양식 넙치에서 분리된 스쿠티카섬모충 Philasterides dicentrarchi의 생물학적 특성 (Biological Characteristics of Scuticociliate, Philasterides dicentrarchi Isolated from Cultured Olive Flounder, Paralichthys olivaceus)

  • 양식넙치에서 분리된 스쿠티카섬모충 P. dicentrarchi에 대한 생물학적 특성을 규명하기 위해 배양, 생육환경, 화합물의 in vitro 구제실험 등을 실시하였다. P. dicentrarchi 배양실험 결과 세포, 세균, 넙치치어 배합사료, 뇌 조직과 로티퍼 조직 등의 유기물을 먹이로 활발히 증식하였다. 생육환경 조건은 염분농도 $5{\sim}45\;ppt,\;pH\;6{\sim}9$ 범위에서 증식이 이루어졌으며, 온도는 $10{\sim}30^{\circ}C$ 범위에서 생존 및 증식이 가능하나 $10{\sim}25^{\circ}C$ 범위에서 활발한 증식이 이루어졌다. P. dicentrarchi의 화합물 구제실험 결과 포르말린 300, 400 ppm에 30분, 200 ppm에 60분, 100 ppm에 90분, 50 ppm에서 120분 만에 구제효과를 보였다. 과산화수소 약욕에 따른 구제효과는 300 ppm에서 60분, 200 ppm에서 90분 만에 구제효과를 보였으며, 150 ppm 처리구와 100 ppm 처리구에서는 150분 만에 각각 운동성을 완전히 상실하여 구제효과를 보였다.

남해지역 자연산 해산어의 Trichodina 감염 현황 (Survey of Trichodina infection in wild populations of marine fish caught from Namhae region, southen coast of Korea)

  • The ectoparasite ciliate Trichodina has been recorded from above 100 species of freshwater and marine fishes. In April 2009, we investigated the trichodia infection in 13 species of marine fishes captured by emplacement net and gill net in order to understand trichodina infection status among the natural fish populations along the Namhae-Gun coast area, the southen coast of Korea. Trichodina infection was observed in 10 out of 10 sea basses Lateolabrax japonicus, 2 out of 5 finespotted flounder Pleuronichthys cornutus and 1 out of a stone flounder Kareius bicoloratus. The sea bass infected with the parasite showed hypertrophy and exfoliation in gill epithelium. In addition, hyperplasia and lamellar fusion of gill filament were noticed among naturally affected sea bass.

Growth and ingestion rates of heterotrophic dinoflagellates and a ciliate on the mixotrophic dinoflagellate Biecheleria cincta

  • To explore the interactions between the mixotrophic dinoflagellate Biecheleria cincta (previously Woloszynskia cincta) and heterotrophic protists, we investigated whether the common heterotrophic dinoflagellates Gyrodinium dominans, Gyrodinium moestrupii, Gyrodinium spirale, Oxyrrhis marina, and Polykrikos kofoidii, and the ciliate Strobilidium sp. were able to feed on B. cincta. We also measured growth and ingestion rates of O. marina and Strobilidium sp. on B. cincta as a function of prey concentration. In addition, these rates were measured for other predators at single prey concentrations at which the growth and ingestion rates of O. marina and Strobilidium sp. were saturated. All grazers tested in the present study were able to feed on B. cincta. B. cincta clearly supported positive growth of O. marina, G. dominans, and Strobilidium sp., but it did not support that of G. moestrupii, G. spirale, and P. kofoidii. The maximum growth rates of Strobilidium sp. and O. marina on B. cincta (0.91 and 0.49 $d^{-1}$, respectively) were much higher than that of G. dominans (0.07 $d^{-1}$). With increasing the mean prey concentration, the specific growth rates of O. marina and Strobilidium sp. on B. cincta increased, but either became saturated or slowly increased. The maximum ingestion rate of Strobilidium sp. (1.60 ng C $predator^{-1}\;d^{-1}$) was much higher than that of P. kofoidii and O. marina (0.55 and 0.34 ng C $predator^{-1}\;d^{-1}$) on B. cincta. The results of the present study suggest that O. marina and Strobilidium sp. are effective protistan grazers of B. cincta.

진해만에서 부유생물을 이용한 해양생태계 건강성평가 예비조사 (A Prelimiary Study for Marine Ecosystem Health Assessment Using the Planktonic Organism in Jinhae Bay)

  • 하계 진해만 16개의 정점에서 해양생태계의 오염 정도를 평가하기 위해서 플랑크톤 건강 지수를 살펴보았다. 해양생태계 건강지수 산출은 부유성 플랑크톤 생물 엔테로박테리아 Escherichia coli, HB, ANF, HNF, HABs 그리고 섬모충류 등과 더불어 무기와 유기 오염원인 DOC, COD, 그리고 Chl.a 등을 각각 조사하였다. 그 중 플랑크톤 지수 산출은 오염압력의 반응에 민감한 Chl.a, HABs, HB, E. coli 등 4개의 지표를 시범적 선정하였다. 또한 점수산정기준은 과거자료를 바탕으로 통계학적 기법으로 해석하였으며, 그들 건강지수 등급도 4개로 나누었다. 그 결과 진해만 해역은 전반적으로 "좋음"의 B등급을 받았다. 본 연구에서 산출한 플랑크톤 건강지수는 진해만의 해양생태계 건강성 평가를 하기 위한 좋은 방법 중 하나로 고려하여도 될 것이다.

