Cyst formation of the marine ciliate, Vorticella sp. using MCCF medium (MCCF 배지를 이용한 해산 섬모충 Vorticella sp.의 cyst형성)

  • The ciliate, Vorticella was often observed in the rotifer mass culture tanks as common co-existing organism. This Vorticella performed as a predator for aquatic bacteria population in the rotifer mass culture tanks. This study was carried out to investigate a cyst formation medium of Vorticella in the laboratory for keeping Vorticella seed. The test organism Vorticella sp. was isolated from culture water of rotifer mass culture tanks. The cyst of Vorticella was formed by dried-method for the formation and maintainance of cyst. MCCF (Marine Ciliate Cyst Formation) medium was used for cyst formation (incystment), preservation and return to moving cell (excystment) of the marine ciliate, Vorticella sp. The cyst shape and size were ellipical type and $30.51 \pm1.98\;\mu$m (Avg. $\pm$ SD) of minor axis and $28.89 \pm2.12\;\mu$m (Avg. $\pm$ SD) of minor axis (n=10), The Vorticella cyst was kept in the room temperature ($10\~35^{\circ}C$) and total dark condition (24D:0L) during 1 year. The preserved cyst was transferred to moving cell state (excystment) only by the addition of fresh sea water in the MCCF medium. The five Vorticella sp. moving cells of excysted from cysts showed the growth up to 912$\pm$64 cells/10 ml in MCCF medium during the culture period of 16 days. This MCCF medium was very useful tool for cyst formation and species preservation of marine ciliate Vorticella.

The Effect of Co-existing Artemia sp. on the Rotifer Brachionus rotundigformis Population Growth (Rotife 배양조에 혼재된 Artemia가 Rotifer 의 증식에 미치는 영향)

  • Artemia often observed as a co-existing organism in the mass culture tank of marine rotifer. The rotifer and Atremia are commonly used as food organisms in the marine fish larvae rearing. In this study, interspecific relation between the rotifer Brachionus rotundiformis (formely called S-type) and anostracan Artemia of the two developmental stages (0 and 19 day old after hatching) were investigated in the larboratory. The population growth of B. rotundiformis and one of the stage (nauplius or adult) of Artemia in mixed culture was compared with that of each single species culture. Culture period was 16 days. Every two days, the number of organisms in each species was counted and transferred to a fresh medium containing $7{\times}10^5$cells/ml of food Nannochloropsis oculata. Culture volume, temperature, salinity and photoperiod were set at 40ml, $25^{\circ}C.$, 22ppt and 24h all dark except to observation time, respectively. The rotifer population growth was greatly decreased by co-existence with Artemia. The coexisting Artemia suppressed the rotifer population growth due to it's high filtering speed for food (N. oculata). This study suggested that contamination by Artemia must be prevented for the stable rotifer production in the rotifer mass culture tank.

Parturition and Early Growth of Crowned Seahorse, Hippocampus coronatus in Korea (한국산 해마, Hippocampus coronatus의 출산과 초기성장)

  • The recent decrease in wild stocks of seahorse, Hippocampus coronatus, has prompted the production of the species in captivity. We here present data on the body color changes of the species according to culture conditions. This investigation examined relationship between adult body size and clutch and growth in 60 days after parturition and survival of juveniles with four fed enriched Artemia nauplii, copepod nauplii and copepod adult. Seahorse adults ($77.77{\pm}9.84mm$, n=12) collected from Kamak Bay and Yeoja Bay from August 2003 to November 2004 gave a parturition of $6\sim75$ juveniles (Mean, 39 juveniles/male/time) for 14 times. A day old seahorses were $14.48{\pm}1.38mm$ (n=15) in standard length (SL) with 14 dorsal fin rays, 12 pectoral fin rays and 4 anal fin rays. Sixty-day old seahorses were $24.65{\pm}0.83mm$ (n=4) in SL. Growth rate of seahorses was 0.18 mm/day at 24 in 60 days after parturition. When offered Artemia nauplii and copepod nauplii, 5 day old sea-horses preferred copepod nauplii to Aremia nauplii with a maximum predation rate of 31 copepod nauplii/sea-horse/h. However 30-day old seahorses preferred selected Artemia nauplii with a maximum predation rate of 14 Artemia nauplii/seahorse/h. Survival was highest (49%) when offered copepod nauplii together with enriched Artemia nauplii.

