Genetic Differentiation in the Mitochondrial Cytochrome b Gene of Korean Brown Frog, Rana dybowskii (Amphibia: Ranidae)

  • The nucleotide sequences of a 504 bp segment of the mitochondrial cytochrome b gene were analyzed to survey the intraspecific variation of the brown frog, Rana dybowskii, collected from nine populations in South Korea. Comparisons of sequence divergence of the cytochrome b gene suggest that the populations examined are clearly classified into two types (type 1 and type 2), diverged from each other by a high value of 14.3-15.9% sequence divergence. The two types are distributed allopatrically in most populations, but only one population occurs sympatrically. In the Tonghae population, their spawning grounds differ in that type 1 spawns in the puddle and type 2 spawns in the mountain creek. Based on the genetic divergences of the cytochrome b gene sequences, the phylogenetic status of Korean R. dybowskii is elucidated by comparing it with related brown frogs distributed in an area adjacent to the Korean Peninsula. Interspecific sequence divergences among type 1, type 2 and other related brown frog species (Russian R. dybowskii, R. pirica, R. ornativentris, R. chensinensis: 2n=24 chromosomes) used in this study ranged from 11.7 to 16.3%. R. dybowskii in Tsushima is very similar to our type 1 (sequence divergence=0-1.6%) and R. chensinensis in western China is closest to our type 2 (sequence divergence=6.8-7.5%).

한국산 쉬리, Coreoleuciscus splendidus (잉어과)의 종내 집단간 분자 유전 변이 (A molecular Genetic Variation among Intra-poplations of Korean shiner, Coreoleuciscus splendidus Mori (Cyprinidae))

  • 한국산 쉬리, Coreuleuciscus splendidus의 종내 집단간 유전자 다양성을 알기 위해 6개 주요강(북한강, 남한강, 금강, 오십천, 낙동강, 섬진강)으로부터 채집된 개체를 대상으로 16S rRNA 유전자와 미트콘드리아 cytochrome b 유전자에 근거하여 비교 분석하였다. 미트콘드리아 cytochrome b 유전자의 657 bp 길이의 염기서열 분석결과, 6개 집단간에 차이는 98.2~99.9%로 나타났으며 지리적으로 격리된 집단간에 높은 유전적 다양성을 보였다. 16S rRNA 유전자는 697 bp의 염기서열을 얻었으며, 종내 변이는 큰 차이가 없이 거의 동일하였다. 16S rRNA 유전자의 6개 집단간에는 97.7%에서 99.7%의 높은 유사성을 보였다.

  • 본 연구는 쥐노래미과(Hexagrammidae)에 속하고 양식산업에서 중요한 위치를 차지하고 있는 노래미, Hexagrammos otakii(Jordan et Starks)와 쥐노래미, H. agrammus(Temminck et Schlegel)의 미토콘드리아 cytochrome b(cyt b)유전자의 염기서열을 서로 비교 분석하였다. 총 489 bp크기의 cyt b 염기서열에서 종간의 변이는 거의 없었다(96%의 유사성).Neighbor-joining방법에 의한 노래미와 쥐노래미에서의 0.0342인 pairwise distance는 두 종이 분자계통유전학적으로 유사함을 시사한다. 이러한 연구 결과들은 쥐노래미(Hexagrammos)속을 대상으로 한 양식, 수산유전학 및 분자계통학 분야에 적용 가능할 것이다.

Simple Assessment of Taxonomic Status and Genetic Diversity of Korean Long-Tailed Goral (Naemorhedus caudatus) Based on Partial Mitochondrial Cytochrome b Gene Using Non-Invasive Fecal Samples

