Hypolipemic and Hypoglycemic Activities of Prunus davidiana in High Fat-Fed Rats

  • Blood lipid levels in rats with high fat-fed hyperlipemia were determined after intraperitoneal administration of a methanolic extract of Prunus davidiana Fr. stems. Administration of the methanolic extract for 6 days produced a significant decrease of blood triglyceride and total cholesterol, and the atherogenic index was also improved. In addition to the hypolipemic effect, the methanolic extract was also shown to be effective in reducing an elevated level of glucose in rats with hyperlipemia resulting from high-fat feeding. On the other hand, blood triglyceride and total cholesterol in rats fed with stock diet were not affected by administration of the methanolic extract even if there was a tendency to decrease. No significant change was also found in the level of glucose. Thus, it is suggested that this methanolic extract probably may increase the metabolic utilization only when fed with excess fat.

갈화(葛花)의 항돌연변이(抗突然變異) 활성(活性)에 관한 연구(硏究) (A Study on Antimutagenic acitivities of from Puerariae Flos Extracts)

  • Antigenotoxicity test (SOS chromotest), antimutagenecity test (Ames test), and antioxidant test (NBT method and xanthine-xanthine oxidase method) were carried out using water-soluble and methanolic extracts from Puerariae Flos. Against the mutagens MNNG and NQO, antigenotoxic activity of methanolic extracts were much more effective than that of water-soluble ones. When the methanolic extract was added to the certain concentration $(100{\mu}{\ell}/tube)$, antigenotoxic acivity against the mutagen MNNG was enhanced. Contrary to the water-soluble extract, the methanolic extract showed high antigenotoxicity against the mutagen NQO with increment of the extract. Against the mutagen MNNG with Ames test, antimutagenic activity of the methanolic extract at $300{\mu}{\ell}/tube$ was 96% as an inhibition ratio of revertant forming CFU/plate. The antioxidant activity of water-soluble extract was comparatively higher than that of the methanolic one.

천궁 메탄올 추출액의 멜라닌 형성 억제 효과 (Inhibitory Effect of Methanolic Extract of Cnidii Rhizoma on the Melanogenesis)

  • Down-regulation of melanin synthesis is required for recovery of pigmentary disorders and it is known that direct inhibitors of tyrosinase suppress melanin synthesis. We screened several oriental medicinal plants using B16/Fl0 cells and found that the methanolic extract of Cnidii Rhizoma down-regulated melanin synthesis effectively. Although the proliferation of Bl6/Fl0 cells was decresed by the methanolic extract of Cnidii Rhizoma, it did not appear necrosis. Bl6/F10 cells incubated with the methanolic extract of Cnidii Rhizoma showed reduced pigmentation and tyrosinase activity. Western blotting revealed that the amount of tyrosinase was decreased by the methanolic extract of Cnidii Rhizoma. These results suggest that the inhibitary effect of the methanolic extract of Cnidii Rhizoma on melanogenesis is due to the suppression of tyrosinase in Bl6/F10 cells and Cnidii Rhizoma is a candidate for an efficient whitening agent.

Comparative antidiabetic activity of different fractions of methanolic extract of Zingiber officinale Roscoe in streptozotocin induced NIDDM rats

  • Earlier we have reported the antidiabetic activity of fresh juice of rhizomes of Zingiber officinale (Z. officinale) and its correlation with 5-HT receptor antagonism. Since 6-gingerol the marker compound of Z. officinale is reported to posses 5-HT anatgonistic activity, the present investigation, was undertaken to find out the concentration of 6-gingerol present in methanolic extract of Z. officinale and its different fractions (petroleum ether, toluene and chloroform). We also evaluated these fractions for antidiabetic activity in streptozotocin (STZ)-induced neonatal type 2 diabetic rats. Fasting glucose and insulin levels in non insulin dependent diabetes mellitus (NIDDM) rats were found to be significantly (P < 0.05) higher than control rats and these were significantly decreased by treatment with methanolic extract of Z. officinale and its fractions. The results of oral glucose tolerance test (OGTT) showed that methanolic extract and its fractions significantly (P < 0.05) decreased both STZ-induced increase in $AUC_{glucose}$ and $AUC_{insulin}$ values in NIDDM groups. Treatment with petroleum ether fraction produced a greater reduction in elevated glucose and $AUC_{glucose}$ levels as compared to treatment with other fractions. Treatment with methanolic extract of Z. officinale and its fractions also produced significant reduction in the elevated lipid, serum glutamate oxaloacetate transaminase (SGOT) and serum glutamate pyruvate transaminase (SGPT) levels in NIDDM rats. The effect of petroleum ether fraction on elevated lipid, SGOT and SGPT levels was significantly greater as compared to treatment with other fractions. The concentration of 6-gingerol was found to be maximum in petroleum ether fraction (11.430%) and minimum in chloroform fraction (0.973%). The methanolic extract and toluene fraction was found to contain 3.080% and 2.191 %, 6-gingerol respectively. In conclusion, our data suggest that methonolic extract and its fractions possess significant antidiabetic activity in NIDDM rats. The extent of activity appears to be dependent on the concentration of 6-gingerol present in the extract or its fractions.

