Establishment of tissue culture and acclimatization method for in vitro mass propagation of Echeveria laui and Echeveria elegans (에케베리아 라우이(Echeveria laui)와 엘레강스(Echeveria elegans)의 대량증식을 위한 조직배양 및 순화 조건 확립)

  • The objective of this study was to investigate the suitable parts for callus induction and optimal concentrations of growth regulators contained in the medium affecting shooting and rooting Echeveria laui and Echeveria elegans for in vitro mass production. To determine the suitable plant parts for callus induction, the leaves were divided into upper, medium and bottom parts and cultured on MS medium at different concentrations with $0{\sim}2mgL^{-1}\;NAA$ and $0{\sim}4 mgL^{-1}BA$. The upper and middle parts of leaves both showed 100% callus formation rate with $NAA\;1\;mgL^{-1}$ and $BA\;1\;mgL^{-1}$ treatment in E. laui. The middle parts of leaves showed 83.3% callus formation rate at $NAA\;2\;mgL^{-1}$ and BA 4 mgL-1 treatment in E. elegans. The shoot induction rate from callus was highest at $NAA\;0.1\;mgL^{-1}$ and $BA\;3\;mgL^{-1}$ treatment in E. laui and $NAA\;0.3\;mgL^{-1}$ in E. elegans. In addition, the number of shoots formation was 10.4 shoots high in $NAA\;1\;mgL^{-1}$ and $BA\;1\;mgL^{-1}$ treatment in E. laui and 12.0 shoots in most effective $NAA\;1\;mgL^{-1}$ and $BA\;0.1\;mgL^{-1}$ treatment in E. elegans. In the case of acclimatization of regenerated plant, growth characteristics did not show any significant difference (35 ~ 55%) shading with respect to the different ratio of substrate mixture, and it was determined that would be appropriate considered plant height and appearance preference of E. laui and E. elegans. It was established that the optimization of culture condition was responsible for the mass propagation in vitro cultures of E. laui and E. elegans.

Changes in oligosaccharide content during the storage period of maesil cheong formulated with functional oligosaccharides (기능성 올리고당으로 제조한 매실청의 저장기간 중 올리고당 함량 변화)

  • This study was carried out to produce the health functional food maesil cheong by replacing sucrose with isomaltooligosaccharide and fructooligosaccharide. The substitution levels of these oligosaccharides were between 10% and 100%. A 1:1 (w/w) mixture of maesil and sugar was adopted for preparing maesil cheong. The pH of maesil cheong remained unchanged (between 2.72 and 3.00) during 90-day storage period, regardless of oligosaccharide content. Citric and malic acids were identified in maesil cheong; citric acid accounted for 71-82% of the total organic acid content. Sucrose was completely liquefied in the sample after 30 days and was hydrolyzed steadily into fructose and glucose over the storage period. More than 75% of isomaltooligosaccharides remained in maesil cheong after 90 days when sucrose was completely replaced with isomaltooligosaccharide. However, fructooligosaccharides were mostly decomposed at the end of storage period. Thus, isomaltooligosaccharides may be suitable for acidic maesil cheong products to expect its health functional effect.

A model for enhancing the academic excellence of adult college students (성인대학생의 학업수월성 강화를 위한 모형)

  • The purpose of this study is to present a model for enhancing the academic excellence of adult college students. For this purpose, 408 adult college students attending 2-year and 4-year colleges in Busan, Daegu, and Gyeongbuk were surveyed and analyzed. The components of the model are curriculum, educational methods, evaluation of education, educational administration, educational environment, and institutional support and the results are as follows. First, the curriculum preferred by adult college students was to acquire diverse academic knowledge for a degree, to acquire knowledge and skills to develop skills for the workplace, and to acquire new information and knowledge regarding issues in society as a whole. Second, the professors' qualification among the educational methods preferred by adult college students was professional competence of the professors based on their theoretical and practical skills. The preferred teaching methods were lecture, discussion, action learning, and the project learning method in that order and video and PowerPoint were preferred as effective teaching mediums. Third, the preferred course for adult college students is operated on weekends, and three years was preferred to get a bachelor's degree. The possible hours of learning per day is 3~6 hours, indicating the necessity of e-learning, B-learning, and prior learning experience recognition systems. Fourth, the education evaluation method preferred by adult college students was a compromise method which is a mixture of absolute evaluation and relative evaluation, and it also showed the need for Pass or Non Pass evaluation method. Fifth, the internal factors of college selection preferred by adult college students were the acquisition of new knowledge and skills, and the external factors were desire to receive many opportunities related to employment and job improvement. The classroom, which provides an effective environment, was a fixed seat classroom and an indoor classroom environment was emphasized for desired educational environment. Sixth, institutional support preferred by adult college students was computer-related programs and learning club support services.

