Triploids are a useful tool for biomass production and molecular breeding of trees with a long life span. Triploids of the poplar 'Hyunsasi' (Populus alba ${\times}$ P. glandulosa) have been developed by crossing between female diploids and a male tetraploid. The tetraploid was developed around the 1970s at Korea Forest Research Institute by colchicine-induced chromosome doubling. Seedlings of the $F_1$ generation were analyzed using flow cytometry to verify their ploidy status. The mean relative fluorescence index of 3 F1 poplars, labeled as Line- 1, Line-17, Line-18, were approximately 1.5 times higher than those of diploid poplars, and the results clearly indicated that they were triploids. The phenotype of the F1 poplars included larger leaves and thicker stem than diploids, and abnormal leaf morphology, especially in the triploid 'Line-18'. Three triploid lines developed roots more slowly and had less roots than diploid. However, 3 poplar cytotypes (2x, Line-1, Line-17) rooted within 10 days on MS medium. In contrast, compared with the 3 cytotypes, the Line-18 showed about 80% and 70% in the rooting rate and the number of roots. The triploid poplars could be directly utilized for biomass production and with their sterility, they could serve as basic material for genetic transformation. In addition, flow cytometric analysis proved to be an effective and reliable method for screening forest trees for their ploidy level.
Plant regeneration system from floral stem of Mymphoides indica via somatic embryogenesis was established. After four weeks of culture onto 1/2MS medium containing 2,4-D, pale-yellow globular structures and calluses were formed on the cut surface of floral stem explants. Upon transfer to 1/2MS basal medium, pale-yellow globular structures were developed into somatic embryos and normal plantlets. These results indicated that pale-yellow globular structures and calluses from floral stem were globular embryos and embryogenic calluses, respectively. The frequency of embryogenic callus formation from floral stem was reached to nearly 100% when floral stem was cultured onto 1/2Ms medium supplemented with low concentration of 2,4-D (0.1 to 0.3 mg/L). However, the higher concentration of 2,4-D resulted in decrease of the frequency of embryogenic callus formation. In this study, low concentration of 2,4-D had a stimulative role in embryogenic callus formation, whereas BA showed inhibitory role in callus formation. In comparison to floral stem, leaf explants showed low frequency of embryogenic callus formation. The highest frequency of embryogenic callus formation from leaf explants was 9.5% when leaf explants were cultured onto 1/2MS medium supplemented with 0.3 mg/L of 2,4-D. The plant regeneration system of Nymphoides indica established in this study, might be applied to mass proliferation, conservation of genetic resources and genetic transformation for molecular breeding.
The importance of biological resources has been gradually increasing, and mollusks have been utilized as main fishery resources in terrestrial ecosystems. But little is known about genomic and transcriptional analysis in mollusks. This is the first report on the transcriptomic profile of Meretrix lusoria. In this study, we constructed cDNA library and determined 542 of distinct EST sequences composed of 284 singletons and 95 contigs. At first, we identified 180 of EST sequences that have significant hits on protein sequences of the exclusive Mollusks database through BLASTX program and 343 of EST sequences that have significant hits on NCBI NR database. We also found that 211 of putative sequences through local BLAST (blastx, E < e-10) search against KOG database were classified into 16 functional categories. Some kinds of immune response related genes encoding allograft inflammatory factor 1 (AIF-1), B-cell translocation gene 1 (BTG1), C-type lectin A, thioester-containing protein and 26S proteasome regulatory complex were identified. To determine phylogenetic relationship, we identified partial sequences of four genes (COX1, COX2, 12S rRNA and NADH dehydrogenase) that significantly matched with the mitochondrial genomes of 3 species-Ml (Meretrix lusoria), Mp (Meretrix petechialis) and Mm (Meretrix meretrix). As a result, we found that there was a little bit of a difference between sequences of Korean isolates and other known isolates. This study will be useful to develop breeding technology and might also be helpful to establish a classification system.

This study was conducted to analyze the reduction of quality and ripening rate of rice due to submergence during ripening stage and identify the physiological cause. Korean japonica type rice cultivars, Nampyeong was used in the experiment. The following 7, 14 day after heading, they were then moved into submergence treatment facility and we conducted the tests under three different submergence conditions - T1(clear water and overhead submergence), T2(Muddy water and exposure of Flag leaf end ), T3(Muddy water and overhead submergence), and the treatment lasted for 4days. The decrease of ripening rate was most severe in T3 treatment at 7days after heading, and the decrease of head rice ratio was most severe in T3 treatment at 14days after heading. Meanwhile the starch synthesis was inhibited, as the supply of assimilation products was inhibited in grain during the submergence treatment. And in stem, sucrose content was increased because the soluble carbohydrates accumulated before heading were decomposed. These changes may be due to the consumption of respiratory substrate in anaerobic conditions and the inhibition of the production of photosynthetic products by light interception. In order to see what kind of reaction occurs at the molecular level, we examined the degree of RNA expression in grain, stem and leaf. First, the expression of rna associated with starch synthesis in grain was decreased in all submergence treatment. and especially Ospul(DBE) was more decreased in 14days after heading treatment than 7days after heading treatment. This difference can explain the result that the decrease of head rice ratio was more severe at 14days after heading. And in stem, the expression of rna associated with the supply of assimilation products was decreased in submergence treatment. Finally in leaf, the expression of rna(ADH, ALDH) associated with anaerobic respiration was increased, while the expression of rna associated with photosynthesis was decreased. These results of physiological analysis can used to develop the cultivation technique and to offer the information for breeding the cultivars with tolerant characteristics to submergence stress during ripening stage in rice.

