Achillea sibirica Ledeb. is widely distributed in Korea and has been often used as folk medicine in peptic and tonic. As one of our searches for bioactive (anti oxidation) compounds from medicinal plants, we studied Achillea sibirica Ledeb. (Compositae). Antioxidant activity of Achillea sibirica was determined by measuring lipid peroxide produced when a mouse liver homogenate was exposed at $90^{\circ}C$ using 2-thiobarbituric acid (TBA) and by evaluation the radical scavenging activity on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical. Whole parts of Achillea sibirica was extracted with methanol and its extracts was fractionated with organic solvent; n-Hexane, methylene chloride, ethyl acetate, n-Butanol. EtOAc fraction exhibited antioxidant activity and From its, two flavonoid glycosides were isolated by silica gel and gel filtration colume chromatography and identified to kampferol 3-O-glucoside and luteolin 7-O-neo-hesperidoside, respectively, by physico-chemical and spectroscopical method. At antioxidant activity test for two compounds isolated, antioxidant activity was showed too. And from hexane fraction sterol was is isolated and identificated to mixture of campesterol, stigmasterol, and ${\beta}-sitosterol$.

Anticarcinogen is one of the major strategies for cancer control. It is well established that dietary factors play an important role in modulating the development of certain types of human cancer. We investiagted the anticarcinogenic effects of Sedum sarmentosum Bunge (SS) with Platycodon grandiflorum A. extracts on HepG2, HeLa and MCF-7 cell lines. By the MTT assay, among the five partition layers of methanol extract of SS (SSM), the ethylether partition layer of SS (SSMEE) showed the strongest cytotoxic effects on all cell lines. We also investigated the synergistic effect of the combination of SS and PG extracts on growth inhibition of the HepG2, HeLa and MCF-7 cell lines compared to the effects of five partition layers of SSM. Combination of SS and PG extracts significantly increased cytotoxic effects on all cell lines. Therefore, we were able to conclude that ethylether partition layer, SSMEE might have potentially useful cytotoxic materials on all the human cancer cells which we used. And we could suggest that the combination of SS with PG enhanced the anticarcinogenic effect on HepG2, HeLa and MCF-7 cell lines. We also determined QR activity of partition layers of SSM, among them, SSMEE on HepG2 cells showed the highest QR activity, 3.21 as control value of 1.0.
Objectives: Hepatocellular carcinoma is the most common primary malignant tumor of the liver worldwide. In-Jin-Ho-Tang(IJHT) has been used as a traditional Chinese herbal medicine since ancient time. and today it is widely applied as a medication for jaundice which is associated with inflammation in liver. In this study, I investigated whether methanol extract of IJHT induced HepG2 cancer cell death. Methods: Cytotoxic activity of IJHT on HepG2 cells was using XTT assay. Apoptosis induction by Ros A in HCT116 cells was verified by the induction of cleavage of poly ADP-ribose polymerase (PARP). and activation of caspase-3, -8 and -9. The release of cytochrome c from mitochondria to cytosol. the level of Bcl-2 and Bax and the expression of p53 and p21 were examined by western blotting analysis. Furthermore, MAPKs activation was analyzed by western blotting analysis. Results: IJHT induced apoptosis in HepG2 cells. And treatment of IJHT resulted in the release of cytochrome c into cytosol, decreased anti-apoptotic Bcl-2, and increased pri-apoptotic Bax expression. IJHT markedly inactivated extracellular signal-regulated kinase (ERK1/2), and activated p38 mitogen-activated protein (MAP) kinase. Sodium orthovanadate (SOV), a phosphatase inhibitor, to reverse IJHT-induced ERK1/2 inactivation and SB203580, a specific p38 MAP Kinase inhibitor efficiently blocked apoptosis of HepG2. Thus, IJHT induces apoptosis in HepG2 cells via MAP kinase modulation. Conclusion: These results indicated that IJHT has some potential for use as an anti-cancer agent.
