Antioxidant, anticoagulant, and α-glucosidase inhibitory effects of mistletoe (Viscum album var. coloratum) extract (겨우살이(Viscum album var. coloratum) 추출물의 항산화, 항응고 및 α-glucosidase 저해 효과)

  • Antioxidant, α-glucosidase inhibition, and anticoagulant effects of 80% ethanolic extract from mistletoe (Viscum album var. coloratum) were investigated. The yield and polyphenol content of the mistletoe extract were 30.9±0.4% and 57.6±1.5 mg gallic acid equivalents/g, respectively. The antioxidant effects of the mistletoe extract such as free and cationic radical scavenging ability, nitrite scavenging ability, and reducing power increased in proportion to its concentration. Also the mistletoe extract inhibited the activity of α-glucosidase, and delayed plasma coagulation mainly by inhibiting the extrinsic and common pathways in blood coagulation system.

The Effects of Mistletoe Extract and Anti-cancer Drugs on the Apoptosis of Gastric Cancer Cells (위암세포 사멸에 미치는 겨우살이 추출물과 항암제의 효과)

  • Purpose: Mistletoe extract was widely used for cancer treatment as complementary or alternative therapy in European area from early twenty century. It is currently used as alternative anti-cancer remedy by piecemeal in domestic medical group, however, the anti-cancer mechanism of mistletoe extract was not known precisely until now. In this study the effect of mistletoe extract on gastric cancer was studied vis cell line experiments. Materials and Methods: The SNU719 gastric cancer cell line was used, and ABNOBAviscum-Q and ABNOBAviscum-F were treated to cells as mistletoe extract, or 5-FU and cisplatin were used with mistletoe extract. The cell viability and cell death rate were estimated by CCK-8 assay kit and lactate dehydrogenase (LDH) assay kit in each. Caspase 3 assay kit was used to measure caspase 3 activity. The protein expression amounts of Bcl2, p53, and PTEN were estimated through Western blot analysis. Results: The co-treatments of mistletoe extract Q/F and 5-FU/cisplatin decreased lesser cell viability than only mistletoe treat. Caspase 3 activity was increased 4~6 times in co-treatment of mistletoe extracts and 5-FU than control. Bcl2 protein expression was reduced by mistletoe extracts or anti-cancer drugs, further more, the co-treatment of mistletoe extracts and 5-FU/cisplatin diminished more the expression than only mistletoe treatment. Mistletoe extracts did not affect the protein expressions of p53 and PTEN. Conclusion: It was concluded that the anti-cancer mechanism of mistletoe extracts was made by caspase 3 activation and lowered Bcl2 expression, and this apoptosis inducing mechanism was independent to p53.

Effects of Dietary Supplementation of Red Ginseng Mare and Korean Mistletoe Powder on Performance and Meat Quality of Broiler Chicken (사료 내 홍삼박과 겨우살이 분말 첨가 급여가 육계의 생산성 및 계육의 품질에 미치는 영향)

  • The objective of this study was to evaluate the effects of different levels of red ginseng mare or Korean mistletoe powder on growth performance and physicochemical properties of thigh meat in broiler. A total of 200 broiler chicks were divided into five groups were fed a basal diet (control), basal diet supplemented with 0.5% red ginseng mare (T1), red ginseng mare 1% added (T2), Korean mistletoe 0.5% added (T3), Korean mistletoe 0.5% added (T4) for five weeks. Growth performance and proximate composition of chicken thigh meat were not influenced by all treatments, except for mortality and crude fat, which was significantly lower in all treatments with red ginseng mare or Korean mistletoe powder compared to control. Dietary red ginseng mare and Korean mistletoe supplementation had no significantly effects on pH, WHC, shear force, CIE $L^*$, $b^*$ values, triglyceride and glucose but TBARS values total cholesterol, and LDL-cholesterol were decreased (P<0.05) by the addition red ginseng mare and Korean mistletoe powder. CIE $a^*$ values increased significantly with added red ginseng mare and Korean mistletoe powder relative to the control (P<0.05). It was concluded that the addition of red ginseng mare and Korean mistletoe powder revealed minor improvements in the lipid oxidative stability of chicken meat.

