Survey of Yogurt Powder Storage in Ambient Export Countries A Safety Evaluation Standard Compliance and Comparative Analysis

  • Yogurt powder is fermented milk processed in the form of dry yogurt, and has advantages such as stability, storability, convenience, and portability. China and Vietnam are important export target countries because of the increased demand for dairy products. Therefore, we surveyed dairy product standardization in order to establish an export strategy. Lactic acid bacteria counts are unregulated in Korea and Vietnam. In China, lactic acid bacteria counts are regulated at $1{\times}10^6$ colonyforming units (CFU)/mL and detected at $6.24{\pm}0.33\;Log\;CFU/mL$. All three countries have regulated standards for total bacterial counts. In China, total bacterial counts of milk powder are regulated to n=5, c=2, m=50,000, M=200,000 and detected at $6.02{\pm}0.12\;Log\;CFU/mL$, exceeding the acceptable level. Lactic acid bacterial counts appeared to exceed total bacterial counts. Coliform group counts, Staphylococcus aureus, Listeria monocytogenes, and Salmonella species were not detected. Acidity is not regulated in Korea and Vietnam. In China, acidity was regulated to over $70^{\circ}T$ and detected $352.38{\pm}10.24^{\circ}T$. pH is unregulated in all three countries. pH was compared to that of general fermented milk, which is 4.2, and that of the sample was $4.28{\pm}0.01$. Aflatoxin levels are not regulated in Korea and China. In Vietnam, aflatoxin level is regulated at 0.05 ppb. Therefore, all ingredients of the yogurt powder met the safety standards. This data obtained in this study can be used as the basic data in assessing the export quality of yogurt powder.

Survey of calcium, fat and lactose contents in processed milks (가공유의 칼슘, 지방, 유당 함량 및 표시기준에 대한 고찰)

  • In this study, we compared nutrient content emphasized on labelling of processed milks with that of whole milks and evaluated how well they conform to labelling standard. A total of 98 samples (33 whole milks, 28 calcium fortified milks, 30 low-fat milks and 7 lactose hydrolyzed milks) were collected in hypermarkets in Seoul from August 28 to August 30 in 2012. Calcium, fat and lactose contents were tested in the samples. Calcium contents ranged 102.2~113.0 mg/100 mL in whole milks and 120.1~337.8 mg/100 mL in calcium fortified milks. The level of calcium contents in fortified milks ranged very broad. Accordingly, the standard of calcium contents on fortified milks will be required. Fat contents ranged 3.1~3.9 g/100 mL in whole milks and 0.1~1.9 g/100 mL in low-fat milks. The average of fats content in low-fat milks was nearly one third than whole milks. Lactose contents was ranged 4.6~5.1 g/100 mL in whole milks and not detected in lactose hydrolyzed milks. All of processed milks were suitable to processing standard and labelling standard. But nutrition claims often used on processed milk such as "High" or "Low" were not adequate to indicate the exact nutrient content, which is a cause of the confusion for milk product labelling to consumers. We need a lot of research about nutrient labelling that can deliver appropriate and understandable information to customers.

Optimization of Lipase-Catalyzed Interesterification for Production of Human Milk Fat Substitutes by Response Surface Methodology (반응표면분석에 의한 모유대체지의 효소적 합성조건 최적화)

  • 1,3-Dioleoyl-2-palmitoylglycerol (OPO)-rich human milk fat substitute (HMFS) was synthesized from tripalmitin (PPP)-rich fraction and oleic ethyl ester by a lipase-catalyzed interesterification. Response surface methodology (RSM) was employed to optimize the presence of palmitic acid at sn-2 position ($Y_1$, %) and of oleic acid at sn-1,3 ($Y_2$, %), with the reaction factors as substrate molar ratio of PPP-rich fraction to oleic ethyl ester ($X_1$, 1:4, 1:5 and 1:6), reaction temperature ($X_2$, 50, 55 and $60^{\circ}C$), and time ($X_3$, 3, 7.5 and 12 h). The optimal conditions for HMFS synthesis were predicted at the reaction combination of $55^{\circ}C$, 3 h and 1:6 substrate ratio. HMFS re-synthesized under the same conditions displayed 70.70% palmitic acid at the sn-2 position and 69.58% oleic acid at the sn-1,3 position. Reaction product was predominantly (90.35%) triacylglycerol (TAG) was observed in which the major TAG species, OPO, comprised 31.24%.

