Microscopic Identification of 'Hwan So Dan' (환소단의 현미감정연구)

  • 'Hwan So Dan(還少丹)' is Chinese patent medicine which has been used for shortage of vitality and asthenia in Korea. It consists of 14 kinds of powdered crude drugs. In powdered crude drugs, it is hard to identify each component by chemical analysis or morphological examination. However, the method of identification of powdered crude drugs has not been clearly established. For the identification of individual crude drugs in such powdery mixtures, microscopic method may advantageously be used as it requires only a small amount specimens. In this study, it is demonstrated that the microscopic method is very effective for the identification of 14 crude drug ingredients in Hwan So Dan.

In powered crude drugs, it is hard to identify each component by chemical analysis or morphological examination. However, the method of identification of powdered crude drugs has not been clearly established. For the identification of individual crude drugs in such powdery mixtures, microscopic method may advantageously be used as it requires only a small amount of specimens. In this study, it is demonstrated that the microscopic method is very effective for the identification of 23 crude drug ingredients in Hwan Myeong Hwan.

In powered crude drugs, it is hard to identify each component by chemical analysis or morphological examination. For the identification of individual crude drugs in such powdery mixtures, microscopic method may advantageously be used, as it requires only small amount of specimens. In this study, it is demonstrated that the microscopic method is very effective for the identification of 24 crude drug ingredients in Dae Jeong Hwan.

Microscopic Analysis of Early Age Cement Paste Axially Compressed 20% (20% 축압축 시킨 초기재령 시멘트 페이스트 미세 분석)

  • Many researches in the past have shown that a majority initial cracking in concrete are caused during early age period. Therefore, the close examination of early age concrete behavior under various stress conditions is necessary to fully understand the cracking mechanism of concrete. In this study early age cement paste specimen is axially strained up to 20% of its original length by laterally reinforcing it. This type of test is called "Tube Squash Test" and has been previously used to apply up to 50% axial strain on concrete. Microscopic analyses (XRD, FESEM, EDS and DSE/TG) are performed on 20% axially strained early age cement paste specimen. The analysis results show that the microscopic structures and material characteristics of 20% axially strained cement paste remained same as the unstrained cement paste.

Accuracy Improvement of Lattice Parameters Measured from Electron Diffraction Data (전자회절을 이용한 격자상수의 측정 정확도 향상)

  • For quantitative analysis of nano-crystal structure, we reported the accuracy improvement method of lattice parameters measured from electron diffraction. For calculation of Au lattice parameters used as a standard crystal structure, it was considered two different acquisition methods (detector and enegy-filter) and three different calculation methods (conventional, least-square and regression fit). As a result, the measurement reliability could be enhanced by using CCD camera which gives higher performance, while energy-filtering did not affect the improvement the camera constant accuracy. Also, the accuracy of lattice parameters could be improved up to $10^{-4}$ order by regression fitting with correction formula. Finally, it is expected that the combination of regression fitting and intensity extraction from energy-filtered precession electron diffraction gives a solution of quantitative structure analysis for unknown nano-crystals.

Circular Fast Fourier Transform Application: A Useful Script for Fast Fourier Transform Data Analysis of High-resolution Transmission Electron Microscopy Image

  • Transmission electron microscope (TEM) is an excellent tool for studying the structure and properties of nanostructured materials. As the development of $C_s$-corrected TEM, the direct analysis of atomic structures of nanostructured materials can be performed in the high-resolution transmission electron microscopy (HRTEM). Especially, fast Fourier transform (FFT) technique in image processing is very useful way to determine the crystal structure of HRTEM images in reciprocal space. To apply FFT technique in HRTEM analysis in more reasonable and friendly manner, we made a new circular region of interest (C-ROI) FFT script and tested it for several HRTEM analysis. Consequentially, it was proved that the new FFT application shows more quantitative and clearer results than conventional FFT script by removing the streaky artifacts in FFT pattern images. Finally, it is expected that the new FFT script gives great advantages for quantitative interpretation of HRTEM images of many nanostructured materials.

