Ecogenetical Characteristics of Dolichospermum in Bukhan River (북한강 수계에서 Dolichospermum의 유전생태학적 특성 연구)

  • This study analyzed the occurrence pattern of Dolichospermum (= Anabaena) in the Bukhan river from March 2012 to December 2014 in order to identify the genotypes of Dolichospermum. Furthermore, 16S rRNA were analyzed to identify the genotypes of Dolichospermum that occurred in 2015 which were then compared to the reference sequence deposited at NCBI. During this period, the occurrence of Dolichospermum was highly correlated to water temperature. In the year 2012 and 2013, Dolichospermum appeared in Lake Cheongpyeong (CP), Sambong (SB), and Lake Paldang (P2) between July and August. However, in 2014, it appeared in SB and P2, but not in CP. This reduction in appearance was attributed to the decreased inflow to Lake Uiam as a result of low rainfall in 2014 as compared to 2012. In July 2015, the Dolichospermum 16S rRNA genotype was confirmed in five locations; Lake Cheongpyeong (CP), Seojong (SJ), Songchon Sewage Treatment Plant (SC), Joan (P4), and Lake Paldang (PD). Anabaena crassa of spiral clone, A. planctonica of linear clone, and A. circinalis of spiral clone exhibited high genetic similarity with the reference sequence. The 16r RNA genotype showed approximately 3 % sequence variation between the locations and were more similar to each other in locations that were closer.

Comparison of Multi-Mode Simulation and SimMan(R) Simulation on Evaluation of Nursing Care for Patients with Dyspnea (호흡곤란환자 간호의 실습교육평가에서 Multi-mode와 SimMan(R) 시뮬레이션 활용 비교)

  • Purpose: The purpose of this study was to compare the effects of nursing simulations between multi-mode simulation and $SimMan^{(R)}$ simulation on nursing students' nursing competency, satisfaction, and simulation participation experience in nursing care for patients with dyspnea. Method: Subjects were 107 junior nursing students in R College of Nursing in Seoul from June to November in 2009. Students were allocated to either a Multi-mode simulation group or a $SimMan^{(R)}$ simulation group. The multi-mode simulation consisted of standardized patients combined with Vital Sim, comparing the $SimMan^{(R)}$ group. Result: The Multi-mode simulation group reported higher levels of nursing competency (p=.017) but lower communication skills (p<.001) compared to the $SimMan^{(R)}$ simulation group. There were no meaningful differences in satisfaction of simulation education between the two groups. The $SimMan^{(R)}$ simulation group received a substantial health assessment and the Multi-mode simulation group experienced reality in caring for standardized patients. Conclusion: The findings of this study demonstrated that Multi-mode simulation has similar educational effects like $SimMan^{(R)}$ simulation and suggest that nurse educators should match simulation fidelity with educational goals for effective education.

  • A method for the identification of Nocardia seriolae, the causative agent of nocardiosis in cultured fishes, using PCR was developed in the study. A PCR primer set specific to N. seriolae was designed based on 16S-23S rRNA sequence of various Nocardia species accessed in GenBank. Designed PCR primer set, Nseri-F (5'-GCA AAC TCT TCG AAC AGT CG-3') and Nseri-R (5'-GGA TAT CAG GAC TTA CCG GC-3'), amplifies the target regions of N. seriolae only, but not 4 other Nocardia species, N. asteroides, N. crassostreae, N. farcinica and N. salmonicida.

Assessing the Age and Growth of the Butter Clam Saxidomus purpuratus in Jinhae Bay Using Transmitted Light (투과광을 이용한 한국 진해만 개조개(Saxidomus purpuratus)의 연령과 성장)

  • The age and growth of butter clams Saxidomus purpuratus were estimated using transmitted light on the shells of 364 samples from January 2017 to December 2017 in Jinhae Bay. Based on monthly variation in the marginal index (MI) of the shell, it is assumed that rings are formed once a year during the period from July to August in this species. The relationship between shell length (SL; mm) and shell height (SH; mm) was expressed by the equation SH=0.8053SL-2.9636 ($R^2=0.94$) and between SL and shell width (SW; mm) by the equation SW=0.5648SL-3.7105 ($R^2=0.90$). The relationship between SL and total weight (TW; g) was expressed by the following equation: $TW=0.00009SL^{3.2141}$ ($R^2=0.96$). von Bertalanffy's growth parameters were estimated using the regression wizard in the SigmaPlot computer program (Systat Software, Inc., v. 10.0). The maximum shell length ($SL_{\infty}$) was 126.16 mm, growth rate was 0.2030/year, theoretical age at shell length 0 ($t_0$) was -0.52 years, and asymptotic total weight ($TW_{\infty}$) was 509.17 g. Growth curves for SL and TW fitted to the von Bertalanffy's equation were expressed as follows: $SL_t=126.16(1-e^{-0.2030(t+0.52)})$, $TW_t=509.17(1-e^{-0.2030(t+0.52)})^{3.2141}$.

