Maternal 3-methylcrotonyl-coenzyme A carboxylase deficiency with elevated 3-hydroxyisovalerylcarnitine in breast milk

  • We report here a case of maternal 3-methylcrotonyl-coenzyme A carboxylase (3-MCC) deficiency in a Korean woman. Her 2 infants had elevated 3-hydroxyisovalerylcarnitine (C5-OH) on a neonatal screening test by liquid chromatography-tandem mass spectrometry (LC-MS/MS), but normal results were found on urine organic acid analysis. The patient was subjected to serial testing and we confirmed a maternal 3-MCC deficiency by blood spot and breast milk spot test by LC-MS/MS, serum amino acid analysis, urine organic acid and molecular genetic analysis that found c.838G>T (p.Asp280Tyr) homozygous mutation within exon 9 of the MCCB gene. Especially, we confirmed marked higher levels of C5-OH on breast milk spot by LC-MS/MS, in the case of maternal 3-MCC deficiency vs. controls.

Milk Yield and Immune Response of Periparturient and Early Lactation Friesian Cows Fed Diets Supplemented with a High Level of Amino-acid Chelated Chromium

  • The trial was carried out on twenty-one Friesian cows at the end of eight months gestation, nine multiparous and twelve primiparous; allocated into three groups (1 control, 2 and 3 experimental). The same diet was administred to all three groups before partum (12.8 kg DM/head/day) and after partum (18.8 kg DM/head/day). The cows in groups 2 and 3 received two different daily quantities of amino-acid chelated chromium (0.6 and 1.2 mg Cr/kg DM) from 4 weeks prior to presumed parturition to 6 weeks after. The milk yield control was carried out at 15, 30, 42 and 60 days. All animals were immunised two weeks prior to the presumed parturition and two weeks after with the following antigens: ovalbumin and brucellergene. Blood samples were collected weekly to monitor humoral and cell-mediated immune responses. When analysing the results of antibody immunity (ovalbumin) in the sixth blood collection both treated groups significantly increased compared to group 1 (0.5230 and 0.4536 vs. 0.1812 OD; p<0.05). The results of the cell-mediated immune response (brucellergene) had significant differences (p<0.10) in correspondence to the third (between group 2 and control) and the fifth (between groups 3 and 2) blood collection. Significant differences in fat corrected milk were observed at 42 days between group 3 and the other two groups (31.01 vs. 26.99 and 28.66 kg/d, p<0.05) and at 60 days between group 3 and control (30.88 vs. 26.69 kg/d, p<0.05). Before partum and at partum a positive immune response was obtained with a lower dose of chromium. After partum a positive immune response, anti-OVA indicator, was obtained with the higher dose of chromium while, $\gamma$-IFN indicator, with the lower dose. A significant increase of the milk yield resulted at both 42 and 60 days with the highest level of chromium.

국산(國産) 조제분유(調製粉乳)와 발효유(醱酵乳)의 영양효과(營養效果)에 관(關)한 연구(硏究) - 1. 유제품(乳製品)의 영양소(營養素) 함량비교(含量比較)를 위한 연구(硏究) - (Studies on the Nutritional Effects of Locally Produced Dried Formula Milk and Fermented Milk -1. Comparative Studies on the Nutrients Content of Dried Formula Milk and Fermented Milk-)

