Engineered nanoparticles in wastewater systems: Effect of organic size on the fate of nanoparticles

  • To verify the fate and transport of engineered nanoparticles (ENP), it is essential to understand its interactions with organic matter. Previous research has shown that dissolved organic matter (DOM) can increase particle stability through steric repulsion. However, the majority of the research has been focused on model organic matter such as humic or fulvic acids, lacking the understanding of organic matter found in field conditions. In the current study, organic matter was sampled from wastewater treatment plants to verify the stability of engineered nanoparticles (ENP) under field conditions. To understand how different types of organic matter may affect the fate of ENP, wastewater was sampled and separated based on their size; as small organic particular matter (SOPM) and large organic particular matter (LOPM), and dissolved organic matter (DOM). Each size fraction of organic matter was tested to verify their effects on nano-zinc oxide (nZnO) and nano-titanium oxide (nTiO2) stability. For DOM, critical coagulation concentration (CCC) experiments were conducted, while sorption experiments were conducted for organic particulates. Results showed that under field conditions, the surface charge of the particles did not influence the stability. On the contrary, surface charge of the particles influenced the amount of sorption onto particulate forms of organic matter. Results of the current research show how the size of organic matter influences the fate and transport of different ENPs under field conditions.

Screening of Anti-microbial and Anti-inflammatory Activity of Common Stalked Barnacle Pollicipes mitella Extract (거북손(Pollicipes mitella) 추출물의 항균 활성 및 항염증 활성 탐색)

  • This study screened the antimicrobial and anti-inflammatory activities of three extracts [1% acetic acid (HAc), distilled water (D.W.), and ethanol] from the common stalked barnacle Pollicipes mitella. Among the extracts, the 1% HAc extract showed the strongest antibacterial activity against several bacteria, but exhibited no activity against Candida albicans. To improve the degree of separation of the 1% HAc extract, solid-phase extraction was performed using a C18 cartridge with three solvents (D.W., 60A, and 100A). The 1% HAc 60A eluate showed the strongest antibacterial activity and enzyme, salt, and temperature stability, with no hemolytic activity. In addition, strong DNA-binding ability but no bacterial membrane permeability was observed. These results indicate that the P. mitella 1% HAc 60A eluate may contain antibacterial organic compounds that target intracellular components but not bacterial membranes. In addition, the 1% HAc 60A eluate exhibited potent inhibitory activity to reduce the production of inflammatory mediators (nitric oxide and prostaglandin E2) and pro-inflammatory cytokines (tumor necrosis factor-α, interleukin (IL)-6, and IL-1β) with no cytotoxicity. Therefore, the P. mitella 1% HAc 60A eluate has anti-inflammatory activity. Collectively, our results suggest that the P. mitella 1% HAc 60A eluate can be used as a bioactive source with antibacterial and anti-inflammatory activities.

Antimicrobial, antifungal effect and safety verification using BCOP assay of extracts from Coptis chinensis (황련(Coptis chinensis) 추출물의 항균, 항진균 효과와 BCOP 분석을 이용한 안전성 검증)

  • Coptis chinensis is used in oriental medicine for soothing, anti-inflammation, antimicrobial and antipyretic properties, and its main ingredient berberine is known to have strong antibacterial activity. In this study, we investigated the anti-microbial effect of hot water extract of Coptis chinensis (CW) on skin related microorganism and the airborne microbe, the antifungal effects of fungi, which are frequently detected in residential environments. CW showed antibacterial effect against Propionibacterium acnes, Staphylococcus aureus and Staphylococcus epidermidis, against the airborne microbe, which was collected in four different places. At the concentration of 100 mg/mL, the antimicrobial activity continued for 42 days, showed heat stability without change in the antimicrobial activity even after heat treatment. The MIC and MBC of CW against S. aureus was 0.03, 0.05 mg/mL, against S. epidermidis was 0.50, 0.75 mg/mL and against P. acne was 0.10, 0.15 mg/mL. As a result of measuring the MIC of four kinds of fungi with high detection frequency in the surrounding environment, Gliocladium virens was 65 mg/mL by determined as MIC which can inhibit one hundred percent of mycelial growth. The concentration 90 mg/mL was determined as MIC against Aureobasidium pullulans and 100 mg/mL against Penicilium pinophilum and Chaetomium globosum. CW was considered a safe extract that showed no irritation even in the ocular mucous membrane irritation evaluation test, a patch test. Therefore, these results suggest that Coptis chinensis has antimicrobial, antifungal and safety on human body and can be applied to the development of materials for cosmetic and residential environment industries.

