Hibiscus syriacus exhibited promising potential as a new source of food and colorants containing various anthocyanins. However, the function of anthocyanins from H. syriacus has not been investigated. In the current study, we evaluated whether anthocyanins from the H. syriacus varieties Pulsae and Paektanshim (PS and PTS) inhibit melanin biogenesis. B16F10 cells and zebrafish larvae were exposed to PS and PTS in the presence or absence of ${\alpha}$-melanocyte-stimulating hormone (${\alpha}$-MSH), and melanin contents accompanied by its regulating genes and proteins were analyzed. PS and PTS moderately downregulated mushroom tyrosinase activity in vitro, but significantly decreased extracellular and intracellular melanin production in B16F10 cells, and inhibited ${\alpha}$-MSH-induced expression of microphthalmia-associated transcription factor (MITF) and tyrosinase. PS and PTS also attenuated pigmentation in ${\alpha}$-MSH-stimulated zebrafish larvae. Furthermore, PS and PTS activated the phosphorylation of extracellular signal-regulated kinase (ERK), whereas PD98059, a specific ERK inhibitor, completely reversed PS- and PTS-mediated anti-melanogenic activity in B16F10 cells and zebrafish larvae, which indicates that PS- and PTS-mediated anti-melanogenic activity is due to ERK activation. Moreover, chromatography data showed that PS and PTS possessed 17 identical anthocyanins as a negative regulator of ERK. These findings suggested that anthocyanins from PS and PTS inhibited melanogenesis in vitro and in vivo by activating the ERK signaling pathway.
Panax ginseng is known for various pharmacological activities mainly due to saponins. Since minor saponins, generated by the decomposition of major saponins, generally exert higher activities than major saponins, the fermentation may increase the minor saponin contents in ginseng products. In this study, we tested fermented ginseng extract whether or not provide a safe cosmetic ingredient for whitening purpose. In this regard, fermented Ginseng extract was prepared and evaluated the inhibitory activity toward tyrosinase and the melanin synthesis suppression. The safety was tested via cell viability and toxicity test. The skin lightening effect was also evaluated by clinical study. The fermented Ginseng extract exerted higher activities in tyrosine inhibition and in suppressing melanin synthesis compared to Kojic acid and arbutin. In the clinical test, skin lightening effecte of the sample was clearly higher than vehicle or Vitamin C. We thus concluded that the fermented Ginseng extract may provide a safe cosmetic ingredient for skin lightening purpose.

In this study, we investigated the melanogenesis inhibitory effects of Rubus crataefifolius Leaf Extract (RCLE) in B16F10 melanoma cells. We examined the effects of RCLE on the melanin contents and tyrosinase activity, as well as the protein expression levels of the melanogenic enzymes TRP-1, TRP-2, and MITF in α-MSH -stimulated B16F10 melanoma cells. RCLE effectively inhibited tyrosinase activity and melanogenesis, suppressed the phosphorylation of PKA and CREB, and expression of MIT involved in the melanogenesis pathway, and down-regulated expression of melanogenesis related proteins. These result suggest that RCLE inhibited α-MSH-stimulated melanin synthesis by suppressing MITF expression. Therefore, our study suggests that RCLE has potential as a safe treatment for excessive pigmentation or as a natural ingredient in cosmetics.
The melanin-concentrating hormone (MCH), a cyclic hypothalamic peptide composed of 17 amino acids, was initially identified in chum salmon (Oncorhynchus keta) as a regulator of pigmentation. Mammalian MCHs are cyclic hypothalamic peptides composed of 19 amino acids that regulate food intake and energy homeostasis. The present study examined not only MCH expression of different tissues but also the melanohore aggregation and intracellular $Ca^{2+}$ influx of fMCH and the other MCH. Real-time qPCR showed that MCH expressed specially in the brain, gonad, and ovary, and expression of MCH was observed during the developmental stages. In the application of synthetic fMCH and both types of synthetic fMCH, dN-fMCH and dC-fMCH, scale melanophore induced significant changes in aggregation activity with various concentrations of MCH. Also, compared to hMCH and sMCH, fMCH exhibited a 36~99.85% increase in relative potency (%), whereas aggregation of dN-fMCH and dC-fMCH remained in a high concentration. However, dispersion was induced rapidly according to be low concentration of dN-fMCH and dC-fMCH. We show that fMCH and its derivates were bound human MCHR1 and rat MCHR expressed in HEK293T cells with nano-molar affinity and are likely to be ligand-induced to mobilize intracellular $Ca^{2+}$. These results may provide new ligands for binding assay with MCHew ligands, as a structure similar to the mammalian MCH structure was discovered in fish. Once the fMCH receptor system is in place, it can be compared to the MCH system of mammals in terms of MCH function.

