보골지 추출물이 B16 melanoma 세포주의 멜라닌 합성에 미치는 영향 (The Effects of Psoraleae fructus Extract on Melanin Synthesis of B16 Melanoma Cells)

  • Objectives : This study was carried out for the development of medicine for vitiligo treatment and focused on the effect of Psoraleae fructus extract on melanin synthesis of B16 melanoma cells. Methods : Activity of tyrosinase playing a vital role in the synthesis and quantity of melanin, which is the final product in cultured B16 melanoma cells, the effects of Psoraleae fructus extract were measured. Results : The results indicated that Psoraleae fructus extract increased beth the amount of melanin and the activity of tyrosinase according to concentration, also supported by western blot analysis. Conclusions : The results suggest that Psoraleae fructus extract has an advantageous effect on the promotion of melanin synthesis and will contribute to the development of vitiligo treatment through further related studies.

B16 흑색종세포에서 로바스타틴에 의한 멜라닌 합성 촉진효과에 미치는 세포내 칼슘의 역할 (Role of Intracellular $Ca^{2+}$ in the Lovastatin-Induced Stimulation of Melanin Synthesis in B16 Melanoma Cells)

  • Although statins, inhibitors of 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase, have been shown to increase melanin synthesis, the exact mechanism of this action is not fully understood. In this study we investigated the possible involvement of intracellular $Ca^{2+}$ signal in the mechanism of stimulation of melanin synthesis induced by lovastatin in B16 cells. Lovastatin stimulated the production of melanin in a dose-dependent manner in the cells. Treatment with mevalonate, FPP and GGPP, precursors of cholesterol, did not significantly suppress the lovastatin-induced melanin production, suggesting that inhibition of cholesterol synthesis may not be involved in the mechanism of the action of lovastatin. In addition, lovastatin did not significantly alter the cAMP concentration and the stimulated production of melanin by lovastatin was not significantly changed by treatment with H89, a potent inhibitor of protein kinase A, which demonstrates that cAMP pathway may not be involved. However, lovastatin increased intracellular $Ca^{2+}$ concentration in a dose-related fashion. Treatment with EGTA, an extracellular $Ca^{2+}$ chelator did not significantly alter the lovastatin-induced intracellular $Ca^{2+}$ increase and melanin synthesis, whereas intracellular $Ca^{2+}$ reduction with BAPTA/AM and intracellular $Ca^{2+}$ release blockers (dantrolene and TMB-8) completely blunted these actions of lovastatin. Taken together, these results suggest that the intracellular $Ca^{2+}$ release may play an important role in the lovastatin-induced stimulation of melanin synthesis in B16 cells. These results further suggest that lovastatin may be useful for the treatment of hypopigmentation disorders, such as vitiligo.

Melanin Synthesis Inhibitory Effect of Eriobotryae Folium Extracts & Eriobotryae Folium and Phreatic Water Mixture

  • Objectives: As interests in the beauty of skin is growing continuously, more people are focusing on white and clean skin. Melanin is the major factor that determines skin color. The abnormal concentration of melanin causes various skin diseases such as vitiligo, freckles, and melasma. This study investigated the inhibitory effect of Eriobotryae Folium extracts (EF) with phreatic water (PW) on the melanin synthesis. Methods: The effect of EF on melanin synthesis was evaluated by using mouse melanoma cells (B16F10). To define the mechanisms, real-time PCR and western blot were used. We also evaluated the inhibitory effects of EF and PW on melanin synthesis by using HRM-2 melanin-possessing hairless mice. After UVB irradiation, melanin differences between the skin parts that were treated and untreated with EF and PW. Levels of mRNA were measured by real-time quantitative PCR and histological analysis of the dorsal skin was conducted by hematoxylin and eosin staining. Results: EF inhibited various mechanisms of melanogenesis, and the effect was increased when combined with PW. In vitro experiments have shown that EF inhibited the expressions of tyrosinase related protein-1 (TRP-1) mRNA, tyrosinase mRNA, microphthalmia-associated transcription factor (MITF) mRNA and the tyrosinase inhibitory activation, but it stimulated the extracellular regulated kinase (ERK) mRNA expression. In vivo experiments have shown that EF prevented melanogenesis in the mice dorsal skin and inhibited TRP-1 mRNA expression. Also these effects were increased when combined with PW. Conclusions: EF and PW might be a new and effective treatment for whitening and treating pigmentation of skin.

