S. mutans에 항균력(抗菌力)을 나타내는 균주(菌株)의 배양학적(培養學的) 성질(性質) (Nutritional and Cultural characterizations of microorganism capable of producing antagonistic activity against Streptococcus mutans)

  • 토양시료로부터 치아우식원인균인 S. mutans에 대하여 항균력을 나타내는 방선균주를 순수분리하여 분리균주를 공시균주로 하여 항생물질 생성에 미치는 환경인자를 규명하였다. 본 공시균주의 항생물질 생성 최적배지조성은 Bacto-soytone 1%, glucose 1%, NaCl 0.5%, $CaCO_3$ 0.1% 였으며 배지의 초기 pH는 7.0이었다. 또한 항생물질은 $28^{\circ}C$에서 진탕배양시 생성의 최적조건이었으며 본 공시균주는 24시간 유도기를 거쳐 배양 72시간째 항생물질의 생성과 더불어 균의 최대증식도를 나타내었다.

식품 섭취의 다양성에 따른 중소도시 노인의 영양 및 식행동, 건강상태에 관한 연구 (A Study on Nutrient Intake, Food Behavior and Health Conditions according to Food Intake Diversity in the Elderly in a Local City)

  • This survey was carried out to study the difference of nutrient intake, food behavior and health condition according to food intake diversity in the elderly(age 65 year over and 74 year under) in a local city. A diet survey with one day 24-recall method was used for 216 subjects(65 male & 151 female). Based on a food intake diversity examination, including an evaluation of KDDS(Korean's Dietary Diversity Score ; counts the number of food groups consumed daily from total five food groups(cereal, meat, vegetable, dairy, and oil groups)), Meal Balance(apply the KDDS at breakfast, lunch and dinner), and DVS(Dietary Variety Score ; total number of foods consumed), subjects were clustered according to food intake diversity. Food intake diversity patterns of male and female clusters were classified each into threes(Male-low group(24.6%), middle group(33.8%), and high group(41.5%)/Female low group(41.7%) middle group(37.1%), and high group(21.2%). The averages of KDDS, Meal Balance and DVS were 3.0, 7.5, 14.8 in the male middle group, and 4.5, 10.2, 25.7 in the male high group, 2.7, 6.3, 14.3 in the female low group, 4.0, 8.6, 18.0 in the female middle group, and 4.5, 10.5, 25.7 in the female high group respectively. The average dairy intake of nutrients below the RDA were energy, protein, Ca, Fe, Vitamin A, Vitamin B$_2$and niacin in the male low group, energy, protein, Ca, Fe, Vitamin A, Vitamin B$_2$in the male middle group and energy, Ca, Vitamin B$_2$in the male high group but a little. Also the female low group had intakes of all nutrients except Vitamin C, the middle group had intakes of energy, protein, Ca, Fe, Vitamin A, Vitamin B$_2$, and the female high group had intakes of energy, Ca, Vitamin A below the RDA respectively. Food intake diversity increasing, nutrient intake, food behavior and health condition proved to be good. This study explored the usefulness of cluster analysis in identifying food intake diversity of three groups of the elderly in relation to their nutrient intake, food behavior and health conditions.

느타리버섯의 액체종균 배양 조건과 생육 특성 (Culture conditions of liquid spawn and the growth characteristics of Pleurotus ostreatus)

  • 느타리버섯의 생산성 향상을 위하여 액체종균 배양액의 조건을 설정하고자 하였다. Potato dextrose 배양액에서 균체 생장을 위한 최적 pH는 6이었다. 대두박 0.3%와 설탕 및 포도당 함량을 달리한 15가지의 배양액('A'~'O')은 $22^{\circ}C$ 및 pH 6의 조건에서 14일동안 배양되었을 때 설탕과 포도당이 각각 함유된 배양액에서 흡광도는 증가되었다. 특히 5~20%의 포도당만 함유된 배양액('C', 'D', E' 및 'F')은 배양기간의 경과에 따라 흡광도가 증가되는 경향이었다. 배양액의 건조 균체량은 20%이하의 설탕이나 포도당이 단독으로 함유된 배양액에서 감소되었으나, 30%의 설탕('A') 또는 포도당 ('B')이 각각 함유된 배양액에서 건조 균체량은 증가되었다. 1100 mL의 톱밥배지 병재배 시 액체종균의 최적 접종량은 15~20 mL이었다. 고체종균 및 액체종균으로 느타리버섯을 재배하여 조직감을 비교한 결과 고체종균에 비해 액체종균으로 재배된 버섯에서 우수한 경향이었다.

