidans. Fucoidans were extracted from the dried edible portions of three forms of U. pinnatifida in low pH condition, mainly by ethanol precipitation and CaCl2 treatment. It was shown that Samcheok form contains 1.8 and 3.5 times more fucoidans than the northern and the southern forms, respectively. The monosaccharide compositions of individual fucoidans were also varied. The fucoidans from the southern and the northern forms were shown to be composed of mainly fucose and galactose with the molar percentage ratios of 83.5%:16.5% and 87.4%:12.6%, respectively, indicating that these are F-type fucoidans. The fucoidan from Samcheok form, however, consisted of fucose (62.7%), galactose (32.9%), and small amount of glucose (4.4%). The results of this study showed that both amount and monosaccharide compositions of fucoidans are variable depending on U. pinnatifida forms.

수 종 상황버섯의 조다당 분획물 조성 (Composition of Crude Polysaccharides in Some Phezllinus Species)

  • 본 연구는 국내에서 재배되거나 수입되어 유통되는 4종 Phellinus속 버섯(구멍쟁이버섯과)의 조다당 분획물의 조성을 비교함으로써 각 종간의 차이에 의한 성분조성의 차이를 검토하기 위하여 수행되었다. 상황버섯의 활성 분획 중의 한 유효성분인 조다당 분획물을 에탄올 냉침법으로 제조한 결과, 조다당 분획물의 취득률은 P. linteus 15.0%, P. baumii 5.1%, P. pini 6.3%, P. igniarius 5.7%로 Phellinus 종에 따라 차이가 있었다. 또한, 그 다당체를 조성하고 있는 단당의 함유량도 버섯의 종에 따라 큰 차이를 보였다. 즉, P. linteus의 경우, 건조중량 1 g 당 함유된 당의 총량은 148.713 mg/g이 었으나, P. baumii는 29.022 mg/g, P. pini는 29.222 mg/g으로 P. lintues의 약 1/5정도의 당 함유량을 나타냈으며, P. igniarius는 P lintues의 1/100정도인 1.546 mg/g의 당을 함유하고 있었다. 모든 Phellinus 속 버섯에서 glucose의 함량이 월등하게 많았으며, 함유되어 있는 각 단당들의 절대량은 종에 따라 차이가 있었다. P. linteus는 mannose와 ribose가 P. baumii와 P. pini보다 적은 양이 함유되어 있는 것을 제외하고는 그 외의 모든 단당을 월등하게 많이 함유하고 있었다. P. baumii와 P pini는 arabinose를 제외하고는 거의 모든 단당의 함량이 유사하였다. P. igniarius는 거의 모든 당의 절대량이 다른 Phellinus 속 버섯에 비하여 매우 적었다.

  • The glycosylation of therapeutic glycoproteins can affect their efficacy, stability, solubility, and half-life. Analyzing the composition of monosaccharides, such as that of neutral and amino sugars, is the first step for elucidating the structure of glycan attached to glycoproteins. In the present study, neutral and amino sugars of lectin obtained from Maackia fauriei were analyzed using an enzyme-linked lectinsorbent assay (ELLA) and high-performance anion exchange chromatography with pulsed amperometric detection (HPAEC-PAD). Peroxidase-labeled lectins such as concanavalin A, Ricinus communis agglutinin, and soybean agglutinin were used for ELLA, since they specifically bind to the monosaccharide residue most frequently encountered in a glycan. The hydrosylate of lectin was prepared by treatment with trifluoroacetic acid, which resulted in the lectin mainly possessing the N-glycan consisting of 98.1 pmol Fuc, 342.1 pmol GlcN, 51.9 pmol Gal, 678.9 pmol Man, and 330.7 pmol Xyl. The present results demonstrate that ELLA and HPAEC-PAD are very effective methods for rapidly estimating the types and relative amounts of monosaccharides in intact glycoproteins.

배양 조건에 따른 상황 버섯의 다당류 생산 및 단당류 구성 변화 (Effect of Cultural Conditions on Polysaccharide Production and its Monosaccharide Composition in Phellinus linteus L13202)

  • 상황 (Phellinus linteus) 의 균사체 생장과 다당류 생산 및 다당류에서의 단당류 구성비에 대한 배양 조건의 영향을 조사하였다. 여러 가지 배지를 사용하여 실험한 결과 GP(glucose-peptone)배지에서 균사 생장이 가장 양호하여, 이 배지를 사용하여 탄소원 및 pH 영향을 조사하였다. 탄소원중 glucose에서 최대 균사체 생장 (0.9 g/100 ml)을 보였고, 다당류의 수율은 $5{\sim}14%$ 로서 mannose에서 최대이었다. 최적 pH는 7 이였으며, 산성 pH에서는 비교적 좋은 생장을 보였으나, 알칼리 pH에서는 생장이 급격히 감소하였다. 한편 pH에 따른 다당류 수율은 $5{\sim}8%$로 큰 변화를 보이지 않았다. 또한 탄소원 및 pH에 따른 단당류 구성비의 변화가 관찰되었다. 즉 탄소원에 따라서 다당류에서 glucose, mannose, 및 galactose 함량은 각각 $80{\sim}95%,\;3{\sim}12%$, 그리고 $2{\sim}10%$이었다. 한편 pH 변화에 따라서, glucose는 $83{\sim}92%$, mannose 는 $4{\sim}8%$, 그리고 galactose는$5{\sim}8%$의 구성비를 보였다. 이 결과는 배양 조건에 의한 항암면역활성 다당류의 구조의 변환 및 생산 증강이 가능함을 시사한다.

