Cultural Characteristics and Fruitbody Formation of Phellinus gilvus (Phellinus gilvus의 배양적 특성과 자실체 형성)

  • For artificial cultivation of Phellinus gilvus (Schw.) Pat we have conducted a study on cultural characteristics and condition of fruitbody formation. The optimum temperature was about $30^{\circ}C$ at pH $6.0{\sim}7.0$ for mycelial growth. Optimum sawdust media were oak sawdust+willow sawdust(5:5, V/V), oak sawdust+willow sawdust+rice bran (4.5:4.5:1, V/V) and oak sawdust+pine sawdust+rice bran(4.5:4.5:1, V/V) and, the spawn incubation period was about $25{\sim}26$ days. Mycelial growth in the inner portion of oak log was 200 mm after 60 days and duration for the first fruitbody primordia were formed about 90 days after inoculation.

  • The EO separation and quantitative determination of polysorbate 20, polysorbate 40, polysorbate 60 and polysorbate 80 was carried out by reversed phase HPLC. The water/acetonitrile was used for the mobile phase of gradient conditions. An YMC Pack Ph ($250mm{\times}4.6mm$ i.d., $5{\mu}m$) and Phenomenex C4 ($250mm{\times}4.6mm$ i.d., $5{\mu}m$) and the selected ELSD detector was applied. The analysis results of HPLC showed good linearity with correlation coefficient of $r^2$=0.997 in the rage of $180.2{\sim}980.5{\mu}g/mL$ and detection limit.

Effect of Water Temperature Condition on Growth and Survival of Juvenile Geoduck (Panopea japonica A. Adams, 1850) (사육 수온이 코끼리조개 Panopea japonica (A. Adams, 1850) 치패의 성장과 생존에 미치는 영향)

  • This study was performed to determine the effect of water temperature condition on survival rate and growth of juvenile geoduck (Panopea japonica). Panopea japonica juveniles were bred for 6 weeks at 12, 15, 18, 21, 24 and $27^{\circ}C$ on incubator. Juveniles median lethal times (LT50) were 29 hours in $27^{\circ}C$, 14.5 day in $24^{\circ}C$, 37.4 day in $21^{\circ}C$. Survival rate of water temperature 12, 15 and $18^{\circ}C$ showed a high survival rate in 95.0%, 95.0% and 93.3% at 6 weeks. As a result of culturing for 6 weeks at 12, 15, 18, 21 and $24^{\circ}C$, a daily growth of shell length and total weight was 0.10 mm, 0.12 mm, 0.13 mm, 0.16 mm and 0.14 mm, and 2.21 mg, 2.65 mg, 2.84 mg, 3.13 mg and 2.93 mg. Juvenile shell length and total weight have significantly increased at $15-21^{\circ}C$. However, mortality rate has significantly increased at more than $21^{\circ}C$. As a result, appropriate water temperature for juveniles culturing considered $15-18^{\circ}C$.

Analysis of Meteorological Elements in the Cultivated Area of Hadong Green Tea (하동녹차 재배지역의 기상요소별 분석)