An Experimental Study on the Hydration Heat of Concrete Using Phosphate based Inorganic Salt (인산계 무기염을 이용한 콘크리트의 수화 발열 특성에 관한 실험적 연구)

  • Whereas the control of the hydration heat in mass concrete has been important as the concrete structures enlarge, many conventional strategies show some limitations in their effectiveness and practicality. Therefore, In this study, as a solution of controling the heat of hydration of mass concrete, a method to reduce the heat of hydration by controlling the hardening of cement was examined. The reduction of the hydration heat by the developed Phosphate Inorganic Salt was basically verified in the insulated boxes filled with binder paste or concrete mixture. That is, the effects of the Phosphate Inorganic Salt on the hydration heat, flow or slump, and compressive strength were analyzed in binary and ternary blended cement which is generally used for low heat. As a result, the internal maximum temperature rise induced by the hydration heat was decreased by 9.5~10.6% and 10.1~11.7% for binder paste and concrete mixed with the Phosphate Inorganic Salt, respectively. Besides, the delay of the time corresponding to the peak temperature was apparently observed, which is beneficial to the emission of the internal hydration heat in real structures. The Phosphate Inorganic Salt that was developed and verified by a series of the aforementioned experiments showed better performance than the existing ones in terms of the control of the hydration heat and other performance. It can be used for the purpose of hydration heat of mass concrete in the future.

Changes in Characteristics of Semi-cured Pig Manure Liquid Fertilizer according to the Storage Duration and Aeration (반부숙상태 돈분뇨 액비의 저장기간 및 폭기여부에 따른 특성 변화)

  • Currently, most of the pig manure generated from pig farms in Korea is in the form of a slurry with a moisture content of about 97%. Pig manure slurry is a mixture of pig manure and cleaning water in the pig house. In this study, changes in properties of pig manure liquid fertilizer according to whether air was supplied or not and with the passage of storage period were analyzed for 120 days. During the experimental period, the degree of maturity of the pig manure liquid fertilizer was higher in the experimental closed batch reactors supplied with air than in the same type reactors not supplied with air. As the liquid fertilizer storage period elapsed, there was a tendency that liquid fertilizer was converted to a state of complete maturity. In the batch reactor in which air was supplied, the moisture content of pig manure slurry, which had a moisture content of 97.90%, was reduced to 96.82% at the end of the experiment. On the other hand, the moisture content in the reactor without air was reduced to 97.33%. The pH of the liquid fertilizer, which was 8.82 at the start of the experiment, changed to 7.57 in the reactor with air supplied and 8.75 in the reactor without air at the completion of the experiment. The nitrogen content in the liquid fertilizer was 0.198 mg/L on average at the start of the experiment and it was lowered to 0.076 mg/L in the air supplied reactor at the end of the experiment. On the other hand, the nitrogen content of the liquid fertilizer was lowered to 0.121 mg/L in the reactor to which air was not supplied. The phosphoric acid (P2O5) concentration in the liquid decreased higher in the liquid fertilizer filled in the reactor without air than the liquid fertilizer filled in the reactor with air supplied as the storage period elapsed. Considering the experimental results, it is considered that the quality of pig manure liquid fertilizer is improved when air is supplied to pig manure slurry and the storage period of pig manure slurry is longer.

