A Literary Study on Bezoar Bovis (우황에 대한 문헌적 고찰)

  • Objectives: Literary investigation of existing data was conducted to verify effects of Bezoar Bovis and its herbal acupuncture, and determine quality management through component analysis. Results: Following results were obtained through literary investigation. 1. Bezoar Bovis is dried cystic stone from a cattle. Its characteristics are cool, no toxicity, and bitter taste. Known actions are: quells heat and detoxifies Fire Poison, extinguishes internal movement of Liver Wind and stops convulsion, vaporizes phlegm, and opens orifice. It is mainly used for treating tremor, stroke, delirium, sore throat, oral furuncle, boil, and others. 2. Bezoar Bovis is effective for eliminating liver toxicity, protecting against brain damage, and has anti-microbial activities. 3. Bezoar Bovis is mixed with bear gall bladder and deer musk to be used as herbal acupuncture, and this mixture is effective is invigorating liver functions as well as treating arthritis, headache, and etc. 4. Principal components of Bezoar Bovis are bilirubin-type pigments and cholic acids. The amount of bilirubin can be used as a standard to determine the quality of Bezoar Bovis.

Bioconversion of flowers waste: Composting using dry leaves as bulking agent

  • At present, in India, handling of solid waste has become a major challenge for the municipal authorities. Composting of solid waste, especially organic waste, can be one of the solutions to tackle the issue of handling solid waste. The present study is focused on agitated piles composting of flower waste (FW). Five combinations of FW with dry leaves (DL) and cow dung (CD) were prepared to conduct the study. Significant changes were observed due to the addition of bulking agent. The bulking material helps to reduce the production of leachate and also to maintain the aerobic condition within the piles. The reduction of total organic carbon was 21% in FW composting which increased by 36.48% during the composting of FW on addition of DL and CD. On the 120th day of composting, the pH of pile five (70 kg FW + 20 kg CD + 15 kg DL) was 7.33, electrical conductivity 2.77 mS/cm, total organic carbon 26.9%, total nitrogen 2.2%, and C:N ratio was 12. Appropriate proportion of waste mixture played an important role in providing favorable conditions for the microbial transformation of flower waste to stabilized compost. Finally, FW with the combination of CD and DL was found to be successful during pile composting.

  • In order to promote the prevention of microbial and enzymatic spoilage and to retain the freshiness, sweet persimmons harvested in Gyeongsangnam-do were treated with graperfruit seed extract(GFSE)-CaCO3 mixture and stored in the proper packaging conditions. A low concentration of GFSE showed effective growth inhibition of plant pathological bacteria and fungi, Enterobacter pyrinus and Fusarium sp., which were involved in the decay of fruits and vegetables. GFSE was stable to heat treatment; its antimicrobial activity was not changed by heat treatment upto 10$0^{\circ}C$. However, when the temperature was raised to 12$0^{\circ}C$, about 90% of total activity was retained within 30 min. GFSE was also highly stable to broad pH changes; its activity was not changed in the range of pH 2.0 to pH 12.0. The physiological function of cell membrane in the spores of Bacillus cereus and the hyphae of Fusarium sp. was destroyed by treating with GFSE. It was observed that treating sweet persimmons with GFSE minxture and storing them in strech-wrapped packages could prolong the greshness of sweet persimmons and reduce quality deterioration.

Studies on the Biological Activity of Synbiotics: A Review (신바이오틱스의 생리활성에 관한 연구 고찰)

  • This paper defines the common features of synbiotics based on the definition of probiotics and prebiotics, and reviews the effectiveness of synbiotic food. The concept of synbiotics is defined as 'a mixture of prebiotics and probiotics that have a beneficial effect on the host, as a dietary supplement that alters living organisms in the gastrointestinal tract and improves their survival.' Synbiotic food contains ingredients with beneficial microbes that are expected to improve interactions between microbial and useful substances. Synbiotic foods may have anti-cancer and immune system-boosting effects. Improved digestion, healthier bowel movements, and overall increased intestinal health has been reported were reported after increasing the healthy microorganisms within the intestinal tract. In addition, depending on the type of food containing the symbiotic ingredients, more consistent weight control, improvement of cardiovascular health, and lower blood glucose levels may also be expected. Unlike previous studies, this review of synbiotics has shown that it is necessary for synergistic effects to take place among microorganisms and components to be further studied. Further research is needed on the safety and ingestion of microorganisms contained in synbiotics.

