Overproduction of a γ-glutamyltranspeptidase from Bacillus amyloliquefaciens in Bacillus subtilis through medium optimization (배지최적화를 통한 재조합 바실러스 서브틸리스에서 바실러스 아밀로리퀴파시엔스 유래 γ-글루타밀펩타이드전달효소의 대량생산)

  • ${\gamma}$-Glutamyltranspeptidase (GGT, EC 2.3.2.2) transfers ${\gamma}$-glutamyl moiety from glutamine to amino acids or peptides and hydrolyzes glutamine to glutamate and ammonia. In order to overproduce ${\gamma}$-glutamyltranspeptidase from Bacillus amyloliquefaciens (BAGGT), the encoding gene was cloned and expressed in Bacillus subtilis. The productivity of BAGGT in Bacillus subtilis was improved by 42-fold by using a dual-promoter system that was generated by combining promoters from B. subtilis ${\alpha}$-amylase and BAGGT genes. Through optimization of medium composition by Plackett-Burman design and central composition design, BAGGT was produced at $18.3{\times}10^7U/L$ of culture in the optimized medium. Compared to previously used Luria-Bertani medium, the optimized culture medium (15 g/L molasses, 60 g/L corn steep liquor, 6 g/L yeast extract, 4 g/L NaCl, 6 g/L $K_2HPO_4$, and 2 g/L $KH_2PO_4$), resulted in a 4.3-fold increase in production of BAGGT.

  • Leuconostoc citreum is one of the representative strains of Leuconostoc spp. that show fast growth rates in fermented vegetables. Sequential experimental designs including the Plackett-Burman design, fractional factorial design, steepest ascent analysis, central composite design and response surface methodology were introduced tooptimize and improve the medium for Leuconostoc citreum. Fifteen medium ingredients were examined and glucose (20 g/l), yeast extract (12.5 g/l), sodium acetate trihydrate (6.12 g/l), potassium phosphate (42.55 g/l) and dibasic ammonium citrate (4.12 g/l)were chosen as the best components to give a critical and positive effect for cell-growth. The biomass was increased to 2.79 g/l (169%), compared to the 1.65 g/l in MRS medium.

  • This study was conducted to examine the possibility of producing poinsettia with various growing media containing used perlite and rockwool slabs as medium components. Pot plants of Euphorbia pulcherrima 'Freedom Red' were grown in 10 media containing used perlite and particles of rockwool slabs at various ratios including 1:1 and 1:3 (v/v). Medium containing used perlite and peatmoss at 1:3 (v/v) was employed as the control treatment. Height, root length, length of the longest branch, number of roots, leaf area, and width and length of the largest bract of plants grown in media containing used perlite were significantly greater than those in the control. Stem diameter and bract count were not affected by the medium composition. From the results, used perlite and rockwool slabs were proved to be a practically useful materials as medium components for pot production of poinsettia plants.

  • The uptake of phenolsulfonphthalein (PSP) and of paraaminohippuric acid (PAH) by cortical slices of the rabbit kidney was investigated while varying the composition of medium. The overall uptake of these substances displayed typical active transport characteristics and was significantly enhanced in presence of acetate. When the phosphate buffer was used the optimal pH was 7.4 for both substances. However, when the tris-buffer was used the optimal pH was 7.4 for PSP and 8.3 for PAH. Removal of $Na^+$ from the medium resulted in a significant reduction in the uptake. Similar results, though lesser in magnitude, were obtained when either $K^+\;or\;Ca^{++}$ was removed from the medium. However, there was no additive effect when $K^+\;and/or\;Ca^{++}$ were additionally removed from the $Na^+-free$ medium. The presence of ${NH_4}^+$ greatly reduced while $Li^+\;and\;Mg^{++}$ moderately reduced the uptake of both substances. However, choline had no effect. In substrate-leached slices, acetate greatly enhance the uptake of organic acids; but this action was not demonstrable in absence of $Na^+,\;K^+\;or\;Ca^{++}$.

