An experiment was carried out to assess the effects of xylanase supplementation on the performance, net energy and gut microflora of broilers fed a wheat-based diet. Day-old male broiler chicks were allocated to two dietary treatments. Each treatment was composed of six replicate cages of seven broilers per cage. The diets were wheat-based and offered as mash. The treatments included i) basal diet deficient in metabolizable energy; and ii) basal diet supplemented with a commercial xylanase added at 4,000 U/kg feed. Bird performance, nutrient utilization and gut microbial populations were measured. Heat production and net energy were determined using an auto-control, open circuit respiration calorimetry apparatus. Results showed that exogenous xylanase supplementation improved feed conversion efficiency (p<0.05) and increased diet AME (+4.2%; p<0.05), as well as heat production (HP), net energy for production (NEp), production of $CO_2$, and consumption of $O_2$. The respiratory quotient (RQ) was also increased (p<0.01) by the addition of xylanase. NEp value was increased by 26.1% while daily heat production per kg metabolizable body weight was decreased by 26.2% when the xylanase was added. Xylanase supplementation numerically increased the ileal digestibility of protein and energy by 3 and 6 percentage units respectively (p>0.05). The ileal digestibility of hemicellulose was significantly improved by xylanase addition (p<0.05).
Two experiments were conducted to evaluate the dietary supplemental fruit extract(Bio-plus) on performance and intestinal microflora of broiler chicks. Seven treatments were allocated into control, 0.05, 0.10, 0.20, 0.10, 0.20, 0.40, 0.80, 1.60% with four replications for five weeks, in both experiments. Diets contained 21.5% CP, 3, 100 kcal/kg ME for the rest two weeks. Weight gain, feed intake, feed conversion ratio(FCR) were weekly measured. Birds were sacrificed to examine the E. coli and Salmonella from cecum and ileum at the end of experiment. In experiment 1, Weight gain of Bio plus treatments tended to increase compared to that of control, but was not consistency between them. Chicks fed Bio-plus showed significantly lower feed conversion ratio(FCR) than control(P〈0.05). In experiment 2, Weight gain of Bio plus treatments was significantly different from control(P〈0.05) for starting period and linearly increased upto 0.80% for the rest of two weeks, but was not significantly different. Feed intake of chicks fed Bio-plus tended to be higher than control. Bio-plus treatments was not statistically different FCR from control. ND titer showed no significance, but tended to be higher in Bio-plus treatments than control. The number of ileum salmonella was significantly decreased in Bio-plus treatments compared to that of control in expt 2 (P〈0.05). Total yeast and Lactobacillus spp. of illeum tended to increase in expt 1 and were significantly higher in expt 2 than control(P〈0.05). The results of these experiment indicated that dietary Bio-plus supplements increased weight gain, the number of ileum yeast and Lactobacillus spp. of broiler chicks.
Various bacterial strains were isolated from water in a paddy field, and their seaweed degrading activities were investigated. As the result, 16 strains were obtained from the microflora sample. They were incubated in a liquid medium of sea tangle powder for 3 weeks. Ratios of reduced sugar to total sugar of the liquid medium were measured once a week during the incubation period. Ratio of reduced sugar to total sugar of 30A412 strain was highest. Accordingly, 30A412 strain was incubated in three different liquid media of sodium alginate, sea tangle powder, and sea mustard powder for 3 or 4 weeks. The ratios of reduced sugar to total sugar and cell growth were measured once a week during the incubation period. Ratios of reduced sugar to total sugar and cell growth were increased with the incubation period. Cell growth and degrading ratio were highest in the liquid medium with sea mustard powder.
Baicalin, a main constituent of the rhizome of Scutellaria baicalensis, is metabolized to baicalein and oroxylin A in the intestine before its absorption. To understand the role of intestinal microflora in the pharmacological activities of baicalin, we investigated its anti-inflammatory effect in mice treated with and without antibiotics. Orally administered baicalin showed the anti-inflammatory effect in mice than intraperitoneally treated one, apart from intraperitoneally administered its metabolites, baicalein and oroxylin A, which potently inhibited LPS-induced inflammation. Of these metabolites, oroxylin A showed more potent anti-inflammatory effect. However, treatment with the mixture of cefadroxil, oxytetracycline and erythromycin (COE) significantly attenuated the anti-inflammatory effect of orally administered baicalin in mice. Treatment with COE also reduced intestinal bacterial fecal ${\beta}$-glucuronidase activity. The metabolic activity of human stools is significantly different between individuals, but neither between ages nor between male and female. Baicalin was metabolized to baicalein and oroxylin A, with metabolic activities of $1.427{\pm}0.818$ and $1.025{\pm}0.603$ pmol/min/mg wet weight, respectively. Baicalin and its metabolites also inhibited the expression of pro-inflammatory cytokines, TNF-${\alpha}$ and IL-$1{\beta}$, and the activation of NF-${\kappa}B$B in LPS-stimulated peritoneal macrophages. Of them, oroxylin A showed the most potent inhibition. Based on these findings, baicalin may be metabolized to baicalein and oroxylin A by intestinal microflora, which enhance its anti-inflammatory effect by inhibiting NF-${\kappa}B$ activation.