Studies on the Manufacture of Peanut Yogurt (땅콩 요구르트 제조에 관한 연구)

  • To increase the nutritional and economic value of commercial peanut yogurt, a peanut yogurt was prepared by 12% skim milk added defatted peanut powder(DPP). The proximate compositions of DPP were moisture 2.3%, crude protein 31.39%, crude lipid 38.84, ash 2.89% and carbohydrate 24.58, respectively. The yogurt product were evaluated for acid production(pH, titratable acidity), number of viable cell, viscosity, color, quality-keeping property and sensory property. By addition of 5% and 10% DPP, the titratable acidity of yogurt was higher than that of yogurt not added DPP. The propagation of lactic acid bacteria was stimulated by adding 5% DPP, and the number of viable cells were about 8.9 log cfu/ml. On the other hand, the number of viable cells in control were 8.3 log cfu/ml. Viscosity of yogurt made from adding 5% and 10% DPP was higher than that of yogurt with only skim milk. When yogurt added with DPP was kept for 15 days at 5$^{\circ}C$ its quality-keeping was relatively good. As the DPP increased, L value(lightness) decreased and a value(redness) increased obviously but the b value(yellowness) of 5% peanut yogurt increased and 10% and 15% again decreased. The overall sensory scores of yogurts added with DPP showed lower than that of yogurt with only skim milk.

Vermicomposting of Sludge from Milk Processing Industry (MPS) (지렁이를 이용한 우유가공 폐수처리장 슬러지의 효율적 퇴비화)

  • This study was conducted to determine the optimal ratio of sludges from milk processing industry (MPS), paper-mill industry (PMS) and night-soil treatment plant (NSS) for vermicomposting. Five different ratios, 0 : 80 : 20 (MPS-0), 25 : 60 : 15 (MPS-25), 50 : 40 : 10 (MPS-50), 75 : 20 : 5 (MPS-75), and 100 : 0 : 0 (MPS-100 : control) MPS : PMS : NSS by wet weight were tested in a small plot experiment. The experiment for each mixing ratio was performed for 2 weeks with the three replications. MPS-100 (100 : 0 : 0) only had the highest decomposition rate with 19.9%, followed by MPS-25, MPS-50, MPS-75 and MPS-0 with 19.5, 19.1, 17.6 and 16.7%, respectively. Except for MPS-100, Vermicomposting resulted in increase in ash, T-P, $NO_2{^-}-N$, $NO_3{^-}-N$, Mg, K, As, Cd and Cu, whereas moisture, VS (Volatile Substance), TKN (Total Kjeldahl Nitrogen), $NH_4{^+}-N$, Ca, Hg and Pb were lower in the final cast than the initial feed mixture. Meanwhile Zn showed very slight difference and Cr and Ni did not show any tendency between the feed mixture and the final cast. In the case of MPS-100, where the decomposition rate was the highest, all the heavy metals in the final cast except for Hg were increased. All the vermicomposts produced from five different mixing ratios of the vermicomposting sludges met the Korea Standard as by-product compost.

The Effect on the Quality of Yogurt Added Water Extracted from Sea Tangle (다시마 추출물이 요구르트 품질에 미치는 영향)

  • For the purpose of making a functional and stable yogurt, new type yogurt were prepared from 12% whole milk and 2% skim milk added hot-water extracted from sea tangle. The yogurt product were evaluated for acid production(pH, titratable acidity), number of viable cell, viscosity, degree of curd sedimentation, quality-keeping property and sensory property. By addition of hot-water extracted from 0.5% sea tangle, the titratable acidity of yogurt(1.89%) was higher than that of yogurt not added hot-water extract of sea tangle(1.53%). The propagation of lactic acid bacteria was stimulated by adding hot-water extracted from 0.5% sea tangle, and the number of viable cells were about 1.4${\times}$10$\^$9/CFU/$m\ell$. On the other hand, the number of viable cells in control were 4.4${\times}$10$\^$8/CFU/$m\ell$. Viscosity of yogurt made from adding hot-water extracted from 0.5% sea tangle(25 CPS) was higher than that of yogurt with only milk(22 CPS). When yogurt made from adding hot-water extracted from sea tangle was kept at 7$^{\circ}C$ for 15 days, its quality-keeping was relatively good. The sedimentation of curd was repressed a little by adding hot-water extracted from 0.5% and 0.1% sea tangle. The overall sensory score of yogurt made from adding hot-water extracted from 1% sea tangle was the best of tested yogurt.

Preparation and Characteristics of Curd Yogurt from Milk Added with Purple Sweet Potato (자색고구마를 첨가한 호상요쿠르트의 제조와 특성)

  • A curd yogurt was prepared by fermenting milk added with skim milk powder and purple sweet potato by culture of 5 types of lactic acid bacteria(Lactobacillus delbruekii sub. sp. lactis, Streptococcus lactis, acidity, number of viable cell, stability of purple sweet potato's pigment and keeping qualify. Among the organisms tested, the acid production and number of viable cell by the culture of L bulgaricus remarkably increased for the first 12 hem which showed 1.04${\times}$10$\^$9/ CFU/mL in number of viable cell and 4.22 In pH where as fermentation by the culture of B. bifidum was slow. After 36 hours of incubation which showed 3.3 ${\times}$ l0$\^$8/ CFU/mL in number of viable cell and 5.1 in pH. In stabilities of purple sweet potato anthocyanin pigment n fermentation, yogurt by B. bifidum was found to be most stable followed by Leuc. lactis, L. delbruekii sub. sp. lactis, L bulgaricus, but yogurt by St. lactis was not stable. When curd yogurt added with Purple sweet Potato was kept at 2∼3$^{\circ}C$ for 14 day, its keeping quality(pH, titratable acidity, number of viable cell) was relative good except product by L. bulgaricus was found to be decreased most of viable cell. After 2 weeks of keeping, pigment of yogurt was decreased by B. bifidum, stable by L. delbruekii sub. sp. lactis.