The effects of green tea (Camellia sinensis) flower extract on melanin synthesis in B16-F10 melanoma cells

  • The present study observed the effects of a green tea (Camellia sinensis) flower extract (GTFE) on melanin synthesis in B16-F10 melanoma cells. GTFE exhibited antioxidant activity on 2,2-diphenyl-1-picrylhydrazyl and inhibited mushroom tyrosinase activity in a dose-dependent manner. Furthermore, GTFE significantly diminished ${\alpha}-melanocyte$ stimulating hormone (${\alpha}-MSH$) stimulated cellular melanin content and tyrosinase activity throughout the concentration range evaluated. Based on RNA sequencing analysis, differential gene expression patterns observed in ${\alpha}-MSH$ stimulated B16-F10 melanoma cells were normalized by the addition of GTFE. In particular, the expression levels of melanoregulin and tyrosinase genes which are key regulating genes in melanin synthesis were up-regulated by 3.5 and 3 fold respectively by ${\alpha}-MSH$, and were normalized to control levels by the addition of GTFE. The results suggest that GTFE inhibits melanin synthesis in ${\alpha}-MSH$ stimulated B16-F10 melanoma cells by normalizing expression of genes that are essential for melanin synthesis. Overall, the results suggest that GTFE could be applied in the development of a whitening agent for the treatment of dermal hyperpigmentation.

오미자(五味子)의 멜라닌 생성 억제와 미백효과에 관한 연구 (A Study on the Melanin Synthesis Inhibition and Whitening Effect of Schizandrae Fructus)

  • Objective : The aim of this study is to assess the effect of Schizandrae Fructus on melanin synthesis inhibition and whitening effect. Methods : We assessed inhibitory effects of Schizandrae Fructus on melanin-release from B16F10, on melanin production in B16F10, on mushroom tyrosinase activity in vitro, on tyrosinase activity in B16F10 and effect of Schizandrae Fructus on the expression tyrosinase, TRP-1, TRP-2, PKA, ERK-1 ERK-2, AKT-1, MITF in B16F10. Results and Conclusion : 1. Schizandrae Fructus inhibited melanin-release, melanin production in B16F10. 2. Schizandrae Fructus inhibited tyrosinase activity in vitro and in B16F10. 3. Schizandrae Fructus suppressed the expression of tyrosinase, TRP-1, TRP-2, PKA, ERK-2 in B16F10.

  • The purpose of this work was to study the effect of extracts from pharbitidis seed and watermelon's inner shell on tyrosinase activity. For this purpose, the effects of plant extracts on total melanin synthesis and tyrosinase activity were measured. The results showed that the extracts effectively inhibited the tyrosinase activity and total melanin synthesis. The non-cytotoxicity of the plant extracts was confirmed by MTT assay.

현미 주정 추출물의 항산화 활성 및 melanin 합성 촉진 효과 (Ethanolic Extract of Oryza sativa Displays Antioxidative Activity and Promotes Melanin Synthesis)