The effects of Magnoliae cortex and Zea Mays L. extract mixtures on experimentally induced periodontitis of beagle dog (후박추출물과 옥수수 불검화 추출물 혼합 경구용 제제가 비글견에서 실험적으로 유발된 치주염에 미치는 영향)

  • It has been reported that Magnoliae cortex extract has antibacterial and antimicrobial activity against pathogenic microbes and Zea Mays L. extract is effective for improving gingival tissue health. The purpose of this study was to examine the anti-inflammatory and antimicrobial effects of Zea Mays L. and Magnoliac cortex extract mixtures through experimental peridontitis induced beagle dog model. Nine beagle dogs with experimentally induced periodontitis were selected. Baseline clinical indices which includes plaque index, gingival index, probing pocket depth, clinical attachment level, gingival fluid flow rate were recorded and microbial assays were done. Magnoliac cortex and Zea Mays L., mixed at 2:l ratio in 105mg capsular dosage, were taken by 3 capsule (Group I) or 6 capsule dosages (Group II) three times a day. After 4,8,12 weeks, clinical indices were recorded. All data of clinical indices were compared through one-way ANOVA with 95% confidence level. Clinical indices of group I and II showed significantly better results than those of control group. There were no significant differences between group I and II. In conclusion, it was confirmed that mixture of Magnoliae cortex and Zea May L. (mix ratio 2:1) possessed clinical improving effects to periodontitis.

PCR-based Identification of Microorganisms in a Kefir Grain

  • Nowadays many people are concerned about being healthy, and many dairy products are taken as health supplementary foods. Among dairy products, kefir, also called as Tibet mushroom, is a yogurt fermented by kefir grain, which is a mixture of lactic acid bacteria and yeasts. Although there are many empirical evidences that kefir is very influential for human body, the exact reason is not definitively discovered. Therefore, it would be useful to understand characteristics of a kefir grain and to categorize bacteria in a kefir grain. In this paper, molecular biological apparatus such as PCR, electrophoresis, PCR purification, DNA sequencing were used to identify and classify the species of lactic acid bacteria and yeast in a kefir grain. We used PCR-based identification method using 16S rRNA primer and Internal Transcribed Spacer (ITS) primer. We identified 6 different species which were selected on different medium. In addition, observation with scanning electron microscope (SEM) enabled us to grasp an external shape of the kefir grain. Although we found a limited number of microbial species, more intensive research are needed for extensive identification of microorganism species in Korean kefir grain.

Biochemical and molecular characterization of a tetrachloroethylene (PCE) dechlorinating Clostridium bifermentans DPH-1

  • The tetrachloroethylene (PCE) dehalogenase of Clostridium bifermentans DPH-1 (a halorespiring organism) was purified, cloned, and sequenced. This enzyme is a homodimer with a molecular mass of ca. 70 kDa and exhibits dehalogenation of dichloroethylene isomers along with PCE and trichloroethylene (TCE). Broad range of substrate specificity for chlorinated aliphatic compounds (PCE, TCE, cis-1,2-dichloroethylene, trans-1,2-dichloroethylene, 1,1-dichloroethylene, 1,2-dichloropropene, and 1,1,2-trichloroethane) for this enzyme was also observed. A mixture of propyl iodide and titanium citrate caused a light-reversible inhibition of enzymatic activity suggesting the involvement of a corrinoid cofactor. A partial sequence (81 bp) of the encoding gene for PCE dehalogenase was amplified and sequenced with degenerateprimers designed from the N-terminal sequence (27 amino acid residues). Southern analysis of C. bifermentans genomic DNA using the polymerase chain reaction product as a probe revealed restriction fragment bands. A 5.0 kb ClaI fragment, harboring the relevant gene (designated pceC) was cloned (pDEHAL5) and the complete nucleotide sequence of pceC was determined. The gene showed homology mainly with microbial membrane proteins and no homology with any known dehalogenase, suggesting a distinct PCE dehalogenase. So, C. bifermentans could play some important role in the initial breakdown of PCE and other chlorinated aliphatic compounds in sites contaminated with mixtures of halogenated substances.