Exploration of Virulence Markers and Genes of Listeria monocytogenes Isolated from Animal Products (축산물유래 Listeria monocytogenes의 virulence marker 및 gene 조사)

  • To investigate the epidemiological characteristics of 68 Listeria monocytogenes isolates, including 11 reference strains and 57 isolates from imported US beef, domestic meats(beef, pork, chicken meat), raw milk, and milk plants. L. monocytogenes was to evaluate the production of virulence proteins, such as hemolysin(LLO) and lecithinase(LCP), the adsorption of Congo red(CRA), and to detect virulence genes using the polymerase chain reaction(PCR). In the study of virulence protein production, 68(100%), 62(91.2%), and 54(79.4%) of the 68 L. monocytogenes strains were positive for LLO production, the LCP test, and the CRA test, respectively, while strains of other species, such as L. innocua, L. gray, L. murrayi, and L. welshimeri, were not. There were no significant differences between L. monocytogenes serotypes and the ability to produce LLO or LCP. L. monocytogenesstrains had very high hemolytic titers(2 to 16 fold), while the other Listeria species, other than L. ivanovii and L. seeligeri, did not. The hemolysin activities of L. monocytogenes, L. ivanovii, and L. seeligeri usually exceeded 1.0 HU/mg, while those of other Listeria spp. were less than 0.04 HU/mg. In the PCR assay, all of the L. monocytogenes strains contained the hlyA, plcA, plcB, inlA, and inlB virulence genes and produced a product of the expected size. In the PCR of the actA gene, the expected 385-bp product was seen in 39(57.4%) L. monocytogenesstrains, while an unexpected 268-bp product was seen in 29(42.6%) strains. Most L. monocytogenes strains isolated from Hanwoo beef produced the 385-bp actA gene product, while strains of imported US beef usually produced the 268-bp actA gene product. By contrast, no virulence gene products were amplified in the other Listeria spp.

Suppressive Effects of By-Product Extracts from Soybean on Adipocyte Differentiation and Expression of Obesity-Related Genes in 3T3-L1 Adipocytes (대두부산물의 지방세포분화 유도유전자의 발현저해 및 전지방세포 분화 억제 효과)

  • Soybean is known to contain various phytochemicals that are related to anti-oxidant, anti-inflammatory and anti-obesity effects in mice and humans. The anti-obesity effect of by-product extracts from soybean on the differentiation of 3T3-L1 pre-adipocytes to adipocytes was investigated by suppressing adipocyte differentiation and lipid accumulation with Oil Red-O assay and quantitative PCR. In inducing differentiation of 3T3-L1 pre-adipocytes in the presence of an adipogenic cocktail, isobutylmethylanthine (IBMX), dexamathasone, and insulin, treatment with filtrated soybean soaked water, soybean milk, and soycurd residue from soybean curd processing significantly decreased mRNA expression of obesity-related gene such as PPAR${\gamma}$, Fabp4, and Scd1, adipsin, apolipoprotein (APOE) and adiponectin (ADIPOQ) without any significant cytotoxicity. We also determined the well-known isoflavones in soybean, such as daidzein and genistein, in the by-product extracts. Taken together, we suggest that soybean by-product extract showed anti-obesity effect by suppressing adipocyte related gene expression, and that by-products collected during soybean curd processing may be a good candidate as an ingredient in health care products.

Effect of HMR Meal Kit Product Selection Attributes on Consumers Satisfaction and Other Recommendation Intention (HMR 밀키트 상품의 선택속성이 소비자만족 및 타인추천의도에 미치는 영향)

  • This study attempted an empirical analysis study on the Meal Kit Product, whose interest and demand continued to increase according to the eating out trend in the Untact era. In addition, this study attempted to investigate the relationship between the factors of Home Meal Replacement Meal Kit Product Selection Attributes, Consumers Satisfaction, and Other Recommendation Intention. Convenience sampling was used for consumers with experience in using Meal Kit Products released by food service companies and start-up companies. The investigation period was conducted for about one month from July 01, 2020, and the final 285 copies were used for analysis. The SPSS 21.0 statistical package program was used to verify the hypothesis. As a result of the analysis, the price (β=.241), convenience (β=.317), and diversity (β=.191) of Hypothesis 1 had a significant effect on Consumers Satisfaction. Price (β=.482), convenience (β=.133), and diversity (β=.342) were found to have a significant effect on the intention to recommend others. It was analyzed that Hypothesis 3's Consumers Satisfaction (β=.443) had a significant effect on Other Recommendation Intention. Finally, through this study, we expect to be provided as basic research data related to Meal Kit Product. It is intended to be presented as a theoretical basis for the use of marketing and direction for the development of milk kit products for catering companies and restaurants.