고지탈묵용 Cellulase 및 Xylanase 생산 (Production of Cellulase and Xylanase for Enzymatic Deinking of Old Newspaper)

  • 효모탈묵용 cellulase와 xylanase 생산을 위한 조건을 조사한 결과 배지초기 pH, Tween 80, 포자접종량, 질소원의 종류 및 탄소원의 종류에 따라 효소 생산량이 크게 변하였다. Cellulase 생산을 위한 최적조건으로는 pH5.0-6.5, Tween 80 0.02, 포자현탄액($1{\times}10^{7}$/ml) 접종량 0.5-1.0, 질소원으로 면실박, 탄소원으로 옥분을 사용하였을 때 가장 효과적 이었다. 또한, xylanase 생산을 위한 최적조건으로는 pH6.5, Tween 80 0.01%, 질소원으로 corn steep liquor, 탄소원으로는 신문고지를 사용하였을 때 효과적이었으며, 이때 포자현탁액의 접종량은 cellulase 생산의 경우와 동일하였다.한편, cellulasedhk xylanase의 동시생산을 위해서는 플라스크 규모에서 질소원으로 면실박 0.5%, 탄소원으로 신문고지 1.0%와 옥분 2.0%를 첨가하였을 때 효과적이었으며, 이때 생상된 배양약의 cellulase의활성은 6.11~7.22IU/ml, xylanase의 활성은 97.7IU/ml이었다. 그러나 fermentor 규모에서는 동일 배지조성에서 효소생산이 다소 감소하였으며, 특히 xylanase 경우 배양액의 효소활성이 낮아서 이에 대한 보완이 요구된다.

  • E-Card Service for Child Nutrition Program is one of the main public policy services nowadays. In case of inconvenience during the use of the e-cards, it is recommended to cooperate with related organizations in order to promptly handle and provide guidance, and thoroughly manage child feeding service such as hygiene, nutrition and kindness etc. To do so, it is very important to provide food service that meets local actual conditions and children's needs in a cost effective manner for the underage who are worried about the poorly-fed by understanding the pattern of child feeding e-card service. Hence. this paper aims to investigate how child feeding e-card service efficiently provides meals according to the local situation and children's needs through big data analysis and to propose a method of identifying welfare conditions according to the purpose of service with actual application examples. The results suggest that, first of all, this study is able to judge appropriateness of public institution's policy in a timely and repetitive manner through non-standard data analysis such as Naver News and transaction data. Secondly, this paper proposes a multi-layered analysis framework, which performs online open data analysis to detect policy issues, visualizes retrieval and preprocessing of real data, and performs abnormal pattern recognition. These will be worthy of reference to other similar projects.

중온성 우모 분해균 Bacillus sp. SE-103의 분리 및 배양 조건 최적화 (Optimization of Conditions for Isolating and Cultivating Bacillus sp. Se-103 with a Mesophilic Feather-Degrading Activity)

  • 본 연구는 박테리아에 의한 우모분의 이용 가능성을 조사 하기 위해 우모 keratin 분해 활성이 높은 균주를 분리한 후 keratinolytic protease 활성이 가장 우수한 효소 생산을 위한 최적 배양 조건을 조사하였다. SE-103 균주로 형태학적 생화학적 특성을 조사한 결과Bacillus sp.로 확인되어 Bacillus sp. SE-103으로 명명하였다. 최적 배지 조성은 glucose 3.0%, urea 0.4%, $NaNO_3$ 0.2%, KCl 0.15%였으며, 배양 조건은 초기 pH 6.0, 배양 온도 $35^{\circ}C$, 24시간 진탕 배양하였을 때, 효소 생산이 가장 우수하게 나타났다.