  • A complex medium was developed for the production of microbial cellulose by Acetobacter xylinum ATCC 23769. The optimum concentration of each nutrient for the production of microbial cellulose was determined to be 10g peptone, 20g yeast extract, 5g glucose, 1.56g Na2HPO4, 1.8g KH2PO4, 0.05g MgSO4, 0.002g FeCl3, 5g citric acid and 10 mL ethanol per liter. With synergistic effects of citric acid and ethanol, cellulose productivity achieved in developed medium was 0.446 gram of cellulose per gram glucose for static culture, which is much higher than reported values. Cell growth and the cellulose production in the developed medium under static culture was also investigated.

  • Stable poly(methyl methacrylate-co-divinylbenzene) (poly(MMA-co-DVB)) microspheres were prepared by precipitation polymerization using acetonitrile as the main medium under various polymerization conditions, including modifications of the agitation speed, monomer and initiator concentrations, DVB content in the monomer mixture, and the use of various cosolvents. Gentle agitation was required to obtain smooth spherical particles. The individually stable microspheres were obtained at monomer concentrations of up to 15 vol% in an acetonitrile medium. The number-average diameter increased linearly with respect to increases in the monomer and initiator concentrations. We found, however, that the uniformity of the microspheres was independent of the variation of the polymerization ingredients because nuclei formation was solely influenced by the crosslinking reaction of the monomers. We obtained higher yields for the polymerization at higher concentrations of monomer and initiator. The concentration of DVB in the monomer mixture composition played an important role in determining not only the size of the microspheres but also the yield of the polymerization. In addition, although we employed various cosolvents as the polymerization medium, we found that acetonitrile/2-methoxyethanol was the only system that provided spherical particles without coagulation. This finding indicates that the precipitation polymerization is strongly dependent on the solvent used as the medium.

  • To increase the productivity of microbial chitosan from Rhizopus oligosporus ATCC22959, production medium and incubation conditions were optimized. The composition of the medium and the incubation conditions were like follows : starch+glucose(1:1) 3.0%, yeast extract 3.0%, KH2PO4 0.05%, MgSO4 0.01%, ZnSO4 0.002%, pH 5.0, incubation temperature 3$0^{\circ}C$, and incubation time 96hours. The productivity of chitosan of optimized medium was about 6.4 times higher than that of basal medium.

  • A total of 120 pigs (Berkshire) were used to investigate the effect of dietary supplementation with artificial culture medium ot wild ginseng (CMWG) on the fatty acid composition, amino acid composition and sensory characteristics or pork About $60{\pm}3kg$ pigs were randomly assigned to one of four experimental diet groups[(both sexes)${\times}$(C: commercial diet feed; T: commercial diet+1 L CMWG per day for 70 days)]. Pigs were slaughtered at approximately 110 kg live weight, and fatty acid composition, amino acid composition and sensory characteristics were measured in pork loin. The monounsaturated and saturated fatty acid contents was greater in barrow fed a diet containing CMWG than those of the gilt pork and control groups, however the polyunsaturated fatty acid composition decreased. The EAA (essential amino acid) content was lower in pigs fed diets containing CMWG than that of the control groups, whereas the SAA (amino acid with sulfide) and FRAA (fragrant amino acid) contents were higher. The EAA and FAA (amino acid in relation to flavor) contents were lower for harrow than for gilt, however the SAAA (amino acid in relation to saccarinity), SAA and FRAA the contents were higher. Regarding the sensory evaluation of fresh meat, the color of gilt pork increased with diets containing CMWG relative to the control group. Drip loss and the marbling score for pigs fed with diets containing CMWG were higher in barrow than in gilt. The flavor and overall acceptability of cooked meat from the control group was higher for barrow than for silt.

  • For the production of riboflavin, strain development of Ashbya gossypii NRRL Y-1056 was attempted by NTG(N-methyl-N'-nitro-N-nitrosoguanidne) treatment. The optimum composition of culture medium and other culture conditions for the production of riboflavin by selected mutant Ashbya gossypii JAG-13 were determined. The optimum composition of medium was 9% of corn oil, 3% of gellatone, 4% of CSL, 0.3% of glycine, 0.2% of S770. The optimum culture temperature and initial pH of medium was $28^{\circ}C$ and 6.5, respectively. oxygen was essential for the production of riboflavin, but excess oxygen inhibit the production of riboflavin. When Ashbya gossypii JAG-13 was cultured under above conditions for 12 days with a bioreactor, 6.9 mg/ml of riboflavin was produced.
