Preparation and Keeping Quality of Vacuum Packed Seasoned-Dried Sardine (진공포장 정어리 조미건제품의 제조 및 품질안정성)

  • As one of trials to process instant sardine foods which can be preserved at room temperature, three kinds of products were prepared as seasoned-dried product (control, C), liquid smoked seasoned-dried product(S) and antioxidant treated seasoned-dried product(E), and their processing conditions and quality stability during storage were examined. Raw sardines were dressed, steamed and then filleted. The sardine fillets were seasoned with the mixed seasoning solution containing $28.0\%$ of sorbitol, $14.0%$ of sugar, $5.6\%$ of table salt, $1.8\%$ of monosodium glutamate, $0.6\%$ of garlic powder and $50.0\%$ of water at $5^{\circ}C$ for 15 hours, and dipped for 45 seconds in $10\%$ Smoke-EZ solution. After liquid smoking, the seasoned and liquid smoked sardine fillets were dried at $45^{\circ}C$ for 4 hours, vacuum packed in laminated plastic film bag(polyester/casted polypropylene= $12{\mu}m/70{\mu}m,\;15{\times}16cm$), and finally pasteurized in water at $95^{\circ}C$ for 30 minutes. The results obtained from chemical and microbial experiments during storage are as follows : the moisture contents, water activity and pH of the products showed little change, and VBN of them slightly increased during storage. The TBA value and POV of the products (E, S) were lower than those of control product(C) considerably. In color values, L value (linghtness) decreased while a and b value (red and yellow) revealed a tendency to increase during storage. The fatty acid composition of the products were similar to those of raw sardine, the predominant fatty acids were 16:0, 20:5, 18:1 and 22:6. The products (E, S) have a good preservative effect on highly unsturated fatty acids during storage. Viable cell counts of those products were negative and histamine contents were less than 2.0 mg/100 g. Among the texture profiles, hardness, elasticity and cohesiveness of the products slightly decreased during storage. Judging from the sensory evaluations, liquid smoked seasoned-dried product(S) was the most desirable, and the products could be preserved in good condition for 40 days at $25{\pm}3^{\circ}C$.

  • Optimal conditions for processing. of seasoned-dried Pacific saury treated with liquid smoke (T2) were evaluated by physicochemical and microbial experiments and sensory evaluation, comparing with control (seasoning only, C) and treatment I $(0.05\%\;Rosemary\;instead\;of\;liquid\;smoke,\;Tl)$. Two hrs of seasoning time was set, and 23 hrs of drying time was determined in all samples. Finally, T2 product was made by soaking treatments (three times of 1 sec, 8 sec and 1 sec) in $5\%$ (v/v) liquid smoke (Scansmoke PB 2110) after 30 min, 4 hrs and 22 hrs of hot-air drying, respectively. The histamine contents in 3 seasoned-dried products were in a 15.33-26.99 mg/l00g range. The water activities of 3 seasoned-dried products were 0.719-0.735 range, and the pH of T2 was lower than the others. In the comparison of POV and TBA values among products, the TBA values and POV of Tl and T2 were significantly low compared to C, and also the viable cell counts of T2 was relatively lower than those of the others. In the color values, significant changes were not found among products, and in the sensory evaluation for odor, taste and overall acceptance, T2 had relatively higher preference on the whole items.

Separation Processes of Biologically Produced 1,3-Propanediol (1,3-프로판디올의 생물학적 생산을 위한 분리공정)

  • As the biodiesel production is increasing rapidly, the crude glycerol, which is principal by-product of biodiesel production, has also been generated in a large amount. Many research studies on value-added utilization of glycerol are under investigation. 1,3-Propandiol is a promising chemical which can be produced from fermentation of glycerol because the application of 1,3-propanediol is mainly in the production of bio-PTT (Poly(trimethylene terephthalate). However, the cost of downstream processes in the biological production of 1,3-propanediol can make a high portion in the total production cost. This review summarizes the present state of separation processes in each step studied for the removal of impurities and the recovery of 1,3-propanediol from its fermentation broth. Furthermore, ATPE (Aqueous Two Phases Extraction) process is suggested as an attractive alternative for the primary separation process of 1,3-propanediol because ATPE is convenient for the simultaneous removal of microbial cells and impurities such as salts of organic acids and the separation of 1,3-propanediol from fermentation broth.

