Parkin Reduces Expression of Monocyte Chemotactic Protein-1 (MCP-1) in TNF-${\alpha}$-stimulated MCF7 Breast Cancer Cells

  • Parkin is a putative tumor suppressor protein and its expression is frequently reduced or absent in several types of tumors. In this study, we examined the role of Parkin in mRNA expression of monocyte chemotactic protein-1 (MCP-1) in the breast cancer cell line MCF7. Expression of MCP-1 mRNA increased after TNF-${\alpha}$ treatment. However, overexpression of Parkin induced a decrease in expression of MCP-1 mRNA in TNF-${\alpha}$-stimulated MCF7. This decrease in MCP-1 mRNA by Parkin overexpression occurred in a dose- and time-dependent manner. Using a wound scratch assay, we found that Parkin overexpression in MCF7 cells also resulted in a decrease in cell migration. These results suggest that Parkin down-regulates MCP-1 synthesis leading to decreased migration of tumor cells. We suggest that one possible mechanism by which Parkin acts as a tumor suppressor is by inhibiting migration or metastasis of cancer cells.

The Effects of Victicis Fructus & Bupleuri Radix on the Allergic rhinitis (만형자(蔓荊子), 시호(柴胡)의 알레르기성 비염에 대한 효과)

  • Background and Objectives : In oriental Medicine, there is a concept that herb works only in it's own specific site called 'Guikyung(歸經)'. If two herbs have same effect and different Guikyung, two herbs would work at different site respectively. so we want to prove that whether Guikyung has significance or not in experimentally. Methods : In order to investigate the effects of Victicis Fructus & Bupleuri Radix on the Allergic rhinitis, BALB/c mouse was used. Control and experimental group 1, 2 were induced allergic rhinitis. Experimental group 1 was administered Victicis Fructus for 28 days and group 2 was administerd Bupleuri Radix for same period. WBC, neutrophil, lymphocyte, monocyte, IL-4 and $IFN-{\gamma}$ were measured and histological examination was performed. The statistical significance was examined by Independent Samples T test. Results : 1. WBC, neuoophil, lymphocyte, monocyte, IL-4 and $IFN-{\gamma}$ showed no statistically significant differences in three groups. 2. Histological examination had no differences in three groups. Conclusion: Victicis Fructus & Bupleuri Radix have no effect on allergic rhinitis and the reason is estimated that Guikymg plays a important roll to determine the effect of herb from the viewpoint of oriental medicine.

Immunoenhancing Effect of 1,2-Benzopyrone on the Oxidative Burst Activity to Phagocytic Response of Canine Peripheral Blood Phagocytes (개 말초혈액 탐식세포의 탐식반응에 따른 순간산소 과소비현상에 있어 1,2-benzopyrone의 면역증강효과)

  • 1,2-benzopyrone can stimulate macrophages to increase the ability of phagocytosis. Peripheral blood polymorphonuclear cells (PMN) and macrophages destroy microbial organisms with reactive oxygen species (ROS), called oxidative burst activity (OBA). This study was undertaken to determine whether 1,2-benzopyrone affects the OBA on the phagocytic response of canine peripheral blood phagocytes. The OBA of phagocytes in the addition or absence of latex beads was analyzed by flow cytometry system using dihydrorhodamine 123 (DHR). The direct treatments of 1,2-benzopyrone have no effect on the OBA of peripheral blood mononuclear cells (PBMC), PMN and monocyte-rich cells regardless of addition of latex beads. When latex beads are added to PMN, the OBA of PMN was remarkably enhanced by culture supernatant from PBMC but not PMN treated with 1,2-benzopyrone. Similary, it was also enhanced by human recombinant (hr) $TNF-\alpha.$ However, when latex beads were not added to PMN, its OBA was not enhanced by culture supernatant from either PBMC or PMN treated with 1,2-benzopyrone. The OBA of latex beads-phagocytized PBMC and monocyte-rich cells was not enhanced by culture supernatant from either PBMC or PMN treated with 1,2-benzopyrone. These results strongly suggested that 1,2-benzopyrone has an immunoenhancing effect on the OBA of PMN when phagocytic response occurred only. This enhanced OBA may be mediated through active humoral substance(s), such as $TNF-\alpha,$ produced by PBMC stimulated with 1,2-benzopyrone.