An Efficient Micropropagation to Obtain the Disease-free Bulbs from Scales for Cryopreservation in Lilium

  • Cryopreservation is one of the ideal and suitable methods for long-term storage of plant germplasm. The plant contaminated with diseases and pathogens are decreased the multiplication rate, survival rate and high quality of plants after cryopreservation. The aim of this work was to improve a micropropagation method for lily in Korea, which are cultivars and indigenous plant. In the last process of rinsing scales after surface-sterilization, we tried to control the diseases and pathogens lived within the tissue by rinsing in 0.03% sodium hypochlorite (NaClO) instead of sterile distilled water. Bulb scales of Lilium were cultured in vitro on MS medium supplemented with Plant Preservative Mixture (PPM). The newly small bulb formed from bulb-scales was transferred to MS medium. We checked the non-contamination and survival rate after 2 weeks in culture. Non-contamination was shown to be 70 to 90% in formed small bulbs. This study will help to mitigate microbial contamination in Lilium species micropropagation for cryopreservation.

Control Efficacy of Entomopathogenic Microagent against Spodoptera exigua on Organic Chinese Cabbage (유기재배 배추포장 내 파밤나방에 대한 곤충병원성 미생물의 방제 효과)

  • This study was carried out to identify the control effect of entomopathogenic microagent against Spodoptera exigua on organic chinese cabbage. In laboratory condition, insecticidal activity of 4 commercial BT pesticides against S. exigua were lower than 10% against second instar S. exigua. The insecticidal activity of entomopathogenic nematode were 33.3%, 83.3% and 100% at the concentration of $1{\times}10^2$, $3{\times}10^2$, $1{\times}10^3nematodes/ml$, respectively. Mixture of BT and nematode showed growth inhibition against S. exigua larvae. S. exigua nucleopolyhedrovirus (SeNPV) of $10^5PIBs/ml$ showed more than 70% insecticial activity. The yield of SeNPV was increased as in higher initial inoculation concentration of NPV, food supply, and growth temperature increased. In greenhouse experiment, the control value of BT and nematode mixture treatment was higher than BT and nematode treatment alone against S. exigua. In treatment of $10^7PIBs/ml$ of SeNPV, S. exigua was controlled completely. In farm condition, mixture of microbial agent and organic agricultural material showed higher control value against lepidopteran pest including S. exigua than BT single treatment.

Rheological Properties of Pork Myofibrillar Protein and Sodium Caseinate Mixture as Affected by Transglutaminase with Various Incubation Temperatures and Times (Transglutaminase를 첨가한 돈육 근원섬유단백질과 카제인염 혼합물의 배양온도와 시간에 따른 물성변화)

  • To investigate the rheological properties of protein mixed gels mediated by microbial transglutaminase (MTGase), pork myofibrillar protein (MFP), sodium caseinate (SC) and their mixture (MS), the various gels were incubated at different temperatures for various times. Extracted MFP, SC and their mixture (MS, 1:1) were incubated at different temperatures ($4^{\circ}C$ vs $37^{\circ}C$) for various times (0, 0.5, 2, 4 hr), and assessed for viscosity, gel strength and other characteristics using differential scanning calorimeter (DSC) and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). DSC measurements showed that incubation at $37^{\circ}C$ rather than $4^{\circ}C$ caused marked changes in thermal transition, and MS displayed similar thermal curves (three endothermic transitions) to MFP and SC alone. After incubation at $37^{\circ}C$ for 2 hrs, the viscosity (cP) of MS increased (p<0.05) due to induction by MTGase, whereas no differences were observed at $4^{\circ}C$. However, gel strength values were no different, regardless of incubation temperatures and times. Future research will address how longer incubation times affect the functionality of protein mixed gels mediated by MTGase.
