Evaluation of Optimum Conditions for Bacteriocin Production from Lactobacillus sp. JB-42 Isolated from Kimchi

  • Bacteriocin-producing microorganism was isolated from Kimchi. The microorganism was identified as a Lactobacillus sp. and named Lactobacillus sp. JB-42. The optimum conditions for the bacteriocin production from the isolated microorganism were evaluated. For the maximum yield of bacteriocin from Lactobacillus sp. JB-42, the cell should be harvested at the early stationary phase and the temperature, pH and NaCl concentration should be $30^{\circ}C$, pH 7 and without the addition of NaCl, respectively. Sucrose, glucose or fructose should be used as a carbon source and organic nitrogen such as tryptone should be used as a nitrogen source for the best yield. The production of bacteriocin was related to the cell growth of Lactobacillus sp. JB-42 indicating the role of Lactobacillus in the Kimchi fermentation process.

  • Purpose: This study is a repetitive comparative analysis of a qualitative case study that carried out a home visiting oral health education program. Method: This study conducted an interview survey through medical examination by interview before and after the home visiting oral health education and measured some pathogenic microorganism counts in the denture. Results: There was a positive change in the self oral care ability of the elderly at home after home visiting oral health education, including the behavior of self management of dentures, and some pathogenic microorganism counts in the dentures. Conclusion: The home visiting oral health education of home care service centers can improve oral health care for the elderly at home.

Storage of Waste-Brown Seaweed and Degradation of Alginate Using Microorganism (미생물을 이용한 미역폐기물의 저장 및 알긴산염 저분자화)

  • We studied a storage of waste-brown seaweed at room temperature and degradation of alginate in seaweed by microorganism DS-02. The seaweeds, mixed with 5.0 wt% DS-02 and sealed in vinyl package without any other treatment, could be stored longer than 1 year without spoilage at room temperature. During the storage process, the alginate of seaweed was decomposed by enzyme of DS-02 and the molecular weight of alginate decreased to about 1/10 of initial quantity. DS-02 growed as fast as it had maximum weight after 24 hour culture and it's enzyme had a maximum activity of alginate degradation at $40^{\circ}C.$ The seaweed sample became particles in DS-02 culture solution and the M. W of alginate decreased to about 1/10 of initial value after 24 hour decomposition. The effect of alginate degradation with DS-02 was similar to that of degradation with 3.0 M HCI solution for 24 hour.

  • Some ocular complications were connected to the contamination by the contact lens related. Especially the most cause of ocular complications was a microorganism from several processing of infections. Therefore the patients stopped the contact lens wear immediately by the side effects of ocular pathology. We had analyzed the microorganism from the laboratory. It was identified as mixed growth of the Chryseamonas meningosepticum and Enterobacter sp.

  • Cytotoxic test was performed by SRB assay on human epidermoid carcinoma HEp-2 cell line for screening the soil microorganism, secreting anticancer agent. One microorganism was selected among two thousand microorganisms for its highest cytotoxicity. And this microorganism was identified with Streptomyces species after performing of diaminopimeric acid and reducing sugar analysis of cell wall and analysing the cultural characteristics and named Streptomyces sp. SM 1119. The effect of anticancer agent in SM 1119 culture extract on the cell cycle was studied by using GG$_{o}$ synchronized NIH 3T3 cells. The extract inhibited the serum stimulation of GG$_{o}$ NIH 3T3 cell only within 1 hour after serum stimulation but not after 6 hours. The extract also reduced the amount of c-myc mRNA in Colo 320 cell. These results suggest that the anticancer agent in the extract inhibits the progression of cell cycle very early stages, probably from G$_{0}$ to G$_{1}$.

