Moderate diet-induced weight loss is associated with improved insulin sensitivity in middle-aged healthy obese Korean women

  • BACKGROUND/OBJECTIVE: The goal of the present study was to investigate the effects of moderate caloric restriction on ${\beta}$-cell function and insulin sensitivity in middle-aged obese Korean women. SUBJECTS/METHODS: Fifty-seven obese pre-menopausal Korean women participated in a 12-week calorie restriction program. Data on total cholesterol (TC), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), triglycerides (TG), and fasting serum levels of glucose, insulin, C-peptide, blood pressure, leptin and anthropometrics were collected. A dietary intake assessment was based on three days of food recording. Additionally, ${\beta}$-cell function [homeostasis model assessment of ${\beta}$-cell (HOMA-${\beta}$), insulinogenic index (ISI), C-peptide:glucose ratio, and area under curve insulin/glucose ($AUC_{ins/glu}$)] and insulin sensitivity [homeostasis model assessment for insulin resistance (HOMA-IR), Quantitative insulin-sensitivity check index (QUICKI) and Matsuda index (MI)] were recorded. RESULTS: When calories were reduced by an average of 422 kcal/day for 12 weeks, BMI (-2.7%), body fat mass (-10.2%), and waist circumference (-5%) all decreased significantly (P < 0.05). After calorie restriction, weight, body fat percentage, hip circumference, BP, TC, HDL-C, LDL-C, plasma glucose at fasting, insulin at fasting and 120 min, $AUC_{glu}$ and the insulin area under the curve all decreased significantly (all P < 0.05), while insulin sensitivity (HOMA-IR, QUICKI and Matsuda index) measured by OGTT improved significantly (P < 0.01). CONCLUSIONS: Moderate weight loss due to caloric restriction with reduction in insulin resistance improves glucose tolerance and insulin sensitivity in middle-aged obese women and thereby may help prevent the development of type 2 diabetes mellitus.

Age-Related $CD4^+CD25^+Foxp3^+$ Regulatory T-Cell Responses During Plasmodium berghei ANKA Infection in Mice Susceptible or Resistant to Cerebral Malaria

  • Different functions have been attributed to $CD4^+CD25^+Foxp3^+$ regulatory T-cells (Tregs) during malaria infection. Herein, we describe the disparity in Treg response and pro- and anti-inflammatory cytokines during infection with Plasmodium berghei ANKA between young (3-week-old) and middle-aged (8-month-old) C57BL/6 mice. Young mice were susceptible to cerebral malaria (CM), while the middle-aged mice were resistant to CM and succumbed to hyperparasitemia and severe anemia. The levels of pro-inflammatory cytokines, such as TNF-${\alpha}$, in young CM-susceptible mice were markedly higher than in middle-aged CM-resistant mice. An increased absolute number of Tregs 3-5 days post-inoculation, co-occurring with elevated IL-10 levels, was observed in middle-aged CM-resistant mice but not in young CM-susceptible mice. Our findings suggest that Treg proliferation might be associated with the suppression of excessive pro-inflammatory Th1 response during early malaria infection, leading to resistance to CM in the middle-aged mice, possibly in an IL-10-dependent manner.

Scanning Electron Microscopic Study of the Sertoli Cell Processes in the Rat (쥐 Sertoli 세포돌기의 주사전자현미경적 연구)

  • The three-dimensional structure of the Sertoli cell in the rat was investigated by scanning electron microscopy. Morphologically, seven types of Sertoli cell processes were evident : Shrot, flat and ramified processes are projected from the lateral side of the basal portion of Sertoli cell. Leaf-like processes are attached to the surface of spermatocytes and spermatids. Slender cord-like processes, flat and irregular shaped processes, sucker-like processes and club-like processes are observated in the middle and apical portion of seminiferous epithelium. The sheet-like processes rest upon more than one-thirds of the surface of each spermatogonium, spermatocyes and spermatids located in the proximity of the Sertoli cell. All Sertoli processes are originated from Sertoli cell column. Just before spermiation, the processes which are attached to the head of maturation spermatid are eliminated. Though the mechanism for elimination of residual body is not known, these observations segget that the Sertoli cell process are thought to have a reciprocity with the germ cells.

  • The polymer electrolyte membrane fuel cell(PEMFC) with the advantages of low-operating temperature, high current density, low cost and volume, fast start-up ability, and suitability for discontinuous operation becomes the most reasonable and attractive power system for transportation vehicle and micro-grid power plant in a household. 200W PEMFC(Polymer electrolyte membrane fuel cell) system applied to middle and small-scaled micro-grid power system was constructed by this study, then the electrical characteristics and diagnosis of the fuel cell were analyzed with thermal variation.