  • 국내(國內)에서 생산(生産)되고 있는 세가지 조제분유(調製粉乳) 및 한가지 발효유(醱酵乳)의 일반성분(一般成分), 비타민, 무기물, 아미노산(酸) 및 지방산(脂肪酸) 함량(含量)을 분석(分析)하여 보증성분(保證成分)과 비교(比較)하고, 아울러 모유(母乳)와 비교(比較)하여 이들 유제품(乳製品)의 영양가(營養價)를 평가(評價)하기 위하여 7주간(週間) 실험(實驗)을 실시(實施)하였던 바 그 결과(結果)를 요약(要約)하면 다음과 같다. 1. 단백질(蛋白質) 및 지방(脂肪) 함량(含量)에 대한 분석치(分析値)는 보증함량(保證含量)보다 약간 낮았고, 당질(糖質) 함량(含量)은 약간 높았으나, 다른 성분(成分)은 대체로 분석치(分析値)와 보증함량(保證含量)이 서로 근사(近似)했고, 분유(粉乳)들간(間)에도 차이(差異)가 없었다. 발효유의 단백질(蛋白質) 및 지방(脂肪) 함량(含量)은 우유보다 낮았으나, 당질(糖質)의 함량(含量)이 높은 것으로 보아 상당한 양(量)의 가당(加糖)이 있는 듯 하였다. 2. 비타민은 거의 모든 분석치(分析値0가 보증(保證)된 함량(含量)보다 높았는데, 특히 비타민A는 $2{\sim}3$배(倍), 비타민E는 $4{\sim}5$배(倍)씩 높았고, 나이아신은 보증함량(保證含量)에 미달하였다. 또한 분유(粉乳)들간(間)에는 약간의 함량차(含量差)가 있었다. 3. 무기질(無機質)은 분석치(分析値)와 보증함량(保證含量)이 비슷했는데 칼슘 외 분석치(分析値)가 보증함량(保證含量)보다 다소 많았다. 이상의 분석(分析)된 조제분유(調製紛乳)들을 15%로 희석하여 우유(牛乳) 및 모유(母乳)와 비교(比較)했을 때, 열량(熱量), 당질(糖質), 단백질(蛋白質)도 우유(牛乳)보다 훨씬 희석되어 모유(母乳)에 유사(類似)함을 알 수 있었다. 또한 철분 및 대개의 비타민이 모유(母乳)나 우유(牛乳)보다 많은 점으로 미루어 이러한 영양소(營養素)가 강화(强化)되어 있음을 짐작할 수 있었다. 이 환원유(還元乳)를 일정(一定)한 조건(條件)에서 한국 영양권장량과 비교(比較)했을 때, 모든 영양소(營養素)를 충분(充分)히 공급(供給)할 수 있었는데 나이아신 만이 권장량에 미달하였다. 또한 분유(粉乳) C에서 철분이 약간 미달했고, 비타민A는 1일(日) 권장량에 6배(倍)나 되어 앞으로 재검토(再檢討)를 요(要)하는 문제라 하겠다. 4. 아미노산(酸) 조성(組成)은 분유간(粉乳間)에 다수 차계(差界)를 보였으며, 필수(必須)아미노산(酸) 조성(組成)이 우유에 가까웠던 점(點)으로 보아 아미노산 조절(調節)은 없었는듯 하였다. 발효유의 아미노산(酸) 조성(組成)은 우유와 거의 같았다. 5. 지방산(脂肪酸)의 조성(組成)은 전체(全體) 포화지방산대(飽和脂肪酸對) 불포화지방산(不飽和脂肪酸)의 비(比)가 3종(種)의 분유간(粉乳間)에 비슷하였고, 특히 필수지방산(必須脂肪酸)의 조성(組成)이 모유(母乳)와 유사(類似)하거나 높아 이들 지방산(脂肪酸)이 첨가(添加)되어 있음을 나타냈다. 이상의 여러 결과(結果)들을 종합(綜合)할 때 3종(種)의 분유간(粉乳間) 영양효과(營養效果)는 비슷하고, 조제분유(調製粉乳)의 일반조성(一般組成), 무기질(無機質) 및 지방산(脂肪酸) 조성(組成)에 있어서 모유(母乳)에 상당히 접근(接近)하는 것으로 믿어진다. 한편 철분, 비타민 등(等)의 강화(强化)로서 단일식품(單一食品)으로서의 효용성(效用性)을 높인 것은 사실이나, 일부 영양소(營養素)의 지나친 강화(强化)문제는 좀더 신중히 다루어져야 할 것으로 생각된다.

Optimizing hormonal and amino acid combinations for enhanced cell proliferation and cell cycle progression in bovine mammary epithelial cells

  • Objective: The number of bovine mammary epithelial cells (BMECs) is closely associated with the quantity of milk production in dairy cows; however, the optimal levels and the combined effects of hormones and essential amino acids (EAAs) on cell proliferation are not completely understood. Thus, the purpose of this study was to determine the optimal combination of individual hormones and EAAs for cell proliferation and related signaling pathways in BMECs. Methods: Immortalized BMECs (MAC-T) were treated with six hormones (insulin, cortisol, progesterone, estrone, 17β-estradiol, and epidermal growth factor) and ten EAAs (arginine, histidine, leucine, isoleucine, threonine, tryptophan, lysine, methionine, phenylalanine, and valine) for 24 h. Results: Cells were cultured in a medium containing 10% fetal bovine serum (FBS) as FBS supplemented at a concentration of 10% to 50% showed a comparable increase in cell proliferation rate. The optimized combination of four hormones (insulin, cortisol, progesterone, and 17β-estradiol) and 20% of a mixture of ten EAAs led to the highest cell proliferation rate, which led to a significant increase in cell cycle progression at the S and G2/M phases, in the protein levels of proliferating cell nuclear antigen and cyclin B1, cell nucleus staining, and in cell numbers. Conclusion: The optimal combination of hormones and EAAs increased BMEC proliferation by enhancing cell cycle progression in the S and G/2M phases. Our findings indicate that optimizing hormone and amino acid levels has the potential to enhance milk production, both in cell culture settings by promoting increased cell numbers, and in dairy cows by regulating feed intake.