Variance characteristics of speaking fundamental frequency and vocal intensity depending on utterance conditions (발화조건에 따른 기본주파수 및 음성강도 변동의 특징)

  • The purpose of this study was to characterize and determine variances of speaking fundamental frequency and vocal intensity depending on gender and three utterance conditions (spontaneous speech, reading, and counting). A total of 65 undergraduate students (32 male students, 33 female students) attending universities in Daegu, South Korea participated in this study. The subjects were all in their 20s. This study used KayPENTAX's Visi-Pitch IV (Model 3950) to measure the variances of speaking fundamental frequency (SFF0) and vocal intensity (VI). As a result, this study came to the following conclusions. First, it was found that both males and females showed no significant difference in SFF0 and vocal intensity among three utterance conditions. Second, this study sought to analyze differences in the variances of SFF0 between males and females. As a result, it was found that females showed significantly higher levels of four measured variances (SFF0 $SD^{**}$, SFF0 $range^{***}$, Min $SFF0^{***}$ and Max $SFF0^{***}$) than males on spontaneous speech. However, it was found that there was no significant difference between males and females in SFF0 range on reading or in SFF0 SD and SFF0 range on counting. It was found that there was no significant difference between males and females in the level of measured variances of vocal intensity depending on utterance conditions. Finally, this study made a comparison and analysis on differences in the variances of SFF0 and vocal intensity among utterance conditions. As a result, it was found that all the measured variances of SFF0 in males were most significantly reduced depending upon spontaneous speech which was followed by reading and counting respectively (SFF0 SD: p<.001, SFF0 range: p<.05, Max SFF0: p<.05). Females however, show no significant difference in the measured variances of SFF0 depending upon three utterance conditions. It was also found that the measured variances of vocal intensity in females were most significantly reduced depending on spontaneous speech that was followed by reading and counting (VI SD: p<.001, VI range: p<.001, Min VI: p<.01 Max VI: p<.05), while males showed no significant difference in the measured variances of vocal intensity depending on three utterance conditions. In sum, these findings suggest that variances of SFF0 in males are affected by three utterance conditions, while variances of vocal intensity in females are affected by three utterance conditions.

Coexisting Fish Fauna in the Seahorse Habitats (해마 서식지 혼재 어류상)

  • Seahorse is an interesting organism for the study of its conservation as well as development as an marine ornament fish. To investigate the ecological characteristics and fish fauna around the habitat of such an endangered species, fishes were collected from the seahorse habitat. A total of 161 fishes were classified into 8 families and 11 species including two species of seahorse, Hippocampus mohnikei and H. coronatus representing 1.9% and 1.2%, respectively, of the population. The most dominant fish species in the area was Takifugu niphobles accounting for 30.4% of the population. The predators of the seahorse such as Acanthopagrus schlegeli, Lateolabrax japonicus and Lateolabrax maculatus were found in the relative abundance of 0.6%, 5.6%, and 11.8%, respectively. The relative abundance of H. coronatus ($0.96\;ind./1,000\;m^2$), H. mohnikei ($0.97\;ind./1,000\;m^2$) and the other coexisting fishes are similar in all the areas investigated (P>0.05).

Cultivation of Tigriopus japonicus by Products of Rotifer Culture Tanks (로티퍼 배양조 부산물을 이용한 Tigriopus japonicus (Copepod : Harpacticoida) 의 배양)

  • Tigriopus japonicus harpacticoida copepod was a common copepoda species as food organism for the marine fish larval rearing. However T. japonicus was difficult to stable culture except for a mixed culture with rotifer. Available food source for the successfully stable culture of T. japonicus was investigated in this study. T. japonicus did not utilization Nannochloropsis oculata instead of that. T japonicus utilized to products from rotifer culture tank. The products from rotifer culture tank was composed of rotifer feces and co-existing aquatic bacteria. The nauplius I stafe and copepodid I stage of T. japonicus showed grown whenproducts from rotifer culture tanks was fed. Specially we observed higher density of nauplii than that of copepodites and ovisac carrying females in the experimental culture populations.

