Flavoral Essential Oil Components in the Stems of Agastache rugosa for Aromatherapy (아로마테라피용 배초향(Agastache rugosa) 줄기의 방향성 정유 성분)

  • This study was carried out to investigate the flavoral essential oil components in the stems of Agastache rugosa. These components were analyzed using gas chromatography-mass selective detector (GC-MSD). The stems of Agastache rugosa were contained alcohols, aldehydes, ketones, fatty acid esters, and terpenoids. The peak area (%) of estragole was highest among its oil components and the next were pulegone and menthone. The terpenoid alcohols found were 1-octen-3-ol, chavicol, spatulenol, 3-hexen-1-ol, 2-cyclohexen-1-ol, methyl eugenol, and octaethyllene glycol. The stems also contained ketones such as pulegone, menthone, cis-isopulegone, 2-cyclohexene-1-one, 3-octanone, 1-cyclohexanone, isoindole-1-one, t-ionone, inden-2-one, as well as the aldehydes of 4-methoxycinnam and benzaldehyde. The following esters were also detected 1-isopulegone-3-yl acetate, caryophyllene oxide, acetate and benzendicarboxylic acid ester. The terpenoids in the stems were identified as caryophyllene, limonene, cyclohexasiloxane-D, germacrene-D, anethole, cadinene, muurolene, and bourbonene. Overall Agastache rugosa contained several functional oil components including phenylpropanoids and terpenoids as flavoral essential oil components for natural aromatherapy.

Chemical Composition and Antitumor Apoptogenic Activity of Methylene Chloride Extracts from the Leaves of Zanthoxylum schinifolium (Zanthoxylum schinifolium잎의 methylene chloride 추출물의 화학적 조성 및 암세포에 대한 세포자살 유도활성과 그 작용기전)

  • To understand antitumor activity of Zanthoxylum schinfolium, which has been used as an aromatic and medicinal plant in Korea, the cytotoxic effect of various organic solvent extracts of its leaves on human tumor cells were investigated. Among these extracts such as methanol extract (SL-13), methylene chloride extract (SL-14), ethyl acetate extract (SL-15), n-butanol extract (SL-16), and residual fraction (SL-17), SL-14 appeared to contain the most cytotoxic activity against leukemia and breast cancer cells tested. The methylene chloride extra.1 (SL-14) possessed an apoptogenic activity causing apoptotic DNA fragmentation of human acute leukemia Jurkat T cells via mitochondrial cytochrome c release into cytoplasm, subsequent activation of caspase-9 and caspase-3, and cleavage of PARP, which could be negatively regulated by antiapoptotic protein Bcl-xL. The GC-MS analysis of SL-14 revealed that the twenty-two ingredients of SL-14 were 9,19-cyclolanost-24-en-3-ol (15.1%), 2-a-methyl-17, b-hop-21-ene (15.1%), 15-methyl-2,3-dihydro-1H benzazepin (11.95%), phytol (10.38%), lupeol (9.92%), 12-methylbenzofuran (8.23%), hexadecanoic acid (5.96%), cis,cis,cis-9,12,15-octadecatrienoic acid-methyl-ester (5.49%), 9,12,15-octadecatrienoic acid-methylester (3.59%), 15-methyl-4-(1-methylethylidene)-2-(4-nitrophenyl) (3.36%), hexadecanoic acid methyl ester (1.93%), vitamine E (1.88%), beta-amyrin (0.96%), and auraptene (0.89%). These results demonstrate that the cytotoxicity of the methylene chloride extract of the leaves of Z. schinifolium toward Jurkat T cells is mainly attributable to apoptosis mediated by mitochondria-dependent caspase cascade regulated by Bcl-xL, and provide an insight into the mechanism underlying antitumor activity of the edible plant Z. schinifolium.