  • South Korea presently harbors less than 800 long-tailed gorals (Naemorhedus caudatus), an endangered species. I report for the first time on the taxonomic status and genetic diversity of the Korean species using non-invasive fecal sampling based on mitochondrial cytochrome b gene sequence analyses. To determine the taxonomic status of this species, I reconstructed a consensus neighbor-joining tree and generated a minimum spanning network combining haplotype sequences obtained from feces with a new goral-specific primer set developed using known sequences of the Korean goral and related species (e.g., Russian goral, Chinese goral, Himalayan goral, Japanese serow, etc.). I also examined the genetic diversity of this species. The Korean goral showed only three different haplotypes. The phylogenetic tree and parsimony haplotype network revealed a single cluster of Korean and Russian gorals, separate from related species. Generally, the Korean goral has a relatively low genetic diversity compared with that of other ungulate species (e.g., moose and red deer). I preliminarily showcased the application of non-invasive fecal sampling to the study of genetic characteristics, including the taxonomic status and genetic diversity of gorals, based on mitochondrial DNA. More phylogenetic studies are necessary to ensure the conservation of goral populations throughout South Korea.

Purification and Comparison of NADH-Cytochrome $b_5$ Reductase from Mitochondrial Outer Membrane of Bovine Heart and Turnip

  • The NADH-cytochrome b5 reductase (NCBR), a mitochondrial external electron carrier, was purified from bovine heart and turnip and their properties were examined. The mitochondrial outer membranes separated were subjected to NCBR isolation through DEAE-Cellulose ion exchange, DEAE-Sephadex gel chromatography, and hydroxyapatite adsorption chromatography. These processes yielded the purification folds of 88 and 42 and the recovery percentages of 0.2%, 5.67% for turnip and bovine heart, respectively. The molecular weight of the NCBR from the two sources was estimated to be 35,000 using SDS polyacrylamide gel electrophoresis. The Michaelis constant Km and maximum velocity Vmax were determined by measuring the NADH-ferricyanide redox system as well as the NADPH-ferricyanide redox system. The kinetics showed that both NCBRs had higher affinities for NADH than artificial electron-acceptor substrate ferricyanide. Although NADPH had a lower affinity for the enzymes than NADH, this study showed the 2'-phosphate dinucleotide could be used as a substrate.

A Phylogenetic Study of Korean Rodents (Muridae, Sciuridae) Based on Mitochondrial and Nuclear DNA

  • The subfamily Murinae is a very controversial group concerning their phylogenetic relationship. Previous studies could not resolve phylogeny among four genera Apodemus, Micromys, Mus and Rattus of the Muridae. In the present study, eight rodent species resident in South Korea were collected and phylogenetically analyzed based on sequence data of five mitochondrial and nuclear DNA regions: 12S rRNA, cytochrome b gene (cyt b), cytochrome oxidase II (COII), control region of mitochondrial DNA, and a thyroglobulin (Tg) of nuclear DNA. According to the phylogeny of the concatenated data, M. musculus separated early in Murinae (ML 100%; BA 1.00 pp) and the genus Rattus grouped with the harvest mouse, M. minutes; these were separated from the genus Apodemus with relatively strong support (ML 74%; BA 0.76 pp). The Siberian chipmunk population was also examined using the five genes to obtain better resolution. The phylogeny for Korean rodents determined using the 12S rRNA, cyt b, COII and control regions discriminated the Siberian chipmunk populations from Korea, Russia, and China.

Molecular Discrimination of Cervidae Antlers and Rangifer Antlers

  • Cervi Parvum Cornu is widely used as a hemopoietic, tonifying, growth-promoting, cardiotonic, and immuno-modulating agent in Korea. In order to develop the quality control method of Cervi Parvum Cornu by the identification of the biological source or origin, the molecular approach was applied using PCR (polymerase chain reaction) and PCR-RFLF (PCR-restriction fragment length polymorphism) analysis. In the PCR analysis of the mitochondrial 12S rRNA gene and cytochrome b gene regions, no distinctive DNA bands from Cervidae (deer) antlers and Rangifer (reindeer) antlers were observed. However, when the amplified products in the mitochondrial cytochrome b gene region were subjected to restriction digestion with TaqI, Cervidae antlers showed an undigested state of 380 by band, differently from two bands of 230 by and 1S0 by from Rangifer antlers. Based on this finding, the base sequences of amplified PCR products in the range of mitochondria) cytochrome b gene from Cervidae antlers and Rangifer antlers were determined and subjected to restriction analysis by various endonucleases. The results showed that antlers from Rangifer species could be simply discriminated with other antlers from 8 Cervidae species (Chinese deer, Russian deer, Hong Kong deer, New Zealand deer, Kazakhstan deer, elk, red deer and Sika deer) by PCR-RFLP analysis using AtuI, HaeIII, HpaII or Sau3AI(MboI) as well as TaqI in the range of the mitochondrial cytochrome b gene.

