Effect of Expanded Rice Husk Medium on Rice Seedling for Machine Transplanting

  • Rice farmers can save labor and expenses by using expanded rice husk (ERH) as a seedling medium since ERH is lighter and cheaper than other commercial seedling media (CSM). This study was carried out to develop a method for rice seedling cultivation using ERH as a seedling medium. It is suggested that a mixture of $60\%$ of ERH and $40\%$ of a CSM could be used as a seedling medium; the planting densities would be 240g per tray for infant seedlings and 200 g for young seedlings; and nitrogen (N) would be applied at a rate of 1g per tray for infant seedlings prior to planting and 2g per tray for young seedlings with division. Great care should be taken to use $CO(NH_2)_2$ as an N-source fertilizer. These results would lay a foundation for the rice seedling cultivation with ERH as a medium.

Effect of Medium Composition on in vitro Propagation and Plantlet Regeneration from Nodal Explants of Cassava (Manihot esculenta Crantz)

  • The Cassava (Manihot esculenta Crantz) is a perennial woody shrub cultivated mainly in the tropics for its starchy tuberous roots. It belongs to the family Euphorbiaceae which also includes rubber (Hevea brasiliensis) and castor bean (Ricinus communis). Among tropical crops, rice, sugarcane, maize and cassava are the most important sources of calories for human consumption. Problems in the propagation of cassava are virus diseases and low rates of seed germination. Thus, a study was undertaken to develop an efficient in vitro mass propagation protocol of Manihot esculenta Crantz. Young and actively growing stem segments were excised from adult plants of cassava. Samples were cut into a 3~4 cm nodal segments with single node after sterilization, and cultivated in the different medium supplemented with various plant growth regulators for 4 weeks. For shoot multiplication, single-node stem segments, approximately 1 cm in length, were taken from in vitro derived shoots and subcultured. After 4~6 weeks, the shoot generation rate was 55.6%, the shoot number and its length were 1.0/explant and 2.3 cm in the most favorable medium composition. Our experiments confirmed that in vitro growth and multiplication of plantlets could depend on its reaction to the different medium composition, and this micropropagation techniques could be a useful system for healthy and vigorous plant production.

  • The Cassava(Manihot esculenta Crantz) is a tropical root crop, originally from Amazonia, that provides the staple food of an estimated 800 million people worldwide. It belongs to the family Euphorbiaceae which also includes rubber (Hevea brasiliensis) and castor bean (Ricinus communis). Among tropical crops, rice, sugarcane, maize and cassava are the most important sources of calories for human consumption. Problems in the propagation of cassava are virus diseases and low rates of seed germination. So we tried to optimize protocols for mass production of somatic embryo amenable to large-scale vegetative propagation of Cassava. After in vitro eight-week culture of leaves of Cassava, the medium which contained the 2,4-D, BAP and IBA showed the highest callus induction rate, embryogenesis callus formation rate and somatic embryo formation in Cassava culture. In the medium with GA3 and myo-inositol, shoots were most vigorously regenerated from somatic embryos of Cassava. Our experiments confirmed that in vitro growth and multiplication of plantlets could depend on its reaction to the different medium composition, and this micropropagation techniques could be a useful system for healthy and vigorous plant production.

  • Embryogenic callus cultures of Korean native Seosanjong of ginger(Zingiber of officinale Rosc.) were induced through stem explants taken from in vitro shoot-tip cultures. Among the four concentrations of 2,4-D tested in Murashige and Skoog medium, 0.5 and 1 mg/L of 2,4-D was most effective in inducing embryogenic callus. Leaf explants did not express any new morphogenetic response in all 2,4-D concentrations tested. Plantlets transferred to hormone-free MS medium were developed and successfully acclimatized under greenhouse.

톱밥배지 침공처리에 따른 표고 '화담' 의 균사배양 및 자실체 생산성 (Cultural Characteristics and Fruiting-body Productivity of New Cultivar 'Hwadam' (Lentinula edodes) by Punching Treatments on the Sawdust Medium)

  • 표고 '화담'의 톱밥배지 안정생산을 위해 침공처리가 균사배양 및 자실체 생산성에 미치는 영향을 조사하였다. 종균 접종후 배지 온도는 배양 23일에 27.0-28.8℃, CO2농도는 배양 16일에 14.0-15.6%까지 높아진 후 감소하였고, O2농도는 CO2농도와 반대의 경향이었다. 배양기간중 침공횟수가 많을수록 배지의 명도(L값)는 감소하고, 색차(∆E)와 중량감모율은 증가하였다. 침공처리에 따른 1주기 재배기간은 큰 차이를 보이지 않았지만, 침공처리 횟수가 늘어날수록 자실체 발이수가 증가하여 3회 침공처리구의 3주기 총수량이 605.7 g으로 가장 우수하였다.