A new analytical method of cyanobacterial toxins, i, e, microcysins was deveeloped using supercritical fluid extraction(SFE). The microcystins mcluded in the study are sparsely soluble in neat supercritical fluid CO$_2$ However, the microcystins were successfully extracted with a temary mixture(90% CO$_2$,9.0% methanol 1.0% water) at 40$^{\circ}$C and 250 atm. The SFE method developed in this study has several advantages over solid-phase extraction(SPE) sample preparation for the analysis of microcystins. Sample handling steps are minimized thus reducing possible losses of analytes and saving analysis time. No clean-up steps are employed in this SFE method. Althouhgh many methods have been described for microcystim RR and LR, the method using solid-phase extraction with ODS cartridges is the most commonly used. However, the adsorbing power of ODS caridges for microcystins is weak, so we have attempted to use a more polar CN cartridge, to increase the adsorbing power for microcystins. Lyophilized cells(100mg) were wxtracted with 5% (v/v) acetic acid. The extract was centrifuged and then the supernatant was applied to a CN cartridge. The cartridge which contained microcystins was rinsed with 5 ml of water and 5 ml of 0.5 M acetic acid. followed by 5 ml of 5% acetonitrile in water , and were determined by HPLC. Better recoveries and chromatogram were observed than with ODS cartridge.

Diospyros lotus has been cultivated for its edible fruits, which are considered to have medicinal importance. The aim of this study was to evaluate the antioxidant and antipruritic activities of water-soluble, methanol extract, and ethyl acetate (EA) fractions from D. lotus leaves. The EA fraction showed the lowest $IC_{50}$ vale (DPPH: $5.3{\mu}g/mL$, ABTS: $53.8{\mu}g/mL$). Therefore, we further investigated anti-inflammatory and antipruritic effects of the EA fraction. TNF-${\alpha}$ production increased by PMA plus A23187 treatment was significantly inhibited by the EA fraction in a dose-dependent manner. The EA fraction also inhibited histamine release from rat peritoneal mast cells stimulated by compound 48/80, which promotes histamine release. Furthermore, EA fraction had inhibitory effects on scratching behavior induced by compound 48/80 in Balb/c mice. These results suggest that the EA fraction from D. lotus leaves has potential as ameliorative agent against oxidative stress and pruritus-related disease.
Histochemical distribution and varietal variation of schizandrin and oil compounds, and the changes of their extraction yield were investigated in fruits of collected Schizandra chinensis including Inje cultivar. In histochemical analysis on the distribution of schizandrin and oil in fruits of Inje cultivar, higher concentrations of them were found in the seed (1.01% and 27.6%, respectively) than in the epicarp and mesocarp of the whole fruit. Average contents of schizandrin in fruits and oil in seeds of collected lines were 0.84% and 27.9%, respectively. The mean composition of fatty acids in seeds oil was 3.6% of palmitic acid, 0.6% of stearic acid, 19.7% of oleic acid, 73.0% of linoleic acid, and 3.1% of linolenic acid, showing high composition(95.8%) of total unsaturated fatty acid. Oil extracted from seeds of Inje cultivar contained 4.29% of schizandrin, indicating that seed oil contained much schizandrin, a bioactive lipid-soluble compound. Compared with 80% methanol extraction in fruits and seeds, yields of schizandrin and oil were lower showing 23.8% and 17.3%, respectively in boiling water extraction of the fruits and seeds without grinding. The seeds soaked with water during four months contained 1.18% of schizandrin and 25.2% of oil, whose contents were similar to those of the seeds stored at room temperature. These results demonstrated that the seed in the whole fruit could be utilized as a source to extract its functional oil and bioactive lipid-soluble compounds like schizandrin, especially after using Schizandra fruits for the beverage manufacture.
Although the vegetable Angelica keiskei (AK) has widely been utilized for the purpose of general health improvement among Korean population, its functionalities are not very well defined. In this study, we examined the effects of methanol extract of AK in rats on the biochemical changes induced by two hepatotoxins, D-galactosamine (GalN) and carbon tetrachloride ($CCl_4$). AK was orally administered once daily for 7 days to male rats at 200 and 500 mg/kg, before hepatotoxins. Effects of AK were assessed 24 hr later. AK pretreatments at 200 and 500 mg/kg significantly blunted GalN-induced elevation in liver lipid peroxidation, plasma aspartate-transaminase (AST) and alanine-transaminase (ALT) activities. AK also prevented, after 500 mg/kg but not after 200 mg/kg, the GalN-induced elevation in triglyceride, total cholesterol and LDL-cholesterol levels. Differently from against GalN-induced toxicity, AK did further elevate the $CCl_4$-induced rise in AST, ALT and lipid peroxidation. These results suggest that AK, when pre-administered prior to GalN, exerted protective effects against GalN-induced hepatotoxicity, in contrast however, AK exacerbated that induced by $CCl_4$. To explore possible mechanism for the toxicity-potentiating effects of AK on $CCl_4$, the activity of hepatic drug metabolism after AK treatment was assessed. It was observed that AK increased the activity of aniline hydroxaylase, a cytochrome P450 isoenzyme responsible for metabolic activation of $CCl_4$. This finding suggests that hepatoprotective effects of AK are not equally expected depending on hepatotoxins employed.