ICM - Trophectoderm Cell Numbers of Mouse IVF/IVC Blastocysts (체외생산된 생쥐 배반포기배의 ICM과 Trophectoderm 세포수에 관한 연구)

  • This work has been carried out to examine the number of Total, ICM and TE cells of F1 mouse blastcysts at day 4 after IVF by differential labelling of the nuclei with polynucleotide-specific fluorochromes and to obtain a fundamental information of preimplantation mouse embryo development. Blastocysts produced by superovulated B6CBA F1(C57BL/${\times}$CBA) eggs were inseminated with $1{\times}10^6$spermatozoa/ml and cultured in M16 medium at $37^{\circ}C$, 5% $CO_2$ incubator for 95hrs. Blastocysts were classified as early, middle, expanded and hatching stage according to the developmental morphology; blastocoel expansion and zona thickness. The results obtained in these experiments were summarized as follows; 1) The development rate of blastocysts at 95hrs after IVF was 86.7% and classified blastocysts to early, middle, expanded and hatching were 16.3%, 18.9%, 10.5% and 40.9%, respectively. 2) The numbers of total blastomere using bisbenzimide in the classified blastocysts to early, middle, expanded and hatching were 35.6${\pm}$1O.4, 49.4${\pm}$8.6, 60.8${\pm}$1O.7 and 62.7${\pm}$13.9, respectively. 3) In ICM and TE cell number by using differential labelling with polynucleotide-specific fluorochrome in the classified blastocysts to early, middle, expanded and hatching; ICM numbers were 9.6${\pm}$3.0, 13.6${\pm}$3.9, 16.0${\pm}$3.3 and 19.5${\pm}$4.6, respectively and TE cell numbers were 30.6${\pm}$5.1, 39.9${\pm}$5.8, 42.2${\pm}$8.1 and 43.7${\pm}$11.1, respectively. These results showed the same increase pattern according to development advance level. Also, when compared with the results of total count were obtained between bisbenzimide only and differential labelling, both of them showed the same increase pattern according to development level and at the same time their cell numbers were almost the same. So, rapid and simple cell count method using differential labelling can be used for the examination of later preimplantation development or as an indicator of embryo quality according to the variables of culture conditions.

  • Crypton is a SP-network block cipher that attracts much attention because of its excellent performance on hardware. Based on Crypton, mCrypton is designed as a lightweight block cipher suitable for Internet of Things (IoT) and Radio Frequency Identification (RFID). The security of Crypton and mCrypton under meet-in-the-middle attack is analyzed in this paper. By analyzing the differential properties of cell permutation, several differential characteristics are introduced to construct generalized ${\delta}-sets$. With the usage of a generalized ${\delta}-set$ and differential enumeration technique, a 6-round meet-in-the-middle distinguisher is proposed to give the first meet-in-the-middle attack on 9-round Crypton-192 and some improvements on the cryptanalysis of 10-round Crypton-256 are given. Combined with the properties of nibble permutation and substitution, an improved meet-in-the-middle attack on 8-round mCrypton is proposed and the first complete attack on 9-round mCrypton-96 is proposed.

A New Structure and Driving Scheme of PDP for High Luminous Efficacy

  • We have developed a new PDP cell structure called MARI(Multi Anode for Reduction of Ionic effect) and new driving scheme achieving a high luminous efficacy. The MARI PDP has middle electrode inserted between X and Y main electrodes. In the MARI PDP, reset and scan voltage is applied to middle electrode and sustain voltage is applied to X and Y electrode. Using a long gap sustain discharge we accomplished a high luminous efficacy. And we developed 42"full panel adopting MARI structure and new driving scheme and attained luminous efficacy of 2.35 lm/W.

  • Structure of the skin in amphibious fish, Periophthalmus modestus, was described in relation to cutaneous respiration. The epidermis has no gland cell. The epidermis consists of three regions: outermost layer of one to five layers of flattened epithelial cells, middle layer of swollen epithelial cells instead of glandular cells and stratum germinativum of cuboidal cells. There are numerous blood capillaries in the outermost layer of the epidermis and diffusion distance between the blood of capillaries and the epidermis is about 1.4 Um. The middle layer of the epidermis appears to be a web-like structure due to the swollen epithelial cells. The stratum germinativum has a well-developed lymphatic space containing lymphocytes. There are numerous blood capillaries and elliptical area with acid mucopolysaccharides in stratum laxum of the dermis. The skin of Periophthalmus modestus may be an accessory respiratory organ for oxygen uptake during terrestrial or aquatic life.

  • This paper was carried out to Investigate changes in the activities of cell-degrading enzymes, cell wall components and cell structure of peach during maturation and storage for valuation of quality. The firmness of peach was decreased during maturation and storage, and was remarkably decreased in Daegubo than Yumyung. Polygalacturonase and $\beta$-galactosidase activities of peach were increased during maturation and storage, and were remarably increased in soft peach and in mature and soft peach, respectively. Contents of alcohol-insoluble substance, cell wall, and total and insoluble pectin of peach were decreased during maturation and storage, but cellulose and soluble pectin were increased. Intracellular space was enlarged during maturation and middle lamella was gradually degraded during maturation.

  • Ultrastructures on the integumentary epidermis of the marbled sole, Limanda yokahamae, were examined by means of the light and transmission electron microscope. Epidermal layer consists of supporting cells, unicellular glands and accessory cells. The supporting cells were classified into superficial cell, intermediated cell and basal cell. The cytoplasm of supporting cells is divided into cortex and medullar part. In the cortex and medullar part, microfilaments and cell organelles are well developed, respectively. Gland cells are present in the superficial and middle epidermis. The cytoplasm of mucous cell reacted to blue in AB-PAS (pH 2.5). Club cell has a roundish central vacuole and well-developed microfilaments in the cytoplasm. Granular cells are occurs in the middle and basal epidermis , and the cytoplasm is occupied with membrane-bounded granules of electron dense. Chloride cells are present in the superficial epidermis , and the cytoplasm is occupied with tubular mitochondria. Three types of pigment cells can be distinguished by electron density of cytoplasmic inclusions.