Distribution Types of the Relict Conifer Community and the Approach for the Ecological Management in Ulleung-Island (울릉도에 자생하는 침엽수류 유존군락의 분포유형과 생태적 관리방안에 대한 연구)

  • Distribution types of native conifers (Juniperus chinensis, Pinus parviflora, Tusga sieboldii and Taxus cuspidata var. latifolia) were studied by phytosociological investigation and ZM method in Ulleung Island, South Korea. Two main types were divided maritime vegetation (Juniperus chinensis forest) and mountain vegetation (Taxus cuspidata var. latifolia forest and Pinus parviflora-Tusga sieboldii forest). The former was divided into sea cliff distribution (J-SC) and sea ridge distribution (J-SR) type. The latter was classified 7 distribution types; Taxus cuspidata var. latifolia forest was rock distribution (Ta-R) and mountain slope distribution (Ta-MS) type, and Pinus parviflora-Tusga sieboldii forest was rock distribution (P T-R), upper and ridge distribution (P T-UR, 3 units sub-types:1sub, 2sub, 3sub), and Mountain slope distribution (P T-MS) type. It was considered that J-SC, Ta- R, and P T-R were maintained by topographic climax, but J-SR, Ta-MS, P T-UR and P T-MS were the process of vegetation succession. Distribution types of topographic climax are entrusted to process of vegetation succession. Types in the process of vegetation succession will be needed tending of forest to promote saplings growth and seedlings germination. Especially in order to restore Tsuga sieboldii forest should be afforest and make forest gap because It is mid shade tolerant tree and purity percentage of its seed is 1~2%. It was considered that the composition of group mixture forest constituted Pinus parviflora, Tsuga sieboldii, Taxus cuspidata, Camellia japonica, Machilus thunbergii and Acer okamotoanum, etc. will be able to restore native vegetation, after take the form of forest gap by strong thinning and pruning of Pinus thunbergii forest.

  • Ground investigation and result analysis generally used to examine all sorts of structures' subsidence or stability can be classified into sandy soil and cohesive soil, and analysis on the liquid limit of soil is utilized to evaluate the physical properties of ground and types or technical behavior of soil. The most widely used method to analyze liquid limit is Casagrande with which liquid limit can be calculated relatively easily; however, it is fairly difficult to apply it to soil equipped with intermediate properties. Therefore, concerning the properties of soil having the intermediate properties of sedimentary ground, this researcher mixed the clay from Yangsan, Gwangyang, and Busan with sandy soil to make intermediate soil and then carried out the test of consistency limit and also evaluated applicability by using the suggested formula of consistency revision. The sample of intermediate soil was the mixture of clay and sandy soil, and to produce intermediate soil, the content (Fc) of fine soil was applied as 50%, 75%, or 100%. Regarding the physical properties of intermediate soil, to maintain the properties of clay in the natural state, bentonite was added at a fixed rate for controlling the properties of clay, and then, consistency was analyzed. By adopting the formula of consistency revision suggested in advanced research, this author analyzed consistency based on the experiment and consistency based on the suggested formula of revision. Also, about intermediate soil collected at the site, consistency based on the experiment and consistency based on the suggested formula of revision were analyzed comparatively, and about intermediate soil collected, this researcher analyzed particle size and calculated the content (Fc) of fine soil to analyze intermediate soil in diverse conditions. Moreover, about intermediate soil collected at the site, the suggested formula of consistency revision was applied to calculate the compression index, and the compression index based on the experiment and the compression index based on the suggested formula were analyzed comparatively to evaluate the applicability of the suggested formula.

Effects of Exhaust Gas Recirculation on Power and Thermal Efficiency of Reactivity Controlled Compression Ignition in Different Load Conditions with a 6-L Engine (6 L급 압축착화 기관에서 천연가스-디젤 반응성 조정 연소 시 부하에 따른 배기 재순환율이 출력 및 열효율에 미치는 영향 분석)

