In Vitro Antagonistic Activity Evaluation of Lactic Acid Bacteria (LAB) Combined with Cellulase Enzyme Against Campylobacter jejuni Growth in Co-Culture

  • The antibacterial effects of nine lactic acid bacteria (LAB) against Campylobacter jejuni were investigated by using agar gel diffusion and co-culture assays. Some differences were recorded between the inhibition effects measured with these two methods. Only two LAB, Lb. pentosus CWBI B78 and E. faecium THT, exhibited a clear anti- Campylobacter activity in co-culture assay with dehydrated poultry excreta mixed with ground straw (DPE/GS) as the only growth substrate source. It was observed that the supplementation of such medium with a cellulase A complex (Beldem S.A.) enhanced the antimicrobial effect of both LAB strains. The co-culture medium acidification and the C. jejuni were positively correlated with the cellulase A concentration. The antibacterial effect was characterized by the lactic acid production from the homofermentative E. faecium THT and the lactic and acetic acids production from the heterofermentative Lb. pentosus CWBI B78. The antagonistic properties of LAB strains and enzyme combination could be used in strategies aiming at the reduction of Campylobacter prevalence in the poultry production chain and consequently the risk of human infection.

Isolation of Bacillus sp. SHL-3 from the Dry Soil and Evaluation of Plant Growth Promoting Ability

  • Excess use of chemical fertilizer causes soil acidification and accumulation of salt, and thus might bring to desertification of soil. To overcome this problem, it needs limited usage of chemical fertilizer and increased usage of natural fertilizer as an alternative. In this study, dry soil-borne Bacillus sp. SHL-3, which was isolated from arid and barren soil, with plant growth promoting activity was isolated for identification and to determine optimal culture condition. A bacterial strain SHL-3 had the IAA productivity ($5.16{\pm}0.10mg\;L^{-1}$), ACC deaminase activity ($0.36{\pm}0.09$ at 51 hours) and siderophore synthesis. It was identified as genus Bacillus sp.. Also, optimal culture condition of SHL-3 were $20^{\circ}C$ and pH 7 in LB medium. Bacillus sp. SHL-3 had up to 4% salt tolerance in the medium. We evaluated the plant growth promotion ability of SHL-3 using yam (Dioscorea japonica Thunb.). As a result, Bacillus sp. SHL-3 was effective on the increase of the shoot length (202.4% increase for 91 days). These results indicate that Bacillus sp. SHL-3 can serve as a promising microbial resource for the biofertilizers of soil.

  • Objective: Our recent findings confirmed the effectiveness of sodium metabisulfite (SMB) in controlling the growth of undesirable microorganisms in fruit and vegetable discards (FVD); however, lactic acid bacteria (LAB) are susceptible to its antibacterial effects. Two series of experiments were conducted to enable the survivability of LAB during silage fermentation of a total mixed ration (TMR) containing SMB-treated FVD. Methods: In Exp. 1, the objective was to isolate a strain of LAB tolerable to the toxic effect of SMB. In Exp. 2, the SMB load was minimized through its partial replacement with a chemical mixture (CM) based on sodium benzoate (57%), potassium sorbate (29%), and sodium nitrite (14%). FVD was treated with SMB + CM (2 g each/kg biomass) and added to the TMR at varying levels (0%, 10%, or 20%), with or without KU18 inoculation. Results: The KU18 was screened as a presumptive LAB strain showing superior tolerance to SMB in broth medium, and was identified at the molecular level using 16S rRNA gene sequence analysis as Lactobacillus plantarum. Inoculation of KU18 in TMR containing SMB was not successful for the LAB development, biomass acidification, and organoleptic properties of the resultant silage. In Exp. 2, based on the effectiveness and economic considerations, an equal proportion of SMB and CM (2 g each/kg FVD) was selected as the optimal loads for the subsequent silage fermentation experiment. Slight differences were determined in LAB development, biomass acidification, and sensorial characteristics among the experimental silages, suggesting the low toxicity of the preservatives on LAB growth. Conclusion: Although KU18 strain was not able to efficiently develop in silage mass containing SMB-treated FVD, the partial substitution of SMB load with the CM effectively alleviated the toxic effect of SMB and allowed LAB development during the fermentation of SMB + CM-treated FVD in TMR.