Carbohydrase inhibition and anti-cancerous and free radical scavenging properties along with DNA and protein protection ability of methanolic root extracts of Rumex crispus

  • The study elucidated carbohydrase inhibition, anti-cancerous, free radical scavenging properties and also investigated the DNA and protein protection abilities of methanolic root extract of Rumex crispus (RERC). For this purpose, pulverized roots of Rumex crispus was extracted in methanol (80% and absolute conc.) for 3 hrs for $60^{\circ}C$ and filtered and evaporated with vacuum rotary evaporator. RERC showed high phenolic content ($211{\mu}g$/GAE equivalent) and strong 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ($IC_{50}$ = 42.86 (absolute methanol) and $36.91{\mu}g/mL$ (80% methanolic extract)) and reduced power ability. Furthermore, RERC exhibited significant protective ability in $H_2O_2/Fe^{3+}$/ascorbic acid-induced protein or DNA damage and percentage inhibition of the HT-29 cell growth rate following 80% methanolic RERC exposure at $400{\mu}g/mL$ was observed to be highest ($10.2%{\pm}1.03$). Moreover, methanolic RERC inhibited ${\alpha}$-glucosidase and amylase effectively and significantly (P < 0.05). Conclusively, RERC could be considered as potent carbohydrase inhibitor, anti-cancerous and anti-oxidant.

Kanahia Laniflora Methanolic Extract Suppressed Proliferation of Human Non-Small Cell Lung Cancer A549 Cells

  • Introduction: Lung cancer is one of the most common cancers worldwide. In certain countries such as United States of America, it is the leading cause of related cancer mortality among both men and women. Natural products play an important role in overcoming the limitations of chemotherapy and radiotherapy. Objectives: In this study, we investigated the antiproliferative and apoptotic activities of Kanahia laniflora methanolic extract against human non-small cell lung cancer cells (A549). Methods: Sulforhodamine B colorimetric assays were used to determine the inhibitory effects of a leaf methanolic extract against A549 cells. Results: The extract showed strong cytotoxic activity against A549 cells with an $IC_{50}$ value of $0.13{\mu}g/ml$ compared to $0.21{\mu}g/ml$ for doxorubicin. The extract also significantly increased the percentage of apoptotic cells to 49.7% as compared to 1.4% and 47.4% for control and doxorubicin respectively. Conclusion: These results showed, for the first time, that a methanolic extract of Kanahia laniflora leaves can inhibit the proliferation of human non-small cell lung cancer cells (A549). Further attention to its potential as a new effective anticancer agent is warranted.

Antioxidant Activities of Chlorophyta and Phaeophyta from Jeju Island

  • Antioxidative activities of Chlorophyta and Phaeophyta in Jeju Island were measured by superoxide anion ($O_2^{{\cdot}-}$), hydroxyl radical ($HO^{\cdot}$), hydrogen peroxide ($H_2O_2$) and DPPH free radical scavenging assays. Methanolic and aqueous extracts of the seaweeds were prepared at both temperatures, higher (70$^{\circ}C$) and room temperature (20$^{\circ}C$), and screened for the construction of an extract bank from seaweeds in Jeju Island. A variety of extracts showed positive effect against reactive oxygen species (ROS). Especially, Sargassum thunbergii methanolic extract at 70$^{\circ}C$ (70ME, 97.41%), S. fulvellum methanolic extract at 20$^{\circ}C$ (20ME, 84.66%), Codium fragile aqueous extract at 70$^{\circ}C$ (70AE, 96.61%) and S. thunbergii 20ME (97.44%) exhibited the highest scavenging activities against $O_2^{{\cdot}-}$, $HO^{\cdot}$, $H_2O_2$ and DPPH free radicals, respectively. Total phenolic contents also examined but did not show a positive correlation with ROS scavenging abilities (except for a few extracts). These results indicate that further investigation is needed to identify and purify the responsible antioxidative components.

Antioxidant and Anticancer Activity of Fractions from Picrasma quassioides (D. Don) Benn. Methanolic Extract

  • The potential antioxidant and anticancer activities of Hexane, EtOAc (Ethyl acetate), BuOH (n-Buthanol) and water fractions from methanolic (MeOH) extract of Picrasma quassioides (D. Don) Benn. were evaluated in vitro. Tested fractions showed strong antioxidant activity, especially EtOAc fraction had the highest activity ($IC_{50}\;=\;114.01\;{\mu}g/mL$), containing high total phenolics and total flavonoids contents, showed $67.59\;Tan\;{\mu}g/mg$ and $64.95\;Que\;{\mu}g/mg$ respectively. Anticancer activity of these fractions was tested by MTT assay on HT-29 (the human colon carcinoma cells) cell line. BuOH fraction not only showed very high anticancer activity, but also had no cytotoxic effect on 293 (the human normal kidney cells) cell line. Considering these results, we used BuOH fraction of MeOH crude extract from P. quassioides (D.Don) Benn. to do assessment of apoptosis by flow cytometry and the mRNA expression levels of widely established apoptotic-related genes on HT-29 cell line. All the experiments showed that BuOH fraction can induce apoptosis on HT-29 cell line strongly. Taken together, methanolic extract of P. quassioides has potential for antioxidant and anticancer activities products.
