In vitro Examination of Chondroitin Sulfates Extracted Midduck (Styela clava) and Munggae Tunics (Halocynthia roretzi) as a Cosmetic Material (In vitro에서 미색류 껍질로부터 추출한 콘드로이틴황산의 기능성 화장품 소재로서의 가능성)

  • With the aim of using a cosmetic material, chondroitin sulfates extracted from midduck tunics (Styela clava) and munggae tunics (Halocynthia roretzi) were examined in vitro with two cell lines for cell toxicity, collagen synthesis, cell growth and recovery ability after U.V. irradiation. Cell toxicity test with A 431 and CCD 1108Sk was able to observe high activity between 400 and 600 $\mu\textrm{g}$/m while standard chondroitin sulfate (CS) purchased from Sigma was showed at 80 $\mu\textrm{g}$/mL. Even fraction 1 and 2 collected from chondroitin sulfates originated from midduck appeared having the highest activity between 600 and 1000 $\mu\textrm{g}$/mL, but slightly lower compared to crude chondroitin sulfates from both mideduck and munggae. In cell growth examination, it was not able to find significant differences between chondroitin sulfates used. Both crude chondroitin sulfates were exhibited the highest activity for two cell lines except that of mideduck which was showed activity for CCD 1108Sk. CS, fraction 1 and 2 from midduck were not able to demonstrate a significant activity in collagen synthesis. On the contrary, crude chondroitin sulfates from both munggae and midduck were showed the highest activity at 100 and 50 $\mu\textrm{g}$/mL with only CCD 1108Sk. The recovery ability after U.V. irradiation with crude chondroitin sulfates from both munggae and midduck were showed high activity at 400 $\mu\textrm{g}$/mL with CCD 1108Sk and A 431. But there were no activity observed in fractions examined, As a consequence, the crude chondroitin sulfates from both munggae and midduck might not only be available as a cosmetic material but also useful for increasing some activity by blending properly.

GIS-based Analysis of Debris-flow Characteristics in Gangwon-do (GIS를 이용한 강원지역 토석류 특성분석)

  • In Korea, there are debris-flow disasters induced by typhoon and localized torrential rainfall annually. There are particularly extensive debris-flow disasters in Gangwon-do because of its geomorphological characteristics; the extensive coverage of mountainous region, steep slope, and shallow soil. In this paper, we constructed a GIS database about topological characteristics of debris-flow basin in Gangwon-do by years of field survey. Also, we conducted frequency analysis based on this database with the digital forest type map and the digital soil map. We analyzed frequencies of debris-flow by simple count for topological characteristics, whereas we analyzed by considering an area ratio based on GIS for physiognomic and geologic characteristics. We used slope, aspect, width, depth and destruction shapes for analysis about topological characteristics of debris-flow basin. Also we used attributes of forest physiognomy, diameter, age, and density about physiognomic characteristics, and i n terms of geologic characteristics, we used attributes of drainage class, effective soil depth, subsoil properties, subsoil grave content, erosion class, parent material of soil, and topsoil properties. In consequence, we figured out topographic, forest physiognomic, and geologic characteristics of debris-flow basin. This result is applicable to establish a rational disaster prevention policy as a fundamental information.

A Study on the Performance of Recycled Asphalt Pavement using Hot Recycling Plant (재생 아스팔트 포장의 공용성능 연구)

  • The recycled asphalt concrete has gotten increasing attention due to the environmental issues. The volume of reclaimed asphalt has increased significantly for last few years because of city remodeling, pavement maintenance, utility excavation, and road widening. Considering the value of reclaimed asphalt, it is rather used for the recycled asphalt concrete than it is used for fill and cover up material instead of soil. This research will be a supplements incomplete issues from existing research results and suggests the quality control guideline for recycled asphalt concrete and upcoming laws. As the first step of research, the trial construction of RAP(Recycled Asphalt Pavement) performed in expressway construction sites. These trial construction sites have been checked every years. And another construction sites studied and selected for more deeper performance check of RAP. For this checks, we used automatic pavement survey equipment and computerized analysis tools. Also, DSR(Dynamic Shear Rheometer) was used for the fatigue life calculation of binder blends(RAP and virgin binder). As a consequence of this research, the application of recycled asphalt provides good enough quality for highway construction. The preceeding literatures reviewed shows that the asphalt rejuvenator are used in many countries but that type of chemical agent are not used in Korea. By using the data of trial construction and mix design in Chongwon-Sangju construction lane, the surface and base courses consisted with the 10% and 30% rap mix asphalt section maintains good performance for up to 7 years. Through the performance check and laboratory tests(DSR), the quality control and mixture's low temperature prevention are the important factor and chemical agent necessary for increasing the fatigue life of RAP binder.

