Anti-oxidant, anti-inflammatory and Whitening effect of Benincasa hispida seed extract (동과자 추출물의 항산화, 항염 및 미백 효능)

  • To investigate the natural cosmetic ingredients of Benincasa hispida seed extract on skin care, we measured anti-oxidant and anti-inflammatory, and whitening effect. DPPH free radical scavenging activity was increased in a dose-dependent manner. The total phenolic content was higher in methanol extract (22.42±0.002 mgGAE/g) than water extract (9.77±0.002 mgGAE/g). MTT assay was demonstrated that the seed extract did not have a cytotoxic effect in RAW264.7 and B16F10 cell lines. We also examined to find out the inhibitory activity on NO production and secretion of TNF-α cytokine in LPS-induced RAW264.7 cells. In B16F10 melanoma cells, the seed extract significantly suppressed α-MSH induced melanin synthesis. Furthermore, westernblot analysis revealed that methanol extract dramatically downregulated the expression level of MITF, TRP-1 and TRP-2. Taken together, the B. hispida seed extract posses anti-oxidant, anti-inflammatory and skin whitening activities, which might provided its functional efficacy in cosmetic materials.

Biological Activities of Cosmetic Material from Ten Kinds of Flower Ethanol Extracts (화장품 소재로서의 꽃 10 종 에탄올추출물 생리활성 특성연구)

  • Background: We investigated the antioxidant, anti-wrinkles, whitening, and moisturizing properties and amounts of phenolic compounds of ethanol extracts from flowers of 10 resource plants from Namwon and Mt. Jiri., Korea. Methods and Results: We measured antioxidant efficacy based on the total polyphenol, and total flavonoid content, and the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. We evaluated the inhibitory effect on melanin synthesis and tyrosinase activity for the whitening effect. Furthermore, we analyzed the elastase and matrix metalloproteinase-1 (MMP-1) inhibition activity for anti-wrinkle capacity. To evaluate the moisturizing effect, we examined hyaluronan synthase (HAS) mRNA expression. In addition, the 19 phenolic compounds were detected using high performace liquid chromatography (HPLC). Among the 10 flowers, the antioxidant effect was high in the order of Rosa multiflora, Nelumbo nucifera, and Elsholtzia splendens. Whitening effect was high in the order of N. nucifera, R. multiflora, and Dendranthema zawadskii. As for the anti-wrinkle property, N. nucifera was the most effective followed by R. multiflora. Taraxacum coreanum was the best for moisturizing effect, followed by D. zawadskii, and E. splendens. Seven phenolic compounds were detected in the extracts of the 10 flowers. Conclusions: Overall, the extracts of five flowers extracts showed strong potential as antioxidant, whitening, anti-wrinkle, and moisturizing functional cosmetic agents.

Whitening Activity of Abeliophyllum distichum Nakai Leaves According to the Ratio of Prethanol A in the Extracts

  • In this study, we evaluated the whitening activity of prethanol A and water extracts from Abeliophyllum distichum Nakai. The extracts were prepared using 0, 50, 70, and 100% prethanol A at $121^{\circ}C$, 1.2 atm for 15 minutes. To confirm effective extraction, the acteoside content of each extract was analyzed with the HPLC-PDA method. The antioxidant activity was evaluated using DPPH and ABTS scavenging activity assays, and the whitening activity was evaluated based on inhibitory activities on the protein and mRNA expression of tyrosinase, tyrosinase-related protein 1 (TRP-1), tyrosinase-related protein 2 (TRP-2), and microphthalmia-associated transcription factor (MITF) in B16 F10 cells. Each extract showed strong antioxidant and whitening activity. $IC_{50}$ values of antioxidant activity from each extract were in order of 100%, 70%, 50%, and 0%. In addition, whitening activity inhibited the protein and mRNA expression of melanin synthesis factor, following the same pattern as antioxidant activity. In conclusion, water and prethanol A extracts of A. distichum showed effective antioxidant and whitening activity and are thus considered to be valuable materials for whitening cosmetics. The results of this study will also provide basic data for the safe and efficient production of A. distichum as a cosmetic material.

Anti-melanogenesis activity of Ecklonia cava extract cultured in tanks with magma seawater of Jeju Island

