Hsa-miR-422a Originated from Short Interspersed Nuclear Element Increases ARID5B Expression by Collaborating with NF-E2

  • MicroRNAs (miRNAs) are a class of small non-coding RNAs that regulate the expression of target messenger RNA (mRNA) complementary to the 3' untranslated region (UTR) at the post-transcriptional level. Hsa-miR-422a, which is commonly known as miRNA derived from transposable element (MDTE), was derived from short interspersed nuclear element (SINE). Through expression analysis, hsa-miR-422a was found to be highly expressed in both the small intestine and liver of crab-eating monkey. AT-Rich Interaction Domain 5 B (ARID5B) was selected as the target gene of hsa-miR-422a, which has two binding sites in both the exon and 3'UTR of ARID5B. To identify the interaction between hsa-miR-422a and ARID5B, a dual luciferase assay was conducted in HepG2 cell line. The luciferase activity of cells treated with the hsa-miR-422a mimic was upregulated and inversely downregulated when both the hsa-miR-422a mimic and inhibitor were administered. Nuclear factor erythroid-2 (NF-E2) was selected as the core transcription factor (TF) via feed forward loop analysis. The luciferase expression was downregulated when both the hsa-miR-422a mimic and siRNA of NF-E2 were treated, compared to the treatment of the hsa-miR-422a mimic alone. The present study suggests that hsa-miR-422a derived from SINE could bind to the exon region as well as the 3'UTR of ARID5B. Additionally, hsa-miR-422a was found to share binding sites in ARID5B with several TFs, including NF-E2. The hsa-miR-422a might thus interact with TF to regulate the expression of ARID5B, as demonstrated experimentally. Altogether, hsa-miR-422a acts as a super enhancer miRNA of ARID5B by collaborating with TF and NF-E2.

  • 전형적인 마이크로 RNA는 주로 해당 마이크로RNA의 호스트 유전자와 동시에 발현하는 형상을 보인다. 마이크로RNA miR-7b와 그 호스트 유전자인 FICT는 유전자 발현 조절부위인 프로모터를 함께 공유할 것으로 추정되며, 이는 이 유전자들의 뇌 특이적인 발현 양상에 기여할 것으로 추정된다. 바이오인포메틱 방법을 이용하여 사람과 마우스의 miR-7혹은 miR-7b의 프로모터 부위가 상호 유사성을 가짐을 확인하였고, 이 부위에 다양한 전자조절 부위가 있는 것을 확인 하였다. 또한 이 가설을 증명하기 위하여 형광발현 리포터 유전자 시스템을 사용하여 형광발현 벡터에 마이크로 RNA miR-7b와 그 호스트 유전자인 FICT의 5' 전부위를 클로링하여 프로모터의 활성정도를 다양한 세포주에서 확인하였다. 이 결과를 통하여 마이크로 RNA와 그 호스트 유전자인 FICT의 프로모터에는 네거티브 유전자 조절인자를 포함하는 것을 확인 할 수 있었다.

LINC00703 Acts as a Tumor Suppressor via Regulating miR-181a/KLF6 Axis in Gastric Cancer

  • Purpose: Long noncoding RNA 00703 (LINC00703) was found originating from a region downstream of Kruppel-like factor 6 (KLF6) gene, having 2 binding sites for miR-181a. Since KLF6 has been reported as a target of miR-181a in gastric cancer (GC), this study aims to investigate whether LINC00703 regulates the miR-181a/KLF6 axis and plays a functional role in GC pathogenesis. Materials and Methods: GC tissues, cell lines, and nude mice were included in this study. RNA binding protein immunoprecipitation (RIP) and pull-down assays were used to evaluate interaction between LINC00703 and miR-181a. Quantitative real-time polymerase chain reaction and western blot were applied for analysis of gene expression at the transcriptional and protein levels. A nude xenograft mouse model was used to determine LINC00703 function in vivo. Results: We revealed that LINC00703 competitively interacts with miR-181a to regulate KLF6. Overexpression of LINC00703 inhibited cell proliferation, migration/invasion, but promoted apoptosis in vitro, and arrested tumor growth in vivo. LINC00703 expression was found to be decreased in GC tissues, which was positively correlated with KLF6, but negatively with the miR-181a levels. Conclusions: LINC00703 may have an anti-cancer function via modulation of the miR-181a/KLF6 axis. This study also provides a new potential diagnostic marker and therapeutic target for GC treatment.

