Production and Characteristics of Environment-Friendly Antimicrobial Substance by Pseudomonas aeruginosa EL-KM (Pseudomonas aeruginosa EL-KM에 의한 환경친화적 항균물질의 생산과 특성)

  • An antimicrobial substance-producing microorganism was isolated from soil samples. Based of the taxonomic characteristics of its morphological, cultural, physiological properties and 16s rRNA sequence alignment, this microorganism was identified as Pseudomonas aeruginosa, and we named Pseudomonas aeruginosa EL-KM. The optimal culture condition for production of antimicrobial substance was 1% mannitol, 0.4% yeast extract, 0.5% Nacl, 0.2% $K_2SO_4$, 100$\mu$M $MgSO_4$.$7H_2O$, 10$\mu$M $CaCl_2$.$2H_2O$, 1$\mu$M $FeSO_4$.$7H_2O$, 1$\mu$M $MnSO_4$.$4-5H_2O$, initial pH 7 and 200 rpm at 3$0^{\circ}C$. The purification of the antimicrbial substance was performed by silica gel column chromatographys, and fraction with TLC $R_f$ 0.77 value represented good antimicrobial activity. The crude antimicrbial substance was stable within a pH range of 3-10 and temperature range of 4$^{\circ}C$-121$^{\circ}C$ autoclaving. This crude antibacterial substance acted as bacteriolytic agent against Vibrio cholerae non-Ol ATCC 25872, and also exhibited excellent properties, when the substance was demonstrated against many other gram-positive, gram-negative bacteria, yeast and fungi.

  • Lactobacillus acidophilus A12 was isolated from chicken feces for use as an immunostimulating livestock probiotic. The purpose of this study was to optimize the production of L. acidophilus A12 using response surface methodology (RSM). Initially, the influence of growth medium was studied in terms of carbon sources (glucose, fructose, lactose, glycerol, sucrose, ethanol, and mannitol), nitrogen sources (beef extract, yeast extract, malt extract, and tryptone), and inorganic salts ($CaCl_2$, $MgSO_4$, $KH_2PO_4$, $(NH_4)_2SO_4$, $FeSO_4$, and NaCl). Through one factor-at-a time experiment, lactose, yeast extract, and $CaCl_2$ were determined to be the best sources of carbon, nitrogen, and inorganic salt, respectively. The optimum composition was found to be 17.7 g/L lactose, 18.6 g/L yeast extract, and 0.9 g/L $CaCl_2$. Under these conditions, a maximum cell density of 9.33 Log CFU/mL was produced, similar to the predicted value.

  • In the present study, ROS detection of L5178Y cells that were treated with twenty test compounds in order to find out hydrogen peroxide ($H_2O_2$) induction for genotoxicity and carcinogenic toxicity. Twenty test compounds were consist of four classes, such as genotoxic carcinogens, genotoxic noncarcinogens, nongenotoxic carcinogens, and nongenotoxic noncarcinogens. Genotoxic carcinogens are 1,2-dibromoethane, glycidol, melphalan, diethylstilbestrol and urethane. Genotoxic noncarcinogens are 8-hydroxyquinoline, emodin, acetonitrile and diallylphthalate, L-ascorbic acid. Nongenotoxic carcinogens are methyl carbamate, O-nitrotoluene, 1,4-dioxane, tetrachloroethylene and 2,3,7,8-tetrachlorodibenzo-p-dioxin. And nongenotoxic noncarcinogens are D-mannitol, 1,2-dichlorobenzene, caprolactam, bisphenol A and chlorpheniramine maleate.

Isolation, Identification and Optimal Cultrul Condition of Antioxidant Producing Bacterium Isolated from the Marine Sources

  • The isolated strain, SC2-1 was Gram-positive, catalase positive, facultatively anaerobic, oxidase negative, motile and small rods. The strain utilized sucrose, dextrose, fructose, mannitol and maltose as a sole carbon and energy source and sodium chloride required for the bacteria growth. The radical scavenging activity of the culture supernatants was determined by DPPH (1,1-diphenyl-2-picrylhydrazyl) method. This bacterium was identified based on cellular fatty acids analysis and 16S rDNA sequencing then named Exiguobacterium sp. SC2-1. The optimum culture conditions for production of antioxidant were $25^{\circ}C,$ pH 7.8 and NaCl concentration were 4%. The modified optimal medium compositions were maltose 2.5% (w/v), yeast extract 1.5% (w/v) and $KH_2PO_4$ 0.05% (w/v). Free radical scavenging activity of under optimal culture conditions were 93%.

  • This experiment was carried out to produce cloned aniraals by nuclear transplantation in rabbits. The ovulated oocytes were collected from the oviducts between 14 and 15 hours after hGG injection. The denuded oocytes were used as nuclear recipient cytoplasm following enucleation by micromanipulation. The blastomeres separated from the 8-cell embryos were used as nuclear donor. The nucleated oocytes receiving a blastomere in the perivitelline space were electrically fused in the 0.28 M mannitol solution at 1.5 kV /cm, 60$\mu$sec for three times. The nuclear transplant embryos which were used and developed to 2- to 4-cell stage in vitro were transferred into the oviducts of synchronized recipient does. A total of 64 nuclear transplant embryos were transferred to 7 recipient does and produced three offspring(4.7%) from a foster mother 31 days after embryo transfer.