한국의 4개 주요항만(부산, 울산, 광양, 인천)에 분포하는 섬모충 플랑크톤의 계절동태 특성 (Characteristic of Seasonal Dynamics of Planktonic Ciliates at Four Major Ports (Busan, Ulsan, Gwangyang and Incheon), Korea)

  • 2007년 2월부터 2008년 11월까지 국내의 4개 주요 항만(부산항, 울산항, 인천항, 광양항)에 분포하는 유종섬모충류의 계절적 출현양상과 선박평형수 벡터종(Coos Bay 벡터종 기준)의 계절적 출현양상을 조사하였다. 조사 기간 중 부산항과 울산항에서 유종섬모충류 난류종인 Ascampbeliella urceolata, Codonellopsis morchella, C. ostenfeldi, Dadayiella ganymedes, Eutintinnus stramentus, Protorhabdonella curta가 8월과 11월에 출현하였다. 인천항과 광양항에서는 전 계절을 통해 연안종이 출현하였고 특히 저수온기와 고수온기에 연안종의 출현이 뚜렷하였다. 광양항에서는 2007년 5월에 난류종 Tintinnopsis ampla의 출현과 2008년 11월에 C. ostenfeldi가 출현을 보였지만 인천항에서는 난류종의 출현이 관찰되지 않았다. 유종섬모충 출현분포의 특성은 연안종이 주로 출현하였으며, 부산항과 울산항에서 8월과 11월에 난류종이 중점적으로 출현하고 있는 것은 쿠로시오 해류(Kuroshio current)의 지류인 대마난류(Tsushima Warm Current)의 영향에 의한 유입되는 것으로 판단된다. 또한 조사 기간 중 주요 항만에서 출현한 유종섬모충류의 벡터종은 총 13종이었다. 광양항에서 13종으로 가장 많은 종이 출현하였고 인천항에서 6종으로 가장 적은 종수가 관찰되었으며 부산항과 울산항에서 각각 10종과 11종이 관찰되었다. 이들 벡터종은 부산항, 울산항, 광양항의 고수온기에 가장 많은 출현종수를 보인 반면, 인천항에서는 저수온기 2월에 가장 많은 출현종수를 보여 항만에 따른 차이를 보였다. 그러나 우리나라에 해역에 영향을 주는 서로 다른 해류에 의한 유입종 연구와 선박 평형수에 의해 유입된 벡터종의 연구가 매우 부족하기 때문에 주요 항만의 유종섬모충류의 종조성 차이에 대한 정확한 결론을 내리기에는 어려움이 있어 앞으로 이에 대한 체계적인 연구가 필요하다고 판단된다.

First Record of Amphisiella milnei (Ciliophora, Stichotrichida) from Korea

  • A marine ciliate Amphisiella milnei (Kahl, 1932) $Horv{\acute{a}}th$, 1950 was discovered from the tidal pool of Baekdo Island, South Korea. The existence of extra cirri between leftmost frontal cirrus and buccal cirrus discriminates this species from its congeners. Its morphological features are described as follows: body size in vivo $110-130{\times}35-45{\mu}m$; elongate rectangular to elliptical in shape; two large and several small ring-shaped structures; yellowish cortical granules arranged irregularly on ventral side but longitudinally along dorsal kineties on dorsal side; 34-40 adoral membranelles, 3 frontal cirri, 1 buccal cirrus, 1 parabuccal cirrus, usually 2 extra cirri behind leftmost frontal cirrus, and 3 frontoventral cirri; amphisiellid median cirral row composed of 25-31 cirri with 27-36 left and 27-44 right marginal cirri; usually 5 transverse cirri and 2 pretransverse cirri with 7 dorsal kineties; two macronuclear nodules. In addition to, 18S rDNA sequence of A. milnei was analyzed to understand its phylogenetic relationship.