보중익기탕으로 호전된 위 운동장애형 기능성 소화불량 환자 2례 (Two cases of the Gastric Dysmotility-like Dyspepsia in functional dyspepsia that were improved by Bojungiki-Tang)

  • Objectives : The aim of this study was to investigate an effect of Bojungiki-Tang in two cases of the Gastric dysmotility-like Dyspepsia in functional dyspepsia Methods : Subjects were 2 patients with Gastric dysmotility-like Dyspepsia. The patients had suffered from a dyspepsia that were not improved by medical treatments. We gave Bojungiki -Tang to the patients 30 minutes after each meal and tried acupuncture and moxibustion 2 times every day. And we had evaluated the condition of patients using a Ryodoraku test and ordinal scales. Results : After the comparison between an admission and discharge date at Ryodoraku test and ordinal scales, we knowed that conditions of two Gastric dysmotility-like Dyspepsia patients had improved. Conclusions : Bojungiki-Tang is effective on the improvement of conditions in the Gastric dysmotility-like Dyspepsia patients.

Production of Blastospore of Entomopathogenic Beauveria bassiana in a Submerged Batch Culture

  • The principal objective of this study was to determine the optimal liquid culture conditions in shake flasks for maximal sporulation of Beauveria bassiana. The optimal initial pH for the spore production of B. bassiana using Potato Dextrose Broth was 5.2. The screening in shake flasks of carbon and nitrogen sources resulted in the identification of an optimal medium based on 3% sucrose and 1% casamino acid, with a C : N ratio of 22 : 4. Using this medium, a production level of $5.65{\times}10^7$ spores per ml was obtained after 5 days of culture. Using 3% corn meal, 2% corn steep powder, and 2% rice bran, the maximum spore concentration of $8.54{\times}10^8$/ml was achieved 8 days after inoculation at $25^{\circ}C$ in a rotary shaking incubator operated at 200 rpm. This represents a yield gain of approximately 2.89 times that of pre-optimization.

Optimization of Indole-3-acetic Acid (IAA) Production by Bacillus megaterium BM5

  • One of the important phytohormones produced by plant growth promoting bacteria is the auxin; indole-3-acetic acid (IAA), with L-tryptophan as the precursor. In this study, we focused on the investigation of optimal conditions for the production of IAA by Bacillus megaterium BM5. We investigated culturing conditions, such as incubation temperature, pH of the culture medium and incubation period, with varying media components such as inoculation volume, tryptophan concentration and carbon and nitrogen source. Besides, optimization study intended for high IAA production was carried out with fermentation parameters such as rpm and aeration. The initial yield of $42{\mu}g\;IAA\;ml^{-1}$ after 24 hr increased to $85{\mu}g\;ml^{-1}$ when 5% (v/v) of L-tryptophan was used in the culture broth. The maximum yield of $320{\mu}g\;IAA\;ml^{-1}$ was observed in trypticase soy broth (TSB) supplemented with starch and soybean meal as C and N sources with a C/N ratio of 3:1 (v/v) at $30^{\circ}C$, pH 8.0 for 48 hrs with 1.0 vvm and 250 rpm in 5 L working volume using 10 L scale fermenter. The bacterial auxin extracted from the culture broth was confirmed by thin layer chromatography and high-performance liquid chromatography and effect on plant growth was confirmed by root elongation test.

Enhanced Antibiotic Production by Streptomyces sindenensis Using Artificial Neural Networks Coupled with Genetic Algorithm and Nelder-Mead Downhill Simplex

  • Antibiotic production with Streptomyces sindenensis MTCC 8122 was optimized under submerged fermentation conditions by artificial neural network (ANN) coupled with genetic algorithm (GA) and Nelder-Mead downhill simplex (NMDS). Feed forward back-propagation ANN was trained to establish the mathematical relationship among the medium components and length of incubation period for achieving maximum antibiotic yield. The optimization strategy involved growing the culture with varying concentrations of various medium components for different incubation periods. Under non-optimized condition, antibiotic production was found to be $95{\mu}g/ml$, which nearly doubled ($176{\mu}g/ml$) with the ANN-GA optimization. ANN-NMDS optimization was found to be more efficacious, and maximum antibiotic production ($197{\mu}g/ml$) was obtained by cultivating the cells with (g/l) fructose 2.7602, $MgSO_4$ 1.2369, $(NH_4)_2PO_4$ 0.2742, DL-threonine 3.069%, and soyabean meal 1.952%, for 9.8531 days of incubation, which was roughly 12% higher than the yield obtained by ANN coupled with GA under the same conditions.