  • Effects of IAA on the elongation and cell wall hlysocidase activities were investigated in excised rape (Brassica napus L. cv. Yongdang) hypocotyl segments. IAA promoted the elongation of rape hypocotyl segments. In rape hypocotyls, the first 10-mm segments from the hook exhibited maximal elongation and the capacity of elongation was gradually decreased with increasing distance of each 10-mm from the hook. A good correlation has been obtained between the magnitude of endogenous growth and the activities of $\alpha$, $\beta$-glucosidase and $\alpha$, $\beta$-galactosidase. However, exogenous application of IAA did not seem to enhance the tissue with IAA resulted in acidification of the incubation medium. From these data, we can conclude that IAA seems to enhance elongation of the tissue segments, at least in part, by releasing hydrogen ion into cell wall, some of which may participate in the cell wall extension process, but does not seem to trigger the activation of $\alpha$, $\beta$-glucosidase and $\alpha$, $\beta$-galactosidase.

Ralstonia pseudosolanacearum 생존에 관여하는 Sigma S 역할 (Sigma S Involved in Bacterial Survival of Ralstonia pseudosolanacearum)

  • Ralstonia pseudosolanacearum은 토양과 물에서 오랫동안 생존하고, 가지과 작물에 심각한 풋마름병을 일으키는 식물병원세균이다. Simga S는 세균의 스트레스 환경에서 반응 또는 정지기 동안 유전자 발현을 조절하는 RNA 중합효소 복합체의 일부인 단백질이다. 본 연구는 스트레스 조건에서 R.pseudosolanacearum의 sigma S의 역할을 조사하기 위해서, R.pseudosolanacearum의 GMI1000 균주의 sigma S를 암호화하는 rpoS 유전자 변이체를 준비하여 야생형 균주와 세균의 특징을 비교하였다. 아울러 rpoS 유전자 역할은 원래 유전자를 변이체에 도입하여 rpoS 유전자 표현형 회복을 확인하였다. 야생형 균주와 rpoS 결여 변이체는 생장 속도, 외피다당류 생산, 식물체에서 병원성, 식물 세포벽 분해 효소 활성에서 차이를 보이지 않았다. 그러나 야생형 균주는 영양분결핍 조건에서 변이체보다 더 민감하게 반응하였고 과산화수소가 첨가된 조건에서 변이체보다 덜 민감하게 반응하였다. 흥미롭게도 영양분결핍 조건에서 rpoS 결여 변이체에서는 장기간 생균수를 유지하지만, 같은 조건에서 야생형 균주 생균수는 빠르게 감소하였다. 그리고 두 균주 배양액 pH를 측정한 결과, 야생형 균주와 변이체 간에 상당한 차이가 나타났다. 야생형 균주는 생장하면서 빠르게 배지의 pH가 감소하여 산성화되었다. 그러므로 야생형 균주의 빠른 사멸은 배지가 산성화되면서 정지기 상태 세균의 산성 pH에 대한 민감도 때문일 것이다. Biolog 분석으로 rpoS 변이체는 acetic acid, D-alanine, D-trehalose, L-histidine을 이용하지 못함을 확인하였다. 본 연구 결과는 R. pseudosolanacearum 세균의 sigma S가 영양분결핍 조건에서 정지기 동안 유기산 생산 또는 이용을 조절하며 정지기 세포사멸도 조절하는 것을 보여준다.

