사염화(四鹽化) 탄소(炭素)에 의한 흰쥐의 간장기능(肝臟機能) 장애(障碍)와 토룡(土龍)엑기스의 투여효과(投與效果) (Effects of Earthworm (Lamnodrilus gotai Hatai) Extract on the Hepatotoxicity of Carbon Tetrachloride in Rats)

  • These studies were attempted to investigate the therapeutic effect earthworm (Lamnodrilus gotai Hatai) extract (LS) on the liver damage induced by carbon tetrachloride ($CCl_4$) in rats. Male rats (Sprague-Dawley) were treated with sub-chronic concentration of $CCl_4$, which was 0.2 ml/kg of $CCl_4$ via peroral administration twice a week. LS administration to rats (6 ml/kg,po) prevented the development of fatty liver, necrosis and fibrosis induced by $CCl_4$. Furthermore, as compared with rats suspended after treated with $CCl_4$ only, LS treatment significantly decreased the increment in serum enzyme activities, liver enlargement and improved growth rate. This protective effect was observed when LS was given monthly for 4 months after $CCl_4$ treated for 2 months. These results suggest that earthworm extract has appreciable therapeutic effect on $CCl_4$-induced hepatotoxicity.

  • In vivo activity of recombinant human erythropoietin (rh-EPO) has been examined using polycythemic model in mice and acute hemorrhage model in rats. The number of reticulocytes in blood stream was increased after a single injection of rh-EPO depending on the dosage of rh-EPO in polycythemy model. It seemed that optimal dose of rh-EPO for polycythemic mice was around 1-10 U/kg. Rh-EPO also showed the effectiveness for increase of reticulocyte numbers both in male and female rats after bleeding. The number of reticulocytes and the change of hemoglobin concentration in the blood stream of normal rats has been examined after injection of rh-EPO. The maximum value of reticulocyte was observed on the 6th day of the injection in these normal rats. In addition, the increase of reticulocyte and the concentration of hemoglobin were dependent on the dosage of rh-EPO. The increase of hemoglobin concentration was continued to the 9th day after injection. In this study, the efficacy of rh-EPO was confirmed in both mice and rats.

Effects of Liquid Culture of Agaricus blazei Murill on Protein, Mineral Levels and Enzyme Activities in Rats

  • This study was performed to investigate the effects of liquid culture of Agaricus blazei Murill on the weight gains, food efficiency ratios, serum protein and mineral levels, and serum enzyme activities in growing male rats. Sprague-Dawley rats (7 weeks old) were given four different types of diets for a succeeding period of 6 weeks, respectively: a normal diet group, a control diet group (normal diet + 15% lard + 0.5% cholesterol), a 30% or 40% A. blazei diet groups (control diet + 30% or 40% A. blazei in water) according to the levels of A. blazei supplementation. The body weight gains, food efficiency ratios, and the liver of the rats fed control diet, 30% or 40% A. blazei diets were significantly increased compared to rats fed the normal diet, but those of rats fed the 30% and 40% A. blazei diets were similar to those of rats fed the control diet. The concentrations of total protein, albumin, glucose, and hematocrit value in serum of rats fed the control diet, 30% or 40% A. blazei diets were similar to those of rats fed the normal diet. The concentrations of urea and creatinine in serum of rats fed the 30% or 40% A. blazei diets were similar to those of rats fed the control diet. but the urea of rats fed the 30% and 40% A. blazei diets were significantly decreased compared to rats fed the normal diet. The concentrations of Ca, P, Fe and Mg, and $Ca^{++}$, $K^+$, $Na^+$ and $Cl^-$ of rats fed the control diet, 30% or 40% A. blazei diets were similar to those of rats fed the normal diet. There were no differences in the activities of aspartate aminotransferase, alanine aminotransferase, $\gamma$-glutamyltranspeptidase and alkaline phosphatase in the serum among the experimental groups. In conclusion, the rats fed the A. blazei maintained normal protein and mineral levels, and enzyme activities of serum. But the A. blazei feeding could not decrease the body and liver weights in the rats fed high cholesterol diets.

대두(생콩)급여가 당뇨쥐의 혈당 및 지질농도에 미치는 영향 (Effect of Raw Soy Flour(yellow and black) on Serum Glucose and Lipid Concentration in Streptozotocin-induced Diabetic Rats)