Wedelolactone Promotes the Chondrogenic Differentiation of Mesenchymal Stem Cells by Suppressing EZH2

  • Background and Objectives: Osteoarthritis (OA) is a degenerative disease that leads to the progressive destruction of articular cartilage. Current clinical therapeutic strategies are moderately effective at relieving OA-associated pain but cannot induce chondrocyte differentiation or achieve cartilage regeneration. We investigated the ability of wedelolactone, a biologically active natural product that occurs in Eclipta alba (false daisy), to promote chondrogenic differentiation. Methods and Results: Real-time reverse transcription-polymerase chain reaction, immunohistochemical staining, and immunofluorescence staining assays were used to evaluate the effects of wedelolactone on the chondrogenic differentiation of mesenchymal stem cells (MSCs). RNA sequencing, microRNA (miRNA) sequencing, and isobaric tags for relative and absolute quantitation analyses were performed to explore the mechanism by which wedelolactone promotes the chondrogenic differentiation of MSCs. We found that wedelolactone facilitates the chondrogenic differentiation of human induced pluripotent stem cell-derived MSCs and rat bone-marrow MSCs. Moreover, the forkhead box O (FOXO) signaling pathway was upregulated by wedelolactone during chondrogenic differentiation, and a FOXO1 inhibitor attenuated the effect of wedelolactone on chondrocyte differentiation. We determined that wedelolactone reduces enhancer of zeste homolog 2 (EZH2)-mediated histone H3 lysine 27 trimethylation of the promoter region of FOXO1 to upregulate its transcription. Additionally, we found that wedelolactone represses miR-1271-5p expression, and that miR-1271-5p post-transcriptionally suppresses the expression of FOXO1 that is dependent on the binding of miR-1271-5p to the FOXO1 3'-untranscribed region. Conclusions: These results indicate that wedelolactone suppresses the activity of EZH2 to facilitate the chondrogenic differentiation of MSCs by activating the FOXO1 signaling pathway. Wedelolactone may therefore improve cartilage regeneration in diseases characterized by inflammatory tissue destruction, such as OA.

Factors Influencing on the Attitude toward withdrawal of life-sustaining treatment of Nursing College Students (간호대학생의 연명치료 중단에 대한 태도에 영향을 미치는 요인)

  • This study was a descriptive study to investigate the factors influencing on attitude toward withdrawal of life-sustaining treatment of nursing students' consciousness of biomedical ethics, good death recognition and meaning of life. The subjects of this study were 293 nursing students attending college at P city. Data were collected for two weeks from May 1, 2018 to May 11, 2018. The statistical method was performed with SPSS WIN 22.0 using a complex sampling analysis, pearson's correlation coefficients, and a multiple regression analysis. The result of this study was significant positive correlation between attitude toward withdrawal of life-sustaining treatment and consciousness of biomedical ethics(r=.266, p<.001), good death recognition(r=.373, p<.001), meaning of life(r=.122, p=.037). The meaning of life was significant positive correlation consciousness of biomedical ethics(r=.294, p<.001), good death recognition(r=.230, p<.001). The good death recognition was significant positive correlation consciousness of biomedical ethics(r=.306, p<.001). Factors Influencing on the attitude toward withdrawal of life-sustaining treatment was good death recognition(${\beta}=.32$, p<.001) and consciousness of biomedical ethics(${\beta}=.16$, p=.004). These factors explained about 16% of the variance. Therefore it is necessary to develop an educational program to positively raise the attitude of consciousness of biomedical ethics and good death recognition.