A Study on Oxygen Reduction Reaction of PtM Electrocatalysts Synthesized by a Modified Polyol Process (수정된 폴리올 방법을 적용하여 합성한 PtM 촉매들의 산소환원반응성 연구)

  • In this research, we evaluated the performance and characteristics of carbon supported PtM (M = Ni and Y) alloy catalysts (PtM/Cs) synthesized by a modified polyol method. With the PtM/Cs employed as a catalyst for the oxygen reduction reaction (ORR) of cathodes in proton exchange membrane fuel cells (PEMFCs), their catalytic and ORR activities and electrical performance were investigated and compared with those of commercial Pt/C. Their particle sizes, particle distributions and electrochemically active surface areas (EAS) were measured by TEM and cyclic voltammetry (CV), while their ORR activity and electrical performance were explored using linear sweeping voltammetries with rotating disk electrodes and rotating ring-disk electrodes as well as PEMFC single cell tests. TEM and CV measurements show that PtM/Cs have the compatible particle size and EAS with Pt/C. When it comes to ORR activity, PtM/C showed the equivalent or better half-wave potential, kinetic current density, transferred electron number per oxygen molecule and $H_2O_2$ production(%) to or than commerical Pt/C. Based on results gained by the three electrode tests, when the PEMFC single cell tests were carried out, the current density measured at 0.6 V and maximum power density of PEMFC single cell adopting PtM/C catalysts were better than those adopting Pt/C catalyst. It is therefore concluded that PtM/C catalysts synthesized by modified polyol can result in the equivalent or better ORR catalytic capability and PEMFC performance to or than commercial Pt/C catalyst.

The Preparation and Physicochemical Properties of Dipalmitoylphosphatidylcholine/Cholesterol/Fluorinated Surfactant Vesicle Incorporated Fatty Acid Salt (불소화지방산염 첨가에 의한 디팔미토일포스파티딜콜린/콜레스테롤/불소화계면활성제 베지클의 제조와 물성 측정 연구)

  • The vesicle system of DPPC(dipalmitoylphosphaticylcholine)/Chol(Cholesterol) has been modified by incorporating various mole fractions of flourinated surfactant($C_8F_{17}(CH_2)_2OCO-CH_2CH(SO_3Na)COO(CH_2)_2C_8F_{17}$. Sodium bis(1H,1H,2H,2H-heptadecaflurododecyl)-2-sulfosuccinate, FS)/fluorinated fatty acid salt ($C_7F_{15}COONH_4$, ammoniumpentadecaflurooctyrate, FFS), and their physicochemical properties have been investigated in an attempt to enhance the stability of phospholipid vesicle system. The ${\zeta}$-potential measurement by use of Zetamaster sub-micron Particle Electrophoresis Analyzer (Malvern Co.) showed that a charged homogeneous DPPC/Chol/FS vesicle has been formed owing to the incorporated FFS effect on the membrane, playing a role as a cosurfactant in the bilayer between DPPC and FS components. With increase in the concentration of FFS, it was found that the particle size and also surface charge of the DPPC/Chol/FS vesicle decreased. The stability of DPPC/Chol/FS/FFS liposome was found to be enhanced significantly compared to that of DPPC/Chol/FS according to the dispersity change as a function of time. The release rate of dye molecule of Methylene Blue from the DPPC/Chol/FS/FFS vesicle was determined to be slower than that of DPPC/Chol/FS system, and it may be attributed to the increase in microviscosity of the hydrophobic region in the bilayer. The affinfinity of DPPC/Chol/FS/FFS vesicles to albumin was found to be slightly lowered compared to that of DPPC/Chol/FS. Based on these findings, it was confirmed that a more stable and homogeneous vesicle system of DPPC/Chol/FS could be prepared by addition of FFS, acting as a cosurfactant in the aggregate formation.