  • Characteristics of meteorological elements were analyzed at Hwagae and Agyang where are the representative areas of Hadong green tea cultivation in Korea. An automatic weather monitoring system (AWS) and a simple data log were employed to measure meteorological data such as temperature, relative humidity, precipitation, and wind direction and speed for 2009. The annual average air temperature of Hwagae and Agyang was 14.5 and 14.2, respectively, showing the warmest month in August ($25.4^{\circ}C$ for Hwagae and $24.9^{\circ}C$ for Agyang) and the coldest month in January ($0.3^{\circ}C$ for Hwagae and $0.2^{\circ}C$ for Agyang). Annual average of daily temperature difference (= daily maximum temperature - daily minimum temperature) was $11.3^{\circ}C$ for Hwagae and $11.1^{\circ}C$ for Agyang. Hwagae and Agyang had 62.7% and 65.3% of the annual average relative humidity, respectively. Annual precipitation was 1387 mm for Hwagae and 1793 mm for Agyang of which were higher of 605mm for Hwagae and 835 mm for Agyang compared to that in 2008. Majority of precipitation occurred between May and August, attributing 77.6% for Hwagae and 76.6% for Agyang to the annual precipitation. The annual total sunshine duration was 2054.3 hrs in Hwagae with the longest monthly sunshine duration in May (235.1 hrs) and the shortest monthly sunshine duration in July (102.5 hrs). Dominant wind direction changed seasonally from northwesterly wind in fall and winter to southeasterly wind in spring and summer. The annual average wind speed was 1.5 m $s^{-1}$ with the highest monthly wind speed of 2.0 m $s^{-1}$ in December and the lowest monthly wind speed of 1.1 m $s^{-1}$ in February. It is expected that continuous observation and assessment of meteorological data will improve our understanding of optimal environmental conditions for green tea cultivation and be used for developing models of green tea cultivation in the Hadong area.

Effect of Low Temperature and Hot-Water Treatment on Germinatino of Seeds in Dendropanax morbifera $L_{EV}$. (저온 및 온탕 침지가 황칠나무 종자의 발아에 미치는 영향)

  • This study was carried out to find out the method of increasing seed germination rate of Dendropanax morbifera LEV. was drupaceous fruit. The length and width of ovarywas 7.8 to 10.7mm, 6.9 to 9.0mm, respectively, and the shape was elliptical. The ovary had 5 loculi with one speed per loculus , but one seed among 5 weeds was exalbuminous seed. The length and width of seed was 6.3 to 7.4mm, 2.0 to2.mm, respectively, and 1,000 seed weight was 14.3 to 18.0g. Seeding after stroring at $10^{\circ}C$ for 60 to 90 days was germinated to 87% . However, fall seeding directly after harvest was increased to 85% at hot-water treatement ($40^{\circ}C$) for 90 to 120 minute.

Brazing characteristics of $ZrO_2$ and Ti-6Al-4V brazed joints with increasing temperature (브레이징 온도 변화에 따른 $ZrO_2$와 Ti-6Al-4V의 접합 특성)

  • Purpose: In this study, brazing characteristics of $ZrO_2$ and Ti-6Al-4V brazed joints with increasing temperature were investigated. Materials and methods: The sample size of the $ZrO_2$ was $3mm{\times}3mm{\times}3mm$ (thickness), and Ti-6Al-4V was $10mm(diameter){\times}5mm(thickness)$. The filler metal consisted of Ag-Cu-Sn-Ti was prepared in powder form. The brazing sample was heated in a vacuum furnace under $5{\times}10^{-6}$ torr atmosphere, while the brazing temperature was changed from 700 to $800^{\circ}C$ for 30 min. Results: The experimental results shows that brazed joint of $ZrO_2$ and Ti-6Al-4V occurred at $700-800^{\circ}C$. Brazed joint consisted of Ag-rich matrix and Cu-rich phase. A Cu-Ti intermetallic compounds and a Ti-Sn-Cu-Ag alloy were produced along the Ti-6Al-4V bonded interface. Thickness of the reacted layer along the Ti-6Al-4V bonded interface was increased with brazing temperature. Defect ratios of $ZrO_2$ and Ti-6Al-4V bonded interfaces decreased with brazing temperature. Conclusion: Thickness and defect ratio of brazed joints were decreased with increasing temperature. Zirconia was not wetting with filler metal, because the reaction between $ZrO_2$ and Ti did not occur enough.

  • Sclerotinia rot of C. japonica caused by Sclerotinia sclerotiorum was observed throught the growing season in the vinylhouse at the Namyangiu, Yangju and Yangpyung areas. The disease was commonly occurred at late autumn after transplanting and continuously increased until spring in next year, The disease was especially severe at the Yangpyung area among three areas. Three isolates of S. sclerotiorum obtained from C. japonica in different geographic areas were evaluated for ability of sclerotia formation and viability under different temperature. Temperature for mycelial growth ranged from 5 to 3$0^{\circ}C$, with optimum temperature at 15~3$0^{\circ}C$. Sclerotia were fewer formed at low temperature, but their size was larger than that at high temperature. Small size of sclerotia was more viable than those of large size at high temperature. Based on the differences in viability of sclerotia, the relative ranks of sclerotia were in order ~2 mm > 3~4 mm > 5 mm~.