  • Infant formula is classified into standard cow's milk-based and special formulas. This review aimed at summarizing the types of special milk formulas currently sold in Korea, and the appropriate indications for the use of these formulas; lactose free formula, soy-based formula, protein hydrolysate formula, amino acid-based formula, preterm formula, medium chain triglyceride formula, low-phosphorus formula, protein-energy-enriched formula, and formulas for inborn errors of metabolism.

  • Prolific females require better nutrition and feeding practice because of larger litter size and the substantial decline in body fat. Life-time pig output will be compromised if body protein and fat are not properly managed. First litter females are especially vulnerable because they can loose ${\geq}15%$ of whole-body protein. Conservation of body protein mass during first lactation minimizes wean to estrus interval and increases second litter size (up to 1.2 pigs). The ability to influence litter-size by amino acid nutrition is a new dimension in our understanding. A P2 fat depth below 12 mm at farrow and below 10 mm at wean compromised wean to estrus interval (>2 d) and next litter size (0.5 to 1.5 pigs) in sows. It is now clear that a 'modest' excess of feed during the first 72 h of pregnancy decreases embryo viability so that the potential for an increased litter size at birth is not realized. The capacity for milk production by prolific young sows is 25% higher than the standard used previously (NRC, 1988). First litter females averaged 9.82 kg milk/d for a 21 d lactation. Second and third litter counterparts averaged 10.35 kg/d. Milk production was 95% of peak by 10 d of lactation and sows were in greatest negative energy and lysine balance during the first 6 d. Nearly 45% of the total loss in body protein occurred within the first 6 d, but this could reduced to 30-35% by using a more aggressive feeding strategy after parturition. There appear to be 2 phases in lactation for lysine need (d 2-12 vs 12-21). Feeding to the higher level alleviates the second litter size decline. The lysine requirement for lactation can be predicted with accuracy, but we are not able to predict the second limiting amino acid. Mammary uptake of valine relative to lysine and recent work with practical diets suggest that the recent NRC (1998) pattern is realistic and that threonine and valine could be co-limiting for corn-soy diets for prolific sows nursing 10-11 pigs. Empirical studies are needed to refine the ideal pattern so that synthetic lysine can be used with more confidence. Milk fat output for the elite sow is extraordinary and poses an unnecessarily high energetic cost. Methods that reduce mammary fat synthesis will benefit the sow and may enhance piglet growth.

Penicillium oxalicum HCLF-34로부터 Acid Proteinase의 부분유전자 Cloning 및 Sequencing (Cloning and Sequencing of Gene Fragment of Acid Proteinase from Penicillium oxalicum HCLF-34)

  • Acid proteinase는Aspergillus niger (acid pretense A)와 Cryphonectria parasitica (acid proteinase EapC)에서 분비하는 단백질로서 치즈를 제조할 때 이용하는 단백질이다. 본 연구에서는 Penicillium oxalicum HCLF-34로부터 acid proteinase의 부분유전자를 기존에 밝혀진 acid proteinase의 homology 정보로부터 degenerate primer를 제작하여 PCR방법을 이용하여 cloning하였다. Cloning된 유전자로부터 438 bp 염기서열을 분석하였으며, 이 염기서 열을 146개의 아미노산 정보로 변환하여 acid proteinase family와 homology를 비교한 결과 acid protease A와 71% 아미노산 서열의 homology를 나타내었고, EapC와 67%의 아미노산 서열 homology가 확인되었다.

전지분유와 발효유를 이용한 치즈 유사품개발 (The Development of Imitated Cheese Using Whole Milk Powder and Fermented Milk)