Effects of Saline Concentrations on the Culture Density and Feeding of Estuarine Cladoceran, Diaphanosoma celebensis (기수산 물벼룩 Diaphanosoma celebensis의 배양 밀도와 섭이에 미치는 염분 농도의 영향)

  • We investigated the effects of salinity on an estuarine cladoceran, Diaphanosoma celebensis as a substitute live food organism of Artemia, which has been used for the next step of rotifer, in the course of seedling production of marine fishes, Culture density for growth, possibility of stable culture (RPGI: Relative Population Growth Index) and food feeding activity (RCN: Remaining Cell Numbers) of D. celebensis were investigated at intervals of 5 ppt under 10 steps of different saline conditions from 0 to 45 ppt. According to the results, the experimental group, which was regarded as the best condition of culture density for growth, stable culture possibility (RPGI) and good food feeding activity (RCN), was salinity culture condition of 20 put, and relative stable culture conditions were observed under saline concentrations $15\~35\;ppt$. Their functions under culture conditions of lower or higher salinities than 20 ppt appeared remarkably to be decreased, while all individuals were dead under the saline condition of 0 ppt after the beginning of the experiment. From these results of this study, it is assumed that estuarine cladoceran, D. celebensis can be cultured easily as a substitute live food organism of Artemia in the course of seedling production of marine fishes.

  • In this study, we investigated that the survival and growth responses of freshwater cladoceran Moina macrocopa and estuarine cladoceran Diaphanosoma celebensis on the saline culture conditions after transferring for using as live food organism. Estuarine cladoceran D. celebensis was survived and grew on the all salines except to saline jump culture condition of 0 ppt. However, freshwater cladoceran M. mcrocopa was died or decline on the over saline jumping culture conditions of 4 ppt within 5 minutes. These suggest the possibility of using the estuarine cladoceran D. celebensis compare with freshwater cladoceran M. macrocopa as a substitute live food organism for Artemia in the marine larval rearing.

  • This study was conducted to monitor residues of 10 veterinary drugs in food products. Various veterinary drugs were examined including enrofloxacin, ciprofloxacin, norfloxacin, oxolinic acid, amoxicillin, ampicillin, oxytetracycline, tetracycline, chlortetracycline, and doxycycline in beef, pork, egg, chicken, eel, flatfish, and rockfish obtained from 6 different regions (Seoul, Incheon, Daejeon, Gwangju, Daegu, Busan). Residues were detected in 21 (6.5%) samples out of 321 samples. In particular, 2 (1.0%) livestock samples had detected residues among 203 products, and 19 (16.1%) aquaculture samples had residues detected among 118 products. The most frequently detected drug residues in aquaculture products were oxytetracycline and amoxicillin, but the levels were mostly below the MRL (Maximum Residue Limit). In only one flatfish sample, amoxicillin was found at a level higher than the MRL (0.05 mg/kg). In livestock products, residues of most veterinary drugs were not detected. But enrofloxacin was detected in 2 chicken (Korean name: Ogolgae) samples at a higher level than the MRL (0.1 mg/kg as the sum of ciprofloxacin).

Selection of Culture Scale for Stable Culture of an Estunrine Cladoceran Diaphanosoma celebensis (기수산 물벼룩 Diaphanosoma celebensis의 안정 배양을 위한 배양 용기의 크기 선택)

  • Cladocera are important food organism for seed production of finfishes. freshwater cladocera such as Daphnia and Moina are well known food organisms for the larval rearing of freshwater fishes and are easy for mass culture. However, mass culture technique for marine cladocera are not yet developed, The only mass produced food organisms available these days for the larval production of marine finfishes are rotifer and Artemia. An estuarine cladoceran, Diaphanosoma celebensis, has a high possibility of being used as a food organism for the larval rearing of marine finfishes because this species is much easier to mass culture than marine ones. Therefore many studies are needed for this species. In this study, the effects of the volumes of culture container, 40, 1,500 and 15,000 ml, on the stable production of this species were tested and results are as follow: The maximum densities of this species in each of the culture volumes were reached after 14 days in 40 ml, 12 days in 1,500 ml, and 21 days in 15,000 ml with values of 3.4 $\pm$ 0.4, 14.2 $\pm$ 2.1 and 2.5 $\pm$ 1.6 per ml, respectively. The relative population growth index (RPGI) was stable in the culture volume of 1,500 ml. Moreover, possible harvesting number(individual/ml/day) was much higher in the 1,500 ml container than the other culture volumes. Therefore, optimum culture volume among the tested volumes for mass production of this species was 1,500 ml.