  • Ecklonia cava is popular in Korea as a marine functional materials. E. cava is generally collected and used on the coast of Jeju Island. However, the continuous use of collected natural E. cava may be limited because difficult to secure throughout the year and may be exposed to environmental pollution. Jeju magma seawater (MSW) was known to be significant advantages such as safety, cleanness, stability, and functional improvement. Attempts have been reported on application of MSW to the culturing of macro- and microalgae and showed improved results. Thus, the objective of the present study was to explore the anti-melanogenesis activity of brown seaweed E. cava (E. cava cultured with MSW [MSWE]) extract cultured in tanks with MSW of Jeju Island to evaluate the possibility of cosmeceutical industrial application. MSWE extract showed the higher polyphenolic and dieckol contents than natural E. cava (NE) extract. Anti-melanogenesis activity of MSWE extract and NE extract are tested and compared using tyrosinase and dihydroxyphenylalanine (DOPA) oxidation inhibition assay. MSWE extracts evidenced more effective tyrosinase and DOPA oxidation inhibition activity than that of the NE extracts and the commercial whitening agent, arbutin. MSWE extracts also markedly inhibited melanin synthesis and decreased the expression of melanogenesis-related protein in ${\alpha}$-melanocyte stimulating hormone-stimulated B16F10 melanoma cells without cytotoxicity. These results suggest that MSW cultivation process would be more effective in releasing bioactive compounds with whitening effect from seaweed such as E. cava at an industrial scale.

Inhibition of adipogenesis and melanogenesis by methanol extract of Codium fragile (Suringar) Hariot in 3T3-L1 adipocytes and B16F10 melanocytes (3T3-L1 및 B16F10 세포에서 청각 메탄올 추출물에 의한 지방 세포 분화 및 멜라닌 생성의 억제 효과)

  • Codium fragile (Suringar) Hariot, a green alga of the Codiales family, has been reported to have several bioactive properties, including antioxidant and anti-inflammatory properties. However, its antiobesity and whitening effects and their underlying mechanisms are unclear. This study aimed to evaluate the antiobesity and melanogenesis inhibitory effects of C. fragile using methanol extracts of C. fragile (MECF). The results of this study revealed that MECF inhibited the accumulation of lipid droplets and triacylglycerol in differentiated 3T3-L1 adipocytes, which was associated with the inhibition of the expression of adipogenesis-related transcription factors, such as peroxisome proliferator-activated receptor γ, CCAAT/enhancer-binding protein-α (C/EBPα), and C/EBPβ, which function as the key regulators of adipogenesis. Also, MECF reduced tyrosinase activity and melanin content in B16F10 cells as well as the expression of tyrosinase, tyrosinase-related protein-1 (TRP-1), TRP-2, and microphthalmia-related transcription factor in the presence of α-melanocyte-stimulating hormone. Taken together, our findings suggest that the extract of C. fragile could be considered a promising functional ingredient for the prevention and treatment of obesity and skin pigmentation in the food and cosmetic industry.

Development of a Skin Index Using Skin Characteristic Factors and Skin Biomarkers of Korean Women According to H igh Temperature and Low Humidity Environments (고온건조 환경에 따른 한국 여성의 피부 특성인자와 피부 바이오 마커를 활용한 피부 지수 개발)

  • In this study, basic skin characteristic data was measured by measuring skin hydration, skin sebum secretion rate, skin melanin index, skin redness index, skin redness image analysis, transepidermal water loss (TEWL), and amount of stratum corneum before and after creating a temporary high temperature and low humidity environments targeting Korean women in their 20s to 50s. Stratum corneum by tape stripping was collected at each measurement and skin biomarkers including total protein content, carbonylated protein, neutral lipid, and lipid peroxidation were analyzed. Based on the results, the differences before and after creating a high temperature and low humidity environments were confirmed, the correlation between skin characteristics and skin biomarkers was confirmed, and a new skin index was created based on this. The new skin index can be used in product efficacy evaluation, and the possibility of constructing a new clinical study method and using skin biomarker discovery research through additional research was confirmed.

Study on the Bioactive Characteristics of Morinda citrifolia as a Cosmetic Raw Material (화장품 소재로서의 노니 추출물에 관한 특성연구)

  • This study attempted to investigate the possibility of the use of Morinda citrifolia (MC) as a cosmetic ingredient from its physiological activities such as antioxidant activity, cytotoxicity and anti-aging effect. MC is a tropical plant that has been used as traditional polynesian foods and medicines for over two thousand years. It has been reported that this shrub can improve antimicrobial, anti-cancer and anti-inflammatory effects and strengthen an immune system. The in vitro antioxidant activity of MC was performed to see the DPPH scavenging activity by measuring total polyphenol content and total flavonoid content. As a result, a lack of any cytotoxicity was confirmed in human dermal fibroblasts (HDF) cell. When MC extract at a concentration of over $50{\sim}100{\mu}g/mL$ was added, MMP-1 expression considerably diminished. In an in vivo test, in addition, cream containing MC extract was prepared and applied to a total of 22 women in their 30 ~ 50s in ages in the morning and in the evening for four weeks. Changes in keratin, melanin index, pore, skin color and wrinkles under the naked eyes were then comparatively measured. Keratin levels slightly increased in the control group but decreased in the experimental group. In addition, wrinkles diminished in the experimental group. This study found that MC extract controls many MMP-1 related mechanisms with great potential for use as a natural ingredient of anti-aging cosmetics.