Antioxidative, and Inhibitory Activities on Melanogenesis of Vitex negundo L. Leaf Extract (목형 잎 추출물의 항산화 활성과 멜라닌 생합성에 대한 저해활성)

  • The aim of this study was to evaluate various aspects of Vitex negundo L. leaf extract, such as the antioxidative activity, tyrosinase inhibitory effects, and inhibitory activities on ${\alpha}$-MSH induced melanogenesis, and active component analysis. The DPPH (1, 1-diphenyl-2-picrylhydrazyl) scavenging activities ($FSC_{50}$) of the ethyl acetate fraction and aglycone fraction of V. negundo L. leaf extract were $14.51{\mu}g/ml$ and $13.96{\mu}g/ml$, respectively. A luminol-dependent chemiluminescence assay revealed that the reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of the aglycone fraction of V. negundo L. leaf extract on ROS generated in an $Fe^{3+}$-$EDTA/H_2O_2$ system was the most prominent at $0.22{\mu}g/ml$. The protective effects of the extracts fractions of V. negundo L. leaf against the rose-bengal sensitized photohemolysis of human erythrocytes were increased in a concentration dependent manner ($1{\sim}50{\mu}g/ml$). In particular, there were greater protective effects of the aglycone fraction on the cellular membrane than that of the fat-soluble antioxidant (+)-${\alpha}$-tocopherol. The inhibitory effects ($IC_{50}$) on mushroom tyrosinase were the highest for the ethyl acetate fraction ($IC_{50}$ = $48.58{\mu}g/ml$). The inhibitory effect on ${\alpha}$-MSH induced melanogenesis in B16 melanoma cells was 41.80% at $50{\mu}g/ml$ of ethyl acetate fraction. Active component analyses by TLC, HPLC and LC/ESI-MS revealed luteolin and isoorientin. These results indicate that V. negundo L. leaf extract can be used as an antioxidant for ROS scavenging. Particularly, the luteolin and isoorientin of the ethyl acetate fraction may be applicable to new whitening cosmetics because of its inhibitory effect on mushroom tyrosinase and ${\alpha}$-MSH induced melanogenesis in B16 melanoma cells.

Characterization of Microsomal $Ca^{2+}$ Uptake in Tomato Root Tissues (토마토 뿌리조직에서 분리한 마이크로솜의 $Ca^{2+}$ 흡수 특성)

  • In order to characterize the property of $Ca^{2+}$ transport in plant cell, microsomes were prepared from the roots of tomato and microsomal $^{45}Ca^{2+}$ uptake was measured. When 1 mM vanadate, a selective inhibitor of P-type ATPases, 50 mM $NO_3^-$, a specific inhibitor of vacuolar $H^{+}-ATPase$, and both of these inhibitors were treated, the microsomal $^{45}Ca^{2+}$ uptakes were inhibited by 20, 33 and 47%, respectively. The inhibitory effects of these two inhibitors were investigated by using a protonophore, gramicidin. When the chemical gradient of $H^{+}$ was relieved by gramicidin, the uptake was decreased by 30%, implying the presence of $Ca^{2+}/H^+$ antiporter in the microsomal membrane. In the $^{45}Ca^{2+}$ uptake experiment, the effect of gramicidin was independent of vanadate-induced inhibition. However, when the activity of vacuolar $H^{+}-ATPase$ was inhibited by $NO_3^-$, the effect of gramicidin was severely decreased. Meanwhile, thapsigargin, a specific antagonist of ER/SR-type $Ca^{2+}-ATPase$, inhibited the microsomal $^{45}Ca^{2+}$ uptake and the maximum inhibitory effect was obtained at $10\;{\mu}M$. The effect of thapsigargin was blocked by $NO_3^-$ and gramicidin, but not by vanadate. These results imply that vanadate directly inhibits the activity of $Ca^{2+}-ATPase$; however, $NO_3^-$ and thapsigargin block the activity of $Ca^{2+}/H^+$ antiporter by inhibiting the vacuolar $H^{+}-ATPase$. In conclusion, the microsomal $^{45}Ca^{2+}$ uptakes are mediated by two major enzymes, $Ca^{2+}-ATPase$ and $Ca^{2+}/H^+$ antiporter in tomato root tissue.