Isolation and Characterization of Plant Growth Promoting Rhizobacteria From Button Mushroom Compost

  • An auxin-producing bacteria (strain 5-1) was isolated from button mushroom compost in Boryeong-Si, Chungcheongnam-Do. The 5-1 strain was classified as a novel strain of Enterobacter aerogenes based on chemotaxonomic and phylogenetic analyses. The isolated E. aerogenes 5-1 was confirmed to produce indole-3-acetic acid (IAA), one of the auxin hormones, using TLC and HPLC analyses. When the concentration of IAA was assessed by performing HPLC quantitative analysis, a maximum concentration of IAA of $109.9mgL^{-1}$ was detected in the culture broth incubated in R2A medium containing 0.1% L-tryptophan for 24 h at $35^{\circ}C$. Acidification of the culture was deemed caused by an increase of IAA because a negative relationship between IAA production and pH was observed. Supplementation with a known precursor of IAA production, L-tryptophan, appeared to induce maximal production at 0.1% concentration, but it reduced production at concentrations above 0.2%. To investigate the growth-promoting effects to crops, the culture broth of E. aerogenes 5-1 was used to inoculate water cultures and seed pots of mung bean and lettuce. In consequence, adventitious root induction and root growth of mung bean and lettuce were two times higher than those of the control.

Microbial Amelioration of Acid Mine Drainage Impaired Soil using the Bacterial Consortia of Klebsiella sp. and Raoultella sp.

  • Acid mine drainage (AMD) resulting from pyrite oxidation in mining areas, subsequently leads to soil acidification accompanied by lowering pH and high concentration of metals and metalloids in its surrounding environment. Regarding to this, the microbial amelioration has been considered as a promising option for a more cost-effective and eco-friendlier countermeasure, compared to the use of alkaline chemicals. This study was aimed to evaluate influencing factors in microbially-mediated amelioration of acidic soil spiked by simulated AMD. For this, microcosm experiments were conducted by acid-neutralizing bacterial consortium (dominated by Klebsiella sp. and Raoultella sp.) under the various conditions of AMD spikes (0-2,500 mg SO42-/L), together with acidic mine soil (0-100 g) or sphagnum peat (0-5 g) in the 200 mL of nutrient medium. The employed bacterial consortium, capable of resisting to high level of sulfate concentration (up to 1,500 mg SO42-/L) in low pH, generated the ammonium while concomitantly reduced the sulfate, subsequently contributing to the effective soil stabilization with an evolution of soil pH up to neutral. Furthermore, it demonstrates that suitable condition has to be tuned for successful microbial metabolism to facilitate with neutralization during practical application.

Heavy Metal Resistant Phosphate Solubilizing Bacteria

  • Soil samples collected from abounded mines of Boryeong area in South Korea were used in isolating bacterial strains and their capacity to solubilize inorganic phosphates and heavy metal tolerance were assessed in vitro. Three different inorganic phosphate sources (Ca phosphate, Fe phosphate, and Al phosphate) and four different heavy metals (Co, Cd, Pb and Zn) each with three concentrations ($100{\mu}g\;mL^{-1}$, $200{\mu}g\;mL^{-1}$, and $400{\mu}g\;mL^{-1}$) were used. The bacterial isolates PSB-1, PSB-2, PSB-3, and PSB-4 solubilized significantly higher amount of Ca phosphate during the first five days of incubation though subsequent drop in soluble phosphorus level in the medium was observed at the later stage (after 5 days) of the incubation. Solubilization of Ca phosphate and Fe phosphate was concomitant with the acidification of the culture medium compared to the control where it remained constant. Isolated strains could solubilize Fe phosphate to certain extent ($25-45{\mu}g\;mL^{-1}$) though solubilization of Al phosphate was found negligible. All the isolates were tolerant to heavy metals (Cd, Pb, and Zn) up to the concentration of $400{\mu}g\;mL^{-1}$ except PSB-1 and PSB-8, which were shown to be vulnerable to Co even at $100{\mu}g\;mL^{-1}$. Heavy metal tolerant strains should be further evaluated for plant growth promoting activities also under field conditions in order to assess their agricultural and environmental significance.