Melanin pigmentation in human skin is a major defense mechanism against ultraviolet light of the sun. Tyrosinase(EC 1.14.18.1) plays a key role in the biosynthesis of ultraviolet of melanin. This is why much researches have been focused on its regulation in controlling the epidermal melanization. We have found that the water-extract of Banha(Pinelliae ternate B.), an oriental medicinal plant, has no tyrosinase inhibitory activity, but does inhibit the melanin biolsynthesis in B16 mouse melanin cells. We also found that Banha extract lowers the tyrosinase activity in cultured cells. To elucidate the action mechanism of Banha extract we have investigated its effect on the tyrosinase mRNA level using reverse transcription-polymerase chain reaction technique. It was revealed that Banha extract reduced the tyrosinase mRNA level in dose dependent manner; when B16 mouse melanoma cells were cultured with 2mg/ml and 5mg/ml of Banha extract, there were 20% and 44% decrease in tyrosinase mRNA level, respectively. These data suggest that the Banha extract exerts its melanogenic inhibitory effect through the transcriptional regulation of tyrosinase mRNA.
Skin pigmentation, also known as complexion coloration, results from the biosynthesis of melanin within the melanocytes of the Stratum basalum and the subsequent transfer, translocation, and degradation of this pigment to, in, and by the neighboring keratinocytes respectively, Melanins are produced and retained in melanosomes synthesized in the cell body that are translocated along the dendrites using microtubules via motor proteins. Melanosomes are eventually captured and retained at the tips of dendrites by attachment to the peripherally localized actin. Melanosomes reaching the dendritic tips are transferred to keratinocytes, primarily via phagocytosis of released melanosomes by keratinocytes. Molecules responsible for cell/cell recognition and interaction that regulate transfer are being identified. Some of these putative mediators appear to be affected by ultraviolet radiation. After the keratinocytes receive melanosomes, the granules are distributed individually or as clusters in dark versus light skin respectively. These melanosomes are then aggregated over the nucleus for photoprotection ofkeratinocyte DNA and eventually degraded.
In this paper, we introduce a digital cross-polarization and fluorescent color imaging system for quantitative evaluation of skin lesions. We describe the characterization of the imaging systems and the quantitative image analysis methods to show the feasibility for quantitative evaluation of skin lesions. The polarization color image was used to compute erythema and melanin index image which are useful for quantitative evaluation of pigmentation and vascular skin lesions, respectively. The fluorescent color image was used to quantitatively evaluate "sebum" and "vitiligo". In quantitative evaluation of various skin lesions, we confirmed the clinical efficacy of the imaging systems for dermatological applications. Finally, we sure that the imaging systems can be utilized as important assistant tools for the evaluation of skin lesions by providing reproducible quantitative result for widely distributed skin lesions.

In the search for novel, natural melanogenesis inhibitors, a new sesquiterpene, inularin, was isolated from the flowers of Inula britannica, and the structure was determined using spectroscopic and chemical methods. The antimelanogenic effects of inularin on B16F10 melanoma cells and zebrafish embryos were evaluated. Inularin dose-dependently reduced melanocyte-stimulating hormone-induced melanin production and L-DOPA oxidation in B16F10 cells. Zebrafish embryos were used to confirm the antimelanogenic activity. Inularin significantly decreased the pigmentation of embryos compared with untreated controls.
Although various ophthalmic imaging methods, including fundus photography and optical coherence tomography, have been applied for effective diagnosis of ocular diseases with high spatial resolution, most of them are limited by shallow imaging penetration depth and a narrow field of view. Also, many of those imaging modalities are optimized to provide microscopic anatomical information, while functional or cellular information is lacking. Compared to other ocular imaging modalities, photoacoustic imaging can achieve relatively deep penetration depth and provide more detailed functional and cellular data based on photoacoustic signal generation from endogenous contrast agents such as hemoglobin and melanin. In this paper, array-based ultrasound and photoacoustic imaging was demonstrated to visualize pigmentation in the eye as well as overall ocular structure. Fresh porcine eyes were visualized using a real-time ultrasound micro-imaging system and an imaging probe supporting laser pulse delivery. In addition, limited photoacoustic imaging field of view was improved by an imaging probe tilting method, enabling visualization of most regions of the retina covered in the ultrasound imaging.
Tyrosinase plays an important role in the process of melanin biosynthesis, and it is a biochemical target enzyme for skin-whitening agents and the remedy for disturbances in pigmentation. We have screened 25 natural plant extracts for inhibitory effects against DOPA oxidase activity of tyrosinase. For the inhibitory effect against DOPA oxidase activity of tyrosinase, the recombinant human tyrosinase and the purified mushroom tyrosinase were used. Each of the dried plants extracted with methanol, and then the extracts were subjected to sequential fractionations with methylene chloride, ethyl acetate, n-butanol and water, respective The methylene chloride fractions of Angelica tenuissima, Nardostachys chinensis, Bombyx mop and Saposhnikovia divaricata, and the n-butanol fraction of Bombyx mori notably inhibited the human tyrosinase activity as well as mushroom tyrosinase activity (more than 90% inhibition). This study suggests that the above extracts have a potential as whitening agents as single ingredients or in combination with other of the extracts.