MiR-26a promotes apoptosis of porcine granulosa cells by targeting the 3β-hydroxysteroid-Δ24-reductase gene

  • Objective: Apoptosis of ovarian granulosa cells (GCs) affects mammalian follicular development and fecundity. This study aimed to explore the regulatory relationship between microRNA-26a (miR-26a) and the 3β-hydroxysteroid-Δ24-reductase gene (DHCR24) gene in porcine follicular granular cells (pGCs), and to provide empirical data for the development of methods to improve the reproductive capacity of pigs. Methods: The pGCs were transfected with miR-26a mimic, miR-26a inhibitor and DHCR24-siRNA in vitro. The cell apoptosis rate of pGCs was detected by the flow cytometry. The secretion levels of estradiol (E2) and progesterone (P) in pGCs were detected by enzyme-linked immunosorbent assay. Double luciferase validation system was used to detect the binding sites between miR-26a and DHCR24 3'-UTR region. Qualitative real-time polymerase chain reaction and Western blotting were used to verify the DHCR24 mRNA and protein expression in pGCs, respectively, after transfecting with miR-26a mimic and miR-26a inhibitor. Results: Results showed that enhancement of miR-26a promoted apoptosis, and inhibited E2 and P secretion in pGCs. Meanwhile, inhibition of DHCR24 also upregulated the Caspase-3 expression, reduced the BCL-2 expression, promoted pGCs apoptosis, and inhibited E2 and P secretion in pGCs. There were the binding sites of miR-26a located within DHCR24 3'-UTR. Up-regulation of miR-26a inhibited DHCR24 mRNA and protein expression in pGCs. Conclusion: This study demonstrates that miR-26a can promote cell apoptosis and inhibit E2 and P secretion by inhibiting the expression of DHCR24 in pGCs.

MiR-199a/b-5p Inhibits Lymphangiogenesis by Targeting Discoidin Domain Receptor 1 in Corneal Injury

  • Discoidin domain receptor 1 (DDR1) is involved in tumorigenesis and angiogenesis. However, its role in lymphangiogenesis has been unknown. Here, we tested whether downregulation of DDR1 expression by miR-199a/b can suppress lymphangiogenesis. We also aimed to identify miRNA target site(s) in the 3' untranslated region (UTR) of DDR1. Transfection with miR-199a/b-5p mimics reduced expression of DDR1 and tube formation in primary human dermal lymphatic endothelial cells, whereas miR-199a/b-5p inhibitors showed the opposite effects. Critically, injection of miR-199a/b-5p mimics suppressed DDR1 expression and lymphangiogenesis in a corneal alkali-burn rat model. The three well-conserved seed matched sites for miR-199a/b-5p in the DDR1 3'-UTR were targeted, and miRNA binding to at least two sites was required for DDR1 inhibition. Our data suggest that DDR1 promotes enhanced lymphangiogenesis during eye injury, and miR-199a/b-5p suppresses this activity by inhibiting DDR1 expression. Thus, this miRNA may be useful for the treatment of lymphangiogenesis-related eye diseases.

Overexpression of Long Non-Coding RNA MIR22HG Represses Proliferation and Enhances Apoptosis via miR-629-5p/TET3 Axis in Osteosarcoma Cells