Butein, a tetrahydroxychalcone, suppresses pro-inflammatory responses in HaCaT keratinocytes

  • Up-regulation of cell adhesion molecules and proinflammatory cytokines contributes to enhanced monocyte adhesiveness and infiltration into the skin, during the pathogenesis of various inflammatory skin diseases, including atopic dermatitis. In this study, we examined the anti-inflammatory effects of butein, a tetrahydroxychalcone, and its action mechanisms using TNF-α-stimulated keratinocytes. Butein significantly inhibited TNF-α-induced ICAM-I expression and monocyte adhesion in human keratinocyte cell line HaCaT. Butein also decreased TNF-α-induced pro-inflammatory mediators, such as IL-6, IP-10 and MCP-1, in HaCaT cells. Butein decreased TNF-α-induced ROS generation in a dose-dependent manner in HaCaT cells. In addition, treatment of HaCaT cells with butein suppressed TNF-α-induced MAPK activation. Furthermore, butein suppressed TNF-α-induced NF-kappaB activation. Overall, our results indicate that butein has immunomodulatory activities by inhibiting expression of proinflammatory mediators in keratinocytes. Therefore, butein may be used as a therapeutic agent for the treatment of inflammatory skin diseases. [BMB Reports 2015; 48(9): 495-500]

  • Effect of Sunbangpaedoktang, which has been used for atopic dermatitis(AD), on the cytokine secretory function of monocyte, which was isolated and cultured from peripheral blood of AD patients, were evaluated. The results were as follows: 1. In AD patients, Sunbangpaedoktang exhibits statistically significant inhibitory effect on IL-4. 2. In AD patients, Sunbangpaedoktang exhibits statistically significant inhibitory effect on IL-5(p<0.05). 3. In AD patients, Sunbangpaedoktang exhibits statistically significant inhibitory effect on $IFN-{\gamma}$(p<0.01). 4. In sample group, comparing the cytokine concentration change rate of AD patients with that of NAD patients IL-4 AD patients exhibits statistically significant decrease(p<0.05).

Matrix Metalloproteinase Inhibitors Attenuate Neuroinflammation Following Focal Cerebral Ischemia in Mice

  • The aim of this study was to investigate whether matrix metalloproteinase (MMP) inhibitors attenuate neuroinflammation in an ischemic brain following photothrombotic cortical ischemia in mice. Male C57BL/6 mice were anesthetized, and Rose Bengal was systemically administered. Permanent focal ischemia was induced in the medial frontal and somatosensory cortices by irradiating the skull with cold white light. MMP inhibitors, such as doxycycline, minocycline, and batimastat, significantly reduced the cerebral infarct size, and the expressions of monocyte chemotactic protein-1 (MCP-1), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), and indoleamine 2,3-dioxygenase (IDO). However, they had no effect on the expressions of heme oxygenase-1 and neuroglobin in the ischemic cortex. These results suggest that MMP inhibitors attenuate ischemic brain injury by decreasing the expression levels of MCP-1, TNF-${\alpha}$, and IDO, thereby providing a therapeutic benefit against cerebral ischemia.

The Effect of Norepinephrine on Immune function in elderly people (노르에피네프린이 노인의 면역기능에 미치는 영향)

  • Purpose: The purpose of this study was to examine how norepinephrine affects immunity in patients over age 65. Methods: We enrolled 25 male and female subjects age 65 or older. A low frequency electroacupuncture (EA) device was used to stimulate acupoint Hogu (L14). The 2 Hz frequency EA was applied to the acupoint for 20 minutes. Leukocyte subtypes-including neutrophils, lymphocytes, monocytes, eosinophils, and basophil-were then measured. The immunoglobulins IgG and IgM were also quantified. The data were finally analyzed using Wilcoxon singed-rank test and regression test as part of the SPSS WIN v. 10.1 program. Results: As norepinephrine levels decreased after EA stimulation, neutrophil, lymphocyte, and monocyte levels increased, and eosinophil and neutrophils levels decreased. Neutrophil and monocyte levels did not change to a statistically significant degree, but eosinophil levels showed a statistically significant decrease (p<0.05). Immunoglobulin IgG showed a statistically significant increase (p<0.05). Conclusion: This study showed that norepinephrine does affect immunity in persons over the age of 65. This indicates that there is an interaction between the nervous system and the immune system, and interaction that plays a crucial roles in the body's immune resistance and homeostasis.

