감광제 광산화에 의한 Bisphenol A 안정성 감소 (Decreased Stability of Bisphenol A by Photosensitization)

  • 내분비교란물질인 BPA는 식품의 포장용기나 젖병 등의 제조에 이용되며, 라이보플라빈 광산화에 의해 안정성이 감소하는 것으로 보고된 바 있다. 본 연구에서는 광산화 시 메틸렌블루, 로즈벵갈, 라이보플라빈, 또는 이산화티타늄 등의 감광제 종류에 따른 BPA안정성을 비교하였고, sodium azide($NaN_3$) 농도에 따른 BPA안정성을 확인하였다. BPA 농도는 라이보플라빈, 로즈벵갈, 메틸렌블루 순서로 유의적으로 감소하였지만(p<0.05), 이산화티타늄 첨가 시료와 대조구인 감광제 무첨가군의 BPA 농도는 유의적인 차이가 없었다(p>0.05). BPA의 안정성은 메틸렌블루 농도 의존적으로 감소하였으며, $NaN_3$ 농도가 증가할수록 BPA 안정성은 증가하였다. 이는 메틸렌블루 광산화에 의한 BPA농도 감소에 일중항산소가 관여함을 의미한다.

천마반하탕이 뇌허혈에 미치는 기전 연구 (Mechanism Study of Cheonmabanhwa-Tang on the Cerebral Ischemia in Rats - Focusing arround Improvement in Changes of Cerebral Hemodynamics -)

  • Cheonmabanhwa-Tang(CBT) has been used in the Oriental Medicine for many centuries as a therapeutic agent for dizziness due to Poong-Dam. This Study was designed to investigate the mechanism of Prescription on cerebral hemodynamics [regional cerebral blood flow(rCBF) and pial arterial diameter(PAD)J in cerebral ischemia rats, The results in cerebral ischemic rats were as follows: Both rCBF and PAD were significantly and stably increased by CBT (10 ㎎/㎏, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. Pretreatment with indomethacin(1 ㎎/㎏, i.p.), an inhibitor of cyclooxygenase significantly but unstably increased the CBT-induced increases in PAD as well as rCBF during the period of cerebral reperfusion. Pretreatment with methylene blue(10 (.1.㎍/㎏, i.p.), an inhibitor of guanylate cyclase significantly but unstably increased the CBT-induced increases in PAD as well as rCBF during 150 minutes of cerebral reperfusion, but decreased stably the CBT-induced increases in rCBF and PAD after 180 minutes of cerebral reperfusion. In conclusion, the present authors thought that CBT caused effect on cerebral hemodynamics via mediation of cyclooxygenase.

Acinetobacter sp.1의 일산화탄소 산화효소의 특성 (Carbon Monoxide Dehydrogenase in Cell Extracts of an Acinetobacter Isolate)

  • 일산화탄소를 이용하여 자가영양적으로 성장한 Acinetobacter sp. 1 의 세포추출액은 혐 기성 실험조건하에서 thionin, methylene blue, 2,6-dichlorophenol-indophenol둥올 일산화탄소의 산회를 위한 전자수용체로 사용할 수 있었으나 NAD, NADP, FAD, 또는 FMN등은 천자수용체로 이용하지 못하였다. 이 세균에 존재하는 일산화탄소 산화효소는 유도효소로 밝혀졌고, pH 7.5와 $60^{\circ}C$에서 최대의 활성을 나타내었다. 이 효소의 활성화에너지는 6.1kcal/mol (25.5 kJ/mol)이며 일산화탄소에 대한 Km값은 $154{\mu}M$로 밝혀졌다. 그리고 잘 알려진 몇가지 금속 chelat tIng agent와 2가의 양이온들은 이 효소의 활성에 거의 영향을 미치지 않았는데 $Cu^{2+}$ 이온만은 이 효소의 활성을 완전히 억제시켰다. 또한 이 효소는 포도당과 숙신산에 의해 활성이 저해되었으며, hydrogenase의 활성도 나타내었다. 그리고 Acinetobacter sp. 1의 일산화탄소 산화효소는 Pseudomonas carboxydohydrogena의 일산화탄소 산화효소와 연역학적인 연관성이 없는 것으로 나타났다.

