Microbial Quality of Street Foods Sold by Season (계절에 따른 길거리 제조 식품의 미생물 오염 특성)

  • This study examined microbiological contamination of street foods(kimbab, fish cake, Korean sausage) by microbiological analysis. A collection of 360 samples of street foods was obtained seasonally in four major cities(Seoul, Daejeon, Busan, Gwangju) in Korea. Aerobic mesophilic counts ranged between 1.0 and 9.9 log CFU/g, with the highest count recorded from Kimbab. Counts of psychrotrophic microorganisms were as high as those of mesophilic microorganisms. Total coliform populations between <1.0 and 7.5 log CFU/g were found in 53.6% of samples. Escherichia coli 4.4%, Staphylococcus aureus 7.8% and Clostridium perfringens 3.3%. Salmonella spp. and Listeria monocytogenes were not detected in any of the samples. Kimbab purchased in spring and summer showed higher S. aureus and Cl. perfringens contamination rates. Microbial contamination levels determined in the present study may be used as the primary data to execute microbial risk assessment of street foods.

Effect of Particulate Matter on the UV-Disinfection of Virus and Risk Assessment (입자성 물질 농도가 바이러스의 UV-처리와 위해성에 미치는 영향 평가)

  • Wastewater reuse for agricultural irrigation needs treatment and control of pathogens to minimize risks to human health and the environment. In order to evaluate the water quality of UV-treated reclaimed water, this study focused on the relationship between micro-pathogens and particulate matters. MS2 was selected as an index organism because it has similar characteristics to human enteric virus and strong resistance to UV disinfection. The turbidity and suspended solid (SS) were selected for test parameters. In this study, it was performed with different UV doses (30 and $60mJ/cm^2$) for estimation of the MS2 inactivation rate using collimated beam batch experiments in the laboratory. The experiment results by turbidity or SS concentration presented that the increased concentration of them lowered MS2 inactivation. At the turbidity (below 4.27 NTU) and SS (below 1.47 mg/L) of the low level range, the inactivation of 60 UV dose is higher than 30 UV dose. However, at the turbidity and SS of the high level, the increasing UV dose did not show apparent increasing the MS2 inactivation. In quantitative microbial risk assessment (QMRA), it can confirm the trend that $P_D$ and turbidity concentrations have positive correlationship at the low concentration, which was also observed in SS. The QMRA can be helpful in communication with public for safe wastewater reuse and be recommended.

Risk assessment for norovirus foodborne illness by raw oyster (Ostreidae) consumption and economic burden in Korea

  • The objective of this study was to evaluate the probability of norovirus foodborne illness by raw oyster consumption. One hundred fifty-six oyster samples were collected to examine the norovirus prevalence. The oyster samples were inoculated with murine norovirus and stored at 4℃-25℃. A plaque assay determined norovirus titers. The norovirus titers were fitted with the Baranyi model to calculate shoulder period (h) and death rate (Log PFU/g/h). These kinetic parameters were fitted to a polynomial model as a function of temperature. Distribution temperature and time were surveyed, and consumption data were surveyed. A dose-response model was also searched through literature. The simulation model was prepared with these data in @RISK to estimate the probability of norovirus foodborne. One sample of 156 samples was norovirus positive. Thus, the initial contamination level was estimated by the Beta distribution (2, 156), and the level was -5.3 Log PFU/g. The developed predictive models showed that the norovirus titers decreased in oysters under the storage conditions simulated with the Uniform distribution (0.325, 1.643) for time and the Pert distribution (10, 18, 25) for temperature. Consumption ratio of raw oyster was 0.98%, and average consumption amount was 1.82 g, calculated by the Pert distribution [Pert {1.8200, 1.8200, 335.30, Truncate (0, 236.8)}]. 1F1 hypergeometric dose-response model [1 - (1 + 2.55 × 10-3 × dose)-0.086] was appropriate to evaluate dose-response. The simulation showed that the probability of norovirus foodborne illness by raw oyster consumption was 5.90 × 10-10 per person per day. The annual socioeconomic cost of consuming raw oysters contaminated with norovirus was not very high.

