Oral streptococci are major constituents of dental plaques, and their prevalence is closely linked with various pathologic symptoms, such as dental caries. To develop natural anticaries agent, we prepared 309 kinds of plant extracts from 215 species of edible or medical plants, and antibacterial activity of the extracts against Streptococcus mutans JC-2 were evaluated based on 96 well microtiter plate assay and disk paper method, subsequently. Among the tested plant extracts, Ailanthus altissima, Paeonia lactiflora, Rubus phoenicolasius, Aralia continentalis, Quercus acutissima, Persicaria hydropiper and Agrimonia pilosa extracts showed strong antimicrobial activity. Determination of minimal inhibitory concentration (MIC) of the selected seven plant extracts showed that Ailanthus altissima, Persicaria hydropiper and Quercus acutissima extracts ($MIC=25\∼30[\mu}g/ml$) has potential as a source of natural anticaries agents.
Vanadium is an essential trace element but has not been identified with a specific biogical role. To study the direct effects of vanadium on osteoblast, we incubated murin osteoblast-like (MC3T3-El) cells with various corcentration of vanadium oxide & sodium orthovanadate. This study was designed to investigate the effect of vanadium on DNA synthesis, alkaline phosphatase (ALP) activity, cAMP formation responsive to parathormone(PTH) and type I $\alpha$ 2 collagen ribonucleic acid (mRNA) level in murin osteoblast-like (MC3T3-El) cells. The cells were cultured in $\alpha-minimal$ essential medium$(\alpha-MEM)$ supplemented with $10\%$ fetal bovine serum (FBS) and then changed to $0.1\%$ FBS with various concenoation of vanadium oxide & sodium orthovanadate. Quiescent cultured MC3T3-El cells incubated for 24 hours with 2,5,10,15,20 ${\mu}M$ vanadium oxide incorporated $[^3H]Thymidine;$ every concentration showed increases in $[^3H]Thymidine$ incorporations dose dependant manner, the greatest response occurred at $20{\mu}M$. Quiescent cultured MC3T3-E1 cells incubated for 3days with 2,5,10,15,20 ${\mu}M$ vanadium oxide, for 2days with sodium orthovanadate and alkaline phosphatase was assayed with disodium phenyl phosphate as substrate. Vanadium oxide increased the alkaline phosphatase content in MC3T3-El cells at $2{\mu}M\;&\;6{\mu}M$ ; the greatest response occurred at $2{\mu}M$. But decreased at other content sodium orthovanadate increased alkaline phosphatase content in MC3T3-El cells at all concenoation ; the greatest response occurred at $4{\mu}M$. Quiescent cultured MC3T3-El cells incubated for 3days with $5,10{\mu}M$ vanadium oxide , with $5,8{\mu}M$ sodium orthovanadate and cAMP formation was measured by Radioimmunoassay(RIA). Vanadium oxide & sodium orthovanadate showed the tendency of inhibitory effects on cAMP responsiveness to PTH in MC3T3-El cells. Quiescent cultured MC3T3-El cells incubated for 24hours with $10,20{\mu}M$ vanadium oxide, with $5,10{\mu}M$ sodium orthovanadate and Type I $\alpha$ 2 collagen ribonucleic acid (mRNA) expression was studied by Nothern blot analysis. Northern blot analysis of vanadium oxide treated cells showed decreasing effects 0& sodium orthovanadate revealed increasing effects in type I $\alpha$ 2 collagen ribonucleic acid (mRNA) level.