Aluminum is the most abundant metallic element in the Earth's crust and considered as the most limiting factor for plant productivity in acidic soils. The inhibition of root growth is recognized as the primary effect of Al toxicity. Seeds of wheat cv. Keumkang (Korean cultivar) were germinated on petridish for 5 days and then transferred hydroponic apparatus which was treated with $0{\mu}M$$AlCl_3$ (control), $100{\mu}M$$AlCl_3$ and $150{\mu}M$$AlCl_3$ for 5 days. The length of roots, shoots and fresh weight of wheat seedlings were decreased under aluminum stress. The concentrations of $K^+$, $Mg^{2+}$ and $Ac^{2+}$ were decreased whereas $Al^{3+}$ and $P_2O_5{^-}$ concentration was increased under aluminum stress. Using confocal microscopy, the fluorescence intensity of aluminum was increased with morin staining. In this study, a proteome analysis was performed to identify proteins, which is responsible to aluminum stress in wheat roots. In 10-day-old seedlings, proteins were extracted from roots and separated by 2-DE, stained by CBB. Using image analysis, a total of 47 differentially expressed protein spots were selected, whereas 19 protein spots were significantly up-regulated such as s-adenosylmethionine, oxalate oxidase, malate dehydrogenase, cysteine synthase, ascorbate peroxidase and 28 protein spots were significantly down-regulated such as heat shock protein 70, o-methytransferase 4, enolase, amylogenin by aluminum stress following protein spots analyzed by LTQ-FTICR mass spectrometry. The results provide the global picture of Al toxicity-induced alterations of protein profiles in wheat roots, and identify the Al toxicity-responsive proteins related to various biological processes that may provide some novel clues about plant Al tolerance.

A new virus-like disease of tomato showing chlorotic spots, malformation and necrosis on leaves, and chlorotic blotching, rings, and necrosis on fruits was observed around Sacheon, Gyeongsangnam-do, Korea in 2004. Host range analysis could not differentiate 4 field isolates collected from tomatoes showing different symptoms but identified them as Tomato bushy stunt virus (TBSV). TBSV-tsf2 isolate induced symptoms in indicator plants similar to those caused by the TBSV-C, -S and -Nf. As the isolate could not systemically infect Chenopodium quinoa, the isolate might belong to the previously described TBSV-S isolate. TBSV-tsf2 isolate caused similar cytological alterations that were similar to that generally caused by previously reported TBSV isolates. TBSV-tsf2 isolate, however, could be considered to belong to new strain of TBSV because masses of small electron-dense patches that were not observed from the previously described TBSV. The complete nucleotide sequences of the genomic RNA of 4739 nt excluding non-translated sequences at both termini have been determined and compared to sequences of other TBSV strains. The complete nucleotide sequence identity among TBSV isolates was 98.9% to 99.7%, and to the other tombusviruses ranged from 80.8% to 94.9%. Comparison of the amino acid sequences all five ORFs with those of other TBSV strains shows a similar genomic organization, and high percentage of amino acid sequence homology with TBSV-Nf than TBSV-S isolate. Since the TBSV symptoms were only observed in Sacheon fields where imported seeds from Japan were planted, the TBSV incidence probably caused by the planting contaminated tomato seeds and thus require more through quarantine procedure to prevent settlement of TBSV in Korea. Altogether, these results support that the Korean isolate of TBSV infecting tomato might be new strain.