Utilization of Multiple Carbon Sources by Plant Cells

  • ;It has been reported that suspension-cultured rice cells grown on mixed carbon sources of glucose (GIc) and acetate exhibited diauxic growth in which acetate was the preferred carbon source (Lee and Lee, 1996). Carrot (Daucus carota L.) suspension cells, showing a diauxic growth very similar to that of rice cells, were used to delineate the mechanisms underlying this preferential use of acetate over GIc. Uptakes of both GIc and 3-0-methylglucose (3-0MG), a non-metabolizable GIc analogue, were similarly inhibited when acetate or butylate, weak acids which are capable of transporting protons into the cytosol, were present in the uptake assay mixture containing cells harvested during the GIc-utilizing second growth phase. Inhibition of GIc uptake by these weak acids was similar when equivalent experiments were carried out with isolated plasma membranes. It was further shown that Glc uptake, which requires a proper proton gradient across the plasma membranes, was inhibited during the first growth phase by acetate-mediated alkalization of growth medium and/or simultaneous acidification of cytosol. This study strongly suggests that Glc utilization in plant cells is inhibited by co-presenting carbon source(s) which can alter the proton gradient across the plasma membrane. We further examined diauxic growth in culture containing GIc and malate. Unlike the case in the culture with GIc and acetate, carrot cells used GIc first. Malate was utilized only after Glc is depleted from medium. These results indicate that GIc can be a preferred or less-preferred carbon source depending on the competing carbon source. It was noted that malate was not directly taken up by cells. Instead it was converted extracellularly into fumarate which was subsequently transported into cells. During the malate-growth phase malate uptake was negligible, and fumarate uptake was active and pH-sensitive. It was shown that fumarase released into medium was responsible for the extracellular conversion of malate into fumarate. An immunoblot experiments showed that fumarase antibody raised against Arabidopsis fumarase provided positive signals only in medium in malate culture, not in fumarate or GIc cultures. This study demonstrates the first example in that fumarase, a mitochondria marker enzyme, can be present in places other than mitochondria.ndria.

Protease Inhibitors in Porcine Colostrum: Potency Assessment and Initial Characterization

  • Porcine colostrum and milk were separated into the acid-soluble and casein fractions by acidification followed by centrifuge. The acid-soluble fraction of porcine colostrum was further separated by liquid chromatography and anisotropic membrane filtration. Trypsin and chymotrypsin inhibitory capacity in porcine colostrum, milk and their components was determined by incubating bovine trypsin or chymotrypsin in a medium containing their corresponding substrates with or without addition of various amounts of porcine colostrum, porcine milk or their components. The inhibition of insulin-like growth factor I (IGF-I) and epidermal growth factor (EGF) degradation in pig small intestinal contents by porcine colostrum was measured by incubating iodinated IGF-I or EGF with the intestinal contents with or without addition of porcine colostrum. Degradation of labeled IGF-I or EGF was determined by monitoring the generation of radioactivity soluble in 30% trichloroacetic acid (TCA). The results showed that porcine colostrum had high levels of trypsin and chymotrypsin inhibitory activity and increased the stability of IGF-I and EGF in pig intestinal contents. The inhibitory activity declined rapidly during lactation. It was also found that trypsin and chymotrypsin inhibitory activity and the inhibition on IGF-I and EGF degradation in the acid-soluble fraction were higher than that in the casein fraction. Heat-resistance study indicated that trypsin inhibitors in porcine colostrum survived heat treatments of $100^{\circ}C$ water bath for up to 10 min, but exposure to boiling water bath for 30 min significantly decreased the inhibitory activity. Compared with the trypsin inhibitors, the chymotrypsin inhibitors were more heatsensitive. Separation of the acid-soluble fraction of porcine colostrum by liquid chromatography and anisotropic membrane filtration revealed that the trypsin and chymotrypsin inhibitory capacity was mainly due to a group of small proteins with molecular weight of 10,000-50,000. In conclusion, the present study confirmed the existence of high levels of protease inhibitors in porcine colostrum, and the inhibition of porcine colostrum on degradation of milk-borne growth factors in the pig small intestinal tract was demonstrated for the first time.