  • 모발의 검정색은 노화됨에 따라 백색으로 변화된다. 이러한 현상은 모낭내 tyrosinase 활성의 감소와 $H_2O_2$에 의한 축적에 의하여 기인된다. 따라서 본 연구의 목적은 현미주정추출물(OREE)를 이용하여 백발화의 원인인 과산화 수소에 대한 항산화 활성과 멜라닌 생성 촉진 효과를 조사한 것이다. 본 연구에서 OREE은 낮은 DPPH radical 소거능과 환원력을 보여주었다. 그러나 B16F1 세포내에서 $H_2O_2$ 소거에 대해서는 높은 항산화효과를 나타내었다. 그리고 OREE는 in vitro에서 DOPA 산화 활성은 나타나지 않았지만 $64{\mu}g/ml$에서 tyrosinase 활성을 증가시켰다. MTT assay에서 OREE는 $32{\mu}g/ml$ 이상의 농도에서 세포독성을 나타내었다. 또한 OREE는 $8{\mu}g/ml$ 이상의 농도에서 멜라닌 합성은 농도에 비례하여 증가하였고, $H_2O_2$로 멜라닌 생성을 저하시킨 세포에서도 멜라닌 합성을 증가시켰다. 멜라닌 합성에 대한 OREE의 효과를 확인하기 위하여 Western blot 분석이 수행되었다. $H_2O_2$로 멜라닌 생성을 저하시킨 세포에서 멜라닌 합성 기전 관여하는 tyrosine hydroxylase와 tyrosinase-related protein-2 (TRP-2) 발현은 OREE의 존재 하에서 증가하였다. 이상의 발견들은 OREE가 멜라닌 합성을 촉진시킬 수 있어 이와 관련된 모발화장품의 개발에 적용시킬 수 있다는 것을 시사하고 있다.

Influence of atmospheric pressure plasma on the melanogenesis in melanoma cells

  • Melanin is a black pigment, responsible for hair and skin color. In order to find the melanin stimulatory technique which prove useful for a gray and a white hair-preventive agent or tanning agent, we developed atmospheric pressure plasma jet (APPJ) and tested for tyrosinase activity and melanin production in melanoma (B16F10) cells in vitro. We found plasma dose dependent increase in melanin production. To explore the contributing mechanism in melanin synthesis, intracellular reactive oxygen species (ROS) and MAP kinase signaling pathways were studied. Furthermore, the development of plasma technology for melanin synthesis and planning for in-vivo future studies will be discussed.

  • Purpose: This study was performed to determine the inhibitory effect of Soyosangagamhwajae(SYG) on melanin synthesis in B16F10 mouse melanoma cell. Methods: The Inhibitory effects of Soyosangagamhwajae(SYG) on melanin synthesis were determined by in-vitro assay. To elucidate inhibitory effects of SYG on melanin synthesis, we determined the melanin release in B16F10 cell. And to investigate the action mechanism, we assessed the gene expression of tyrosinase, TRP-1, TRP-2. PKA, $PKC{\beta}$ in B16F10 cell. Results: 1. SYG significantly inhibited melanin-release in B16F10 cell. 2. SYG significantly inhibited mushroom tyrosinase activity in vitro. 3. SYG significantly suppressed the expression of tyrosinase in B16F10 cell. 4. SYG significantly suppressed the expression of TRP-1, TRP-2 in B16F10 cell. 5. SYG significantly suppressed the expression of PKA, $PKC{\beta}$ in B16F10 cell. Conclusion: From these results, it may be concluded that SYG has the antimelanogenetic effect.

Enhancement of Melanin Synthesis by the Branch Extracts of Vaccinium oldhamii through Activating Tyrosinase Activity in B16F10 Melanoma Cells

  • This study was conducted to investigate the effect of branch extracts of Vaccinium oldhamii (VOB) on melanin synthesis in B16F10 cells. VOB promoted melanin production in absence or presence of ${\alpha}$-melanocyte-stimulating hormone (${\alpha}-MSH$) in B16F10 cells. However, VOB did not affect the expression of tyrosinase and TRP-1 associated with melanin synthesis at the mRNA and protein levels in B16F10. But, VOB decreased TRP-2 protein level and induced tyrosinase activation in B16F10 cells. Inhibition of tyrosinase activity and tyrosinase knockdown attenuated VOB-mediated melanin synthesis. In conclusion, VOB may stimulate melanin synthesis through activating tyrosinase activity.
