본 연구에서는 에어컨 내에서 서식하는 세균을 분리하여 동정하였고 이들 에어컨에 대한 위생 관리에 대하여 조사하였다. 20개의 에어컨 냉각기에서 8종의 세균이 분리, 동정되었는데 그것들은 가장 많이 분리 동정된 Pantoea sp.를 비롯하여 Bacillus circulans, Bacillus pumilus, Corynebacterium, Flavimonas oryzihabitans, Ochrobacterum anthropi, Micrococcus sp., non fermented bacilli (NFB)이었다. 한편 에어컨 사용자 1,322명을 대상으로 에어컨의 위생관리 상태에 대하여 조사하였는데 설문자의 1,138명(86%)이 에어컨 사용시 환기를 하는 것으로 나타났다. 에어컨 청소에 있어서 1,128명(85%)이 청소를 하는 것으로 나타났지만 에어컨의 오염원들이 되는 에어컨의 필터 청소는 864명(66%)이 하지 않는 것으로 나타났고 에어컨의 열교환기는 1089명(82%)이 하지 않는 것으로 나타났다. 이런 결과로 상기에서 언급된 세균들과 같은 에어컨 오염원이 증가하면 호흡기 질환과 같은 인간의 질병을 초래할 수 있다는 것을 알 수 있었고 따라서 에어컨 사용시 에어컨의 오염원제거와 위생관리는 에어컨내 세균으로부터 발생되는 인간의 질병을 예방할 수 있을 것이다.
산업화의 영향으로 발생한 토양 지하수 내 중금속 및 유류 오염정화를 위해 다양한 시도가 이루어지고 있다. 그러나 미세토양이 상대적으로 많이 포함되어 있는 오염토양 정화시 오염물질의 rebound나 tailing 현상이 발생되어 정화기간이 장기화 되어왔다. 이러한 문제점은 미세토양과 미세공극이 존재하는 미세환경과 오염물질의 각각의 특성, 존재형태, 그리고 상호간의 다양한 반응 등으로부터 기인한다고 보고되었다. 따라서, 본 연구에서는 토양 지하수 미세환경과 오염정화 효율성과의 상관성에 대해 고찰하고 미세환경의 샘플링, 분석, 평가 기법을 소개 및 제안하며, 이를 활용한 오염정화효율 향상과 최적의 정화공법선정을 위한 정보를 제공하고자 하였다. 오염토양의 물리 화학 생물학적 특성 그리고 오염물질 종류 및 특성에 따른 정밀 평가를 통해 특정 정화공법 적용시 달성 가능한 정화기간과 정화수준을 예상할 수 있을 것이다. 따라서, 미세환경의 정밀 분석, 평가 기술을 바탕으로 정화 기술의 효율성과 타당성 검토가 가능할 것으로 판단되며 오염된 토양 지하수 복원을 위한 최적의 정화공법을 선정하는 기초자료로써 활용될 수 있을 것으로 사료된다.
본 연구에서는 성체 한우의 도축과정 중에 채혈된 혈액으로부터 분리 정제한 혈청의 생물학적 안전성을 검증하기 위하여 세균, 마이코플라즈마 및 바이러스의 검출과 동정을 실시하였다. 검사를 실시한 대부분의 혈청에서 전염성 병원체가 검출되지 않았고 두 개의 혈청에서 토양 유래의 것으로 추정되는 Bacillus thuringiensis가 검출 되었다. 본 연구의 결과는 국내에서 도축되는 한우는 생물학적 안전성을 담보할 수 있는 최소한의 기본적인 안전장치를 통해 도축된다는 사실을 보여주며 한우의 도축과정에서 부산물로 얻을 수 있는 혈액을 안전한 생물학적 제제로 개발할 수 있다는 가능성을 보여주는 것으로 생각된다.
Aspergillus awamori(A)와 Zymomonas mobilis(Z)로 구성된 혼합고정화 배양계(A-Z 36계)의 특성을 이용하여 생전분으로 부터 직접 에탄올생산을 행할 때 투입되는 에너지를 절약할 목적으로 비가열살균 배양법을 검토하였다. A-Z 36계에 의한 비가열살균 배양은 배지에 함유된 세균 때문에 어려웠다. Sorbic acid, benzoic acid, dehydroacetic acid, p-hydroxybenzoic acid, Vantocil IB, Neupectin-L 등의 여러 가지 살균제 중에서 Vantocil IB와 Neupectin-L은 잡균의 오염없이 대조구와 비슷한 속도로 기질을 분해하였다. 0.1% Neupectin-L은 대조구보다 약간 높은 6.9 g/l(대조구 : 6.4 g/l)의 에탄올을 생산하였다$(Y_{p/s}:0.39)$. 0.1% Neupectin-L을 첨가한 유가배양에서는 2%기질을 기본으로 5회분의 기질을 첨가하여 34 g/l 에탄올을 얻어 2.0 g/l/day의 생산성을 나타내었다.