  • An optical sensing membrane has been fabricated to measure the concentration of dissolved oxygen(DO) and pH value simultaneously. It has employed HPTS as a pH sensitive dye and a ruthenium(II) complex as a DO sensitive dye. The sensing membrane has been applied to wells in a 24-well microtiter plate. Using the 24-well microtiter plate the concentrations of dissolved oxygen and pH values have been on-line monitored during the cultivations of E.coli DH5${\alpha}$, B.cereus 318 and P.pastoris X-33. On-line monitoring of DO and pH in microorganism cultivation processes showed good performance of the sensing membrane containing 5 mM HPTS and 2 or 5 mg/mL Rudpp.

Biological Control of Fusarium Wilt of Carnation Plants by Antagonistic Microorganism in Greenhouse (카네이션의 시설재배에서 길항성 세균을 이용한 Fusarium Wilt 의 생물학적 방제)

  • This study was carried out to screen and select the effects of antifungal bacterial strains which inhibit the growth of plant pathogen, Fusarium oxysporum(fusarium wilt) occurred in carnation plants in greenhouse. We isolated an effective bacterial strains and investigated into the antifungal activity of the antagonistic microorganism and it's identification. Twenty bacterial strains which strongly inhibited Fusarium oxysporum were isolated from roots of carnation plants and the soil in greenhouse, and the best antifungal bacteria designated as C121, was finally selected. Antagonistic bacterial strain, C121 was identified to be the genus Bacillus sp. based on the morphological, biochemical and cultural characterizations. The Bacillus sp. C121 showed 58.1% of antifungal activity against the growth of Fusarium oxysporum. By the bacterialization of the cultural broth and the heat bacterialization culture filtrate of it, Bacillus sp. C121 was shown 92.1% and 21.0% of antifungal activity, respectively.

Monitoring of Indicator Microorganism Concentrations of River Sediment and Surface Water in the Geum River Basin (금강 수계 내 하천퇴적물 및 지표수의 지표미생물 농도분포)

  • Characterization of sediment quality is important for the proper management of surface water quality, yet sediment has not been monitored sufficiently. In this study, fecal indicator microorganism concentrations of sediments in the Geum River Basin were monitored. Sampling was carried out at one paddy field, one lakeshore and five monitoring stations in the lower reach of the Geum River Basin. Surface waters and sediments were sampled four times during rainy season. Total coliform concentrations of sediments were 12 times higher in average to those of surface waters while E. coli concentrations of sediments were six times higher. No correlation found between indicator microorganism concentration between surface waters and sediments.

Biological Control of Fusarium Wilt of Tomato Plants by Antagonistic Microorganism in Greenhouse (온실재배 토마토에서 발생하는 위조병의 미생물학적 제어)

  • This study was conducted to screen the antagonistic bacteria which inhibit the growth of plant pathogen, fusarium wilt(Fusarium oxysporum) occurred in tomato plants in greenhouse. Ten bacterial strains which strongly inhibited Fusarium oxysporum were isolated from the nature, and the best antagonistic bacterial strain designated as KC175, was selected. The antagonistic strain KC175 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. The Bacillus sp. KC175 showed 58.2% of antifungal activity against the growth of Fusarium oxysporum. By the bacterialization of the culture broth and the heat bacterialization culture filtrate of it, Bacillus sp. KC175 showed 91% and 18% of antifungal activity, respectively.

  • The purpose of this study was to evaluate the growth of lettuce last for 36 days. Lettuce planted on the same medium with different treatment. The first treatment watering using bio-fertilizer (farmer) and second watering using effective microorganism. Lettuce grown in greenhouse with temperature $18-32^{\circ}C$.Characteristic observed that growth in height and weight of the plants. From the observations that can be generated at high observation, treatment 1 had a higher than treatment 2. On the last day treatment plant with treatment 1 had a 99.8 mm high while treatment 2 has height of 90.7 mm. Observation on severe treatment 1 has a heavier weight than treatment 2. Plants with treatment 1 have a weight on the last day of 0.15665 grams while in plants with treatment 2 weights are 0.11365 grams.
