Quinone profile를 이용한 하천생태계의 미생물군집구조 해석 (Analysis of Microbial Community Structure in River Ecosystem Using Quinone Profiles)

  • The differences in microbial community structures between planktonic microorganism and biofilm in rivers were investigated using respiratory quinone profiles. The compositions of microbial quinone for 4 tributaries of the Kyongan Stream located in/flowing through Yongin City, Gyeonggi-Do were analyzed. Ubiquinone(UQ)-8, UQ-9, menaquinone(MK)-6 and Plastoquinone(PQ)-9 were observed in all samples of planktonic microorganism and biofilm for the sites investigated, Most planktonic microorganism and biofilm had UQ-8(15 to 30%) and PQ-9(over 30%) as the dominant quinone type. These results indicated that oxygenic phototrophic microbes(cyanobacteria and/or eukaryotic phytoplankton) and UQ-8 containing proteobacteria constituted major microbial populations in the river. The quinone concentration in the river waters tested, which reflects the concentration of planktonic microorganisms, increases with increasing DOC. Further research into this is required. The microbial diversities of planktonic microorganism and biofilm calculated based on the composition of all quinones were in the range from 4.2 to 7.5, which was lower than those for activated sludge(ranging from 11 to 14.8) and soils(ranging from 13.4 to 16.8). The use of quinone profile appears to be a useful tool for the analysis of microbial community structure in river.

Influences of New Azo Dyes to the Aquatic Ecosystem

  • The influences of a series of new azo direct dyes including copper-complexes based on benzidine congeners, 2,2'-dimethyl-5,5'-dipropoxybenzidine and 5,5'-dipropoxybenzidine, were examined using microorganism, Daphnia magna. The purpose of the research described in this paper was to use bioassays with daphnids to determine the aquatic toxicity of new azo dyes in which copper was incorporated. The results clearly show that copper has negative effects to aquatic ecosystem as expected. The study also suggested that the assay with Daphnia magna was an excellent method to evaluate the influences of dyes to the aquatic environment.

Temporal and Spatial Change in Microbial Diversity in New-developed Wetland Soil Covered by Tamarix chinesis Community in Chinese Yellow River Delta

  • Soil samples were collected from new-developed wetland soil ecosystem of Tamarix chinesis plantation in Chinese Yellow River Delta in different months of 2003. Soil characteristics, temporal change and spatial distribution of microbial community composition and their relationship with nitrogen turnover and circling were investigated in order to analyze and characterize the role of microbial diversity and functioning in the specific soil ecosystem. The result showed that the total population of microbial community in the studied soil was considerably low, compared with common natural ecosystem. The amount of microorganism followed as the order: bacteria> actinomycetes>fungi. Amount of actinomycetes were higher by far than that of fungi. Microbial population remarkably varied in different months. Microbial population of three species in top horizon was corrected to that in deep horizon. Obvious rhizosphere effect was observed and microbial population was significantly higher in rhizosphere than other soils due to vegetation growth, root exudation, and cumulative dead fine roots. Our results demonstrate that microbial diversity is low, while is dominated by specific community in the wetland ecosystem of Tamarix chinesi.

Environmental toxicity and biodegradation of Pseudomonas sp. EL-G527 producing biosurfactant

  • A biosurfactant-producing microorganism, .Pseudomouos sp. EL-G527 was isolated from activated sludge by enrichment culture when grown on mineral salt medium containing n-hexadecane as a carbon source. The biosurfactant from .Pseudomonar sp. EL-G527 exhibited lesser toxicity to bacterial population than synthetic surfactants and in the biodegradation test, biosurfactant was rapidly degraded and lost its activity as surface active material after 1 day incubation. In this study, the biosurfactant from Pseudomonas sp. EL-G527 was effective surface-active compound, more biodegradable and less toxic to microbial ecosystem than various synthetic surfactants.

Prospects of Activated Sludge Process in Japan - Its Past, Present, and Future -

  • Our life totally depends on activated sludgeprocess for treatment of wastewater: sewage and industrial wastewater. Activated sludge process was the epoch-making technology in Environmental field. One century has been almost passed since the process was developed in England, and the process is still on the development of improvement. Here, history of activated sludge process, its mechanismsof treating the wastewater, expectations that we had on the process in the past, and future image and possibility on the process were presented. By reviewing the events related to the process, we can foresee potentials for new possibility of activated sludge process.