  • Reactivity controlled compression ignition (RCCI) combustion is one of dual-fuel combustion systems which can be constructed by early diesel injection during the compression stroke to improve premixing between diesel and air. As a result, RCCI combustion promises low nitrogen oxides (NOx) and smoke emissions comparing to those of general dual-fuel combustion. For this combustion system, to meet the intensified emission regulations without emission after-treatment systems, exhaust gas recirculation (EGR) is necessary to reduce combustion temperature with lean premixed mixture condition. However, since EGR is supplied from the front of turbocharger system, intake pressure and the amount of fresh air supplementation are decreased as increasing EGR rate. For this reason, the effect of various EGR rates on the brake power and thermal efficiency of natural gas/diesel RCCI combustion under two different operating conditions in a 6 L compression ignition engine. Varying EGR rate would influence on the combustion characteristic and boosting condition simultaneously. For the 1,200/29 kW and 1,800 rpm/(lower than) 90 kW conditions, NOx and smoke emissions were controlled lower than the emission regulation of 'Tier-4 final' and the maximum in-cylinder pressure was 160 bar for the indurance of engine system. The results showed that under 1,200 rpm/29 kW condition, there were no changes in brake power and thermal efficiency. On the other hand, under 1,800 rpm condition, brake power and thermal efficieny were decreased from 90 to 65 kW and from 37 to 33 % respectively, because of deceasing intake pressure (from 2.3 to 1.8 bar). Therefore, it is better to supply EGR from the rear of compressor, i.e. low pressure EGR (LP-EGR) system, comparing to high pressure EGR (HP-EGR) for the improvement of RCCI power and thermal efficiency.

Confirmation of Enzymatic Synthesis of 1, 2-Octanediol Galactoside using Mass Spectrometry and NMR Spectroscopy (Mass spectrometry와 NMR Spectroscopy를 이용한 1, 2-Octanediol Galactoside의 효소합성 확인)

  • 1, 2-Octanediol galactoside (OD-gal) has been synthesized from 1, 2-octanediol (OD), as a safer cosmetic preservative, using recombinant Escherichia coli β-galactosidase (β-gal). To confirm the molecular structure of synthesized OD-gal, mass spectrometry and NMR (1H- and 13C-) spectroscopy of OD-gal were carried out. In the reaction mixture, a sodium adduct ion of OD-gal (m/z=331.1732) was identified using mass spectrometry analysis. In addition, 1H NMR spectrum of OD-gal showed multiple peaks corresponding to the galactosyl group, which is evidence of galactosylation on OD. Downfield proton peaks at δH 4.39 ppm and multiple peaks from δH 3.98~3.55 ppm were indicative of galactosylation on OD. Up field proton peaks at δH 1.52~1.26 ppm and 0.89 ppm showed the presence of CH2 and CH3 protons of OD. 13C NMR spectrum revealed the presence of 24 carbons suggestive of α- and β-anomers of OD-gal. Among 14 carbon peaks from each anomer, the 4 peaks at δC 31.4, 29.0, 22.3, and 13.7 ppm were assigned to be overlapped showing only 24 peaks out of a total of 28 peaks. The mass value from mass spectrometry analysis of OD-gal, and 1H and 13C NMR spectral data were in good agreement with the expecting structure of OD-gal. Finally, we identified a galactose molecule from the hydrolysate of OD-gal using β-gal. We are expecting that through future study it will eventually be able to develop a safe cosmetic preservative.

The Acaricidal Effects of Slaked Lime and Plant Extracts on Poultry Red Mites (소석회와 식물 추출물의 닭진드기에 대한 구충 효과)

  • This study evaluated calcium hydroxide (slaked lime) and a combination of plant extracts ('natural product'; clove, cinnamon, and saponin; 1:1:1 ratio) as acaricidal control mechanisms for poultry red mites. Red mite susceptibility was evaluated after treatments with 10% slaked lime, 20% slaked lime, and 1% natural product. The duration of the acaricidal effect was also tested at 0, 10, 30, and 60 min after treatment using 20% slaked lime, 1% natural product, or a mixture of both. In the in vitro experiment, the slaked lime treatments were 73.2% (10% slaked lime) and 85.1% (20% slaked lime) effective on red mites. In acaricidal effect of control materials over times, with 20% slaked lime, the acaricidal effect decreased to 50.7% after 30 min, and 12.7% after 60 min (P<0.05). With 1% natural product, there was no acaricidal effect after 30 min (P<0.05). With 20% slaked lime +1% natural product, all of poultry red mites died until 30 min, and 92.9% after 60 min (P<0.05). On the farm, poultry red mites were observed that the number of poultry red mites increased 7,923 from 36 to 45 weeks, but then decreased to 483 after 20% slaked lime plus 1% natural product treatment. These results indicate that combining slaked lime and plant extracts effectively control poultry red mites.
