Comparison of Scanning Electron Microscopic Specimens Dried with Different Methods (건조기법을 달리한 SEM 시료상의 비교검토)

  • To compare the quality of ultrastructural preservation of Scanning Electron Microscopic specimens dried with different methods; pure air-drying, air-drying with using Tetramethylsilane(TMS), four kind of air-drying using Hexamethyldisilazane(HMDS) and critical point drying(CPD), we conducted scanning electron microscopic observation on liver, skeletal muscle and intestinal tissues from laboratory rat treated with each method. In pure air drying group, severe distortion of tissue surface was observed, and in HMDS treated group, only liver tissue showed slight distortion. But in TMS treated group, each tissue showed a good presentation comparable to CPD group. The results suggest that the method of air-drying using TMS may be the former is less expensive and simple be and also time-saving.

  • The purpose of this study was to observe microscopic change of salivary gland tissue, which is a cause of xerostomia in diabetic condition; for this target, the author injected streptozotocin 0.1ml/100 gm b.w. on the rat, Sprague Dawley, to induce diabetes, and then observed microscopic changes in parotid gland tissue using light microscopy and electron microscopy. The results were as follows : 1. Parotid gland tissue of the diabetic rat was atrophied or degenerated in lapse of experimental time, but began to repair from 14 days after diabetic induction. 2. In the basal lamina of the vessel of parotid gland tissue in the diabetic rat, lamina lucida was discontinued and lamina densa was increased in thickness, but the number of capillary was gradually increased and dilated. 3. In acinic and intercalated ductal cells of parotid gland in the diabetic rat, changes of mitochondria, RER, secretory granule, free ribosome were prominent. In conclusion, the present study demonstrated that degenerative changes of the parotid gland tissue were due to not completely thickening of the basal lamina of vessels, but many other causal factors, because thickness of the basal lamina of vessels was not related with degenerative changes.

Usefulness of Microscopic Procedures in Composite Grafts for Fingertip Injuries

  • Purpose: Fingertip amputations are the most common type of upper limb amputations. Composite grafting is a simple and cost-effective technique. Although many factors have investigated the success of composite grafting, the success rate is not high. Therefore, this study was conducted to investigate whether the microscopic procedure process during composite grafts improves the success rate. Materials and Methods: Thirteen cases of unreplantable fingertip amputation underwent a microscopic resection procedure for composite graft in the operating room. The principle of the procedure was to remove the least devitalized tissue, maximize the clean tissue preservation and exact trimming of the acral vessel and to remove as many foreign bodies as possible. Results: All fingertips in the thirteen patients survived completely without additional procedures. Conclusion: Composite grafting allows for the preservation of length while avoiding the donor site morbidity of locoregional flaps. Most composite grafts are performed as quickly as possible in a gross environment. However, we take noticed the microscopic resection. This process is thought to increase the survival rate for the following reasons. First, the minimal resection will maximize the junction surface area and increase serum imbibition. Second, sophisticated trimming of injured distal vessels will increase the likelihood of inosculation. Third, accurate foreign body removal will reduce the probability of infection and make it possible to increase the concentration and efficiency in a microscopic environment. Although there is a need for more research into the mechanisms, we recommend using a composite graft under the microscopic environment.

Microscopic Identification of the Chinese Patent Medicine 'Geum Ryeo Hwan' (금려환(錦儷丸)의 현미감정 연구)

  • Geum Ryeo Hwan(錦儷丸) is Chinese patent medicine which has been used for women's diseases and climacteric syndrome in Korea. It consists of 12 kinds of powdered crude drugs. In powdered crude drugs, it is hard to identify each component by chemical analysis or morphological examination. However, the method of identification of powdered crude drugs has not been clearly established. Therefore, it is of interest to establish the microscopic method for identification of powdered crude drugs of Chinese patent medicines. The effectiveness of microscopic method is exemplified by the identification of tissue and contents of crude drugs by comparison with standard drugs. Moreover, this method has advantage as a microscale analysis, since it requires only a small amount of specimens. In this study, it is demonstrated that the microscopic method is very effective for the identification of 12 crude drug ingredients in Geum Ryeo Hwan.