  • In this study, we evaluated the mechanism of long non-coding RNA MIR22 host gene (LncRNA MIR22HG) in osteosarcoma cells. Forty-eight paired osteosarcoma and adjacent tissues samples were collected and the bioinformatic analyses were performed. Target genes and potential binding sites of MIR22HG, microRNA (miR)-629-5p and tet methylcytosine dioxygenase 3 (TET3) were predicted by Starbase and TargetScan V7.2 and confirmed by dual-luciferase reporter assay. Cell Counting Kit-8, colony formation and flow cytometry assays were utilized to determine the viability, proliferation and apoptosis of transfected osteosarcoma cells. Pearson's analysis was introduced for the correlation analysis between MIR22HG and miR-629-5p in osteosarcoma tissue. Relative expressions of MIR22HG, miR-629-5p and TET3 were measured by quantitative real-time polymerase chain reaction or Western blot. MiR-629-5p could competitively bind with and was negatively correlated with MIR22HG, the latter of which was evidenced by the high expression of miR-629-5p and low expression of MIR22HG in osteosarcoma tissues. Overexpressed MIR22HG repressed the viability and proliferation but enhanced apoptosis of osteosarcoma cells, which was reversed by miR-629-5p upregulation. TET3 was the target gene of miR-629-5p, and the promotive effects of upregulated miR-629-5p on the viability and proliferation as well as its repressive effect on apoptosis were abrogated via overexpressed TET3. To sum up, overexpressed MIR22HG inhibits the viability and proliferation of osteosarcoma cells, which was achieved via regulation of the miR-629-5p/TET3 axis.

  • 미만성 거대 B 세포 림프종(DLBCL)은 비호지킨 림프종에서 가장 흔한 형태이다. DLBCL에서 약물치료에 대한 연구가 많은 진전을 보였지만, 아직 많은 환자의 경우 DLBCL로 인한 사망률이 상당하다. 따라서 DLBCL에 대한 이해와 새로운 표적 치료제의 개발이 필요하다. PDE (인산이에스테르 가수분해효소)4B는 최근 시행된 유전자 발현 프로파일링에서 약제내성을 가지는 DLBCL에서 과발현 되는 유전자로 밝혀졌다. PDE4B의 주된 역할은 이차전달자인 고리형 AMP (cylclic AMP, cAMP)를 5'AMP로의 가수분해를 촉진시켜 cAMP를 비활성화 시키는 것이다. cAMP는 B 세포에서 세포증식 저해와 세포사멸을 유도하고 PDE4B는 B 세포에서 이러한 cAMP의 기능을 소멸시키는 것으로 알려져 있다. 그러나 PDE4B의 과발현이 어떤 기작에 의한 것인지는 연구가 미비하다. 본 논문에서는 비정상적으로 발현된 마이크로 RNA (microRNA, miRNA)가 PDE4B의 과발현에 관련되어 있을 것이라는 가정하에 실험을 진행하였다. PDE4B 3'-UTR에는 세 개의 miR-23b 예상 결합부위가 존재하고, 이는 luciferase reporter assay를 통해서 확인하였다. 흥미롭게도, miR-23b 결합 부위들은 인간에서부터 도마뱀에 이르기까지 진화적으로 보존되어 있었고, 이는 세포 생리학적 측면에서 PDE4B-miR-23b 사이의 상호작용이 중요한 역할을 수행함을 암시하고 있다. miR-23b의 과발현은 PDE4B의 mRNA 발현을 감소시키고 세포내의 cAMP의 농도를 증가시켰다. 뿐만 아니라, miR-23b의 발현은 아데닐산고리화효소(adenylyl cyclase)의 활성약제인 forskolin이 처리된 경우에만 DLBCL 세포들의 증식과 생존을 억제하였다. 이는 miR-23b는 PDE4B 발현을 감소시킴으로써 세포증식과 생존을 조절함을 보여주는 것이다. 이를 통해 생각해 볼 때, miR-23b는 PDE4B를 억제함으로써 DLBCL에서 나타나는 항암제 내성을 극복할 수 있고, 따라서 miR-23b는 잠재적 종양 억제자로서 효과적인 치료적 타겟으로 예상된다.