Photodynamic Therapy에 의한 산화적 스트레스 조건에서 Helicobacter pylori의 Fur 단백질의 역할 (The Role of Helicobacter pylori's Fur Protein in the Oxidative Stress Induced by Photodynamic Therapy)

  • Helicobacter pylori균의 ferric uptake regulator (Fur) 단백질의 산화적 손상에 대한 역할을 연구하였다. H. pylori균의 fur 유전자를 제거한 돌연변이체를 만들고 wild type H. pylori균과 돌연변이체 균의 산화적 스트레스에 대한 반응을 비교하였다. 산화적 스트레스는 methylene blue와 660 nm 파장의 빛을 이용하는 광역학적 치료방법으로 유도하였다. 산화적 스트레스를 가한 실험조건에서 wt H. pylori와 돌연변이체의 생존력, DNA 손상의 정도를 비교 검토하였다. 그 결과 fur 유전자가 제거된 돌연변이체의 생균수가 wt에 비해 10,000배 가량 감소한 것을 알 수 있었으며 DNA의 산화적 손상의 marker인 8-hydroxy-2-deoxyguanosine (8-OHdG)의 양도 fur 유전자 제거된 돌연변이에서 wild type에 비해 3배 정도 더 생성됨을 확인하였다. 따라서 본 실험결과 H. pylori균의 fur 유전자가 PDT법으로 유도한 산화적 스트레스에 방어 기작을 하는 것으로 사료된다.

콜라겐을 첨가한 퍼머넌트웨이브제의 개발 (The Study of Development of permanent wave for Additives of Collagen)

  • 본 연구에서는 콜라겐의 처방이 퍼머넌트웨이브 시술시 미치는 영향을 알아보기 위하여 정상모에 콜라겐을 농도별 처방한 퍼머넌트웨이브제로 퍼머넌트웨이브를 시술한 후 모발에 컬의 형성력, 굵기 관찰, 인장 강도, 메틸렌블루 흡광도, 수분 감소량 등의 변화를 측정하여 비교분석하였다. 결론적으로 퍼머넌트웨이브제의 콜라겐 처방 여부에 따라 모발의 굵기, 모발의 인장강도, 모발의 수분 감소량, 흡광도가 처방하지 않은 환원제에 대비하여 감소하였으며, 처방된 콜라겐의 농도가 증가할수록 모발의 손상도가 줄어드는 결과를 보였다. 그러나 콜라겐의 농도의 크기가 커질수록 퍼머넌트웨이브 시술시 컬의 형성력이 떨어지는 결과가 나타났다. 그러므로 퍼머넌트웨이브 시술시 모발의 손상도를 줄이기 위한 콜라겐 처방 농도는 2%가 적당하였다.

노궁(勞宮)($PC_8$) 직자(直刺)가 백서(白鼠)의 뇌혈류력학(腦血流力學)에 미치는 영향 (The Effects of Perpendicular Needling Laogong ($PC_8$) on the Improvement of Cerebral Hemodynamics in Rats)