Prolonged Exposure to Lipopolysaccharide Induces NLRP3-Independent Maturation and Secretion of Interleukin (IL)-1β in Macrophages

  • Upon sensing of microbial infections or endogenous danger signals in macrophages, inflammasome signaling plays a significant role in triggering inflammatory responses via producing interleukin (IL)-$1{\beta}$. Recent studies revealed that active caspase-1, a product of the inflammasome complex, causes maturation of inactive pro-IL-$1{\beta}$ into the active form. However, the underlying mechanism by which this leaderless cytokine is secreted into the extracellular space remains to be elucidated. In this study, we demonstrated that prolonged lipopolysaccharide (LPS) treatment to macrophages could trigger the unexpected maturation and extracellular release of IL-$1{\beta}$ through a nucleotide-binding oligomerization domain-like receptor family, pyrin domain-containing 3 (NLRP3)-independent manner. Short-term treatment (less than 6 h) of LPS induced robust production of the IL-$1{\beta}$ precursor form inside cells but did not promote the maturation and secretion of IL-$1{\beta}$ in bone marrow-derived macrophages or peritoneal macrophages. Instead, prolonged LPS treatment (more than 12 h) led to a significant release of matured IL-$1{\beta}$ with no robust indication of caspase-1 activation. Intriguingly, this LPS-triggered secretion of IL-$1{\beta}$ was also observed in NLRP3-deficient macrophages. In addition, this unexpected IL-$1{\beta}$ release was only partially impaired by a caspase-1 and NLRP3 inflammasome inhibitor. Collectively, our results propose that prolonged exposure to LPS is able to drive the maturation and secretion of IL-$1{\beta}$ in an NLRP3 inflammasome-independent manner.

A revaluation of algal diseases in Korean Pyropia (Porphyra) sea farms and their economic impact

  • As with land crops, cultivated algae are affected by various diseases ranging from large outbreaks of a disease to chronic epiphytes, which may downgrade the value of the final product. The recent development of intensive and dense mariculture practices has enabled some new diseases to spread much faster than before. A new disease is reported almost every year, and the impact of diseases is expected to increase with environmental change, such as global warming. We observed the incidence of diseases in two Pyropia sea farms in Korea from 2011 to 2014, and estimated the economic loss caused by each disease. Serious damage is caused by the oomycete pathogens, Pythium porphyrae and Olpidiopsis spp., which decreased the productivity of the Pyropia sea farms. In Seocheon sea farms, an outbreak of Olpidiopsis spp. disease resulted in approximately US $1.6 million in loss, representing approximately 24.5% of total sales during the 2012-2013 season. The damage caused by green-spot disease was almost as serious as oomycete diseases. An outbreak of green-spot disease in the Seocheon sea farms resulted in approximately US $1.1 million in loss, representing 10.7% of total sales in the 2013-2014 season in this area. However, the causative agent of green-spot disease is still not confirmed. "Diatom felt" is regarded as a minor nuisance that does not cause serious damage in Pyropia; however, our case study showed that the economic loss caused by "diatom felt" might be as serious as that of oomycete diseases. Bacteria and cyanobacteria are indigenous members of epiphytic microbial community on Pyropia blades, but can become opportunistic pathogens under suitable environmental conditions, especially when Pyropia suffers from other diseases. A regular acid wash of the Pyropia cultivation nets is the most common treatment for all of the above mentioned diseases, and represents approximately 30% of the total cost in Pyropia sea farming. However, the acid wash is ineffective for some diseases, especially for Olpidiopsis and bacterial diseases.