Physiological Activity of Supercritical Poria cocos back Extract and Its Skin Delivery Application using Epidermal Penetrating Peptide (초임계 복령피 추출물의 생리활성 및 경피투과 펩티드를 이용한 경피 약물전달의 응용)

  • In this study, Poria cocos bark were extracted by supercritical process, and anti-inflammatory, whitening, and antioxidant effects were measured in comparison with ethanol extract. Also, An effective percutaneous permeation method using a selected formulation of the extract and a drug delivery peptide was proposed. Pachymic acid, known as the anti-cancer and anti-inflammatory compound of the ventricle, is an indicator component and the HPLC analysis shows that the supercritical extract of the pericardium is more than twice that of the Poria cocos bark extract. In order to confirm antioxidative effect of Bombyx mori, DPPH scavenging ability and ABTS scavenging ability test showed that the ethanol extract of Poria cocos Back had lower concentration than the supercritical extract of Poria cocos back. However, RAW 264.7 Measurements of Nitric oxide (NO) production in cells showed lower NO production at the same concentration than the Poria cocos back ethanol extract. In addition, after 72 hours of processing of $20{\mu}g/mL$ of the Poria cocos back extract in B16 melanoma cells, both the intracellular and extracellular melanin extract were effective and the supercritical extract was lower melanin content. No toxicity was observed at the concentration of $800{\mu}g/mL$ in RAW 264.7 cells used in NO production experiments. However, in B16 melanoma cells, even at $50{\mu}g/mL$, both Poria cocos back ethanol extract and supercritical extract showed a survival rate of less than 60%. The liposome formulation and drug delivery peptides were shown to be useful for percutaneous permeation of Supercritical Extract of Poria cocos back using a liposome formulation and a drug delivery peptide. it is expected that there will be great potential for development as a variety of cosmetic materials for Poria cocos back.

Loss of EMP2 Inhibits Melanogenesis of MNT1 Melanoma Cells via Regulation of TRP-2

  • Melanogenesis is the production of melanin from tyrosine by a series of enzyme-catalyzed reactions, in which tyrosinase and DOPA oxidase play key roles. The melanin content in the skin determines skin pigmentation. Abnormalities in skin pigmentation lead to various skin pigmentation disorders. Recent research has shown that the expression of EMP2 is much lower in melanoma than in normal melanocytes, but its role in melanogenesis has not yet been elucidated. Therefore, we investigated the role of EMP2 in the melanogenesis of MNT1 human melanoma cells. We examined TRP-1, TRP-2, and TYR expression levels during melanogenesis in MNT1 melanoma cells by gene silencing of EMP2. Western blot and RT-PCR results confirmed that the expression levels of TYR and TRP-2 were decreased when EMP2 expression was knocked down by EMP2 siRNA in MNT1 cells, and these changes were reversed when EMP2 was overexpressed. We verified the EMP2 gene was knocked out of the cell line (EMP2 CRISPR/Cas9) by using a CRISPR/Cas9 system and found that the expression levels of TRP-2 and TYR were significantly lower in the EMP2 CRISPR/Cas9 cell lines. Loss of EMP2 also reduced migration and invasion of MNT1 melanoma cells. In addition, the melanosome transfer from the melanocytes to keratinocytes in the EMP2 KO cells cocultured with keratinocytes was reduced compared to the cells in the control coculture group. In conclusion, these results suggest that EMP2 is involved in melanogenesis via the regulation of TRP-2 expression.

Verification of Biological Activities and Tyrosinase Inhibition of Ethanol Extracts from Hemp Seed (Cannabis sativa L.) Fermented with Lactic Acid Bacteria (대마씨 발효 추출물의 생리 활성 및 미백 활성 검증)

  • Hemp seed (Cannabis sativa L.; HS), an annual herbaceous plant in the Cannabis genus, has been reported to play various biological functions in immunity increase, atherosclerosis, constipation, hyperlipidemia prevention, anti-inflammatory, and anti-cancer. In recently years, as superfood, the growing interest in the health care benefits of hemp seed has led to increased consumption. In this study, we investigated the effect of an ethanol extract of HS fermented with lactic acid bacteria (Lactobacillus plantarum KCTC 3107, L. plantarum KCTC 3108, L. brevis BHN-LAB128, L. paracasei BHN-LAB129). An antibacterial activity against Staphylococcus aureus and Bacillus cereus were 13.99 mm and 15.17 mm, respectively. The ethanol extracts of fermented hemp seed by lactic acid bacteria that the contents of total polyphenol, total flavonoid content, DPPH radical scavenging activity, SOD-like activity, and ${\alpha}$-glucosidase inhibitory activity were increased compared to non-fermented hemp seed. Also, tyrosinase inhibitory activity of the fermented hemp seed (FHS), known to melanin increasing substance was increased. In these results, we suggested that FHS have effects of anti-oxidant, ${\alpha}$-glucosidase inhibitory activity, and tyrosinase inhibitory activity. Hence, we proposed that FHS has possible to development as functional foods and cosmetics.
