Evaluation of reference genes for RT-qPCR study in abalone Haliotis discus hannai during heavy metal overload stress

  • Background: The evaluation of suitable reference genes as normalization controls is a prerequisite requirement for launching quantitative reverse transcription-PCR (RT-qPCR)-based expression study. In order to select the stable reference genes in abalone Haliotis discus hannai tissues (gill and hepatopancreas) under heavy metal exposure conditions (Cu, Zn, and Cd), 12 potential candidate housekeeping genes were subjected to expression stability based on the comprehensive ranking while integrating four different statistical algorithms (geNorm, NormFinder, BestKeeper, and ${\Delta}CT$ method). Results: Expression stability in the gill subset was determined as RPL7 > RPL8 > ACTB > RPL3 > PPIB > RPL7A > EF1A > RPL4 > GAPDH > RPL5 > UBE2 > B-TU. On the other hand, the ranking in the subset for hepatopancreas was RPL7 > RPL3 > RPL8 > ACTB > RPL4 > EF1A > RPL5 > RPL7A > B-TU > UBE2 > PPIB > GAPDH. The pairwise variation assessed by the geNorm program indicates that two reference genes could be sufficient for accurate normalization in both gill and hepatopancreas subsets. Overall, both gill and hepatopancreas subsets recommended ribosomal protein genes (particularly RPL7) as stable references, whereas traditional housekeepers such as ${\beta}-tubulin$ (B-TU) and glyceraldehyde-3-phosphate dehydrogenase (GAPDH) genes were ranked as unstable genes. The validation of reference gene selection was confirmed with the quantitative assay of MT transcripts. Conclusions: The present analysis showed the importance of validating reference genes with multiple algorithmic approaches to select genes that are truly stable. Our results indicate that expression stability of a given reference gene could not always have consensus across tissue types. The data from this study could be a good guide for the future design of RT-qPCR studies with respect to metal regulation/detoxification and other related physiologies in this abalone species.

PET Fabric Supported Fixed Site Carrier Membrane for Selective Metal ion Transport

  • Development of a novel fixed site carrier membrane (FCM), supported by PET fabric for metal ion separation is reported. The membranes were prepared by dipping PET fabric into the methylene chloride solution of Poly(5-vinyl-m-phe-nylene-m'-phenylene-32-crown-10) (P(VCE)), a polymeric metal ion carrier. It was found that the flux of mono-valent metal ion transported across the membrane is signif=cantly differed from each other and the flux decreases in the order $Cs^+$>$Rb^+$>$K^+$>$Na^+$>$Li^+$ irrespective to the anion except perchlorate anion. It was explained in terms of the stability of the complex, formed by crown ether unit of the P(VCE) and the various metal ions, meanwhile, the lower rate of transport in the presence of perchlorate anion was ascribed to its low hydrophilicity.

An Efficient Preparation of 4-Nitrosoaniline from the Reaction of Nitrobenzene with Alkali Metal Ureates

  • This paper describes the synthesis of alkali metal salts of urea (ureates) and their application to the direct preparation of 4-nitrosoaniline from nitrobenzene via nucleophilic aromatic substitution of hydrogen. Sodium and potassium ureates were readily prepared from the reaction of urea with sodium hydride, metal methoxides, and metal hydroxides. The effect of ureates as nucleophiles on the conversion of nitrobenzene to 4-nitrosoaniline was investigated and compared with that of a urea-metal hydroxide mixture. It was found that the ureates were superior for producing 4-nitrosoaniline owing to their higher thermal stability of the ureate. The ureate obtained from the treatment of urea with sodium hydride gave the highest yield for the preparation of 4-nitrosoaniline. The ureates generated from the reaction of urea with metal hydroxide also gave high yields of 4-nitrosoaniline. Catalytic hydrogenation of 4-nitrosoaniline afforded polymer-grade 1,4-benzenediamine in quantitative yield.

도재-금속의 결합 강도에 미치는 비금속 합금의 열처리 효과 (The effect of preheat treatment on ceramic to metal bond strength)

  • In dental prosthetics, the application of metal-ceramic restorations has steadily increased since their introduction. This is due to excellent esthetics in combination with high mechanical stability. In order to optimum bond strength between metal and ceramics, controlled oxidation of metal substructure is essential factor. Beryllium containing and beryllium free Ni-Cr alloys for metal-ceramic restorations were evaluated for the metal-ceramic bond strength by changing heat treatment for oxide formation. A mechanical three-point bending test was employed to evaluate the interfacial bond strength of metal-ceramic. In each metal, plate type specimens were used for mechanical three-point bending test. With Ni-Cr alloys for metal ceramics, mechanical three-point bending test showed that double degassing was more available preheat treatment method than another. It was found that beryllium containing Ni-Cr alloys are more effective than beryllium-free for metal-ceramic bond strength.