토양에 투입된 미생물의 거동 및 활성 (Fate and Activity of Microorganism introduced into the Soil)

  • 농작물에 영양분을 공급하거나 식물의 생장촉진, 식물 병원균의 억제 등을 위하여 토양에 미생물이 투입된다. 대표적인 예가 질소고정 공생세균과 Pythium, Rhizobium 등이다. 이들이 투입되어 성공적으로 작용하려면 생존하여 충분한 밀도로 집략을 형성하여야 한다. 이들의 생존과 집략화에 미치는 영향은 생물적인 인자와 비생물적 인자에 의해 좌우된다. 생물적 인자중에서 중요한 것은 포식과 경쟁, 비생물적 인자 중에서 중요한 것은 물의 장력 유기탄소, 무기영양분(N, P), pH 등이다. 토양의 토성과 소공극 분포도 투입된 미생물의 생존과 활착에 중요한 역할을 담당한다. 또한 미생물에 대한 토양생태계의 선택성은 접종 세포 개체군의 활착에 있어서 중요하다. 예를 들면 항생제의 사용에 의한 토착미생물의 제어, 계면활성제를 분해하는 Psendomonas종을 투입하는 경우이다. 투입미생물의 활착을 증진시키기 위하여 이탄, 점토 등의 수송체를 동시에 첨가할 수 있는데 이들은 투입미생물에게 보호적인 미소서식지를 만듦으로서 토양중에서 세균을 보호할 수 있다. 또한 세균이나 공업용 효소를 부동화시키기 위한 수송체로서 유기성 중합체가 사용되고 있다. 이러한 물질들의 예를 들면 아질산칼륨, agarose, k-carrageenan 등이다. 이들도 미소서식처를 제공하므로서 투입미생물의 활착을 돕는 역할을 한다.

미생물혼합제제 처리가 토양의 미생물상과 화학적 특성 및 시설 채소 생육에 미치는 영향 (Effect of Microorganism Mixture Application on the Microflora and the Chemical Properties of Soil and the Growth of Vegetables in Greenhouse)

  • BACKGROUND: The urgency of feeding the world's growing population while combating soil pollution, salinization and desertification requires suitable biotechnology not only to improve crop productivity but also to improve soil health through interactions of soil nutrient and soil microorganism. Interest in the utilization of microbial fertilizer has increased. A principle of nature farming is to produce abundant and healthy crops without using chemical fertilizer and pesticides, and without interrupting the natural ecosystem. Beneficial microorganisms may provide supplemental nutrients in the soil, promote crop growth, and enhance plant resistance against pathogenic microorganisms. We mixed beneficial microorganisms such as Bacillus sp. Han-5 with anti-fungal activities, Trichoderma harziaum, Trichoderma longibrachiatum with organic material degrading activity, Actinomycetes bovis with antibiotic production and Pseudomonas sp. with nitrogen fixation. This study was carried out to investigate the mixtures on the soil microflora and soil chemical properties and the effect on the growth of lettuce and cucumber under greenhouse conditions. METHODS AND RESULTS: The microbial mixtures were used with each of organic fertilizer, swine manure and organic+swine manure and compared in regard to changes in soil chemical properties, soil microflora properties and crop growth. At 50 days after the treatment of microorganism mixtures, the pH improved from 5.8 to 6.3, and the EC, $NO_3$-Na and K decreased by 52.4%, 60.5% and 29.3%, respectively. The available $P_2O_5$ and $SiO_2$ increased by 25.9% and 21.2%, respectively. Otherwise, the population density of fluorescent Pseudomonas sp. was accelerated and the growth of vegetables increased. Moreover, the population density of E. coli and Fusarium sp., decreased remarkably. The ratio of bacteria to fungi (B/F) and the ratio of Actinomycetes bovis to fungi (A/F) increased 2.3 (from 272.2 to 624.4) and 1.7 times (from 38.3 to 64), respectively. Furthermore, the growth and yield of cucumber and lettuce significantly increased by the treatment of microorganism mixtures. CONCLUSION(S): These results suggest that the treatment of microorganism mixtures improved the chemical properties and the microflora of soil and the crop growth. Therefore, it is concluded that the microorganism mixtures could be good alternative soil amendments to restore soil nutrients and soil microflora.