  • Recent years have witnessed the increased usage of flammable metals, such as aluminum or magnesium, in wide range of high-tech industries. These metals are indispensable for the improvement of physical properties of materials as well as the design capability of the final product. During the process, unwanted metal dusts could be released to the environment. This can lead to an occupational health and safety issues. Due to their flammable nature, more serious problem of an explosion can happen in extreme cases. The explosion is the combustion of tiny solid particles and vapor mixture, caused by pyrolysis. This complex composition makes engineering analysis more difficult, compared to simple gas explosions or vapor cloud combustions. The study was conducted to assess this light metal dust explosion in an effort to provide the bases for a risk assessment. Dust explosion characteristics of each material was carefully evaluated and an appropriate analysis tool was developed. A comprehensive database was also constructed and utilized for the calibration of the developed response model and the verification for its accuracy. Subsequently, guidelines were provided to prevent dust explosions that could occur in top-notch industrial processes.

Preparation and Characterization of Iron Phthalocyanine Thin Films by Vacuum Sublimation (진공증착법을 이용한 철프탈로시아닌 박막의 합성과 그 특성)

  • In this experiment the Iron phthalocyanine (FePc) films on Si-wafer and alumina pallet were prepared using vacuum sublimation with conditions of changing reaction time, temperature, and deposition rate. Then, some samples were annealed following annealing. Techniques such as XRD, SEM, and resistance measurement method, were dedicated to characterize the changes of surface structure, phase transformation and electric resistance sensitivity in accordance with change of film thickness. In proportion to the decrease of deposition temperature from $370^{\circ}C$ to $350^{\circ}C$, intensities of (200), (011), (211) and (114) planes of $\alpha$-phase were decreased and (100) plane of $\beta$-phase were appeared. The film thickness were controlled by regulating the volume of precursor material during rapid deposition. As a result, it was observed that crystalline particle size had been increased according to the increase of film thickness and $\alpha$-phase transformed to $\beta$-phase. In consequence of measuring the crystallinity of films annealed between $150^{\circ}C$ and $350^{\circ}C$, $\alpha$- to $\beta$-phase transformation was appeared to begin at $150^{\circ}C$ and completely transformed to $\beta$-phase at $350^{\circ}C$. Electric resistance sensitivity of FePc film to $NO_x$ gas along temperature change of FePc films was observed to be more stable with the decrease of the film thickness.

Augmented Plasticity: Giving Morphological Editability to Physical Objects (증강가소성: 물리적 오브젝트에 형태적 편집가능성 부여하기)

  • Product designers sketch various ideas of foreground figures(detail design) onto background figures(basic form) and evaluate numerous combinations of them in the late stages of design process. Designers have to test their ideas elaborately with a high-fidelity physical model that looks like a real product. However, due to the requirements of time and expense in making high-fidelity design models, it is impossible to evaluate such a number of combinatorial solutions of background and foreground figures. Contrary to digital models, physical design models are not easily modifiable and so designers cannot easily develope ideas through iterative design-evaluation process. To address these problems, we proposed a new concept 'Augmented Plasticity' that gives morphological editability to a rigid physical object using Augmented Reality technology and implemented the idea as Digital Skin system. Digital Skin system figures out the position and orientation of object surface with ARToolKit visual marker and superimposes a deformed surface image seamlessly using differential rendering method. We tried to apply Digital Skin system to detail design, redesign of product, and material exploration task. In consequence, it was found that Digital Skin system has potential to allow designers to implement and test their ideas very efficiently in the late stages of design process.

Determination and Generation Factor of Acrylamide Content in the French Fries and Snacks by HPLC/MS/MS (HPLC/MS/MS를 이용한 French Fries와 스낵류에서의 Acrylamide 함량 분석 및 생성요인)

  • The formed acrylamide in many fried and baked starchy foods is in consequence of its formation during frying and preparation methods. Being acrylamide established to be a toxic substance, the implications to public health from amounts found in food are not clear. So this study was carried out to investigate the contents of acrylamide of French fries and snacks in addition to acrylamide formation in relation to water and lipid in foods. The raw materials of foods used in the experiments were wheat, potato, com and rice. The preparation of foods was modified in a little to raise the recovery ratio and contents of water and lipid were analyzed to the public food method. The contents of acrylamide were measured by the liquid chromatography tandem mass spectrometry. The average level of acrylamide produced in snacks is $236{\pm}322$ ppb and potato snacks came out the highest value at $521{\pm}403$ ppb in 104 snacks. The formation of acrylamide in fried food was found to depends on the composition of raw material not on water and lipid contents in food and not on storage period and temperature.