Immunostimulating Effect of 1,2-Benzopyrone on Phagocytic Response of Canine Peripheral Blood Phagocytes (개 말초혈액 탐식세포의 탐식반응에 대한 1,2-benzopyrone의 면역자극 효과)

  • 1,2-benzopyrone has been shown to affect on the activation and stimulation of macrophage. To examine the immunostimulating effect of 1,2-benzopyrone on the phagocytic response of canine peripheral blood mononuclear cells (PBMC) as well as polymorphonuclear cells (PMN), the phagocytic activity of phagocytes was analyzed by flow cytometry system using FITC-labelled latex. The 1,2-benzopyrone did not show any direct effect on phagocytic response of PBMC and PMN. But it showed an enhanced effect on the phagocytic response of monocyte-rich cells fractioned by cell size from dot plot profile in flowcytometric cytography of PBMC. The phagocytic activity of these cells was also enhanced by addition of culture supernatant from PBMC treated with 1,2-benzopyrone. Similarly, the phagocytic activity of PMN but not PBMC in the same procedures was enhanced by culture supernatant from PBMC treated with 1,2-benzopyrone. However, the culture supernatant from PMN treated with 1.2-benzopyrone did not show the enhancing effect on phagocytic activity for monocyte-rich cells and PMN. These results, therefore, suggested that enhanced phagocytic activity of canine peripheral blood PMN and monocytes may be mainly mediated by humoral factor(S) released from PBMC treated with 1,2-benzopyrone.

Study on the Mechanism of Radiation-induced MCP-1 Expression in RAW264.7 Macrophage Cells (RAW264.7 대식세포에서 방사선에 의한 MCP-1 발현 기작 연구)

  • The purpose of this study was to investigate the expression mechanism of MCP-1 in gamma-irradiated RAW 264.7 macrophage cells. MCP-1 plays an important role in attracting monocyte to injured site at the early inflammation stage. However the production mechanism of MCP-1 by gamma-irradiation in RAW 264.7 macrophage cells was almost undiscovered. We found that MCP-1 was produced in RAW 264.7 macrophage cells by irradiation with 5 Gy. And these inceases were attenuated by specific inhibitors treatment, such as $NF-{\kappa}B$, JNK, ERK, JAK2, and Pyk2. These results indicate that radiation-induced MCP-1 production is mediated by MyD88- and TRIF-dependent pathways in RAW 264.7 macrophage cells. Furthermore, gamma-irradiation induced heme oxygenase-1 (HO-1) expression in RAW 264.7 macrophage cells. However this induction level was reduced before MCP-1 and $IFN-{\beta}$ production.

Side effects of treatment with busulfan at high doses in dogs

  • Busulfan is the most commonly used drug for preconditioning during the transplantation of hematopoietic stem cells and male germ cells. Here, we describe side effects of high doses of busulfan in male mongrel dogs. Busulfan was intravenously administered to three groups of dogs at doses of 10, 15, and 17.5 mg/kg body weight. The total white blood cell, neutrophil, eosinophil, lymphocyte, monocyte, and platelet counts steadily reduced in a dose-dependent manner following busulfan treatment. The white blood cell, neutrophil, and monocyte counts recovered after 6 weeks of busulfan treatment, however, the eosinophil, lymphocyte, and platelet counts remained unaltered. Additionally, there was one fatality in the each of the groups that were administered 15 and 17.5 mg/kg busulfan. The gross lesions included severe hemorrhage in the stomach, intestinal tracts, mesentery and urinary bladder. Microscopic investigation revealed severe pulmonary edema and hemorrhage in the lungs, and severe multifocal to coalescing transmural hemorrhage in the intestines and urinary bladder. These results indicated that treatment with busulfan at doses higher than 15 mg/kg initiates severe bleeding in the internal organs and can have fatal results.