Genetic Variation of the Mitochondrial Cytochrome b Sequence in Korean Rana rugosa (Amphibia; Ranidae)

  • Nucleotide sequences of a 501 base-pair (bp) fragment in the mitochondrial cytochrome b (cyt b) gene were analyzed for 12 populations of Rana rugosa from Korea and Japan using a polymerase chain reaction (PCR) and direct silver sequencing. Two genetically distinct groups (type-A and type-B) were found in Korea. Type-A was found throughout most of South Korea and type-B was restricted to the mid-southeastern regions (Samchok, Yongdok, Chongsong and Pohang). But in the Tonghae population, both types were found. The level of mitochondrial DNA (mtDNA) sequence differences ranged from 0% to .0.8% among six populations of type-A, and 0 to 1.0% among 4 populations of type-B. However, sequence differences between type-A and type-B ranged from 5.4% to 6.6%, Using Kimura's two-parameter distance, the level of genetic sequence divergence between type-A and type-B was 6.7%. The Japanese R. rugosa was clustered very far from the Korean R. rugosa with 14.7%. In the neighbor-joining and UPGMA tree, all Korean samples were grouped, but subdivided into two types in 99% of the bootstrap iteration.

한국산 짱뚱어(Boleophthalmus pectinirostris)와 남방짱뚱어(Scartelaos gigas) (Gobiidae)의 분자유전학적 계통연관과 DNA 다형화 (Phylogenetic Relationship and DNA Polymorphism of Boleophthalmus pectinirostris and Scartelaos gigas (Teleostei: Gobiidae) of Korea)

  • 우리나라 망둑어과 어류 중 말뚝망둥어아과에 속하며 형태학적으로 흡사한 짱뚱어 (B. pectinirostris)와 남방짱뚱어(S. gigas)를 대상으로 12S rRNA, 16S rRNA 및 mitochondria cytochrome b 유전자의 염기서열 분석을 통해, 종내 및 종간의 분자유전학적 특성을 파악하고자 하였다. 짱뚱어 종내 개체간 유전적 차이를 밝히기 위해 mitochondria내에 3가지 유전자를 분석한 결과, 순천지역과 군산지역간의 염기서열은 12S rRNA는 총 434 bp (base pair)의 염기서열을 얻었으며 이를 비교 분석한 결과, 순천지역과 군산지역의 집단 간에는 100% 동일한 염기서열을 보였고, 16S rRNA 유전자는 총 484 bp의 염기서열 중 99.6%의 차이로 단지 2 bp가 치환된 것으로 나타났고, mitochondria cytochrome b 유전자의 경우, 444 bp의 염기서열을 얻었으며 두 지역 간 염기서열은 100%의 일치도를 보였다. 한국산 짱뚱어를 대상으로 한 동일한 종내에서 서식지 (서해, 남해)에 따른 유전적 차이나 지리적 거리의 차이가 있을 것으로 예상하였으나 이상의 3가지 유전자를 비교해 본 결과를 종합해 보면, 종내 개체 변이와 지역별 변이는 일어나지 않은 것으로 밝혀졌다. 또한 짱뚱어와 남방짱뚱어 2종간 비교 (inter-species)에서는 순천지역 짱뚱어와 해남지역 남방짱뚱어를 12S rRNA 유전자와 16S rRNA 유전자 및 mitochondria cytochrome b 유전자 염기서열을 비교 분석한 결과, 각각 96.1% (17 bp), 94.0 (29 bp), 그리고 82.9% (76 bp)로 나타나 두 종의 유전학적 차이는 크게 나타났다. 따라서 본 연구에서 나타난 짱뚱어와 남방짱뚱어 두 종간에 3.9~17.1%를 보여 서로 다른 종으로 볼 수 있을 것으로 사료되었다.