  • The objective of the present study was to evaluate in vitro antioxidant activity, antioxidant content and pancreatic lipase inhibitory activities of Makgeolli supplemented with 0, 1, 2, and 4% (w/v) Jeju camellia mistletoe during fermentation. Total phenolic and flavonoid contents tended to increase as content of Jeju camellia mistletoe increased. Supplementation with Jeju camellia mistletoe resulted in a significant increase in the scavenging capacities of 1,1-diphenyl-2-picrylhydrazyl, hydrogen peroxide, nitric oxide and superoxide anion radicals, and reducing power activity. Moreover, pancreatic lipase inhibitory activity was significantly elevated by Jeju camellia mistletoe addition. These results suggest that Jeju camellia mistletoe is considered to be a good material to improve antioxidant and pancreatic lipase inhibitory activities of makgeolli.

  • A simple and rapid homogeneous enzyme-linked binding assay for mistletoe lectin I(ML I) was developed using a coupled enzyme system of malate dehydrogenase (MDH) and D-galactose. A highly substituted MDH-galactose conjugate was prepared by employing an isothiocyanate method for formation of thiourea bond. In the presence of ML I, ML I inhibits the activity of the conjugate based on the ML I/D-galactose specific interaction. Thus, the concentration of ML I can be related to the homogeneous inhibition of the MDH-galactose conjugate. Using this method. ML I can be measured at the level of microgram per milliliter within 10 minutes.

  • The antioxidant activity of various solvent extracts from Jeju camellia mistletoe (Korthalsella japonica (Thunb.) Engl.) was investigated using various in vitro assays as the 1,1-diphenyl-2-picrylhydrazyl radical scavenging, ferrous ion chelating and reducing power assays. Methanol and ethanol extracts showed the most potent antioxidant activity in all assays tested followed by water extract. The inhibitory effect of the Jeju camellia mistletoe extracts on pancreatic lipase and $\acute{a}$-glucosidase was also evaluated and the results showed that methanol and ethanol extracts markedly reduced both enzyme activities. Therefore, the methanol and ethanol extracts of Jeju camellia mistletoe is definitely worthy of further investigation for these beneficial effects on nutraceutical medicine.

  • Viscum album var. coloratum (mistletoe) is considered as one of the endangered plant species in Korea. Our objective is to restore its population and multiplication of plant by ex situ method. In this research we explored the maximum germination (in vitro) of freshly harvested and stored seeds of mistletoe collected in different time intervals. Ex vitro germination after artificial inoculation on the branches of Prunus mume in different physiological conditions was also monitored. The research revealed that the lately harvested seeds (Feb. and March 2014) were superior over early harvested seeds (Nov. 2013 and Jan. 2014) of mistletoe due to the higher percentage of germination (above 93%). According to the data, it is also revealed that the survival and germination rate of mistletoe seeds decreased with the increase in storage duration. In ex vitro germination, the fluctuated temperature of a glass house in natural condition enhanced (four fold) the rate of germination on the branches of Prunus mume than the constant temperature condition in the glass house.

  • The mistletoe lectins are major active components in the extrct of Viscum album var. coloratum. This study was performed to investigate the anti- skin cancer effect of Korean mistletoe extract intensified with mistletoe lectin (KI 2103S). B16F10 melanoma cells were allografted in C57BL/6 mice and F344 rats. The effect of KI 2103S on melanoma was measured by monitoring tumor index. The KI 2103S was injected intra-tumorally and tumor index was decreased in dose dependent manner.

  • The mistletoe lectins are major active components in the extract of European mistletoe (Viscum album L) that have been widely used in adjuvant chemotherapy of cancer. This study was performed to investigate the antitumor activity of extract of Korean mistletoe (Viscum album var. coloratum) modified with Korean mistletoe lectin (Viscum album var. coloratum agglutinin, VCA). Compared with the results of VCA, survival rate was increased and experimental lung metastasis was reduced by treatment of modified extract (VCM). In addition, the treatment of VCM reduced angiogenesis and VCA-induced toxicity measured by a CAM assay. And VCM inhibited proliferation and induced apoptosis in vitro in tumor cells originated from tissues which are possible to apply topically without surgery. Taken together, the antitumor activities of VCM-treated group outperformed the activities of the VCA-treated group.

  • Velutin and betulinic acid were isolated as a cytotoxic principle from the dichloromethane extract of Korean mistletoe (Viscum album var. coloratum) by repeated silicagel chromatography and recrystallization. In in vitro analysis of cytotoxic activity using NIH-3T3 cells, dichloromethane extract of Korean mistletoe was shown to be highly cytotoxic against tumor cells. And we obtained active compound (reffered to VD-2 and VD-6) from the dichloromethane extract. (omitted)