Supplementing Vitamin E to the Ration of Beef Cattle Increased the Utilization Efficiency of Dietary Nitrogen

  • The objectives of the trial were to investigate the effects of supplementing vitamin E (VE) on nutrient digestion, nitrogen (N) retention and plasma parameters of beef cattle in feedlot. Four growing Simmental bulls, fed with a total mixed ration composed of corn silage and concentrate mixture as basal ration, were used as the experimental animals. Four levels of VE product, i.e. 0, 150, 300, 600 mg/head/d (equivalent to 0, 75, 150, 300 IU VE/head/d), were supplemented to the basal ration (VE content 38 IU/kg dry matter) in a $4{\times}4$ Latin square design as experimental treatments I, II, III and IV, respectively. Each experimental period lasted 15 days, of which the first 12 days were for pretreatment and the last 3 days for sampling. The results showed that supplementing VE did not affect the nutrient digestibility (p>0.05) whereas decreased the urinary N excretion (p<0.01), increased the N retention (p<0.05) and tended to increase the microbial N supply estimated based on the total urinary purine derivatives (p = 0.057). Supplementing VE increased the plasma concentrations of VE, glucose and triglycerol (TG) (p<0.05) and tended to increase the plasma concentration of total protein (p = 0.096) whereas did not affect the plasma antioxidant indices and other parameters (p>0.05). It was concluded that supplementing VE up to 300 IU/head/d did not affect the nutrient digestibility whereas supplementing VE at 150 or 300 IU/head/d increased the N retention and the plasma concentrations of VE and TG (p<0.05) of beef cattle.

  • Chlorine disinfection has been used in drinking water supply to disinfect the water-borne microbial disease which may cause to serious human disease. As Chlorination is still the least costly, relatively easy to use, chlorination is the primary means to disinfect portable water supplies and control bacterial growth in the distribution system. However, chlorine also reacts with natural organic matter (NOM), which presents in nearly all water sources, and then produces disinfection by-product (DBps), which may have adverse health effects. Although the existent DBPs have been reported in drinking water supplies, it is not feasible to predict the levels of the various DBPs due to the complex chemistry reaction involved. The objectives of this study were to investigate seasonal variation of DBPs formation and difference of DBPs concentration in the plant to tap water. The average concentration of THMs was 20.04 ${\mu}g/{\ell}$, HAAs 8-15 ${\mu}g/{\ell}$, HANs 2-4.5 ${\mu}g/{\ell}$ respectively. Distant variation of DBPs formation is that THMs concentration increase by 17% at 2 km point from the plant and by 28% at 7 km and HAAs, HANs also increase each by 16%, 32%, at 2 km from the plant and 35%, 56%, at 7 km. DBPs increase in water supply pipe continually. The seasonal occurrence of DBPs is that in May and August DBPs concentration is very high then in March, in May DBPs concentration is highest. The temperature is main factor of DBPs formation, precursor also. Precursor which was accumulated for winter flowed into the raw water by flooding in spring and summer and produced DBPs. Therefore for the supply of secure drinking water, it is required to protect precursor of flowing into raw water and to add to BCAA and DBAA to drinking water standards.

Structural Basis for Recognition of L-lysine, L-ornithine, and L-2,4-diamino Butyric Acid by Lysine Cyclodeaminase