새로운 질소-산소계 여러 자리 리간드의 합성 및 전이금속(Ⅱ)이온 착물의 안정도상수 (Syntheses of New Nitrogen-Oxygen Multidentate Ligands and Their Stability Constants of Transition Metal(Ⅱ) Ions)

  • 여러자리 산소-질소계 리간드들은 시프염기 리간드인 Bis(salicylidene)-ethylendiamine(BSED), Bis(salicylidene)-propylenediamine(BSPD), Bis(salicylidene)-diethylenetriamine(BSDT), Bis(salicylidene)-triethylenetetraamine(BSTT) 및 Bis(salicylidene)-tetraethylenepentaamine(BSTP)의 이민기를 수소환원장치에서 백금촉매를 이용하여 네자리 리간드인 N,N'-bis(2-hydroxybenzyl)-ethylenediamine(BHED)과 N,N'-bis(2-hydroxybenzyl)-propylenediamine(BHPD), 다섯자리 N,N'-bis(2-hydroxybenzyl)-diethylenetriamine(BHDT), 여섯자리 N,N'-bis(2-hydroxybenzyl)-triethylenetriamine(BHTT), 일곱자리 N,N'-bis(2-hydroxybenzyl)-tetraethylenepentaamine(BHTP) 리간드들을 합성하였다. 리간드들의 양성자 해리상수와 전이금속(Ⅱ)과 착물의 안정도상수는 전위차 적정법으로 측정하였다. 리간드별 전이금속(Ⅱ)이온에 대한 착물 안정도상수값$(logK_{ML})$의 크기는 BHED아연(Ⅱ)의 순서로 이 결과는 Irving-Williams 서열과 잘 일치하여 증가하였다.

치환기를 가진 질소-산소(N3-O2)계 리간드 합성과 중금속(II)이온 착물의 안정도상수 결정 (Synthesis of Pentadentate Nitrogen-Oxygen(N3-O2) Ligands with Substituents and the Determination of Stability Constants of Their Heavy(II) Metal Complexes)

  • A new $N_3-O_2$ pentadentate ligand, H-BHPT, was synthesized. Hydrochloric acid salts of Br-BHPT, Cl-BHPT, $CH_3O$-BHPT and $CH_3$-BHPT, having Br-, Cl-, $CH_{3-}$ and $CH_3O-$ substituents at the para position of the phenol hydroxyl group of H-BHPT were synthesized. Hydrochloric acid salts of 3OH-BHPT and 4OH-BHPT, having different position of the phenol hydroxyl group of H-BHPT were also synthesized. The synthesis of each ligand was confirmed by C. H. N. atomic analysis and $^1H$ NMR, $^{13}C$ NMR, UV-visible, and mass spectra. The calculated proton dissociation constants ($log{K_n}^H$) of the phenol hydroxyl group and secondary amine group of the synthesized $N_3-O_2$ ligands showed five steps of the proton dissociations. The order of the overall proton dissociation constants ($log{\beta}_p$) of the ligands was Br-BHPT < Cl-BHPT < H-BHPT < $CH_3O$-BHPT < $CH_3$-BHPT. The order agreed with that of Hammett substituent constants (${\delta}_p$). However, dissociation steps of 3OH-BHPT were four and that of 4OH-BHPT was three. The calculated stability constants ($logK_{ML}$) between the ligands and transition metal ions agreed with the order of $log{\beta}_p$ values of the ligands. The order of the stability constants between the transition metal ions with the synthesized ligands was Co(II) < Ni(II) < Cu(II) > Zn(II) > Cd(II) > Pb(II). The order agreed well with that of the Iriving-Williams.

  • Introduction: To evaluate the skeletal stability after a bilateral sagittal split osteotomy (BSSO) setback of the mandible fixed with a biodegradable internal fixation device or metal internal fixation device. Materials and Methods: Thirty consecutive patients underwent mandibular setback via BSSO. Fifteen patients were fixed with a biodegradable internal fixation device or metal internal fixation device respectively. Posteroanterior (PA) and lateral cephalograms were taken preoperatively and at two days, 5.5 months and 14.5 months postoperatively. The relevant skeletal points were traced and digitized to evaluate the skeletal changes postoperatively. The relapse rates were analyzed and compared statistically. Results: There was no statistically significant differences in postoperative stability between the two groups.(P<0.05) Conclusion: The biodegradable internal fixation device may make an effective device alternative to a metal internal fixation device for setback BSSO.

  • Ni-Cr-Al metal-foam-supported catalysts for steam methane reforming (SMR) are manufactured by applying a catalytic Ni/Al2O3 sol-gel coating to powder alloyed metallic foam. The structure, microstructure, mechanical stability, and hydrogen yield efficiency of the obtained catalysts are evaluated. The structural and microstructural characteristics show that the catalyst is well coated on the open-pore Ni-Cr-Al foam without cracks or spallation. The measured compressive yield strengths are 2-3 MPa at room temperature and 1.5-2.2 MPa at 750℃ regardless of sample size. The specimens exhibit a weight loss of up to 9-10% at elevated temperature owing to the spallation of the Ni/Al2O3 catalyst. However, the metal-foam-supported catalyst appears to have higher mechanical stability than ceramic pellet catalysts. In SMR simulations tests, a methane conversion ratio of up to 96% is obtained with a high hydrogen yield efficiency of 82%.

Paper Ionophoretic Technique in the Study of Mixed Complexes

  • Stability constants of complexes of aluminium(III) and thorium(IV) with methionine and cysteine have been determined by modified paper electrophoretic technique at $\mu$ = 0.1 M. The proportion of ionic species of methionine and cysteine were varied by changing pH of background electrolyte. The stability constants of the complexes metal-methionine-cysteine have been found to be 4.31 ± 0.12 and 5.40 ± 0.19 (log K values) for $Al^{3+}\;and\;Th^{4+} $ complexes, at temperature 35 ${^{\circ}C}$, respectively.

  • Complexation reactions of nitrilotriacetate (NTA) and penicillamine with $Cu^{2+}$ and $Co^{2+}$ have been studied in solution phase using paper electrophoresis technique. The stability constants of the complexes Cu(II)-nitrilotriacetate-penicillamine and Co(II)-nitrilotriacetate-penicillamine have been found to be $6.64{\pm}0.03\;and\;5.86{\pm}0.05$ (logarithm stability constant values), respectively at 35$^{\circ}C$ and ionic strength 0.1 M.