In the prevention of root surface caries, antimicrobial therapy for the control of subgingival and supragingival plaque is seriously considered as a long term suppression of pathogenic microflora. Recently, varnishes containing antimicrobial agents have been developed to control the supragingival microflora. The purpose of this study was to determine the antimicrobial effects of 20% chlorhexidine varnish and 2.6% silane fluoride varnish with sealant. In clinical experiments, 12 subjects were selected from the periodontally treated patient and divided into 3 groups. After a dental prophylaxis, the subjects were treated with single application of placebo varnish (group I), 20% chlorhexidine varnish (group II), and 2.6% silane fluoride varnish (group III). Root surface plaque samples were taken before (baseline) and one, two, four, and 8 weeks after the treatments. Microbiological examinations of root surface plaque were performed with culture study and indirect immunoflorescence (I.I.F.) study, and immunological examination of gingival crevicular fluid antibody titers was performed with ELISA study. The results were as follows: 1. Pathogenic microflora on the root surface including S. mutans, S. sanguis, S. mitis, A. naeslundii, A. viscosus were 24 - 37% on I.I.F. study. 2. S. mutans, S. sanguis, S. mitis, A. naeslundii, A. viscosus of the root surface plaque was significantly reduced from 1 week to 8 weeks after antimicrobial varnish treatment, but showed generally increasing tendency in control group. 3. Gingival crevicular fluid antibody titers were significantly reduced from 1 or 2 weeks to 4 weeks after antimicrobial varnish treatment.
Biohydrogen production from organic wastewater by anaerobically activated sludge fermentation has already been extensively investigated, and it is known that hydrogen can be produced by glucose fermentation through three metabolic pathways, including the oxidative decarboxylation of pyruvic acid to acetyl-CoA, oxidation of NADH to $NAD^+$, and acetogenesis by hydrogen-producing acetogens. However, the exact or dominant pathways of hydrogen production in the anaerobically activated sludge fermentation process have not yet been identified. Thus, a continuous stirred-tank reactor (CSTR) was introduced and a specifically acclimated acidogenic fermentative microflora obtained under certain operation conditions. The hydrogen production activity and potential hydrogen-producing pathways in the acidogenic fermentative microflora were then investigated using batch cultures in Erlenmeyer flasks with a working volume of 500 ml. Based on an initial glucose concentration of 10 g/l, pH 6.0, and a biomass of 1.01 g/l of a mixed liquid volatile suspended solid (MLVSS), 247.7 ml of hydrogen was obtained after a 68 h cultivation period at $35{\pm}1^{\circ}C$. Further tests indicated that 69% of the hydrogen was produced from the oxidative decarboxylation of pyruvic acid, whereas the remaining 31% was from the oxidation of NADH to $NAD^+$. There were no hydrogen-producing acetogens or they were unable to work effectively in the anaerobically activated sludge with a hydraulic retention time (HRT) of less than 8 h.

In order to improve funtionality of kochujang, red ginseng powder(1, 2 and 5% red ginseng on the total weight basis) was added to the raw material of kochujang. Changes in moisture content, microflora and enzyme activities were investigated in control kochujang(Sunchang kochujang) and red ginseng added kochujang during fermentation at 25oC for 120 days. Kochujang samples fremented for 90 and 120 days were subjected to sensory evaluation test. As the fermentation progressed, the moisture content and titratable acidity were gradually increased whereas pH was decreased. However, red ginseng had no effects on those changes. Bacterial count was maintained at the level of 107 CFU/g during fermentation. The level of yeast count was 104 CFU/g at the early stage of fermentation and increaed to 107 CFU/g after 60 days of fermentation and then maintained at 107 CFU/g. Red-ginseng had no effects on bacterial and yeast counts of kochujang. Amylase activity was gradually reduced during fermentation. Amylase activity was increased until 60 days of fermentation and then reduced by 90 days of fermentation. Acidic protease activity was slightly reduced at the initial stage of fermentation and then rapid increased. Acidic protease activity was at the highest level after 120 days of fermentation. Red ginseng did not alter those enzyme activities of kochujang. There were no differences in sensory properties between kochujang samples fermented for 90 and 120 days of fermentation. The addition of red-ginseng did not affect such sensory properties of kochujang as color, odor, hot taste and salty taste, but overall preference appeared to be improved by red ginseng addition.