The rs61764370 Functional Variant in the KRAS Oncogene is Associated with Chronic Myeloid Leukemia Risk in Women

  • Background: Chronic myeloid leukemia (CML) is one of the most frequent hematopoietic malignancies in the elderly population; however, knowledge is limited regarding the genetic factors associated with increased risk for CML. Polymorphisms affecting microRNA (miRNA) biogenesis or mRNA:miRNA interactions are important risk factors in the development of different types of cancer. Thus, we carried out a case-control study to test the association with CML susceptibility of gene variants located in the miRNA machinery genes AGO1 (rs636832) and GEMIN4 (rs2740348), as well as in the miRNA binding sites of the genes BRCA1 (rs799917) and KRAS (rs61764370). Materials and Methods: We determined the genotype of 781 Mexican-Mestizo individuals (469 healthy subjects and 312 CML cases) for the four polymorphisms using TaqMan probes to test the association with CML susceptibility. Results: We found a borderline association of the minor homozygote genotype of the KRAS_rs61764370 polymorphism with an increased risk for CML susceptibility (P = 0.06). After gender stratification, this association was significant only for women (odds ratio [OR] = 13.41, P = 0.04). The distribution of the allelic and genotypic frequencies of the four studied SNPs was neither associated with advanced phases of CML nor treatment response. Conclusions: To the best of our knowledge, this study is the first to show a significant association of the KRAS_rs61764370 SNP with CML. To further determine such an association of with CML susceptibility, our results must be replicated in different ethnic groups.

MicroRNA Analysis in Normal Human Oral Keratinocytes and YD-38 Human Oral Cancer Cells

  • MicroRNAs (miRNAs) are small non-coding RNAs that mediate gene expression at the post-transcriptional level by degrading or repressing targeted mRNAs. These molecules are about 21-25 nucleotides in length and exert their effects by binding to partially complementary sites in mRNAs, predominantly in the 3'-untranslated region (3'-UTR). Recent evidence has demonstrated that miRNAs can function as oncogenes or tumor suppressors through the modulation of multiple oncogenic cellular processes in cancer development, including initiation, cell proliferation, apoptosis, invasion and metastasis. In our present study, we examined the expression profile of miRNAs related to oral cancer cell growth inhibition using normal human oral keratinocytes (NHOK) and YD-38 human oral cancer cells. By miRNA microassay analysis, 40 and 31 miRNAs among the 1,769 examined were found to be up- and down-regulated in YD-38 cells compared with NHOK cells, respectively. Using qRT-PCR analysis, the expression levels of miR-30a and miR-1246 were found to be increased in YD-38 cells compared with NHOK cells, whereas miR-203 and miR-125a were observed to be decreased. Importantly, the overexpression of miR-203 and miR-125a significantly inhibited the growth of YD-38 cells. This finding and the microarray data indicate the involvement of specific miRNAs in the development and progression of oral cancer.

MicroRNA-200a/210의 인체 지방 유래 중간엽 줄기세포 골분화 및 증식 조절 기전 (MicroRNA-200a/210 Controls Proliferation and Osteogenic Differentiation of Human Adipose Tissue Stromal Cells)

  • MicroRNA는 인체 지방 유래 중간엽 줄기세포(hADSC)의 분화와 증식을 조절할 수 있으나 hADSC에서 miR-200a와 miR210의 역할은 아직까지 보고된 바가 없다. 표적 mRNA의 3' UTR 에 결합할 수 있는 construct를 이용한 luciferase assay를 통하여 microRNA가 직접적으로 표적 mRNA에 결합하는 것을 확인 하였다. hADSC에서 miR-200a 과발현은 ZEB2의 발현 감소를 통해 hADSC의 분화와 증식을 증가시키는 것을 확인할 수 있었으며, miR210의 과발현은 IGFBP3의 발현 감소를 통해 hADSC의 증식을 감소시키는 반면, 분화는 증가시키는 것을 확인 할 수 있었다. hADSC에서 miR210의 발현 억제는 표적유전자의 발현을 증가시킴으로써 hADSC의 증식을 증가시키는 반면, 분화를 억제시키는 것을 확인할 수 있었다. luciferase assay통하여 microRNA-200a/210의 과발현이 대조군에 비해 표적 유전자인 ZEB2/ IGFBP3의 luciferase 활성화를 감소시키는 것을 확인하였으며, hADSC에서 ZEB2/ IGFBP3 발현 억제는 microRNA-200a/210 과발현과 유사한 효과를 나타내는 것을 확인할 수 있었다. 이러한 결과는 microRNA-200a/210가 hADSC의 분화와 증식을 각각의 표적 유전자인 ZEB2/ IGFBP3을 통해 조절한다는 것을 나타낸다.