Anti-inflammatory Effects of the Extracts of Prunus pendula for. ascendens (Makino) Ohwi Leaves and Identification of Active Constituents (올벚나무 잎 추출물의 항염 효능 및 유효성분 규명)

  • In this study Anti-inflammatory activity was investigated for the extract of Prunus pendula for. ascendens (Makino) Ohwi by monitoring nitric oxide (NO) production in LPS-stimulated RAW 264.7 murine macrophage cells. The P. pendula ethyl acetate (EtOAc) fraction showed to decrease the NO synthesis by 76.3% at $100{\mu}g/mL$ concentration. The inhibition occurred in a dose-dependent manner without causing cell toxicity. The EtOAc fraction also inhibited the production of $PGE_2$, $IL-1{\beta}$, IL-6 and expression of iNOS, COX-2 protein in dose-dependent manner. From the phytochemical study to isolate the active constituents, five known compounds were identified, which are ursolic acid (1), prunasin (2), methyl p-coumarate (3), kaempferol (4), astragalin (5). All of the compounds 1 - 5 were isolated for the first time from the P. pendula. Among the isolates, the flavonoids 4 and 5 were verified to inhibit NO production with high efficiency. These results suggested that extract of P. pendula leaves could be useful as anti-inflammatory agents in pharmaceutical or cosmetic applications.

  • Water vapor sorption of corn starch in various protective film coated capsules in 100% RH chamber for 14 days were as follows: The percent of water vapor sorption were 28.63% in uncoated capsule, 25.16% in hydroxy propylcellulose(HPC) coated capsule, 15.59% in 2-methyl-5-vinyl pyridine-methyl acrylated-methacrylic acid (MPM) coated capsule and 15.50% in polyvinyl acetal diethyl amino acetate(AEA) coated capsule. 2. Water vapor sorotion of magnesium trisilicate in various protective film coated capsules in 100% RH chamber for 14 days were as follows. The percent of water vapor sorption were 13.91% in uncoated capsule, 13.30% in HPC-coated capsule, 10.87% in MPM-caated capsule and 9.9% in AEA-coated capsule. 3. Water vapor sorption of magnesium carbonate in various protective film coated capsules in 100% RH chamber for 14 days were as follows: The percent of water vapor sorption were 12.48% in uncoated capsule, 10.72% in HPC-coated capsule, 8.10% in MPM coated capsule and 7.8% in AEA-coated capsule. 4. MPM-coated capsules and AEA-canted capsules were mere effective to protect water vapor sorption than HPC-coated capsules.

Flavonoid Constituents of Acacia catechu (Acacia catechu의 플라보노이드 성분)

  • Ten compounds were isolated from the ethanolic extract of the leaves of Acacia catechu (Fabaceae), and their structures were identified as nine flavonoids [(+)-catechin (1), (-)-epicatechin (2), (+)-afzelechin (3), (-)-epiafzelechin (4), (+)-mesquitol (5), kaempferol (6), quercetin (7), quercetin 3-methyl ether (8), and caryatin (9)] and an ellagic acid (10). The chemical structures of these compounds were identified on the basis of spectroscopic methods (mass spectrometry, 1D and 2D nuclear magnetic resonance) and comparison with literature values. This is the first time that the isolation of caryatin (9) has been reported from Acacia genus.

Physicochemical and Microbial Properties of the Korean Traditional Rice Wine, Makgeolli, Supplemented with Banana during Fermentation

  • The objective of the present study was to evaluate the physicochemical and microbial properties of the Korean traditional rice wine Makgeolli, supplemented with banana during 6 day fermentation. The alcohol contents of the control and banana Makgeolli were 17.0 and 16.5%, respectively. The pH values decreased while total acidity, total soluble solids, and color values increased throughout the fermentation process. An increase in microorganism counts throughout the 6-day fermentation period was noted in all samples. The major free sugar and organic acid detected in all samples were glucose and succinic acid, respectively. There were 39 volatile compounds detected in the control and banana Makgeolli. The major ester detected was ethyl acetate (20.037 and 22.604% for the control and banana Makgeolli, respectively). The major alcohol compounds detected were 3-methylbutanol (20.933%) and 3-methyl-1-butanol (34.325%) in the control. 2-mtehyl-1-propanol (22.289%) and 3-methyl-1-butanol (39.851%) were the highest alcohol compounds detected in the banana Makgeolli.