Establishment of Sample Preparation Method for PCR Detection of Clostridium perfringens from Agricultural Products (PCR 법을 이용한 농산물 중 Clostridium perfringens 검출을 위한 전처리법 확립)

  • This study was undertaken to compare the efficacy of different sample preparation (stomaching, pulsifying, and sonication) and DNA extraction methods (boiling and commercial kit) for detection of enterotoxin-producing Clostridium perfringens from produce by polymerase chain reaction (PCR). Each produce type was inoculated at concentrations of 102, 103, 104, 105, 106, and 107 spores/g. Produce inoculated with spores was treated with three sample preparation methods, and DNA was extracted by boiling method and a commercial kit, followed by PCR. The detection limit of stomached samples was lower than that of pummeled and sonicated samples by 10-100 times. Moreover, the DNA extraction efficiency of the commercial kit was found to be superior to that of boiling. In particular, the PCR efficiency of cherry tomato and perilla leaf samples was greatly affected by sample preparation and DNA extraction method. These data suggest that DNA extraction with a commercial kit after pulsification is an optimum sample preparation method for detection of C. perfringens by PCR.

Investigation of Microbial Contamination Levels of Leafy Greens and Its Distributing Conditions at Different Time - Focused on Perilla leaf and Lettuce - (시기별 엽채류의 미생물 오염도와 유통 조건 조사 - 들깻잎과 상추를 중심으로 -)

  • The objective of this study was to investigate and evaluate microbial contamination levels of leafy greens (perilla leaf and lettuce) and its distributing conditions at different seasons (Feb, May, Aug, and Nov of the year 2011) in order to provide insight into any potential health hazards associated with consumption of these commodities. Leafy greens were collected from a farm located in Geumsan, Chungnam and wholesale markets (WM) and traditional markets (TM) located in Suwon. At the same time, temperature and relative humidity fluctuations experienced by the leafy greens during distribution from the farm to the distribution center were measured by a data logger. The contamination levels of perilla leaf and lettuce were determined by analyzing total plate count. Coliform groups, Bacillus cereus, Escherichia coli, Escherichia coli O157:H7, Salmonella spp., Listeria monocytogenes and Staphylococcus aureus were determined. The contamination levels of total aerobic bacteria, coliform groups and B. cereus in both vegetables sampled during May and August found to be higher than those sampled during February and November. E. coli O157:H7, Salmonella spp., L. monocytogenes were not detected in the vegetables analyzed in this study. There were no significant trends between samples at WM and TM in the contamination levels. Relative humidity of vegetables distributed from the farm to the distribution center showed over 90% during distribution regardless of measured seasons. In the case of background microflora on leafy greens, the density was significantly increased at 20, 30 and $37^{\circ}C$ during storage of 24h. E. coli O157:H7 and B. cereus inoculated on the leaves also showed similar increases in the storage tests. The microbial contamination levels determined in this study may be used as the fundamental data for microbial risk assessment.

Biotransformation of a Fungicide Ethaboxam by Soil Fungus Cunninghamella elegans

  • Metabolism of a new fungicide ethaboxam by soil fungi was studied. Among the fungi tested, Cunninghamelia elegans produced metabolites from ethaboxam, which were not found in the control experiments. M5, a major metabolite from ethaboxam was firmly identified as N-deethylated ethaboxam by LC/MS/MS and NMR. N-Deethylated ethaboxam has been found as a single metabolite in in vitro metabolism with rat liver microsomes. Ml was proved to be 4-ethyl-2-(ethylamino)-1,3-thiazole-5-carboxamide (ETC) by comparing with the authentic compound. In addition, M2, M3, and M4, and M6 were tentatively Identified by LC/MS/MS as hydroxylated and methoxylated ethaboxams, respectively. Production of the major metabolite, N-deethylated ethaboxam, by the fungus suggested that C. elegans would be an efficient eukaryotic microbial candidate for evaluating xenobiotic-driven mammalian risk assessment.

