  • Object : This study was designed to asses the effect of Kunnuetang and herbs on Mouse neuroblastoma 2a cells damaged by hypoxia-reoxygenation. Method : Mouse neuroblastoma 2a (N2a) cells were measured by MTT assay and LDH assay after 48h hypoxia and 6h reoxygenation. Mouse neuroblastoma 2a (N2a) cells were treated by Kunnuetang, Duchang and Daejo. Result : 1. Kunnuetang, was effective on LDH assay of hypoxia and reoxygenation. 2. Duchang and Daejo were generally effective on LDH assay of hypoxia and reoxygenation. 3. In MTT assay of hypoxia Kunnuetang was not effective. Duchung and Daejo were not generally effective on MTT assay, but in certain condition Herbs were effective. 4. In MTT assay of reoxygenation Kunnuetang and Daejo were not effective. But Duchung was effective in certain condition. Conclusion : The results suggest that Kunnuetang, Duchang and Daejo may have protective effect on vascular dementia and ad patient.

  • Recently, a traditional medicine called Gouteng-san, which consists of eleven herbs, was reported to be effective in treating vascular dementia with a double-blind, placebo-controlled study. Gout-eng-san is also used for patients with vascular dementia in combination with Si-Wu-Tang. The effect of Gouteng-san and Si-Wu-Tang on deficit of learning behavior was investigated using step-down passive avoidance task in mice. Hot-water extract of Gouteng-san (1.5 and 6 g/kg, p.o.) significantly prolonged the step-down latency shortened by scopolamine. The extract of Uncaria hook (150 mg/kg, p.o.), one of the component herb of Gouteng-san, significantly prevented the decrease in the latency after scopolamine. Hot-water extract of Si-Wu-Tang (1.5 and 6 g/kg of dried herbs, p.o.) prevented dose-dependently scopola-mine-induced disruption qf learning behavior. Si-Wu-Tang also prevented the ischemia-induced deficit of learning behavior. Both hot water extract of peony and angelica (1.5 g/kg, p.o.), which are component herbs qf Si-Wu-Tang, prevented the scopolamine-induced learning behavior deficit. Scopolamine (10 uM) suppressed long-term potentiation (LTP) of population spike in the CA1 region of the rat hippocampal slices. Peoniflorin (0.1~ 1uM) extracted from paeony root significantly ameliorated scopolamine-induced inhibition of LTR These results suggest that improvement of deficit of learning behavior by Gouteng-san and Si-Wu-Tang is mediated by direct and/or indirect activation of the cholinergic system in the brain.

  • Purpose: To screen candidate oriental herb material for antimelanogenics. Methods: Oriental herbs (n=100) were screened for mushroom tyrosinase inhibitory activity in vitro using the HM3KO human melanin cell line cultured in DMEM supplemented with 10% fetal bovine serum. Cytotoxicity was assessed by a cell viability assay involving 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), Trypan Blue exclusion, and cell enumeration. Results: Tyrosinase inhibitory effects on 100 oriental herbs was evident. Of these, 11 herbs inhibited tyrosinase activity by 40% without being cytotoxic to HM3KO cells. Three herb varieties significantly decreased melanin synthesis in HM3KO cells. Conclusions: Oriental herb can have antimelanogenic effects indicating their potential for functional therapeutic use in dermatological whitening.

The result of this stqdy are as follows: 1. The 17 medicatlons(63% of 27 medicine) of Sogal part decreased' the glucose level significantly.(Singihwan, Baekhotang, Jowiseunggitang, Yukamijihwangwon, Palmihwan, Kamijeonssibaekchultang, Insamsukgotang, Sacngjinyanghyultang, Insambokryungsan, Hwalhyulyunjosaengjineum, Chungsinbogitang, Hwanggitang, Hwangryunjihwangtang, Ojeupokchunhwan, Yongbongwon, Joosahwangryunwon, Kagambaekchulsan) 2. Dry herbs like Banha, Namsung are the contraindicated herbs in the Sogal part of Donguibogam but Ijintang and Dodamtang which include those herbs also decreased the blood glucose level significantly. 3. The three medications-Bangpungtongsungsan, Bohyulansintang, Bogantang from the six me dications which were used to treat Diabetes in the 6th I.M. of Kyung Hee Oriental Hospital also decreased the blood glucose level significantly.

  • This study was conducted to investigate the effect of medical herbs extracts on physicochemical characteristics of cooked rice. Three types (CLP 1, CLP 2, CLP 3) of extracts were prepared using 11 medicinal herbs, respectively. Polished rice was cooked with different concentration of medicinal herbs extracts by electric cooker. The moisture contents in cooked rice were decreased by increased concentration of herbs extracts, while total polyphenol contents were significantly increased. DPPH radical scavenging activities of cooked rice obviously were increased as herb extracts level increased. The consistency and setback value of the rice flour was reduced at all of the extracts, while breakdown and initial pasting temperature was increased. Color of cooked rice was shown different comparing to the control. However, the hardness, adhesiveness, springiness, cohesiveness and gumminess of cooked rice was increased depending on the concentration of extracts. These results showed that medicinal herbs extracts, which can benefit human health, is inducing the changes of physicochemical characteristics of cooked rice.