Material Characteristics and Clay Source Interpretation of Crucibles in Baekje Kingdom Excavated from the Ssangbukri Site in Buyeo, Korea (부여 쌍북리 유적 출토 백제 도가니의 재료학적 특성과 원료의 산지해석)

  • The crucibles of Baekje Kingdom from the Ssangbukri Site which were used for glass and metal melting had light brown, grayish blue and grayish brown colored bodies. In thin section, the crucibles contained numerous quartz grains and pottery fragments. The surface was covered with fine grained quartz for thermal resistance. Based on decomposition of mica group minerals and formation of mullite detected by X-ray diffraction analysis, it was inferred that all crucibles have been fired over $1,000^{\circ}C$. It was also found that firing temperature has exceeded $1,100^{\circ}C$ in some crucibles because feldspar was not detected. The maximum temperature was assumed at $1,200^{\circ}C$. The magnetic susceptibility values and geochemical characteristics sorted out the crucibles into two groups that differed from the characteristics of the local soils. This reflected geological setting of the site where the alluvium was formed from two kinds of surrounding rock masses, granite gneiss and biotite granite. However, the local soils had similarities with the crucibles in weathering degree and geochemical behavior of major elements. In consequence, it was considered that the raw clay of the crucibles was supplied from the local area of the site.

A Study on the Contact Resistance according to the Tightening and Overlapping area of Bus Bar (부스 바의 체결 및 중첩 구간에 따른 접촉 저항에 관한 연구)

  • The bus bar is an electrical connection widely used for the power supply of skyscrapers and power distribution and industrial equipment electrical panels in industrial plants. There are various materials to be considered in the design of the bus bar, such as material based on the use environment, the sectional area according to the power capacity, the length of the surface circumference, and the tightening method. Even with a bus bar manufactured to a size of sufficient power capacity in the design, if the actual tightening is incorrect, it may lead to fire due to deterioration. For these reasons, a variety of research on the temperature rise of the electrical contact point has continued. However, the temperature rise of the contacts is a consequence of the result, not a direct cause. In this paper, the influence of contact resistance on the fastening force and the overlapping section of the bus bar are investigated by measuring the change in resistance from building the specimen. A total of eight bus bar specimens were manufactured and measured. Resistance was measured by varying the clamping force and the interval between overlapping sections when the specimens were crossed. We propose a safe power connection model by analyzing the contribution of these factors to the actual contact resistance change.

Screening the level of cyanogenic glucosides (dhurrin) in sorghum accessions using HPLC analysis

  • Sorghum (Sorghum bicolor (L.) Moench.) is one of the most important crops for human and animal nutrition. Nonetheless, sorghum has a cyanogenic glucoside compound which can be degraded into hydrogen cyanide, toxic to humans and animals even with tiny amount. In consequence, breeding materials with a low cyanide level has been a top priority in sorghum breeding programs. To fulfill our long-term goal, we are screening sorghum accessions with low cyanide level, which would be an important breeding material for food safety. We collected seeds of various sorghum accessions and analyzed relevant metabolites to find useful breeding materials of sorghum accessions containing low cyanide. Fourteen wild relatives were obtained from the University of Georgia in US, a reference accession BTx623, and three local varieties from National Agrobiodiversity Center of Rural Development Administration in Korea, and one wild species from the Wild Plant Resources Seed Bank of Korea University in Korea. Sorghum plants were grown in plastic greenhouse under natural conditions. After growing, leaf samples were harvested at different developmental stages: seedling phase, vegetative phase (right before flowering), and reproductive phase (ripening). Using collected samples, quantification analysis were performed by an HPLC system for three metabolites (dhurrin, 4-hydroxybenzaldehyde, and 4-hydroxyphenylacetic acid) in sorghum plants. Prior to metabolome analysis, specific experimental condition for HPLC system was set to be able to separate three metabolites simultaneously. Under this condition, these metabolites were quantified in each accession by HPLC system. We observed that the metabolite contents were changed differently by developmental stages and accessions. We clustered these results into five groups as patterns of their contents by developmental stages. Most of accessions showed that 4-hydroxybenzaldehyde content was very high at seedling stage and decreased rapidly at vegetative phase. Interestingly, the patterns of dhurrin content were very different among clusters. However, 4-hydroxyphenylacetic acid content was maintained at low levels by developmental stages in most accessions. The results would demonstrate how dhurrin and alternative degradation pathways are differentiated in each accession.