  • This study investigated the effect of raw soybean yellow(RSY) and raw soybean black(RSB) on body weights, pancreas weights, serum glucose and lipid concentrations of boty normal and diabetic male rats(Sprague-Dawley, body weight 338.8$\pm$19.2g). For each experimental, some rats were injected with streptozotocin intraperitoneal(i.P.) to induce diabets, and other rats were injected with buffer i.P. as a control group. Body weight gain(BWG) was lower in rats fed both RSY and RSB diet(3.4~5.0g BWG) than in those fed control diet(58.1g BWG). The diabetic rats showed significant decrease of bo요 weight (-50~-67g) compared with the control rats. RSY and RSB feeding resulted in greater pancreatic weight relative to body weight, and diabetic rats fed RSY and RSB had heavier pancreatic weights relative to body weight than control rats. The levels of fasting serum glucose in diabetic rats fed both RSY and RSB decreased by 42 and 31%, respectively, compared with the diabetic control rats, but they increased by 80 and 110% compared with the normal control rats. The concentrations of total lipid, triglyceride, phospholipid and total cholesterol in serum of diabetic rats fed RSB diet were lower than those in control rats, but serum HDL-cholesterol, HDL-cholesterol/total cholesterol ratio were not affected by raw soybeans feeding. Atherogenic index wa lower in diabetic rats fed RSY and RSB diet than those in control rats. The concentrations of total lipid, triglyceride and phospholipid in liver were lower in raw soybean groups than those in control group. The concentrations of cholesterol in liver of groups treated raw soybean were decreased compared with the control group.

3-MCPD의 생식ㆍ발생독성에 관한 연구 (Study on the Reproductive and Developmental Toxicity of 3-MCPD)

  • 3-Monochloro-1,2-propanediol(3-MCPD) is a toxic compound, often present in different foods containing acid hydrolyzed(AH) protein, like seasonings and savory food products. The purpose of the present study was to investigate the effects of 3-MCPD on male fertility, sperm and testosterone secretion. In vivo male fertility test was performed for observing the adverse effects of 3-MCPD on the function of male reproductive system and pregnancy outcome. 0.01, 0.05, 0.25, 1 and 5 mg/kg b.w. of 3-MCPD was given daily by gavage to groups of 15 adult male SD rats for 4 weeks. At the end of pre-treatment period, males were mated overnight with normal females. Following morning, males demonstrating successful induction of pregnancy were sacrificed on that day to assess sperm parameters and histopathology of reproductive organs. The resulting pregnant females were sacrificed on day 20 of gestation to evaluate pregnancy outcome. As a result, four-week paternal administration with 3-MCPD resulted in adverse effects on male fertility and pregnancy outcome without remarkable histopathological changes in testes and epididymides; sperm motility, copulation index and fertility index were markedly decreased in the treated group and numbers of live fetuses showed steep dose-response curves. Also, spermatogenesis was investigated in this experiment. However, no effect was observed on production of sperm in testes treated with 3-MCPD for 4 weeks. Hormone assay was performed for observing the effects of 3-MCPD on testosterone and luteinizing hormone (LH) in blood and testes of male SD rats and cultured primary Leydig cell. In result, significant changes of related hormones did not observed by treatment of 3-MCPD. These results indicated that paternal treatment with 3-MCPD induced spermatotoxic effect, which caused an antifertility on male.

당뇨 유발 흰쥐에서 여주열매 첨가 시 혈당과 콜레스테롤 수준에 미치는 영향 (Effect of Bitter Melon on Plasma Blood Glucose and Cholesterol Levels in Streptozotocin Induced Diabetic Rats)

  • This study was designed to examine the effects of bitter melon (BM) on the plasma blood glucose and cholesterol levels in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats through an injection of streptozotocin (STZ) dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight. Sprague-Dawley rats were then fed for four weeks, with the experimental groups receiving a modified diet containing 5% or 10% powder derived from BM. The experimental groups were divided into 4 groups, consisting of the normal control group, STZ-control group and diabetic fed with BM 5% & 10% treated groups. The rats' body weight, blood glucose and cholesterol values were measured along with the hematocrit (Hct) values and aminotransferase activities. Body weight losses were observed in the diabetic groups, whereas the control rats gained weight. There were significant differences in kidney weight between the control group and the diabetic groups. The Hct levels of the diabetic BM-treated group were significantly higher than the STZ-control group. Aspartate aminotransferase activity was lower in the non-diabetic group compared to the diabetic experimental groups. Further, the blood glucose was significantly decreased in the 5% & 10% BM of the diabetic group. There were no significant difference in cholesterol levels among the diabetic groups. These results indicate that the supplementation of bitter melon may have a favorable influence on reducing the blood glucose level in STZ-induced diabetic rats.

백서(白鼠)에 있어서 식이배합(食餌配合)이 혈액내(血液內) Cholesterol에 미치는 영향 (The Effect of Serum Cholesterol Levels of Experimntal Rats fed by Vit. E, Garlic and different the Levels of Proteins in their Diet)

  • Biochemical studies such as growth rate, blood cholesterol and ascorbic acid contents in various organs of albino male rats were studied in the Garlic, Vitamin E, high and low protein diet fed groups. The results of this study were summarized as follows : 1) Either single 10% or 30% protein diet fed rats were not shown growth rate properly. In supplementation of Vitamin E or Garlic on 30% protein growth rate was a more increased than that of the 10% protein diet group. 2) The rate of food consumption of rats fed a 10% or 30% protein diet supplemented with Vitamin E and Garlic was more increased than that of the control Group receving 10% or 30% protein diet alone. 3) No essential difference was observed between the cholesterol level in blood of rats fed 10% protein diet supplemented with Vitamin E and Garlic and that of rats Receiving 10% protein diet alone. But the cholesterol level in blood of rats fed 30% protein diet supplemented with Vitamin E and Garlic was lower than that of control receiving 30% protein diet alone. 4) The Vitamin C contents in various organs of rats fed the diet supplemented with Vitamin E and Garlic seems to be a little higher than that of the control group fed the protein diet alone.