  • Objectives : This study was designed to investigate the effects of acupuncturing $PC_8$ used perpendicular needling method determine the mechanism of action of acupuncturing $PC_8$ by measuring the changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats. Methods : This study also investigated the effects of acupuncturing $PC_8$ on the change of rCBF in cerebral ischemic rats, and revealed the mechanism of its action. In addition, the effects of acupuncturing $PC_8$ on focal ischemic brain injury was studied in cerebral ischemic rats. Results : 1. Acupuncturing $PC_8$ significantly increase rCBF but decreased MABP in normal rats. 2. Acupuncturing $PC_8$ increased of rCBF was significantly inhibited by pretreatment with indomethacin (1mg/kg, i.p.), an inhibitor of cyclooxygenase in normal rats. 3. Acupuncturing $PC_8$ increased of rCBF was significantly inhibited by pretreatment methylene blue (10 ${\mu}g$/kg, i.p.), an inhibitor of guanylate cyclase in normal rats. 4. Acupuncturing $PC_8$ was significantly improved the rCBF than control group increased unstable in cerebral ischemic rats. 5. Acupuncturing $PC_8$ was not significantly improved the rCBF than control group by pretreatment with indomethacin (1mg/kg, i.p.), an inhibitor of cyclooxygenase in cerebral ischemic rats. 6. Acupuncturing $PC_8$ was significantly increased the rCBF than control group by pretreatment methylene blue ($10{\mu}g$/kg, i.p.), an inhibitor of guanylate cyclase in cerebral ischemic rats. Conclusions : In conclusion, our study suggested that acupuncturing $PC_8$ can increase rCBF in normal state, and improve stability of rCBF in ischemic state. In addition, we suggested that mechanisms related with acupuncturing $PC_8$ was involved in the guanylate cyclase pathway.

시근과 시엽이 Phenylephrine으로 유발된 가토의 수축혈관에 미치는 영향 (Effects of Diospyros kaki L. Radix or Diospyros kaki L. Folium on Arterial Contraction Induced by Phenylephrine in Rabbit)

  • Objectives : This study was undertaken to define the effect of Diospyros kaki L. Radix or Diospyros kaki L. Folium on phenylephrine-induced arterial contraction and the mechanism of Diospyros kaki L. Radix or Diospyros kaki L. Foliuminduced relaxation. Methods : In order to investigate the effect of Diospyros kaki L. Radix or Diospyros kaki L. Folium on contracted rabbit carotid arterial strips, transverse strips with intact or damaged endothelium were used for the experiment using organ bath. Diospyros kaki L. Radix or Diospyros kaki L. Folium extract was infused into contracted arterial strips induced by phenylephrine. To analyze the mechanism of Diospyros kaki L. Radix or Diospyros kaki L. Folium-induced relaxation, Diospyros kaki L. Radix or Diospyros kaki L. Folium extract was infused into contracted arterial strips induced by phenylephrine after treatment with indomethacin, $N{\omega}-nitro-L-arginine$, methylene blue or tetraethylammonium chloride, and $Ca^{2+}$ was infused into contracted arterial strips induced by phenylephrine after treatment of Diospyros kaki L. Radix or Diospyros kaki L. Folium in a $Ca^{2+}$-free solution. Results : Diospyros kaki L. Radix or Diospyros kaki L. Folium showed relaxation effect on arterial strip with endothelium contracted by phenylephrine, but in the strips without endothelium, Diospyros kaki L. Radix or Diospyros kaki L. Folium-induced relaxation was significantly inhibited. The endothelium-dependent relaxation induced by Diospyros kaki L. Radix or Diospyros kaki L. Folium was decreased by pretreatment with $N{\omega}-nitro-L-arginine$ or methylene blue but it was not observed in the strips pretreated with indomethacin or tetraethylammonium chloride. When $Ca^{2+}$ was applied to the strips which were contracted by phenylephrine in a $Ca^{2+}$-free solution, arterial contraction was increased. However, pretreatment with Diospyros kaki L. Radix or Diospyros kaki L. Folium inhibited contractile response to $Ca^{2+}$. Conclusions : Diospyros kaki L. Radix or Diospyros kaki L. Folium may suppress influx of extra- cellular $Ca^{2+}$ through the formation of nitric oxide in the vascular endothelial cells.