  • The biochemical properties of strains of Staphylococcus aureus isolated from cattle with peracute gangrenous mastitis, acute systemic mastitis and chronic mastitis were examined. Of 261 strains of Staphylococci isolated from quarters with clinical mastitis, 140 (53.6%) were classified as Staphylococcus aureus and 121 (46.4%) were coagulase-negative staphylococci. All the strains of Staphylococcus aureus isolated from quarters with peracute gangrenous mastitis and acute systemic mastitis showed production of alpha heamolysin, coagulase, lipase, phosphatase, nuclease and gelatinase, and fermentation of mannitol. However, of 114 strains of Staphylococcus aureus obtained from quarters with chronic mastitis, 64 (56.1%) possessed alpha lysin, and 43 (37.7%) produced lipase in egg yolk medium. The most common hemolytic pattern of the strains associated with chronic mastitis was ${\beta}{\delta}$ type.

  • Actinomycetes CS0707 has been isolated in soil sample from location in the Jeju province. Korea, and produces thermostable extracellular proteases. Actinomycetes CS0703 showed the highest protease activity at late exponential phase when grown in OSYM medium (oatmeal 2.0%, soybean meal 1 %, dried yeast 1 %, mannitol 1 %) at $48^{\circ}C$. Three forms of protease(Ta-1, TA-2 and Ta-3) were fractionated by Ultrogel AcA 54 column chromatography, and further purified through ammonium sulfate fractionation, ultramembrane filtration, and DEAE-sepharose CL-6B column chromatography.   • Peatlands are marginal agricultural lands due to highly acidic soil conditions and poor drainage systems. Drought stress is a big problem in peatlands as it can affect plants through poor root development, so technological innovations are needed to increase the productivity and sustainability of upland rice on peatlands. Rhizobacteria can overcome the effects of drought stress by altering root morphology, regulating stress-responsive genes, and producing exopolysaccharides and indole acetic acid (IAA). This study aimed to determine the ability of rhizobacteria in upland rice to produce exopolysaccharides and IAA, identify potential isolates using molecular markers, and prove the effect of rhizobacteria on viability and vigor index in upland rice. Rhizobacterial isolates were grown on yeast extract mannitol broth (YEMB) medium for exopolysaccharides production testing and Nutrient Broth (NB)+L-tryptophan medium for IAA production testing. The selected isolates identify using sequence 16S rRNA. The variables observed in testing the effect of rhizobacteria were germination ability, vigour index, and growth uniformity. EPS-1 isolate is the best production of exopolysaccharides (41.6 mg/ml) and IAA (60.83 ppm). The isolate EPS-1 was identified as Klebsiella variicola using 16S rRNA sequencing and phylogenetic analysis. The isolate EPS-1 can increase the viability and vigor of upland rice seeds. K. variicola is more adaptive and has several functional properties that can be developed as a potential bioagent or biofertilizer to improve soil nutrition, moisture and enhance plant growth. The use of rhizobacteria can reduce dependence on the use of synthetic materials with sustainable agriculture.

Evidence for Singlet Oxygen Involvement in Cell-free Myeloperoxidase/$H_2O_2$/ Chloride Sytem: Exclusion of Hydroxyl Radical Involvement (Cell-free Myeloperoxidase/$H_2O_3$/Chloride System에서 Singlet Oxygen이 관여한다는 실험적 증거)

  • The present study was performed to determine oxygen metabolites involved in cell-free MPO/$H_2O_2/Cl^-$ system by observing the effects of their scavengers on NADH oxidation and ethylene production from methional by the action of MPO prepared from human leukocytes. It was clearly demonstrated that NADH was oxidized by the cell-free MPO/$H_2O_2/Cl^-$ system as evidenced by complete inhibition of the oxidation of the substrate in the presence of eiher azide or catalase, or by omitting $Cl^-$. The MPO-mediated oxidation of NADH was completely abolished by a $^1O_2$ quencher, DABCO but not by $OH{\cdot}$ scavengers, mannitol, benzoate, formate and methanol. In ethylene assay, no ethylene was detected from methional in the MPO/$H_2O_2/Cl^-$ system with evident production of the gas by xanthine-oxidase and $Cu^{++}-H_2O_2$ systems which are suggested to generate $OH{\cdot}$. From the results obtained, it is concluded that $^1O_2$ is a major mediator with exclusion of $OH{\cdot}$ involvement in the cell-free MPO-mediated oxidation.

Optimization of Cell Culture Condition for Erythritol Production by Penicillium sp. KJ8l (Penicillium sp. KJ 81에 의한 Erythritol 생산 최적 배양 조건)

  • Erythritol is of interest as a low calorie sweetner. Penicillium sp. KJ8l was screened for erythritol producer in nature. The effect of culture conditions on erythritol production by Penicillium sp. KJ81 was examined. This strain produced about 12 g/l erythritol and a small amout of glycerol. Erythritol was not produced from mannitol, arabinose, sorbitol, and xylose but from glucose, sucrose, fructose, mannose, lactose, maltose, and galactose. This strain was able to produce erythritol in a medium containing 60% sucrose but demonstrated the highest productivity of erythritol in a 30% sucrose medium. The highest yield in Penicillium sp. KJ8l was obtained when 0.5% ammonium sulfate was added to the medium containing 30% sucrose and 0.5% yeast extract. Penicillium sp. KJ81 produced 28.2 g/l erythritol when this strain was cultured in the medium containing 30% sucrose, 0.5% yeast extract, 0.5%$(NH_{4})_{2}SO_{4}$ 0.1% $KH_{2}PO_{4}$ and 0.01% $MgCl_{2}$ under the condition of 1 vvm aeration and 200 rpm agitation at $37^{\circ}C$ for 10 days in 5ι jar fermentor.