海岸 鹽濕地 生態系의 에너지 流轉 (Energy Flow in a Coastal Salt Marsh Ecosystem)

  • Energy flow through the trophic levels was studied at a salt marsh ecosystem distinguished into low and high marsh. Gross primary productions of Suaedeto-Salicornietum and Artemisieto-Limonietum at low marsh were 8, 299 and 13, 154kca/$m^2$/yr, and those of Calama-grostetum and Sonchuso-Setaetum at high marsh were 17, 899 and 15, 177kca/$m^2$/yr, respectively. Efficiencies of solar energy utilization of plants were 1.7 and 2.6% at the former, and were 3.6 and 3.2% at the latter. Of gross productions, net primary productions were 3, 977 and 5, 280kca/m2/yr at low marsh and were 6, 354 and 5, 329kca/$m^2$/yr at high marsh, and the remainder, 52~67%, was consumed by respiration of plants. A small amount (0.03~0.04%) of the net primary production was flowed through grazing food chain and most amout was transferred into dead parts. Of dead parts, 40% was accuulated as litter and the rest was decomposed into detritus. In the detritus food chain, a little energy was utilized by detritus feeder, and a major by microorganism. The amounts of energy flowed through grazing and detritus feeders at high marsh were much more than those at low marsh, but tertary production as spider was Vice versa.

해양 자유생활형 선충류 Enoplolaimus sp. (Enoplida: Thoracostomopsidae)와 Bathylaimus sp. (Enoplida: Tripyloididae)의 배양용 적합배지 선정 및 초기 생활사 연구 (Sutdy of Appropriate Media Selection and Early Life Cycle of Marine Free-Living Nematodes, Enoplolaimus sp. (Enoplida: Thoracostomopsidae) and Bathylaimus sp. (Enoplida: Tripyloididae))

  • 실험실 내에서의 해양 선충류 최적 배양 조건을 찾기 위해 한천배지로 다양한 배지를 제작하여 선충류를 대상으로 한 연구를 수행하였다. 선충류 채집은 태안 만리포 조간대 중 하부의 사질 퇴적물 표층에서부터 약 5 cm의 퇴적물에서 이루어졌다. 배양에 이용한 한천 배지는 Killian 배지의 조성을 다소 변형하고, 한천 농도에 차이를 두었으며, 이 한천배지들을 직경 60 mm 페트리디쉬에 배지 높이가 2.0 mm가 되도록 분주하였다. 선충류의 먹이 생물로서 순수 배양한 미생물 5종을 혼합하여 각각의 배지에 첨가하였다. 각 배지에 Enoplolaimus sp. 5개체를 접종하여 $20^{\circ}C$, 암배양으로 설정된 배양기 내에서 배양하였다. Enoplolaimus sp.에 대하여 여러 조건의 실험을 반복 수행한 결과, Killian 배지에 소고기 육즙만을 첨가한 배지의 한천농도 1.0%에서 평균 생존시간이 246.8시간으로 가장 높은 생존율을 나타냈다. 가장 낮은 생존시간은 Killian 배지에 bacto peptone과 소고기 육즙(beef extract)을 첨가한 한천농도 0.4%의 먹이 공급을 한 배지에서 평균 27.6시간으로 가장 낮은 생존시간을 나타냈다. Bathylaimus sp.는 Killian 배지 한천농도 1.0%의 먹이 공급을 하지 않은 배지에서 가장 높은 생존시간을 나타내었고, 산란량과 부화율 실험에서는 평균 7개의 알을 산란하였으며 평균 99.5시간 후에 94.7%의 부화율을 보이는 결과를 나타냈다

일본 가가와현 산불피해지의 복구대책 (The Restoration of Forest Fire Area in Kagawa Prefecture, Japan)

  • The forest seemed apparently to die on the forest fire area in Honjima, Kagawa Prefecture, Japan. However, the soil that became growing basic of vegetation hardly suffered damage, and the forest recovery was started by the sprout, etc. in the next year. For restoration of forest fire area, the fascine mulching works and log barrier works using the damaged trees were used for the upper-stream, and chack dam and erosion control dam were set up in the downstream. Also, the forest restoration was tried with the plants and the microorganism that inhabit in Honjima to preserve a peculiar forest ecosystem.