Reliability Test of the TEM Rotation Holder for 3-D Structure Analysis (3차원적 구조분석을 위한 TEM Rotation Holder의 신뢰도 점검)

  • Accuracy and precision of the goniometer and the specimen holder should be measured and corrected to improve reliability of 3-D structure analysis using transmission electron microscopy (TEM). In this study, we described the operation principle and performance of the Gatan rotation holder. Through analysis of the images taken inside the microscope, rotation angles were measured within the accuracy of ${\pm}0.42^{\circ}$. For comparison the rotation angles were measured outside the microscope using a home-made measurement tool, which resulted in the accuracy of ${\pm}0.6^{\circ}$. Additionally, we found abnormal specimen drifts during rotation probably due to the unstable engagement between the specimen cup and the rotation belt.

  • The multi-scale analysis is more proper and precise for composite materials because of considering the individual microscopic structure and properties of each material for composite materials. The purpose of this study is to verify the validity of using palladium particles in carbon/fiber composites by multi-scale analysis. The palladium is a material for itself to detect leaking hydrogen by using the property of adsorbing hydrogen. The macroscopic model material properties used in this study are homogeneous material properties from microstructure. Homogenized material properties that are calculated from periodic boundary conditions in the microscopic representative volume element model of each macroscopic analysis model. In this study, three macroscopic models were used : carbon fiber/epoxy, carbon fiber/palladium, palladium/epoxy. As a result, adding palladium to carbon/epoxy composite is not a problem in terms of strength.

Open Kyphoplasty Combined with Microscopic Decompression for the Osteoporotic Burst Fracture

  • Objective : The purpose of this retrospective clinical study was to describe a treatment for osteoporotic burst fracture in the setting of severe fractures involving fragmentation of the posterior wall and neural compromise with symptoms of cord compression. Methods : Indication for microscopic decompression and open kyphoplasty were intractable pain at the level of a known osteoporotic burst fractures involving neural compression or posterior wall fragmentation. A total of 18 patients [mean age, 74.6 years] with osteoporotic thoracolumbar burst fractures [3 males, 15 females] were included in this study. In all cases, microscopic decompressive laminectomy was followed by open kyphoplasty. Clinical outcome using VAS score and modified MacNab's grade was assessed on last clinical follow up [mean 6.7 months]. Radiological analysis of sagittal alignment was assessed preoperatively, immediately postoperatively, and at final follow up. Results : One level augmentation and 1.8 level microscopic decompression were performed. Mean blood loss was less than 100 ml and there were no major complications. The mean pain score before operation and at final follow up was 7.2 and 1.9, respectively. Fourteen of 18 patients were graded as excellent and good according to the modified MacNab's criteria. Overall, 6.0 degrees of sagittal correction was obtained at final follow-up. Conclusion : The combined thoracolumbar microscopic decompression and open kyphoplasty for severe osteoporotic fractures involving fragmentation of posterior wall and neural compromise provide direct visualization of neural elements, allowing safe cement augmentation of burst fractures. Decompressive surgery is possible and risk of epidural cement leakage is controlled intraoperatively.

A comparison of Modified One Layer Vasovasostomy with Optical Loupe and Microscope (Optical Loupe와 현미경을 이용한 단층정관정관문합술의 비교)

  • Objective: The objective of this study was to compare results of the macroscopic one-layer vasovasostomy with those of microscopic one-layer vasovasostomy and to analyze the change of semen parameters according to the interval of vasal obstruction. Method and Materials: Between March 1987 and December 1997, we performed 121 vasovasostomies using modified one-layer technique with loupe magnification (macroscopic vasovasostomy) or microscope. Among the 68 could be followed post-operatively, 37 patients were treated by macroscopic technique with loupe, and 31 patients by microscopic technique. We compared rates of anatomical patency (sperm count above than $10{\times}10^{6}$/ml) and pregnancy of macroscopic vasovasotomy with those of microscopic vasovasostomy. Patency and pregnancy rates according to vasal obstructive interval were also examined. Results: The patency rates of macroscopic and microscopic technique were 86.5% and 87.1%, and pregnancy rates of macroscopic and microscopic technique were 64.9% and 67.7%. There was no statistical significance between these two methods (p>0.05). The pregnancy rates and sperm motility were decreased if more than 10 years had elapsed following vasectomy (p<0.05). Conclusion: We found little difference in success rates resulting from macroscopic and microscopic vasovasostomy and the former had the advantage of reduced cost and a lower operator skill level. In post-operative semen analysis, the sperm motility was the most probable factor associated with vasal obstructive interval.