Fermented Rhus verniciflua (FRV) as feed additives act as excellent anti-diarrheal drugs as they increase the intestinal absorption rate therefore being indirectly associated with enhancing growth performance and increasing digestibility in livestock. A total of 80 weaned pigs with an average initial body weight (BW) of $6.82{\pm}1.11kg$ were used to evaluate a diet supplemented with FRV meal in a 6 week feeding trial with two dietary treatments [CON - basal diet; TRT - CON + 0.2% FRV] on growth performance, apparent total tract digestibility (ATTD), blood profiles, fecal microflora, and fecal score. Our results showed that the dietary supplementation of FRV improved (p < 0.05) average daily gain and gain : feed (G : F) ratio during days 15 - 42 and the overall experiment period and also increased the ATTD of dry matter (p < 0.05) at days 14 and 42. On the contrary, there was no effect (p > 0.05) on average daily feed intake, ATTD of nitrogen and energy, and blood profiles during the entire experiment. Moreover, dietary inclusion of FRV significantly increased fecal Lactobacillus (p < 0.05) counts and reduced the diarrhea during days 22 - 42. Thus, the results suggest that FRV can be used as a potential additive to improve growth performance and dry matter and to reduce diarrhea while having beneficial effects on fecal microflora in weanling pigs.
The diversity of microflora according to growth stage of Pleurotus ostreatus and the correlation between microbe and medium fermentation were investigated. In farmhouse I, the aerobic bacteria and fungi as longer of growing period were increased. And, thermophilic bacteria and fluorescent Pseudomonas sp. showed high density at the early stage of spawn inoculation. The thermophilic actinomycetes were distributed evenly during all the growing period, but mesophilic actinomycetes were not observed. In farmhouse II, thermophilic actinomycetes were not observed in fermented medium and density of fungi were suddenly increased at 60 days after spawn inoculation. And also, mushrooms can hardly be harvested due to Penicillium spp. After medium fermentation, density of aerobic bacteria, thermophilic bacteria, and fluorescent Pseudomonas sp. was higher at farmhouse I than those of farmhouse II. In farmhouse I, Bacillus sp. and Pseudomonas sp. dominated at early stage of mushroom growth but as time goes by, density of Bacillus sp. was higher than the others. And also, the kind of microbe showed a few at early stage of mushroom growth but increased as time goes by. In farmhouse II, Bacillus sp. was dominated at early stage of mushroom growth. And the growth of Bacillus sp. and Pseudomonas sp. showed intersect aspect each other in the farmhouse I but Bacillus sp. dominated during all growth periods in the farmhouse II.

A variety of nuruk were collected from various provinces in Korea, and their microflora profiles were analyzed at the species level. A total of 42 nuruk samples were collected and when the viable cell numbers in these nuruk were enumerated, the average cell numbers of bacteria, fungi, yeast, and lactic acid bacteria from all nuruk were 7.21, 7.91, 3.49, and 4.88 log CFU/10 g, respectively. There were no significant differences in viable cell numbers of bacteria or fungi according to regions collected. Bacillus amyloliquefaciens and B. subtilis were the predominant bacterial strains in most samples. A significant portion, 13 out of 42 nuruk, contained foodborne pathogens such as B. cereus or Cronobacter sakazakii. There were various species of lactic acid bacteria such as Enterococcus faecium and Pediococcus pentosaceus in nuruk. It was unexpectedly found that only 13 among the 42 nuruk samples contained Aspergillus oryzae, the representative saccharifying fungi in makgeolli, whereas a fungi Lichtheimia corymbifera was widely distributed in nuruk. It was also found that Pichia jadinii was the predominant yeast strain in most nuruk, but the representative alcohol fermentation strain, Saccharomyces cerevisiae, was isolated from only 18 out of the 42 nuruk. These results suggested that a variety of species of fungi and yeast were distributed in nuruk and involved in the fermentation of makgeolli. In this study, a total of 64 bacterial species, 39 fugal species, and 15 yeast species were identified from nuruk. Among these strains, 37 bacterial species, 20 fungal species, and 8 yeast species were distributed less than 0.1%.