강활(羌活)이 가토(家兎) 혈관평골근(家兎 血管平滑筋)의 긴장성(緊張性) 조절(調節)에 미치는 영향(影響) (The Effects of Notopterygii Rhizoma on the Carotid Arterial Tension in Rabbit)

  • The purpose of this study was to analyze the Rhizoma on the blood pressure, heart rate and to define the mechanism of Notopterygii Rhizoma-induced relaxation in rabbit common carotid arterial contracted by agonists. Method : In order to explore the effect of Notopterygii Rhizoma on the blood pressure and heart rate, Notopterygii Rhizoma extract was injected in vein of rabbit ear. In order to investigate the effect of Notopterygii Rhizoma on norepinephrine(NE)-induced contracted rabbit carotid arterial strips, transverse strips with intact or damaged endothelium were used for the experiment using organ bath. To analyze the mechanism of Notopterygii Rhizoma-induced relaxation, Notopterygii Rhizoma extract infused into NE-induced contracted strips induced by agonists after treatment of methylene blue, propranolol, ouabain and it infused into serotonin, potassium chloride-induced contracted strips. Result : The blood pressure was significantly decreased by Notopterygii Rhizoma, but heart rate was insignificantly. In addition, Notopterygii Rhizoma significantly relaxed the norepinephrine, serotonin, potassium-induced contracted strips with intact endothelium or damaged endothelium. The relaxing effect of Notopterygii Rhizoma In NE-induced contracted strips with damaged endothelium by pretreatment of methylene blue, propranolol was not changed, but Ouabain was significantly decreased. Conclusion : These results were shown that Notopterygii Rhizoma affected the NE -induced contracted smooth muscle without the participation of endothelium, and demonstrated that the mechanism of NotoDtervgii Rhizoma-induced relaxation was the obstruction of receptor-operated Ca2+ channel.

  • CheonghunHwadam-tang(CHT) have been used in oriental medicine for many centuries as a therapeutic agent of vertigo by wind, fire and phlegm. CHTS was CHT adding Schizonepetae Herba. The effects of CHTS on the cerebral blood flow and blood pressure is not known. The purpose of this Study was to investigate effects of CHTS on the regional cerebral blood flow(rCBF) and mean arterial blood pressure(BP), action-mechanism of CHTS-induced changed-rCBF and BP. The changes of rCBF and BP was determinated by Laser-Doppler Flowmetry(LDF). The results were as follows ; CHTS extract was increased significantly rCBF in a dose-dependent, but was not changed BP compared with CHTS non-treated group. Pretreatment with propranolol, indomethacin and methylene blue were inhibited CHTS induced increase of rCBF, propranolol(all CHTS-treated group) and indomethacin(CHTS 0.01 mg/kg) of them were significantly decreased. Pretreatment with propranonol and indomethacin were inhibited CHTS induced increase of BP, but pretreated with methylene blue was significantly accelerated BP in high dosage. This results suggest that CHTS increased rCBF by dilating pial arterial diameter and the action of CHTS is also mediated by adrenergic β -receptor and cyclooxygenase.

당귀와 백수오 추출물이 뇌혈류 변화에 미치는 영향 (Effects of Angelicae Gigantis and Cynanchum wilfordii Hemsley Extract on the Changes of Cerebral Flow)

  • This study was designed to investigate the effects of Angelicae Gigantis and Cynanchum wilfordii Hemsley extract (AAC) on the changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, and in the rats with cerebral ischemia induced by middle cerebral artery occlusion, and further to determine the mechanisms. The results were as follows. AAC significantly increased rCBF but significantly decreased MABP in a dose-dependent manner in normal rats. The increase of AAC-induced rCBF was significantly inhibited by pretreatment with methylene blue (0.01 mg/kg, i.p.), an inhibitor of guanylate cyclase, and AAC-induced MABP was decreased by pretreatment with methylene blue. In cerebral ischemics, rCBF was stably improved by AAC (10 mg/kg, i.p.) during the period of cerebral reperfusion, which was contrasted with the findings of rapid and marked increase in the control group. These results suggest that AAC can increase rCBF in the normal state, as well as improve the stability of rCBF in cerebral ischemic state.