  • L-pipecolic acid is a non-protein amino acid commonly found in plants, animals, and microorganisms. It is a well-known precursor to numerous microbial secondary metabolites and pharmaceuticals, including anticancer agents, immunosuppressants, and several antibiotics. Lysine cyclodeaminase (LCD) catalyzes ${\beta}$-deamination of L-lysine into L-pipecolic acid using ${\beta}$-nicotinamide adenine dinucleotide as a cofactor. Expression of a human homolog of LCD, ${\mu}$-crystallin, is elevated in prostate cancer patients. To understand the structural features and catalytic mechanisms of LCD, we determined the crystal structures of Streptomyces pristinaespiralis LCD (SpLCD) in (i) a binary complex with $NAD^+$, (ii) a ternary complex with $NAD^+$ and L-pipecolic acid, (iii) a ternary complex with $NAD^+$ and L-proline, and (iv) a ternary complex with $NAD^+$ and L-2,4-diamino butyric acid. The overall structure of SpLCD was similar to that of ornithine cyclodeaminase from Pseudomonas putida. In addition, SpLCD recognized L-lysine, L-ornithine, and L-2,4-diamino butyric acid despite differences in the active site, including differences in hydrogen bonding by Asp236, which corresponds with Asp228 from Pseudomonas putida ornithine cyclodeaminase. The substrate binding pocket of SpLCD allowed substrates smaller than lysine to bind, thus enabling binding to ornithine and L-2,4-diamino butyric acid. Our structural and biochemical data facilitate a detailed understanding of substrate and product recognition, thus providing evidence for a reaction mechanism for SpLCD. The proposed mechanism is unusual in that $NAD^+$ is initially converted into NADH and then reverted back into $NAD^+$ at a late stage of the reaction.

Anthocyanins from Clitoria ternatea Attenuate Food-Borne Penicillium expansum and its Potential Application as Food Biopreservative

  • Clitoria ternatea or Commonly known blue pea, is a perennial climber crop native to Asian countries. The current study was aimed to evaluate the antimicrobial activity C. ternatea extract on food borne microorganisms and its antifungal effect on Penicillium expansum. The extract showed significant antimicrobial activity against 3 Gram positive bacteria, 2 Gram negative bacteria and 1 filamentous fungus on disc diffusion assay. The extract also showed good biocidal effect on all Gram positive bacteria tested and P. expansum. However, the kill curve analysis revealed that the fungicidal activity of the extract against P. expansum conidia was depend on the concentration of the extract and the time of exposure of the conidia to the extract. The scanning electron micrograph of the extract treated P. expansum culture showed alterations in the morphology of fungal hyphae. The germination of P. expansum conidia was completely inhibited and conidial development was totally suppressed by the extract, suggesting the possible mode of action of anthocyanin. Besides, the extract also exhibited 5.0-log suppression of microbial growth relative to control in the rice model. The results indicate the potential use of the C. ternatea anthocyanin as food biopreservative.

Removal of RDX using Lab-scale Plug Flow Constructed Wetlands Planted with Miscanthus sacchariflorus (Maxim.) Benth (물억새를 식재한 플러그 흐름 습지에서의 RDX 제거동역학)

  • RDX (hexahydro-1,3,5-trinitro-1,3,5-triazine) is the most important explosive contaminant, both in concentration and in frequency, at military shooting ranges in which green technologies such as phytoremediation or constructed wetlands are the best option for mitigation of explosive compounds discharge to the environment. A study was conducted with two identical lab-scale plug flow constructed wetlands planted with Amur silver grass to treat water artificially contaminated with 40 mg/L of toxic explosive compound, RDX. The reactor was inoculated with or without RDX degrading mixed culture to evaluate plant-microorganism interactions in RDX removal, transformation products distribution, and kinetic constants. RDX and its metabolites in water, plant, and sediment were analyzed by HPLC to determine mass balance and kinetic constants. After 30 days of operation, the reactor reached steady-state at which more than 99% of RDX was removed with or without the mixed culture inoculation. The major transformation product was TNX (Trinitroso-RDX) that comprised approximately 50% in the mass balance of both reactors. It was also the major compound in the plant root and shoot system. Acute toxicity analysis of the water samples showed more than 30% of toxicity reduction in the effluent than that of influent containing 40 mg/L of RDX. In the Amur silver grass mesocosm seeded with the mixed culture, the specific RDX removal rate, that is 1st order removal rate normalized to plant fresh weight, was estimated to be 0.84 kg−1 day−1 which is 16.7% higher than that in the planted only mesocosm. Therefore, the results of this study proved that Amur silver grass is an effective plant for RDX removal in constructed wetlands and the efficiency can be increased even more when applied with RDX degrading microbial consortia.