Mycelial Yield of Pleurotus ostreatus Using Thinned Apple, Pear, and Peach on Submerged Culture

  • The effect of thinned fruits, apple, pear and peach, on the mycelial growth of mushrooms was investigated. The growth of mycelia with the addition of thinned fruit was clearly better than that in the control for all the tested mushrooms. The growth rate of Pleurotus ostreatus was faster than any other mushroom. The optimal concentrations of thinned apple, pear, and peach in a solid culture were 1.0%, 1.0%, and 3%, respectively, while in a liquid culture the optimal concentrations were 5,0%, 3.0%, and 5.0%, respectively. When Pleurotus ostreatus was incubated in a 20-L pilot scale fermenter with 10 L of a liquid medium containing 3% thinned fruit at 25$^{\circ}C$ and 6 vvm for 10 days, the mass-production of mycelia was 74.2 g/10 L (apple), 96.2 g/10 L (pear), and 86.3 g/10 L (peach). The mycelial yield of Pleurotus ostreatus in a medium containing thinned fruit was 2 ∼ 3 times higher than that in the control.

표고 수출용 완성형 배지의 적합 배지조성 선발 (Evaluation of medium composition for developing an export-quality complete medium for Lentinula edodes)

  • 수입되는 중국산 완성형 배지를 대체하고, 수출에 적합한 표고 완성형 배지를 개발하고자 본 연구를 수행한 결과 참나무톱밥+미송톱밥+파옥쇄(40:40:20, v/v) 혼합배지인 T13에서 pH 5.3으로 대조구와 비슷하였으며 C/N율은 82로 대조구 52 대비 높게 나타났다. 배지조성별 균사 생장을 조사한 결과 영양원으로 파옥쇄 혼합 시 균사 생장이 빨라지는 것을 확인할 수 있었으며, 대조구 대비 균사 생장이 빠르고 밀도가 높은 3가지 처리를 1차 선발하였다. 1차로 선발된 3가지 처리를 본 배지에 접종하고 배양특성을 조사한 결과 T12, T13, T18 3처리 모두 대조구 대비 배양기간이 단축되었고, 그 중 T13에서 대조구 대비 '농진고'는 12일, '참아람'은 10일 배양기간이 단축되었으며 특히 2품종 모두 갈변 소요일수가 크게 단축되었다. 1차 선발한 3처리를 재배생육한 결과 '농진고'는 T13에서 대조구 대비 갓이 크고 두꺼웠으며, 대도 길고 굵어 개체중이 높게 나타났다. '참아람' 품종에서도 비슷한 경향을 나타냈으나 대 직경은 대조구 대비 가는 형태를 나타내었다. 1주기부터 5주기까지의 배지조성에 따른 혼합배지별 수량은 파옥쇄가 영양원으로 혼합된 T12, T13, T18 배지조성에서 대조구 대비 모두 수량이 증가하였으며, 그 중 T13에서 대조구 대비 '농진고'는 5.9%, '참아람' 12.6% 증수되었다. 이러한 연구 결과를 바탕으로 참나무톱밥+미송톱밥+파옥쇄(40:40:20, v/v) 혼합배지를 표고 완성형 배지의 적합 배지로 최종 선발하였다.

  • Apple (Malus×domestica Borkh.; Rosaceae) is an important fruit crop grown mainly in temperate regions of the world. Tissue culture in vitro is a biotechnological technique that has been used to genetically improve cultivars (scions) and rootstocks. This could be important in the production of genetically uniform scions and rootstocks for commercial apple production. In nurseries, apple plants are produced by grafting scions onto rootstocks. The Cornell-Geneva (Geneva® series) breeding program has bred several dwarf rootstocks that are resistant to diseases and pests and are also cold hardy. This study was conducted to determine the optimal medium strength to improve sprouting shoot rate of apical meristem of the apple rootstocks of fire blight resistance. The apple rootstocks apical meristem at size (0.2 mm to 0.3 mm) with axillary buds were cultured on the MS(Murashige & Skoog) medium supplemented with plant growth regulators. The sprouting ratio and growth characteristics was evaluated after eight weeks in vitro culture. The highest rate of bud differentiation and shoot formation were 23.8% and 55.6%, respectively. After 6 weeks, shoots were regenerated from apical meristem, and their growth characteristics was significantly varied on the respective basal medium with different plant growth regulators. Our studies showed that the apple rootstocks the apple rootstocks of fire blight resistance plantlets could be successfully produced from apical meristem differentiated out of young twigs via organogenic regeneration.