  • 재고 분유를 이용하기 위하여 유청 배출없이 전지 분유와 발효유를 혼합하여 모조 치즈를 제조하였으며, 숙성 환경과 숙성 기간의 차이를 관찰하여 자연 치즈와 유사한 성질을 나타내는 최적의 숙성 조건을 규명하기 위하여 수분함량 변화, 일반성분, 비카제인태 질소화합물, 비단백태 질소화합물 그리고 유리 아미노산 등의 변화를 관찰하였다. 본 모조 치즈는 제조 후 건조 1일째가 수분함량이 40.27%로 가장 적합하였고, 이들은 진공 포장한 후 $12^{\circ}C$에서 숙성하는 방법과 P. camemberti 곰팡이를 분무한 후 $12^{\circ}C$, 95% RH에서 숙성하는 두 가지 방법으로 숙성하였다. 곰팡이 숙성 시 유당 함량이 24.64%에서 4주 숙성 시 5.43%로 급격히 감소하는 현상이 있었으나 진공 포장 숙성 시에는 유당의 변화가 심하지 않았다. 또한 진공 포장 숙성 시 비카제인태 질소 및 비단백태 질소는 숙성 4주 이후에 각각 28.10 mg/mL에서 50.61 mg/mL으로, 6.58 mg/mL에서 21.59 mg/mL으로 증가하였으며, 곰팡이 숙성 시 비카제인태 질소 및 비단백태 질소는 숙성 4주 이후에 각각 28.10 mg/mL에서 54.05 mg/mL으로, 6.58 mg/mL에서 23.06 mg/mL으로 증가하였다. 진공 포장 숙성 시 4주간의 유리 아미노산 차이를 측정한 결과, arginine, tyrosine, leucine이 두드러진 증가를 보였다. 그리고 총 유리 아미노산이 7.73 nmol/mg에서 9.29 nmol/mg으로 증가하였고, bitter amino acid는 1.36 nmol/mg에서 2.43 nmol/mg으로 증가하였다. 또한 곰팡이 숙성 시 4주간의 유리 아미노산을 측정한 결과, asparagine을 제외한 모든 유리 아미노산이 두드러진 증가를 보였다. 총 유리 아미노산이 8.40 nmol/mg에서 34.87 nmol/mg로 증가하였고, bitter amino acid는 1.53 nmol/mg에서 10.02 nmol/mg로 증가하였다. 치즈 유사품 제조 시에 P. camemberti를 이용한 치즈가 P. camemberti 를 이용하지 않는 치즈보다 단백질 분해 정도나 풍미가 더 우수한 것으로 나타났다.

Principal Milk Components in Buffalo, Holstein Cross, Indigenous Cattle and Red Chittagong Cattle from Bangladesh

  • The aim of the present study was to get a total physical and chemical characterization and comparison of the principal components in Bangladeshi buffalo (B), Holstein cross (HX), Indigenous cattle (IC) and Red Chittagong Cattle (RCC) milk. Protein and casein (CN) composition and type, casein micellar size (CMS), naturally occurring peptides, free amino acids, fat, milk fat globule size (MFGS), fatty acid composition, carbohydrates, total and individual minerals were analyzed. These components are related to technological and nutritional properties of milk. Consequently, they are important for the dairy industry and in the animal feeding and breeding strategies. Considerable variation in most of the principal components of milk were observed among the animals. The milk of RCC and IC contained higher protein, CN, ${\beta}$-CN, whey protein, lactose, total mineral and P. They were more or less similar in most of the all other components. The B milk was found higher in CN number, in the content of ${\alpha}_{s2}-$, ${\kappa}$-CN and ${\beta}$-lactalbumin, free amino acids, unsaturated fatty acids, Ca and Ca:P. The B milk was also lower in ${\beta}$-lactoglobulin content and had the largest CMS and MFGS. Proportion of CN to whey protein was lower in HX milk and this milk was found higher in ${\beta}$-lactoglobulin and naturally occuring peptides. Considering the results obtained including the ratio of ${\alpha}_{s1}-$, ${\alpha}_{s2}-$, ${\beta}$- and ${\kappa}$-CN, B and RCC milk showed best data both from nutritional and technological aspects.

Rapid Analysis of Melamine Content in Powdered and Liquid Milk Using Fourier Transform Infrared Spectroscopy

  • Melamine is a chemical intermediate to manufacture amino resins and plastics, which cannot be used as food additive since it can cause kidney stones. A qualitative determination method of melamine residue in powdered and liquid milk was developed using Fourier transform infrared (FTIR) spectroscopic technique. The calibration sets consisted of 21 standard melamine solutions, in which 1% trichloroacetic acid aqueous solution and acetonitrile (3:1, v/v) were used as solvent. The model was validated using 10 standard melamine solutions which were unused to build up the calibration set. Infrared (IR) absorbance peaks specific to almost all chemical groups in melamine molecule were shown in the spectral range between 1,100 and 1,800/cm. Combined partial least squares (PLS)-$2^{nd}$ derivative calibration model coupled with mean centering (MC) mathematical enhancement showed the highest correlation coefficients ($R^2$>0.99). In brief, the FTIR technique can be used for quantitative analysis of melamine in milk samples.