Chemical Composition and Active Antioxidants of Eucommia ulmoides Oliv. Bark

  • Chemical composition of the Eucommia ulmoides bark, including extractives, proximate, mineral, fatty acid and monosaccharide compositions, was studied The most abundant mineral was calcium (533.17 mg/l00 g). $\alpha-linolenic$ acid (24.7%) and linoleic acid (24.3%), showed higher contents among the fatty acids. Major monosaccharides of E. ulmoides balk were arabinose (13.94 mg/g), xylose (18.91 mg/g) and glucose (119.7 mg/g). From the bark of E ulmoides, four compounds were isolated and their structures were elucidated as caffeic acid (I), kaempferol (II), quercetin (III) and isoquercitrin (IV) by spectroscopic analysis such as NMR and MS, including cellulose TLC and other chemical evidence such as hydrolyzation and acetylation. The antioxidant activities of four isolated compounds were evaluated by DPPH free radical scavenging, hydroxyl scavenging and reducing power assays. The results indicated that all the isolated compounds showed higher DPPH radical scavenging activity than $\alpha-tocopherol$ and BHT that were used as positive controls and these four compounds exhibited considerable reducing power and hydroxyl radical (OH) scavenging activity. Considering from the results above, it suggests that the E. ulmoides bark is a potential natural source of antioxidant material.

Isolation and Characterization of a Water-Soluble Polysaccharide from the Mycelia of Solid Cultured Agaricus blazei Murill

  • A fraction (CMEx-AH-${\beta}$) of water-soluble polysaccharides, showing selective antitumor activities, was isolated from the mycelia of solid cultured Agaricus blazei Murill by hot-water extraction, ethanol precipitation, and series of chromatography. Chemical characteristics of CMEx-AH-${\beta}$, were as follows: carbohydrate content, 48%; monosaccharide composition, Man:Glu:Gal (2:93:5); molecular weight, $2{\times}10^5$; uronic acid content, 6.2%. Fundamental structure of CMEx-AH-${\beta}$, was deduced as ${\beta}-(1\;{\rightarrow}\;6)$-D-glucan with ${\beta}-(1\;{\rightarrow}\;3)$-D-glucosidic side chains based on results of methylation and $^{13}C$ NMR spectroscopic analyses.

  • Fraction (PMEx-AH-${\beta}$) of water-soluble polysaccharide, showing stimulating activity against macrophages, was isolated from mycelia of solid cultured Phellinus linteus by hot-water extraction, ethanol precipitation, and chromatography. Chemical characteristics of PMEx-AH-${\beta}$ were as follows: carbohydrate content, 71%; monosaccharide composition, Man:Glu:Gal (9:64:27); molecular weight, $1-7{\times}10^4$; uronic acid content, 6.8%. Fundamental structure of PMEx-AH-${\beta}$ is deduced as ${\beta}$-($1{$\rightarrow}6$)-D-glucan with ${\beta}$-($1{\rightarow}3$)-D-glucosidic side chains based on methylation analysis.

Determination of Mono- and Oligosaccharides Derivatized with p-Aminobenzoic Ethyl Ester by Reverse Phase HPLC

  • Mono- and oligosaccharides are derivatized with p-aminobenzoic ethyl ester (ABEE), strongly absorbs UV light at 254 nm, in the presence of sodium cyanoborohydride. C18-bonded silica column is used for the separation of sugar-ABEE derivatives in an isocratic mode. RP-HPLC conditions are optimized by using ternary mixture as mobile phase and $45^{\circ}C$ as a column temperature. Sugar-ABEE derivatives are separated well within a short run time (ca. 25 min) by reverse-phase partition chromatographic mode. The ($1{\rightarrow}6$) linkage type of dihexose-ABEE derivatives has shorter retention time than ($1{\rightarrow}4$)-linkage type. After acid hydrolysis of glycoproteins with 2M trifluoroacetic acid, monosaccharide composition and contents are determined. This procedure is very useful for the simultaneous analysis of neutral and amino sugars in a single chromatographic step using RP-HPLC without reacetylation of deacetylated amino sugars, which are produced by acid hydrolysis.

매생이 유래 올리고당의 추출 분리 및 Angiotensin I Converting Enzyme 저해능 분석 (Analysis of Angiotensin I Converting Enzyme Inhibitory Activity of Oligosacchride Extracted from Capsosiphon fulvescens)

  • The hydrolysates prepared with various enzyme digestion of Capsosiphon fulvescens were used to measure the inhibitory effects against angiotensin I converting enzyme (ACE). The commercially available enzymes such as Celluclast, Viscozyme, Lysing enzyme, Flavourzyme, Alcalase and Pectinex were used to digest C. fulvescens and produce hydrolysates. The maximum ACE inhibitory activity was observed using Alcalase hydrolysis (72.9%). The optimal conditions of Alcalase extraction were pH 8.0 and extraction time for 12 hr. The hydrolysates were fractionated using preparative-LC and anion-exchange chromatography on DEAE-cellulose and the fraction B and B-2 were isolated. The ACE inhibitory activity of fraction B-2 by anion-exchange chromatography was 82.6%. The molecular weight of fraction B-2 estimated using size exclusion chromatography was about 1 kDa. The monosaccharide composition of the fraction B-2 was determined to be mannose (1.1%), glucuronic acid (1.3%), galactose (1.3%) and glucose (96.3%).