Effects of Dietary Activated Charcoal Mixed with Wood Vinegar on Quality and Chemical Composition of Egg in Laying Hens (목초액 함유 활성탄의 급여가 산란계에서 계란의 품질과 화학적 조성에 미치는 영향)

  • This experiment was conducted to investigate the effects of dietary supplementation of activated charcoal (AC) mixed with wood vinegar on performance and egg quality in laying hens. A total of one hundred ninety two, 56-wk-old Lohmann brown laying hens were divided into four groups, consisting four replicates of 12 birds each and fed one of the four experimental diets containing AC 0%, AC 0.5%, AC 1%, AC 2% for 5 wks. The blood profiles and contents of total lipid and cholesterol, fatty acid profiles in egg yolk were determined. There were no significant differences in laying performance, feed intake and vitelline membrane strength among the treatments. The eggshell breaking strength and Haugh unit were significantly increased in 2% AC group compared to the other groups (P<0.05). Egg yolk index in 2% AC group was significantly higher (P<0.05) than those of the control and 0.5% AC group and yolk color was significantly improved by the dietary AC (P<0.05). The levels of serum total cholesterol, HDL-cholesterol and enzyme activities were not influenced by feeding AC. Total lipid of egg yolk in 1 % and 2% AC groups was significantly decreased (P<0.05) compared to that of the control, although there was no significant difference in yolk cholesterol. In conclusion, the addition of activated charcoal with wood vinegar in layer diet resulted in improving egg quality by means of enhancing eggshell breaking strength, Haugh unit, yolk index and yolk color.

Antioxidative Activity and Component Analysis of Phellinus linteus Extracts (상황버섯 추출물의 항산화 및 성분분석)

  • In this study, the antioxidative effect, antibacterial, inhibitory effects on tyrosinase, inhibitory effects on elastase and component analysis of Phellinus linteus (P. linteus) extracts were investigated. The ethyl acetate fraction of P. linteus extracts ($2.94\;{\mu}g/mL$) showed the highest free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of P. linteus extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction ($0.0072\;{\mu}g/mL$) showed the most prominent ROS scavenging activity. The protective effects of extract/fractions of P. linteus extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The P. linteus extracts showed cellular membrane protective effects in a concentration dependent manner ($5{\sim}50\;{\mu}g/mL$). The inhibitory effect ($IC_{50}$) on tyrosinase of P. linteus extract was the highest at 50 % ethanol extract ($6.34\;{\mu}g/mL$), and the inhibitory effect ($IC_{50}$) on elastase of P. linteus was the highest at ethyl acetate fraction ($14.08\;{\mu}g/mL$). TLC, HPLC chromatogram and LC/ESI-MS of the ethyl acetate fraction obtained from P. linteus extracts were identified interfungin A (PL RPT-1a). These results indicate that extract/fractions of P. linteus can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging ROS, and protect cellular membranes against ROS. Extract/fractions of P. linteus can be applicable to new cosmeceuticals for antioxidant, antiaging, antiwrinkle and whitening.

Development of Natural Seasoning using Desalinated Tuna Boiled Extract (탈염된 참치 자숙액을 이용한 천연조미료 개발)

  • The hydrolysate of desalinated tuna boiled extract (TBE) were prepared by continuous hydrolysis of TBE using a membrane reactor. TBE and tuna boiled extract hydrolysate (TBEH) were isolated depending on molecular weights. The major molecular weight distributions of TBEH-l0K, TBEH-5K and TBEH-lK were 9,800Da, 3,000Da and 990Da, respectively. The amounts of nucleotides and their related compounds of TBE were 3.47 $\mu$mole/g AMP, 23.75 $\mu$mole/g IMP, 9.07 $\mu$mole/g inosine and 1.89 $\mu$mole/g hypoxanthine. Total content of amino acids having desirable taste (glycine, glutamic acid, alanine, proline, aspartic acid, serine) was about $63\%$ of total amino acid from TBE and about $62\%$ from TBEH. The natural seasoninings were prepared with TBE and TBEH. From the results of sensory evaluations, complex seasoning containing TBEH-1K was almost equal to the shellfish complex seasoning obtained from the market. The mixed sauce which was made by mixing of $50\%$ TBEH sauce and $50\%$ fermented soy sauce was similar to the tradition soybean sauce in product quality and it showed the possibility to be used for the substitute product for acid hydrolyzed soysauce.