Mechanism of the Hydrolysis of 2-Phenyl-4H,5H,6H-3-methyl-3-thiazinium Perchlorate Derivatives

  • Hydrolysis reactions of 2-phenyl-4H,5H,6H-3-methyl-3-thiazinium perchlorate (PTP) and its derivatives at various pH have been investigated kinetically. The hydrolysis is quantitative, producing N-3-mercaptopropyl-N-methylbenzamide as the only product in the all pH ranges. The observed rate of hydrolysis of PTP was always of the first-order. For hydrolysis from PTP, Hammett ρvalues were 0.53, 0.84 and 1.13 for pH 5.0, 8.0, and 10.0, respectively. Bronsted βvalue was 0.53 for general base catalysis. This reaction is catalyzed by general w acetate concentration. However, as the amount of base becomes larger, the rate of hydrolysis reaction approaches the limiting values. The plot of log k vs. pH shows that the rate constants (kt) are two different regions in the profile; one part is directly proportional to hydroxide ion concentration and the other is not. On the bases of these result, the plausible hydrolysis mechanism and a reaction equation were proposed: Below pH 4.5, the hydrolysis was initiated by the addition of water to α-carbon. Above pH 9.0, the hydrolysis was proceeded by the addition of hydroxide ion to α-carbon. However, in the range of pH 4.5-8.0, these two reactions occured competitively.

Antioxidant Compounds from Distylium racemosum Leaves

  • The leaves of D. racemosum showed strong DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity and the order of the radical scavenging activity against DPPH radical is ethyl acetate (EtOAc) fraction>crude extracts>residue fraction>hexane fraction>ether fraction, under the experimental conditions. Since EtOAc fraction has highest antioxidative activity among these fractions, the isolation was performed from the EtOAc fraction of the leaves of D. racemosum and four phenolic compounds were isolated and identified as follows: methyl gallate, kaempferol, quercetin and quercitrin. The free radical scavenging activities of these compounds were 79.9%, 93.1%, 93.6% and 66.7% at 10 ㎍/ml, respectively. The IC50 of compound 1, compound 2, compound 3 and compound 4 were 6.1, 4.1, 3.6 and 6.5 ㎍/ml, respectively. These compounds have higher antioxidative activity compared with reference compounds, ascorbic acid (IC50 = 9.6 ㎍/ml).

Experimental Study and Correlation of the Solid-liquid Equilibrium of Some Amino Acids in Binary Organic Solvents

  • Under ordinary atmospheric circumstances, the gravimetric technique was used to measure the solubility of L-cysteine (L-Cys) and L-alanine (L-Ala) in various solvents, including methyl alcohol, ethyl acetate, and mixtures of the two, in the range o 283.15 K to 323.15 K. Both individual solvents and their combinations showed a rise in the solubility of L-Cys and L-Ala with increasing temperature, according to the analyzed data but when analyzed at a constant temperature in the selected mixed solvents, the solubility declined with decreasing of initial mole fractions of methyl alcohol. To further assess, the relative utility of the four solubility models, we fitted the solubility data using the Jouyban-Acree (J-A), van't Hoff-Jouyban-Acree (V-J-A), Apelblat-Jouyban-Acree (A-J-A), and Ma models followed by evaluation of the values of the RAD information criteria and the RMSD were. The dissolution was also found to be an entropy-driven spontaneous mixing process in the solvents since the thermodynamic parameters of the solvents were determined using the van't Hoff model. In order to support the industrial crystallization of L-cysteine and L-alanine and contribute to future theoretical research, we have determined the experimental solubility, correlation equations, and thermodynamic parameters of the selected amino acids during the dissolution process.

Antimutagenic Effects and Compounds Identified from Hexane Fraction of Persimmon Leaves (감잎 핵산획분의 항돌연변이 효과와 항돌연변이 물질의 GC-MS를 이용한 동정)

  • Methanol extract of dried persimmon leaves was fractionated to hexane, chloroform, ethyl acetate, butanol, and aqueous tractions. Hexane, butanol, and aqueous fractions had high yields of extracts. Hexane fraction among these fractions showed the highest inhibition rate on the mutagenicities of aflatoxin (AFB$_1$), dimethyl-amino-bi-phenyl (DMAB), N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), and 4-nitroquinoline-1-oxide (4-NQO) in Salmonella typhimurium TA100. Hexane fraction was further fractionated into eight fractions by silica gel column c-hromatography and thin layer chromatography (TLC). The fraction 5 on TLC exhibited the highest antimutagenic activity on AFB$_1$, DMAB, and MNNC. 1'-oxocannabinol, 3B-acetoxy-17-methyl-5a-18 (13-17) abeoardrost-13-one, 4-methoxy-2'6'-dinitro-3, 5-di-t-butylbiphenyl, 8, 9-dihydro-5, 6-dimethoxy-dibenz [c, h]isoquino [2, 1, 8-1 ma]carbazole-11, 16-dione were tentatively identified from this antimutagenic fraction by GC-MS.