This study was conducted to determine the incidence of microorganisms associated with dressed broiler with intact skin and without skin at different frozen storage periods such as 0, 10, 20, 30 days and to demonstrate the role of packaging and pretreatment chilling on the changes of carcass quality. The values of total viable count (TVC), total coliform count (TCC), total streptococcal count (TStC) and total staphylococcal count (TSC) were determined for meat samples of thigh and breast and swab samples of visceral surfaces of the broilers with intact skin and without skin. It was observed that the values of TVC, TCC, TStC and TSC in both cases of dressed broiler with intact skin and without skin exceeded the International Commission on Microbiological Specification for Foods. However, numbers of microorganisms were considerably decreased during the frozen storage. Packing and prechilled conditions were generally better effective in decrease of the loads of microorganisms than without packing and prechilled conditions, and lower bacterial numbers were also found in dressed broiler with intact skin than that without skin. The highest sensory panel score was obtained at 10 days of frozen storage. These results, thus, indicate that usages of appropriate periods and conditions of frozen storage and packaging systems can minimize the potential health hazards associated with contaminants gaining access to the dressed or processed broilers and improve the quality and shelf life of dressed broilers.
Estuarine sediments are frequently polluted with hydrocarbons from fuel spills and industrial wastes. Polycyclic aromatic hydrocarbons (PAHs) are components of these contaminants that tend to accumulate in the sediment due to their low aqueous solubility, low volatility, and high affinity for particulate matter. The toxic, recalcitrant, mutagenic, and carcinogenic nature of these compounds may require aggressive treatment to remediate polluted sites effectively. In petroleum-contaminated sediments near a petrochemical industry in Gwangyang Bay, Korea, in situ PAH concentrations ranged from 10 to 2,900 ${\mu}g/kg$ dry sediment. To enhance the biodegradation rate of PAHs under anaerobic conditions, sediment samples were amended with biostimulating agents alone or in combination: nitrogen and phosphorus in the form of slow-release fertilizer (SRF), lactate, yeast extract (YE), and Tween 80. When added to the sediment individually, all tested agents enhanced the degradation of PAHs, including naphthalene, acenaphthene, anthracene, fluorene, phenanthrene, fluoranthene, pyrene, chrysene, and benzo [a] pyrene. Moreover, the combination of SRF, Tween 80, and lactate increased the PAH degradation rate 1.2-8.2 times above that of untreated sediment (0.01-10 ${\mu}g$ PAH/ kg dry sediment/day). Our results indicated that in situ contaminant PAHs in anoxic sediment, including high molecular weight PAHs, were degraded biologically and that the addition of stimulators increased the biodegradation potential of the intrinsic microbial populations. Our results will contribute to the development of new strategies for in situ treatment of PAH-contaminated anoxic sediments.

PURPOSE. Computer-aided design and manufacturing (CAD-CAM) of implant abutments has been shown to result in surface contamination from site-specific milling and fabrication processes. If not removed, these contaminants can have a potentially adverse effect and may trigger inflammatory responses of the peri-implant tissues. The aim of the present study was to evaluate the bacterial disinfection and cleaning efficacy of ultrasonic reprocessing in approved disinfectants to reduce the microbial load of CAD-CAM abutments. MATERIALS AND METHODS. Four different types of custom implant abutments (total N = 32) with eight specimens in each test group (type I to IV) were CAD-CAM manufactured. In two separate contamination experiments, specimens were contaminated with heparinized sheep blood alone and with heparinized sheep blood and the test bacterium Enterococcus faecium. Abutments in the test group were processed according to a three-stage ultrasonic protocol and assessed qualitatively and quantitatively by determination of residual protein. Ultrasonicated specimens contaminated with sheep blood and E. faecium were additionally eluted and the dilutions were incubated on agar plates for seven days. The determined bacterial counts were expressed as colony-forming units (CFU). RESULTS. Ultrasonic reprocessing resulted in a substantial decrease in residual bacterial protein to less than 80 ㎍ and a reduction in microbiota of more than 7 log levels of CFU for all abutment types, exceeding the effect required for disinfection. CONCLUSION. A three-stage ultrasonic cleaning and disinfection protocol results in effective bacterial decontamination. The procedure is reproducible and complies with the standardized reprocessing and disinfection specifications for one- or two-piece CAD-CAM implant abutments.