The inhibitory effects of radish juice on the mutagenicities of cigarette smoke condensate (CSC), methanol extract of charred part of fried or broiled saury pike (MECS) and 2-aminofluorene (2-AF) were examined by the use of Ames assay toward Salmonella typhimurium TA 98 strain. Radish juice exhibited inhibition percentage of about 100, 100 and 87 on the mutagenicities of CSC, two kinds of MECS and 2-AF, respectively. Except for juices of cabbage and leek, radish juice has inhibited more effectively the mutagenicity of CSC than other fruit or vegetable juices studied. Inhibitory effect of radish juice might be originated from the components with molecular weight above 50,000 and decreased sharply in 5 min by heat treatment at $100^{\circ}C$, but hardly changed at low and moderate storage temperatures such as $4^{\circ}C,\;10^{\circ}C,\;25^{\circ}C\;and\;35^{\circ}C$ for about 2 weeks. Precipitate obtained from ammonium sulfate saturation from 30 to 80% had inhibitory effect on the mutagenicity of CSC. Extracts from 3 bands of non-denaturing gel of $30{\sim}80%$ amnonium sulfate precipitate have exhibited the inhibitory effects on the mutagenicity of CSC.
The present study investigated the anti-proliferative and chemosensitizing effects of Crinum asiaticum var. japonicum against multi-drug resistant (MDR) cancer cells. The 80% methanol extract, chloroform ($CHCl_3$) fraction and butanol (BuOH) fraction of C. asiaticum inhibited the growth of mitoxantrone (MX) resistant HL-60 (HL-60/MX2) cells. When HL-60/MX2 cells were treated with the $CHCl_3$ and BuOH fractions, DNA ladder and sub-G1 hypodiploid cells were observed. Furthermore, the fractions reduced BcI-2 mRNA levels, whereas Bax mRNA levels were increased. These results suggest that the inhibitory effect of C. asiaticum on the growth of the HL-60/MX2 cells might arise from the induction of apoptosis. Treatment of HL-60/MX2 cells with the fractions markedly decreased the mRNA levels of the multi-drug resistance protein-1 and breast cancer resistance protein. The $CHCl_3$ fraction and hexane fraction increased MX accumulation in HL-60/MX2 cells. These results imply that the $CHCl_3$ fraction of C. asiaticum plays a pivotal role as a chemosensitizer. We suggest that components of C. asiaticum might have a therapeutic potential for the treatment of MDR leukemia.
The present study was conducted to monitor the level of triflumizole residues in fruits (apple and pear) and vegetable (cucumber) samples in order to assess risk posed by the presence of such residues to the consumer. Triflumizole was applied at a recommended dose rate to apple and pear pulps and to a cucumber sample. The samples were collected at harvesting time following several treatments (three and/or four treatments). Triflumizole was extracted with methanol and re-extracted into dichloromethane. The presence of triflumizole was determined by HPLC with UV detection at 238 nm following the cleanup of the extract by open preparative chromatographic column with Florisil. The versatility of this method was evidenced by its excellent linearity (> 0.999) in the concentration range between 0.2 and 4.0 mg/kg. The mean recoveries evaluated from the untreated samples spiked at two different fortification levels. 0.1 and 0.4 mg/kg, and ranged from 87.5${\pm}$0.0 to 93.3${\pm}$2.6 for the tested fruits and vegetable, respectively, and the repeatability (as relative standard deviation) from three repetitive determinations of recoveries were no larger than 6%. The calculated limit of detection was 0.02 mg/kg and the minimum detectable level of 4 ng for triflumizole was easily detected. When triflumizole was sprayed onto the apple trees three times at 50-40-30 and 40-30-21 days prior to harvesting and four times onto the pear trees at 40-30-21-14 days prior to harvesting, the mean residual amounts of 0.05 and 0.06 mg/kg for apples and pears, respectively, were not detected in all of the treatments. When the cucumber sample was fumigated four times at 7, 5, 3 and 1 day prior to harvesting, the mean residual amount was not detectable. Triflumizole can be used safely when sprayed (wettable powder, 30% active ingredient) and fumigated (10%) 4 times at 14 and 1 day prior to harvesting to protect the fruits and vegetable, respectively.