The Processing Technology of Soy Protein Meat Analog Using Twin-Screw Extruder - Heat Transfer Analysis of Cooling Die -

  • Soy protein meat analog was produced using a twin-screw extruder attached with a cooling die. Heat transfer analysis was performed for cooling dies with various die sizes at the four different moisture contents of feed during extrusion process. The experimental design consisted of two cooling die widths (30 and 60 mm), three cooling die lengths (100, 200, and 300 mm), four product moisture contents (71.2, 67.0, 61.6 and 55.8%), and water and water plus ethylene glycol as cooling material. When water was used as cooling medium, the values of equivalent overall heat transfer coefficient $(U_e)$ for each die width of 30 and 60 mm were in the range of 187.0 - 341.4 and $358.5-191.6W/m^2^{\circ}C$ depending on the size of die length. Convective heat transfer coefficients between cooling water and inside die wall of cooling channel $(h_c)$ for both die widths of 30 and 60 mm were 588.5, 416.1, and $339.8W/m^2^{\circ}C$ for each die length of 100, 200, and 300 mm. Convective heat transfer coefficients between product and inside die wall of product channel $(h_p)$ for each die width of 30 and 60 mm were in the range of $434.6-888.1W/m^2^{\circ}C$ and $460.7-1014.5W/m^2^{\circ}C$ depending on the size of die length. When water plus ethylene glycol was used as cooling medium, the values of $U_e$ were in the range of $143.9-319.6W/m^2^{\circ}C$ and $177.8-332.7W/m^2^{\circ}C$ for each die width of 30 and 60 mm depending on the size of die length.

Breeding and characteristics of a low-temperature variety oak mushroom (Lentinula edodes) 'Sanjo 708 ho' (표고 중온성 품종 '산조708호' 육성 및 특성)

  • In 2010, a new variety for sawdust cultivation,was produced by monokaryotic-monokaryotic crossing between Sanjo 701ho and FMRI0995. The optimum temperature for Sanjo 708ho mycelial growth was $25^{\circ}C$; mycelial growth on potato dextrose agar was 51.6 mm over 7 days. The fruiting pattern was sporadic, and the optimal temperature range for fruiting was $5{\sim}20^{\circ}C$. Regarding fruiting body characteristics, the pileus was 65.8 mm in size, 16.1 mm in thickness, and hemispherical in shape. The stipe was 42.6 mm in and 19.6 mm in thickness. Mproductivity was good during thelow-temperature period from autumn to spring, with an average productivity of 310.7 g/bag.

Bi2212 Tube Characteristics for SCFCL (한류기용 Bi2212 튜브의 특성)

  • For the practical application on SCFCL, Bi2212 tubes were fabricated by Centrifugal Forming Process (CFP) in terms of many different processing parameters. Typical sizes of tubes were 60, 150 mm in length and 2.5, 3.5, 4.8 mm in thickness. Initially powder was melted by induction heating. The optimum range of melting temperatures and preheating temperature were $1100^{\circ}C$ and $500^{\circ}C$ for 30min respectively. The nominal mold rotating speed was around 1000 RPM. A tube was annealed at $840^{\circ}C$ for 80 hours in oxygen atmosphere. The tube of 50mm x 70mm x 2.5mm, rotated with 1000 RPM showed $I_c=890\;A\;and\;T_c=80$. It was found that the tube processed with faster rate of mold rotation speed, thinner tube thickness and shorter tube length shows better electric characteristics as compared with the tube normally processed. In order to study the uniformity heat and fluid flow analysis tool was adopted along tube.