Five Alexandrium species lacking mixotrophic ability

  • Mixotrophy in marine organisms is an important aspect of ecology and evolution. The discovery of mixotrophic abilities in phototrophic dinoflagellates alters our understanding of the dynamics of red tides. In the phototrophic dinoflagellate genus Alexandrium, some species are mixotrophic, but others are exclusively autotrophic. There are differences in the ecological roles of autotrophic and mixotrophic Alexandrium in marine food webs. However, of the 34 known Alexandrium species, the mixotrophic ability of >20 species has yet to be explored. In this study, the mixotrophic capabilities of Alexandrium insuetum CCMP2082, Alexandrium mediterraneum CCMP3433, Alexandrium pacificum CCMP3434, Alexandrium tamutum ATSH1609, and Alexandrium margalefii CAWD10 were investigated by providing each species with 22 diverse prey items including bacterium-sized microbeads (1 ㎛), the cyanobacterium Synechococcus sp., algal prey species, and the ciliate Mesodinium rubrum. None of the 5 Alexandrium species fed on any of the prey items. These results increase the number of Alexandrium species lacking mixotrophic abilities to 9, compared to the 7 known mixotrophic Alexandrium species. Furthermore, the Alexandrium phylogenetic tree based on the large subunit ribosomal DNA contained 3 large clades, each of which had species with and without mixotrophic abilities. Thus, the acquisition or loss of mixotrophic abilities in Alexandrium might readily occur.

Morphological Redescriptions of Three Condylostoma Ciliates (Heterotrichida: Condylostomatidae) New to Korea

  • Heterotrichous ciliates were isolated from marine and brackish habitats in Korea, and their morphology, infraciliature and morphometrical characters were investigated using silver impregnated methods. These three Condylostoma species were identified as Condylostoma curva Burkovsky, 1970, C. minutum Bullington, 1940 and C. spatiosum Ozaki and Yagiu in Yagiu, 1944. The species of Condylostoma based on morphology are highly variable in most of characters and distinguished as following. Condylostoma curva is oval to elongate ellipsoidal, size about $245{\times}100{\mu}m$ in vivo, and conspicuously dark greenish brown with cortical granules on their surface, buccal cavity relatively large about 40% of body length, 5-11 macronuclear nodules, 4-8 frontal cirri serially arranged and 22-38 somatic kineties. Condylostoma minutum is elongate ellipsoidal and the size about $310{\times}55{\mu}m$ in vivo, buccal field about 35% of body length, 12-20 macronuclear nodules, adoral zone consisted of 82-107 membranelles, large and long one frontal cirrus and 38-44 somatic kineties. Condylostoma spatiosum is large elongate ellipsoidal, size about $670{\times}105{\mu}m$ in vivo, buccal cavity about 25% of body length, 11-25 macronuclear nodules, 111-144 adoral membranelles, membrane-like 2 frontal cirri, 49-74 somatic kineties. These three Condylostoma species are described here for the first time in Korea.

First Record of Two Euplotes Ciliates (Ciliophora: Spirotrichea: Euplotida) from Korea

  • Two marine euplotid ciliates, i.e. Euplotes cristatus Kahl, 1932 and E. minuta Yocom, 1930, were collected from the public waterfront of Incheon on the Yellow Sea and from the Songjeong Beach, Busan, in the Strait of Korea, respectively. These two species were verified as unrecorded species in Korea. These species were described based on live observation, protargol impregnation, and silver nitrate impregnation. In addition, the small subunit ribosomal DNA (SSU rDNA) sequences of the two species were compared with previously known sequences of the Euplotes species. Euplotes cristatus has an elongated oval form, size in vivo of $60-84{\times}38-68\;{\mu}m$, 35-50 adoral zone of membranelles (AZM), 10 frontoventral cirri (FVC), 5 transverse cirri (TC), 4-5 caudal cirri (CC), 8 dorsal kineties (DK), 10-16 dorsal cilia of middle DK, and silverline system of single-vannus type. Euplotes minuta has a small ovoid form ($44-53{\times}26-35\;{\mu}m$ in vivo), 31-41 AZM, 10 FVC, 5 TC, 4 CC, 9 DK, 10-12 dorsal cilia of middle DK, and silverline system of single-vannus type.

New Record of Two Apokeronopsis Species (Ciliophora: Urostylida: Pseudokeronopsidae) from Korea

  • The morphology of the two marine hypotrichous ciliates Apokeronopsis bergeri and A. ovalis, isolated from the Yellow Sea, Korea, are described based on live and protargol-impregnated specimens. It is the first time that these species have been recorded in Korea. In addition, the small subunit ribosomal RNA gene was sequenced for comparison with the public database. The genus Apokeronopsis has recently been established in the family Pseudokeronopsidae, and the two congeners of the Korean population share the following characteristics: one row of one or more buccal cirri; usually two frontoterminal cirri; midventral complex composed of two distinctly separated rows; one left and one right marginal row; number of transverse cirri, more than eight; absence of caudal cirri; two types of cortical granules. Apokeronopsis bergeri differs from A. ovalis primarily in body shape (fusiform vs. oval form), size (usually $260{\times}80{\mu}m$ vs. $160{\times}55{\mu}m$), type II cortical granules (oval vs. round shape; yellow-green vs. mostly colourless and only a few yellow-green in colour), and morphometric data (75-106 vs. 53-70 in adoral membranelles; 37-47 vs. 24-36 in frontal cirri; 9-15 vs. 1-2 in buccal cirri), as well as molecular data (2.87% of pairwise distance).