  • The full sequence analysis of 16 different $\beta-actin$ genes isolated from a single Rivulus marmoratus was performed. The numbers of isolated $\beta-actin$ genes varied from 1764 to 1769. They showed different amino acid residues at the exon 1 region. We named this new gene R. marmoratus $\beta-actin$ 2 gene. Intraspecific variation of R. marmoratus $\beta-actin$ 2 gene was also examined. Major differences of 16 isolated $\beta-actin$ genes, compared to already reported R. marmoratus $\beta-actin$ gene (GenBank AF168615), were observed in both deduced amino acid sequences (1~2%) and intron 2 sequences (4~5%). In this paper, we confirmed the intraspecific variation of $\beta-actin$ gene in this species.

Effects of Feeding Fermented Colostrum Feed on the Growth to Piglets (발효초유사료 급여가 자돈의 성장에 미치는 영향)

  • This study was carried out to assess the fermentation properties of lactic acid bacteria (LAB) isolated from bovine colostrum and effects of feeding fermented colostrum feed on the growth to piglet. A total of 427 colonies were isolated from bovine colostrum on the BCP plate count agar. These LAB isolated were subcultured in 10% reconstituted skim milk, and seven strain thereof were selected for their highest acid productions, and two strain thereof were finally selected for their excellent sugar utilization. These strains were identified as Streptococcus thermophilus and Streptococcus macedonicus based on l6S rDNA sequencing data, named S. thermophilus CNB-11 and S. macedonicus CNB-11 respectively. For fermentation profiles, sugar utilization, acid production and viable cell counts were excellent in S. thermophilus CNB-11 as compared with S. macedoniclts CNB-11 after 48 hour. The effect of feeding fermented colostrum feed 0.5% using S. thermophilus CNB-11 was investigated for growth rate, analysis of blood and incidence of diarrhea. 24 heads of piglets were divided into two groups: the experimental and the control of 12 animals each. The average growth rate in the pigs fed fermented colostrum feed was higher 16.73% compared with control diet (p<0.05). There were no differences in the concentrations of blood glucose, cholesterol, albumin and globulin in pigs fed fermented colostrum feed as compared with control piglets. Incidence of diarrhea was no in pigs fed fermented colostrum feed as compared with control piglets.

Mortality Analysis of Open Heart Surgery (75 Cases) (개심수술후의 사망예에 대한 임상적 고찰)

  • From 1976 through June 1980, 75 patients underwent Open heart operation at Korea University Hospital.Of the 75 patients, 39 were congenital heart cases and 36 were acquired heart disease cases. 39 cases of congenital heart disease were consisting of 16 T.O.F.,4 A.S.D., 10 V.S.D., 3 P.S., 1 P.D.A., 1 V.S.D. + Mi, 1 Truncus arteriosus, 1 Ebstein, 1 D.C.R.V., 1 Single ventricle. Among 36 valvular replacement cases, 18 cases of MVR, 3 cases of AVR, 6 cases of Double valve replacement, and 10 cases of Open Mitral commissurotomy, were performed. Postoperative mortality rate of congenital heart disease was 25.6% and that of acquired heart disease was 8.3%. Overall mortality rate of open heart surgery was 17.3%. Among 16 cases of postoperative death cases, 5 cases of autopsy were performed. Postoperative cause of death of our series were intracranial bleeding, pacemaker failure, low output syndrome, protamine anaphylaxis, bleeding, prosthetic valve embolism, C V A, miliary tuberculosis, hypothermia due to pump failure.

Indirect Evidence on Sex Reversal with Sex Ratio of Tegillarca granosa (Bivalvia: Arcidae) and Ruditapes philippinarum (Bivalvia: Veneridae)

  • This study attempts to propose the possibility of the sex reversal in Tegillarca granosa and Ruditapes philippinarum by confirming the changes in the sex ratio with the shell length (SL) in the same population level. For analysis of sex ratio with SL, 1500 individuals of T. granosa (SL 10.1-45.0 mm) and 712 individuals of R. philippinarum (SL 15.1-70.0 mm) were used. Sex was analyzed histologically. The average sex ratios (F:M) of T. granosa and R. philippinarum were 1:1.22 and 1:0.96, respectively. However, sex ratio was found to differ when the clams were divided into groups according to SL in 5.0 mm intervals. Both species displayed the tendency of increase in the proportion of female with increase in SL. In this study, changes in the sex ratio in accordance with the growth of T. granosa and R. philippinarum are determined to be indirect evidence that signifies their sex reversal.