In this review, we described the catalogues of the rice proteome which were constructed in our program, and functional characterization of some of these proteins was discussed. Mass-spectrometry is the most prevalent technique to rapidly identify a large number of proteome analysis. However, the conventional Western blotting/sequencing technique has been used in many laboratories. As a first step to efficiently construct protein cata-file in proteome analysis of major cereals, we have analyzed the N-terminal sequences of 100 rice embryo proteins and 70 wheat spike proteins separated by two-dimensional electrophoresis. Edman degradation revealed the N-terminal peptide sequences of only 31 rice proteins and 47 wheat proteins, suggesting that the rest of separated protein sports are N-terminally blocked. To efficiently determine the internal sequence of blocked proteins, we have developed a modified Cleveland peptide mapping method. Using this above method, the internal sequences of all blocked rice proteins(i, e., 69 proteins) were determined. Among these 100 rice proteins, thirty were proteins for which homologous sequence in the rice genome database could be identified. However, the rest of the proteins lacked homologous proteins. This appears to be consistent with the fact that about 45% of total rice cDNA have been deposited in the EMBL database. Also, the major proteins involved in the growth and development of rice can be identified using the proteome approach. Some of these proteins, including a calcium-binding protein that tuned out to be calreticulin, gibberellin-binding protein, which is ribulose-1.5-bisphosphate carboxylase/oxygense active in rice, and leginsulin-binding protein in soybean have functions in the signal transduction pathway. Proteomics is well suited not only to determine interaction between pairs of proteins, but also to identify multisubunit complexes. Currently, a protein-protein interaction database for plant proteins(http://genome.c.kanazawa-u.ac.jp/Y2H)could be a very useful tool for the plant research community. Also, the information thus obtained from the plant proteome would be helpful in predicting the function of the unknown proteins and would be useful be in the plant molecular breeding.
This study was performed to survey prevalence of Copper metabolism domain containing 1 (COMMD1) mutation using molecular diagnostic method in a population of Bedlington terriers in Korea. COMMD1 gene (formerly MURR1) functions as a regulator of sodium transport and copper metabolism. The deletion of exon 2 of the COMMD1 gene causes copper toxicosis in Bedlington terriers. Bedlington terriers with this autosomal recessive disorder were shown to have the elevated liver copper levels due to genetic derangement in the biliary copper excretion pathway. DNA samples were extracted from whole blood collected from 257 Bedlington terriers (109 males, 148 females) of pet dog clubs in Korea. A multiplex PCR was carried out to detect of exon 2 deletion of COMMD1 gene. In this study, it was possible to know the existence and prevalence of exon 2 deletion of COMMD1 in Bedlington terriers in Korea. Of the 257 samples, 131 (51%) were wild type homozygous for the normal COMMD1 gene, 108 (42%) were heterozygous, having both normal and mutated copy of the COMMD1 gene. The eighteen (7%) were mutant type homozygous. The results of genetic analysis could help establish proper management strategy and selective breeding program to prevent COMMD1 mutation in Bedlington terriers in Korea.
Xenotransplantation may help to overcome the critical shortage of human tissues and organs for human transplantation, Swine represents an ideal source of such organs owing to their anatomical and physiological similarities to human besides their plentiful supply, However, the use of organs across the species barrier may be associated with the risk of transmission of pathogens, specially porcine endogenous retroviruses (PERVs).• Although most of these potential pathogens could be eliminated by pathogen-free breeding, PERVs are not eliminated by this treatment. PERVs are integrated into the genome of all pigs and produced by normal pig cells and infect human cells. They belong to gamma retroviruses and are of three classes viruses: A, B and C. In the present study, PCR based cloning was performed with chromosomal DNA extracted from pigs from domestic pigs in Korea. Amplified PCR fragments of about 1.5 Kb, covering the partial env gene, were cloned into pCR2.l-TOPO vectors and sequenced. A total of 91 env clones were obtained from domestic pigs, Berkshire, Duroc, Landrace and Yorkshire in Korea. Phylogenetic analysis of these genes revealed the presence of only PERV class A and B in the proportion of 58 % and 42 %, respectively. Among these, 28 clones had the correct open reading frame: 18 clones in class A and 10 clones in class B. Since both these PERV classes are polytropic and have the capacity to infect human cells, our data suggest that proviral PERVs have the potential to generate infectious viruses during or after xenotransplantation in human.
Radiation technique in agriculture was initiated to develop mutant rice. Seeds of Daechungbyeo rice were irradiated with 250 Gy gamma ray for the purpose of inducing and selecting rice variants. Some quantitative traits of the variants in M$_{8}$ generation were evaluated and RAPD analysis was carried out. Variants showed a wider range of agronomic characteristics in both a positive and a negative direction compared with their original variety. The new mutants were characterized by an increased or decreased in plant height, lodging resistance and shorter panicle. RAPD analysis showed that polymorphic bands were presented in most of the primers. In comparison with the original variety, variants were classified into four groups through UPGMA analysis. Among mutants no. 91, 139, 140 and 141 was ranked as salt tolerance and the proline content of these mutants was more increased than that of original variety. The lines of 139, 140 and 141 had the highest genetic distance as compared to original variety in the dendrogram. It is expected that such variants will be useful not only for studying molecular genetics but also for breeding research and genetic analysis.s.