The supply of microbiological risk-free water is essential to keep food safety and public hygiene. And removal, inactivation, and destruction of microorganisms in drinking water are key for ensuring safety in the food industry. Ultraviolet-C (UV-C) irradiation is an attractive method for efficient disinfection of water without generating toxicity and adversely affecting human health. In this study, the disinfection efficiencies of UV-C irradiation on Shigella flexneri (Gram negative) and Listeria monocytogenes (Gram positive) at various concentrations in drinking water were evaluated using a water purifier. Their morphological and physiological characteristics after UV-C irradiation were observed using fluorescence microscopy and flow cytometry combined with live/dead staining. UV-C irradiation (254 nm wavelength, irradiation dose: 40 mJ/cm2) at a water flow velocity of 3.4 L/min showed disinfection ability on both bacteria up to 108 CFU/4 L. And flow cytometric analysis showed different physiological shift between S. flexneri and L. monocytogenes after UV-C irradiation, but no significant shift of morphology in both bacteria. In addition, each bacterium revealed different characteristics with time-course observation after UV-C irradiation: L. monocytogenes dramatically changed its physiological feature and seemed to reach maximum damage at 4 h and then recovered, whereas S. flexneri seemed to gradually die over time. This study revealed that UV-C irradiation of water purifiers is effective in disinfecting microbial contaminants in drinking water and provides basic information on bacterial features/responses after UV-C irradiation.
멸균 초콜릿 우유로부터 분리한 내열성 균주에 대해 열, pH, 전해수, 오존처리, microwave 및 감마선 처리를 하여 균주의 사멸효과에 대해 알아보았다. 균주의 지방산 분석과 API kit를 통하여 균주를 동정한 결과, Bacillus lentus로 동정되었으며, 잠정적으로 Bacillus lentus M1으로 명명하였다. B. lentus M1에 110$^{\circ}C$, 15분간 열처리하였을 경우 생육이 억제되었으며, pH 처리 시 pH 5 이하, 10 이상에서 생육이 억제된 것으로 나타났다. B. lentus M1에 대한 전해수의 항균활성을 paper disc법으로 측정한 결과, 높은 생육억제를 보였으며, 오존 처리의 경우 초기 균수가 $10^2$ CFU가량의 균을 10분 동안, $10^3$ CFU가량의 균을 30분 동안 처리 시 균의 생육이 억제되는 것으로 나타났다. Microwave를 1분간 처리 시 B. lentus M1이 모두 사멸한 것으로 나타났다. 감마선 조사의 경우, 1 kGy 조사 시 생균수가 $1.61{\times}10^3 $CFU로 초기 균수에 비해 4 log cycle 가량 균수가 감소하였으며 7 kGy에서 완전히 사멸하였다. 이상의 결과를 통해 열, pH, 전해수, 오존 처리 및 방사선 처리 방법이 멸균 초콜릿 우유의 생존 오염균인 B. lentus M1을 효과적으로 사멸시킬 수 있을 것으로 사료된다.
Lake Shiwha, an artificial lake located near metropolitan Seoul, offers a unique water environment and has been suspected to have high levels of chemical and microbiological contaminations. Lake Shiwha was originally connected to the sea but currently has four major surface water inputs from agricultural, municipal, industrial areas and in addition an occasional inflow from the sea. The objectives of this study are to investigate the relative contribution of microbial contaminants from each of the inflowing surface waters and to identify appropriate microbial indicator organisms in this unique water environment. We measured the levels of microbial contaminations in the four inflowing surface waters. A number of microbial indicator organisms including total coliform (TC), fecal coliform (FC), E. coli, Enterococci, somatic and male-specific coliphages were analyzed. Bacterial indicator microorganisms were detected and quantified by the $Colilert^{(R)},\;Enterolert^{(R)}$ kit. Surface water (50 l) was sampled by $ViroCap^{TM}\;5"$ cartridge filters and analyzed by the single agar layer method for detecting coliphages. The concentrations of TC, FC, E. coli, and Enterococci were 1543 CFU/100 ml${\sim}1.99{\times}10^6$ CFU/100 ml, 0 CFU/100 ml${\sim}202$ CFU/100ml, 0 CFU/100 ml${\sim}1.80{\sim}10^5$ CFU/100ml, 74 CFU/100 ml${\sim}3408$ CFU/100 ml, respectively. The male-specific and somatic coliphages were detected in three different inflowing surface waters. Isolated E. coli and Enterococci strains were further analyzed by 16s rDNA amplification and subsequent phylogenetic analysis from Jungwang-chun, Ansan-chun, Banwol-chun and penstock of inflowing surface water. Our results indicated that the concentrations of different fecal indicator microorganisms might not be highly correlated with each other. Multiple microbial indicator organisms should be used for monitoring microbial contamination and microbial source tracking methods.