The Effects of Sodium Chloride on the Physiological Characteristics of Listeria monocytogenes

  • Sodium chloride is used to improve various properties of processed meat products, e.g., taste, preservation, water binding capacity, texture, meat batter viscosity, safety, and flavor; however, many studies have shown that sodium chloride increases the resistance of many foodborne pathogens to heat and acid. Listeria monocytogenes has been isolated from various readyto- eat (RTE) meat and dairy products formulated with sodium chloride; therefore, the objective of this paper was to review the effects of sodium chloride on the physiological characteristics of L. monocytogenes. The exposure of L. monocytogenes to sodium chloride may increase biofilm formation on foods or food contact surfaces, virulence gene transcription, invasion of Caco-2 cells, and bacteriocin production, depending on L. monocytogenes strain and serotype as well as sodium chloride concentration. When L. monocytogenes cells were exposed to sodium chloride, their resistance to UV-C irradiation and freezing temperatures increased, but sodium chloride had no effect on their resistance to gamma irradiation. The morphological properties of L. monocytogenes, especially cell elongation and filament formation, also change in response to sodium chloride. These findings indicate that sodium chloride affects various physiological responses of L. monocytogenes and thus, the effect of sodium chloride on L. monocytogenes in RTE meat and dairy products needs to be considered with respect to food safety. Moreover, further studies of microbial risk assessment should be conducted to suggest an appropriate sodium chloride concentration in animal origin foods.

  • 상수도 관망의 수질 관리는 매우 중요한 목표이지만, 2019년도 적수 사태를 비롯하여 여러 수질사고를 경험하고 있다. 이러한 수질 사고는 내부의 각종 부착물 및 침전물들로 인해서 발생하거나 파손된 관로를 통해 외부에서 유입될 수도 있다. 수질 사고는 다양한 경로를 통해 발생할 수 있으며 외부에서의 바이러스 유입의 가능성 역시 늘 존재하고 있다고 할 수 있다. 본 연구에서는 상수도 관망으로의 잠재적인 바이러스 유입에 따른 위험도 분석 체계를 구현하였으며, 다양한 바이러스 유입에 따른 상수도 관망 위험도의 민감도 분석을 수행하였다. 제안한 분석 체계는 상수도 관망의 주요 소독 물질인 염소와 바이러스의 반응을 모의할 수 있도록 EPANET-MSX 모형을 탑재하였으며, 위험도 분석을 위한 QMRA(Quantitative Microbial Risk Assessment)를 적용하였다. 바이러스 유입 시나리오 구축을 위해 상수도 관망 내 유입되거나 인체에 유해 한 영향을 줄 수 있는 바이러스를 우선 선별 하였고, 인체에의 영향 및 염소와의 반응에 대해 정리하였다. 또한 바이러스는 모든 절점에서 유입이 가능한 것으로 가정하였으며, 최악의 상황 모의를 위해 바이러스가 지속적으로 유입되는 시나리오를 구축하였다. 적용 관망은 미국 내 실제 관망 중 반응계수에 대한검토가 완료된 관망으로 선정하였으며, 관망의 구조 및 탱크의 유무에 따라 분류하여 적용한 인자들의 불확실성 및 경향을 파악하기 위한 MCS(Monte Carlo Simulation)를 통해 민감도 분석을 수행하였다. 제안한 분석 체계는 향후 수질 사고에 대한 위험도를 최소화할 수 있는 운영에 근거를 제공해 줄 수 있을 것이다.

Predictive model and quantitative microbial risk assessment of enterohemorrhagic Escherichia coli and Campylobacter jejuni in milk (우유에서 장출혈성 대장균과 캠필로박터균의 행동예측 모델 개발 및 정량적 미생물 위해성 평가 연구)

  • We prepared the growth and survival models of enterohemorrhagic Escherichia coli (EHEC) and Campylobacter jejuni in milk as a function of temperature and assessed the microbiological risks associated with the consumption of whole milk. EHEC and C. jejuni were not detected in whole milk (n=195) in the retail market. The minimum growth temperature of EHEC in milk was 7℃. The lag time of EHEC in whole milk was longer than that in skim milk. The survival ability of C. jejuni in milk was better at 4℃ than at 10℃. Lower delta values were observed in whole milk than in skim milk, indicating that C. jejuni survived better in skim milk. The probability of foodborne illness from whole milk consumption was 5.70×10-5 for EHEC and 9.86×10-9 for C. jejuni. Sensitivity analysis results show that the market temperature of EHEC and the dose-response model of C. jejuni are correlated with the probability of foodborne illness.