Ozonized olive oil was tested for its mutagenic potential in a Salmonella/microsome assay. Additionally, antimicrobial activity was tested against Propionibacterium acnes, Staphylococcus epidermidis, Staphylococcus aureus and Pseudomonas aeruginosa, pathogenic strains related to acne, using the paper disk and agar dilution method. Ozonized olive oil showed antimicrobial activities against all the strains tested, with minimal inhibitory concentrations (MICs) values in a range of 2${\sim}$10 mg/mL. Mutagenicity of ozonized olive oil was evaluated with Salmonells typhimurium TA98, TA100 and Ta1535, with and without addition of S9 mixture. No increase in the number of $his^{+}$ revertants over the negative control (solvent and non-ozonized olive oil) values was observed with TA98 (1,000 ${\mu}g/plate$), TA100 (1,500 ${\mu}g/plate$) and TA1535 (1,500 ${\mu}g/plate$) strains. The results from this study suggested that ozonized olive oil does not show any mutagenic potential.

This study was carried out to evaluate cytotoxic effects of Atractylodes macrocephala Koidz. (A. macrocephala Koidz.) extract on NIH 3T3 fibloblast. SK-MEL-3 (HBT 69) and KB (ATCC No, OCL 17) cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-y1)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. 10.2 mg/ml Concentration of A. macrocephala Koidz. extracts in SK-MEL-3 showed that their susceptibility (sensitivity) to these compounds decreased in the following order ; adriamycin > H₂O > ethyl acetate > ethyl alcohol > chloroform > n-hexane in SK-MEL-3 cell lines ; 5-FU > H₂O > n-hexane > ethyl acetate > ethyl alcohol > chloroform in KB cell lines. In order to develop an antimicrobial agent, A. macrocephala Koidz. was extracted with solvents. The minimal inhibitory concentrations (MICs) of each solvent extract of A. macrocephala Koidz. against microogranisms were also examined. Antimicrobial activities of ampicillin and ketoconazole as references were compared to those of each solvent extract of A. macrocephala Koidz. The antimicrobial activity of the ethyl acetate soluble extract of A. macrocephala Koidz. had growth inhibition activity against S. mutans and P. putida (MICs. 500 ㎍/ml). These results suggest that the ethyl acetate soluble extract of A. macrocephala Koidz. possessed antitumorous and antimicrobial agents
Fusarium head bight (FHB) is a devastating disease on major cereal crops worldwide which causes primarily by Fusarium graminearum. Synthetic fungicides are generally used in conventional agriculture to control FHB. Their prolonged usage has led to environmental issues and human health problems. This has prompted interest in developing environmentally friendly biofungicides, including botanical fungicides. In this study, a total 100 plant extracts were tested for antifungal activity against F. graminearum. The crude extract of Pterocarpus santalinus heartwood showed the strongest antifungal activity and contained two antifungal metabolites which were identified as ${\alpha}$-cedrol and widdrol by GC-MS analysis. ${\alpha}$-Cedrol and widdrol isolated from P. santalinus heartwood extract had 31.25 mg/l and 125 mg/l of minimal inhibitory concentration against the spore germination of F. graminearum, and also showed broad spectrum antifungal activities against various plant pathogens. In addition, the wettable powder type formulation of heartwood extract of P. santalinus decreased FHB incidence in dose-dependent manner and suppressed the development of FHB with control values of 87.2% at 250-fold dilution, similar to that of chemical fungicide (92.6% at 2,000-fold dilution). This study suggests that the heartwood extract of P. santalinus could be used as an effective biofungicide for the control of FHB.