Role of Tyrosine Kinases in Vascular Contraction in Deoxycorticosterone Acetate-Salt Hypertensive Rats

  • It has been known that activation of tyrosine kinases is involved in signal transduction. Role of the tyrosine kinase in vascular smooth muscle contraction was examined in deoxycorticosterone acetate (DOCA)-salt hypertensive rats. Male Sprague-Dawley rats underwent uninephrectomy, one week after which they were subcutaneously implanted with DOCA (200 mg/kg) and supplied with 1% NaCl and 0.2% KCl drinking water for $4{\sim}6$ weeks. Control rats were treated the same except for that no DOCA was implanted. Helical strips of carotid arteries were mounted in organ baths for measurement of isometric force development. Genistein was used as a tyrosine kinase inhibitor. Concentration-response curves to 5-hydroxytryptamine (5-HT) shifted to the right by genistein in both DOCA-salt hypertensive and control rats. Although the sensitivity to genistein was similar between the two groups, the maximum force generation by 5-HT was less inhibited by genistein in arteries from DOCA-salt hypertensive rats than in those from controls. Genistein-induced relaxations were attenuated in arteries from DOCA-salt rats. Genistein affected the contraction to phorbol 12, 13-dibutyrate (PDBu) neither in DOCA-salt nor in control arteries. These observations suggest that tyrosine kinase is involved in 5-HT-induced vascular contraction, of which role is reduced in DOCA-salt hypertension.

음곡 상응부위에 시술한 방기 약침이 LPS로 유도된 흰쥐의 염증성 신손상에 미치는 영향 (The Effect of Stephaniae tetrandrae Radix Herbal-acupuncture at KI10 on LPS Induced Nephritis in Rats)

  • Objectives : This study aimed to evaluate the effects of Stephaniae tetrandrae Radix Herbal-acupuncture(SR-HA) at KI10(Umgok) on nephritis induced by lipopolysaccharide(LPS) in rats. Methods : Male SD rats (n=32) were allocated to four groups; normal, LPS, saline, and SR-HA. The rats in the LPS, saline, and SR-HA groups were injected intra-peritoneally with LPS (2 mg/kg) to induce nephritis. The rats in the saline and SR-HA groups were treated with saline injection and SR-HA respectively at KI10 three times a week. To evaluate the effects of SR-HA at KI10 on nephritis in rats, the authors evaluated the numbers of WBC and neutrophils in blood, and the levels of creatinine, TNF-${\alpha}$, CINC-1 in serum, creatinine, total protein in urine and renal MPO. Results : SR-HA at KI10 significantly inhibited the increase of WBC and neutrophils in blood, TNF-${\alpha}$, CINC-1 in serum, creatine, total protein in urine and MPO in kidney of LPS-stimulated rats. Conclusions : SR-HA at KI10 may have therapeutic effects on nephritis in LPS-stimulated rats.

두릅추출물이 정상쥐 및 당뇨쥐에 미치는 영향 (Effect of Saponin from the Shoot of Aralia elata in Normal Rats and Steptozotocin Induced Diabetic Rats)

  • The purpose of this study was to investigate the effect of saponin in the shoot of Aralia elata on serum lipid level and glucose in streptozotocin(STZ) induced diabetic rats. Sprague Dawley normal male rats weighing 70$\pm$5g were randomly assigned to normal(control group), crude saponin(S group), and shoot of Aralia elata(D group). Experimental diets have been fed for 6 weeks. STZ induced diabetic rats were classified to diabetic control(DC group) and crude saponin(DS group). Diabetic rats were experimentally induced by intravenous injection of STZ(65mg/kg of body weight) dissolved in citrate buffer(pH 4.5). DS group has been i.p. injected with crude saponin solved in phosphate buffer(pH 7.0, 10mg/100g body weight) and DC group fed for 10 days. Body weight decreased significantly in crude saponin group. Feed intakes and feed efficiency ratio were not significantly different among C, S, and D group. The crude saponin group has indicated the lowest values of serum total cholesterol, glucose, and triglyceride. However, the values of serum glucose and triglyceride were not significant. Insulin levels among the crude saponin group, the shoot powder group, and the control group were not significantly different. When STZ induced diabetic rats have i.p. injection of crude saponin, the crude saponin has reduced the serum glucose but it is not been significant.