  • Ah Myeong Dan (兒明丹) is Chinese patent medicine which is used for congenital fever and congenital boils in Korea. It consists of 11 kinds of powdered crude drugs. In powdered crude drugs, it is hard to identify each component by chemical analysis or morphological examination. However, the method of identification of powdered crude drugs has not been clearly established. Therefore, it is of interest to establish the microscopic method for identification of powdered crude drugs of Chinese patent medicines. Bone Formation by rhBMP-7 Transduced HEK 293 Cells in Nude Mouse (재조합 BMP-7 유전자가 전달된 HEK 293 세포에 의한 누드 마우스에서의 뼈형성)

  • To induce bone formation at ectopic site by tissue engineering and gene therapy, we transplanted collagen sponges containing rhBMP-7 transduced HEK 293 cells in the hypodermis of nude mice. Bone formation was investigated by histological and electron microscopic method at 3, 6, and 9 weeks after transplantation. At 9 weeks after transplantation, eosinophilic bony tissue was observed in the implanted collagen sponge and was confirmed as bone tissue by Von Kossa stain. In the transmission electron microscopic observation, the cells in newly formed bone tissue had eccentrically located nucleus and well developed rough endoplasmic reticulum (rER). Therefore, the cells were evaluated as osteoblasts. Those results suggest that it is possible to form a bone tissue in the ectopic site by transplantation of rhBMP-7 transduced HEK 293 cells. This will be contributed to push more advanced gene therapy for bone formation. However, the HEK 293 cell is unable to apply to the clinical gene therapy. Therefore it is worth to find more compatible cells for clinical application. In addition, collagen sponge is considered as an excellent scaffold and/or carrier for gene therapy and a good biomaterial for tissue engineering.

  • The purpose of this study was to observe the effects of early loading on the surrounding bone tissue of the implants. 12 ITI HS implants (8 mm) were inserted on the mandible at 3 mongrel dogs. The implants were divided into two groups; the functional group was functioned with crown, and nonfunctional group was functioned without crown. After 3 and 5 months animals were sacrificed and specimens were examined using radiography, light microscopy, scanning electron microscopy. The following results were obtained. 1. On light microscopic and scanning electron microscopic examination, the difference in the amounts of bone attachment between 3 months loaded and unloaded implants were a little. Sometimes more bone attachment were observed in loaded implants. 2. Many osteoids surrounded the fibrous connective tissues indicating of bone remodeling were observed along the outer surface of the implants. 3. On clinical, radiographic, microscopic examination, epithelial downgrowths were not observed and bone attachments to the outer surface of the implants were observed in 3 months loaded and unloaded implants with good oral hygiene. 4. Epithelial downgrowths and crater-like bone resorptions were observed on 5 months loaded and unloaded implants with poor oral hygiene. 5. Many inflammatory cells in the epithelial tissue were packed in the hollow on 5 months loaded implants.

  • The purpose of this study was to observe the effect of ${\beta}GP$ in the remaining dental pulp tissue after pulpotomy in the dogs' teeth. For vital pulpotomy, 72 dogs' teeth were used and class V cavities were prepared and the pulps were amputated. ZOE and Dycal (Caulk Co., USA) were placed over the amputated tissue and cavities were sealed with ZOE cement in the control group. In the experimental group, ${\beta}GP$, ${\beta}GP$-ZOE, ${\beta}GP$-Dycal were placed over the exposed pulp tissues respectively. Dogs were sacrificed after 1, 2 and 4 weeks following the operations and the teeth were decalcified in the nitric acid, sectioned and stained with HE for light microscopic examination. For electron microscopic examination, specimens were made after 2 and 4 weeks following the operation. A comparative microscopic examination revealed as follows. 1. The dentin bridge was formed continuously due to osteodentin in the ${\beta}GP$-Dycal group at the 2nd week, the dentin bridge composed of osteodentin and tubular dentin was observed at the 4th week. 2. Osteodentin formation was vigorously in the ${\beta}GP$-Dycal than in the Dycal group. 3. In the surface of osteodentin the osteodentinoblasts showing vivid synthetic activity were observed and the matrix vesicles were presented during calcification of osteoid dentin matrix. 4. The dentin bridge formation was not observed in ${\beta}GP$ group and ${\beta}GP$-ZOE group.

THYMOLIPOMA(Report of one case) (흉선지방종-1례 보고-)

  • We experienced thymolipoma which is a rare benign mediastinal tumor, an admixture of atrophic thymic tissue and matured adipose tissue. The patient was, 13 years old boy, admitted due to anterior mediastinal mass on routine chest X-ray and complained of mild exertional dyspnea. The physical examination revealed percussion dullness, decreased breathing sound at right anterior hemithorax. The chest PA and lateral views showed both paracardiac mass density and obliteration of retrosternal clear space. The chest CT Scan showed tubular structure containing of thymic tissue and enhanced fatty tissue, suggested thymolipoma. Operation was performed under the impression of thymolipoma. Through median sternotomy, the huge mass was resected. Grossly, the mass was yellowish fatty colored, measured 15X12X10cm, weighed 600gm. Light microscopic examination showed that thymic tissue containing of Hassal`s corpuscles were surrounded by matured adipose tissue, as the characteristic finding of thymolipoma. The postoperative course was uneventful.