This study was carried out to treatment test for bovine mastitis by the determination of adenosine triphosphate (ATP) based on firefly bioluminescence. The results obtained are followed; 1. In the susceptibility test, cephalothin which looks the most effective were sensitive to Staphylococcus sp. (72.3%), Micrococcus sp. (84.2%), Streptococcus sp. (72.7%) and Gram positive bacilli (72.7%), Gram negative bacilli were sensitive to gentamicin (92.3%) and Yeast-like-fungi was the most sensitive to clotrimazole, and nystatin in order. 2. When the number of bacteria, such as Staphylococcus aureus, Candida tropicalis isolated from the mastitis milk were counted by conventional agar plating technique, and compared with the concentration of bacterial ATP, it gave a good linear relationship. The content of ATP per Staphylococcus aureus, cell was 3.1fM and Candida tropicalis showed the high level of A TP (90fM). 3. The ATP assay was applied to the determination of minimal inhibitory concentration (MIC) of various antibiotics. When Staphylococcus aureus was incubated in the presence of different concentration of tetracycline, erythromycin, kanamycin and streptomycin sulfate and the growth was monitored by the conventional agar plating technique and ATP assay, both methods shown the same results that they were 1mcg/ml, 2mcg/ml, 6.25mcg/ml and 8mcg/ml, respectively. 4. For the determination of susceptibility of sensitive and resistant Staphylococcus au reus isolated for the milk with mastitis to tetracycline, erythromycin kanamycin and streptomycin sulfate, the minimum time required for the test was determined by the assay of ATP every 30 minutes during incubation of 3 hours at $37^{\circ}C$. ATP concentration time curve calculated on both resistant and sensitive strains incubated 3 hours as the optimum time for the determination of susceptibilities of various antibiotics exemed. The ATP concentration of each test broth (antibiotic containing), expressed as a percentage of its own control brith (antibioticfree) indicated values of 30% to be indicative of each antibiotic sensitivity. Single time point ATP assay carried out on the various sensitive and resistant of Staphylococcus aureus to antibiotics examined after 3 hours at $37^{\circ}C$ correlated exactly with disc diffusion and MIC. 5. In the cure of intramammary treatment of bovine mastitis in lactating quarters, the cure rate of Staphylococcal mastitis showed to cephalexin (80%), cloxacillin and gentamicin (70%), ampicillin and oxytetracycline (60%), and Streptococcal mastitis showed to cephalexin (85%), penicillin (80%), cloxacillin and oxytetracycline (75%), and ampicillin (70%), but intramammary antimycotic drug (clotrimazol) were only a little effect about fungal mastitis.

The 30% resistant frequencies of pathogenic bacteria were identified against generally utilizing disinfectants. Among the used disinfectants, foodsef, Taego, and Iodo 175 were dictated by lower sensitivities against pathogenic bacteria, as well as higher resistant frequencies when compared with other disinfectants. The resistant frequencies against antibiotics were also dictated by 30% through MIC (minimal inhibitory concentration) and paper disc methods. Especially, the used bacteria exhibited resistances against gentamycin, kanamycin, and streptomycin, which included in all aminoglycoside group. The MBT-01108 material, which extracted and purified from a powder obtained by processing of Opunita ficus-indica var. saboten Makino trunk, did not develop or grow resistant bacteria. Interestingly, the multi-drug resistant bacteria such as MRSA, resistant Pseudomonas aeruginosa, VRE, and E. coli 0157 did not resistant against MBT-01108 material. These results suggest that MBT-01108 material uses as an anti-microbial agent.
Objectives: The aim of this study was to investigate deodorizing effectsof medicinal herbs (Mume Fructus, Eriobotryae Folium, Acanthopanacis Cortex, Angelicae Dahuricae Radix) for development of a gargle solution. Methods: 1. The antimicrobial effects of medicinal herbs were evaluated with the minimal bactericidal concentration (MBC) and the change of the number of viable cells in the herb extracts(1%) for 48 hrs against P. gingivalis 2561 and Pr. intermedia ATCC 25611. 2. Deodorizing activity of each herb and Garglin $Mint^{(R)}$gainst methyl mercaptan were analyzed by gas chromatography (GC). 3. We used the malodor modeling of the salivary sediment system with a Halimeter. 4. In the preliminary clinical study, the baseline concentration of VSC in the oral cavity of each subject was measured by Halimeter. Subjects would gargle for 30 seconds with cysteine. After 4 minutes subjects would gargle for 30 seconds with Garglin and herb extracts (2%). Subsequently, concentration of VSC were measured at 0, 4, 8, 12 and 20 minutes. Results: 1. MBC of Mume Fructusfor P. gingivalis 2561 was determined to be <1% and MBCs of Eriobotryae Folium for P. gingivalis 2561 and Pr. intermedia ATCC 25611 were determined to be <2% and <1%, respectively. Mume Fructus (1%) completely suppressed the P. gingivalis cell viability from 5 hrs and Eriobotryae Folium (1%) completely suppressed the Pr. intermedia cell viability from 48 hrs. 2. In GC analysis, deodorizing activities were 91.54% with Mume Fructus, 87.97% with Eriobotryae Folium, 100% with Acanthopanacis Cortex, 72.36% with Angelicae Dahuricae Radix and 40.54% with Garglin $Mint^{(R)}$. 3. In malodor modeling of the salivary sediment system, each of the medicinal herbs had significantly inhibitory effect on malodor formation (p<0.05). 4. In the preliminary clinical study, the concentration of VSC of the herb groups was significantly lower than of the control group, but not in Garglin $Mint^{(R)}$. Conclusions: Mume Fructus, Eriobotryae Folium, Acanthopanacis Cortex and Angelicae Dahuricae Radixhave deodorizing activities and potential as an effective mouthwash against oral malodor.
The five short-chain fatty acids such as isobutyric(C-4), butyric(C-4), isovaleric(C-5), valeric(C-5) and caproic (C-6) acids obtained from the extract of common purslane showed wide antifungal spectra against spore germination and mycelial growth of the twenty five phytopathogenic fungi tested in vitro, although there were differences in antifungal potency among them. The antifungal potency of each fatty acid varied significantly against different fungi in spore germination and mycelial growth. The seventeen fungi used for spore germination test and the sixteen fungi used for mycelial growth test can be divided into three groups depending upon differences in minimal inhibitory concentration of each fatty acid for them, respectively. Caproic acid was significantly more toxic to germination than to mycelial growth of the test fungi, while the other four fatty acids did not show such a significant differences in toxicity with a few of exceptions as shown in valerie acid. The longer the chain-length of fatty acid was, the higher the antifungal potency was shown. The normal fatty acids such as butyric and valerie acid were more toxic than their isomers to spore germination and mycelial growth of the test fungi. Each fatty acid was more toxic to spore germination of obligate parasites and some of facultative parasites, and mycelial growth of facultative parasites than to spore germination and mycelial growth of saprophytes, respectively.
The fruits of sweet cherry are highly appreciated by the consumer due to their precocity and quality, such as their sweetness, color and sourness. In this study, the hot-water extract and its sequential organic solvent fractions were prepared from domestic Napoleon sweet cherry (Prunus avium L.) to investigate antimicrobial and antioxidant activity. The hot-water extract contained about 40% sugars, and the solvent fraction yields for hexane, ethylacetate (EA), butanol, and water residue were 0.01%, 3.45%, 16.30%, and 80.24%, respectively. Contents of total polyphenol and total flavonoid of the fractions were 1.24~5.24%, and 0~3.76%, respectively. Among the fractions, EA fraction showed the highest total polyphenol and total flavonoid concentrations. Evaluation of antimicrobial activity of the extract and fractions revealed that EA fraction and butanol fraction contained strong antibacterial activity against Listeria monocytogenes, Staphylococcus aureus, and Salmonella typhimurium with minimal inhibitory concentration (MIC) of 0.5~1.0 mg/mL. But the extract and fractions tested were not active to Pseudomonas aeruginosa. In a while, only hexane fraction showed anti-Candida activity with 0.5~1.0 mg/mL of MIC. The fraction showed strong activity against different multi-antibiotics resistant strains such as C. albicans CCARM 14020. Antioxidative activity assay showed that EA fraction has a strong DPPH scavenging activity and a reducing power. The $IC_{50}s$ of vitamin E and EA fraction were 15.5 and $195.5\;{\mu}g/mL$, respectively. Our results suggest that the fruit of P. avium L. has high potentials with anti-Candida and antioxidative activity.
