A 4-Week, Repeated, Intravenous Dose, Toxicity Test of Mountain Ginseng Pharmacopuncture in Sprague-Dawley Rats

  • Objectives: Mountain ginseng pharmacopuncture (MGP) is a pharmacopuncture made by distilling extract from mountain cultivated ginseng or mountain wild ginseng. This pharmacopuncture is injected intravenously, which is a quick, lossless way of strongly tonifying Qi function. The present study was undertaken to evaluate a 4-week, repeated, intravenous injection, toxicity test of MGP in Sprague-Dawley (SD) rats. Methods: Twenty male and female 6-week-old SD rats were used as subjects. We divided the SD rats into 4 groups: the high-dosage (10 mL/kg), medium-dosage (5 mL/kg), low-dosage (2.5 mL/kg) and control (normal saline) groups. MGP or normal saline was injected intravenously into the caudal vein of the rats once daily for 4 weeks. Clinical signs, body weights, and food consumption were monitored during the observation period, and hematology, serum biochemistry, organ weight, necropsy, and histological examinations were conducted once the observations had been completed. Results: No mortality was observed in any of the groups during the observation period. No changes due to MGP were observed in the experimental groups regarding clinical signs, body weights, food consumption, hematology, serum biochemistry, organ weight and necropsy. No histological changes due to MGP were observed in any of the male or female rats in the high-dosage group. Conclusion: During this 4-week, repeated, intravenous injection, toxicity test of MGP in SD rats, no toxic changes due to MGP were observed in any of the male or female rats in the high-dosage group. Thus, we suggest that the high and the low doses in a 13-week, repeated test should be 10 mL/kg and 2.5 mL/kg, respectively.

Effect of Estrone Treatment on Proteinuria in Maturing Lean and Obese Male SHR/Mcc-cp Rats (Estrone 투여가 SHR/Mcc-cp 계통의 정상체중과 비만한 성장기 숫쥐들에서 단백뇨 증상에 미치는 영향)

  • Lean and obese male spontaneously hypertensive(SHR/Mcc-cp) rats were fed a ground rat chow diet with or without 0.001% estrone added from 6 to 18 weeks of age. Urine samples were collected weekly with 24 hour fasting. Both control lean and obese rats showed significantly higher urinary protein than their estrone treated counterparts. Treatment with 0.001% estrone diet was found to reduced the proteinuria in the maturing lean and obese SHR/Mcc-cp rats. Peaks in urinary protein level were noted at 16 weeks of age in both lean and obese control rats. Both control and estrone treated lean rats showed higher proteinuria than the obese rats. Therefore, obesity does not appear to be a contributing factor to the proteinuria in young male SHR/Mcc-cp rats.

The Effect of Flavonoid Fraction Extracted from Rhus verniciflua Stokes on Sexual Behavior in SD Male Rats (옻나무 플라보노이드가 수컷 백서의 성행동에 미치는 영향)

  • Rhus verniciflua Stokes (RVS) has been used as a food supplement and a traditional herbal medicine for a men's sexual enhancement. In this study, we prepared a flavonoid fraction (RWE) from a hot water extract of RVS and its influence on sexual behavior was studied in male rats which were orally administered varying doses of RWE for 2 weeks. All doses of RWE stimulated sexual behavior in male rats such as reducing mounting latency and enhancing mount frequencies. However, intermission numbers were not changed and none of group can observe the ejaculation during behavioral testing time 5 min. Testosterone levels were Increased about 0$\∼$53$\%$ and 92$\∼$164$\%$ by the treatment of RWE for 1 and 2 weeks, respectively. However, estrogen levels in male rats tended to decrease in a dose dependent manner of RWE. At the dose of RWE 200 mg/kg, estrogen level was reduced to 77.5$\%$ and 70.3$\%$ of control after 1 and 2 weeks treatment. These findings suggest that the Rhus flavonoid fraction can stimulate the androgen-dependent male sexual behavior and it can be applied to the material of functional food for enhancing the sexual function.

  • Zearalenone (ZEA), a nonsteroidal estrogenic mycotoxin, is known to cause toxicity of testis in male rats. To investigate whether apoptosis is involved in ZEA-induced testicular toxicity and to identify the stage and target germ cell type, 10-weeks-old Sprague-Dawley male rats were treated with a single intraperitoneal (i.p.) dose of ZEA (5mg/kg) and euthanized at 3, 6, 12, 24, and 48 h subsequently.(omitted)

Effects of Cervi Pantotrichum Cornu and Cervi Cornu on Longitudinal Bone Growth in Adolescent Male Rats (녹용(鹿茸)과 녹각(鹿角)의 성장기 흰쥐 장골 길이성장에 대한 효과)

  • Objectives : This study was designed to investigate the effects of Cervi Pantotrichum Cornu and Cervi Cornu on the growth of longitudinal bone in the adolescent male rats. Methods : Longitudinal bone growth was measured by fluorescence microscopy. To examine the effects on the growth plate metabolism, the heights of growth plate and the induction of local bone morphogenetic protein-2 were measured. Results : Treatment of Cervi Pantotrichum Cornu significantly enhanced longitudinal bone growth compared with control group. However, Cervi Cornu did not show the significant effects. Conclusion : Cervi Pantotrichum Cornu enhanced longitudinal bone growth and promoted the induction of local bone morphogenetic protein-2 of growth plate in adolescence male rats.

Rat Single Oral Dose Toxicity Test of Armeniacae Semen (Including Endocarp) (행인(杏仁)의 랫트 단회 경구 투여 독성시험)

  • Objectives : The object of this study was to obtain acute toxicity information (single oral dose toxicity) of Armeniacae Semen (AS), a medicinal herb used for treating constipation and various respiratory diseases, in rats. Methods : In order to observe the $LD_{50}$ (50% lethal dose), approximate lethal dosage (ALD) and target organs, AS aqueous extracts were orally administered once to female and male Sprague Dawley rats at dose levels of 2,000, 1,000, 500 and 0 (control) mg/kg (body weight). The mortality, changes in the body weight, clinical signs and gross observation were monitored for 14 days after single oral treatment of AS extracts, and the organ weights and histopathological findings of principle organs were observed after sacrifice. Results : After single oral treatment of AS 2,000 and 1,000 mg/kg, all (5/5; 100%) female and male rats died within 30 minutes after treatment, while no mortalities were observed in the female and male rats treated with 500 mg/kg of AS extract. Therefore, $LD_{50}$ in female and male rats was calculated as 741.95 mg/kg. Seizure, loss of locomotion, salivation, increases of respiration and heart-beat were observed after AS extract treatment, which were observed in all rats including the lowest dosage group, 500 mg/kg in the present study. In addition, lung congestion was visible in all mortal rats with AS 2,000 and 1,000 mg/kg, respectively. Conclusions : The results obtained in this study suggest that AS extract ranges in Class III, because the $LD_{50}$ and ALD in both female and male rats were calculated as 741.95 mg/kg and 500~1,000 mg/kg, respectively. However, AS extract should be carefully treated at clinical applications, because salivation, increase of respiration and heart-beat were also observed in the lowest dosage group, 500 mg/kg in the present study.

Repeated Intramuscular-dose Toxicity Test of Water-soluble Carthami Flos (WCF) Pharmacopuncture in Sprague-Dawley Rats

  • Objectives: Water-soluble carthami flos (WCF) is a new mixture of Carthami flos (CF) pharmacopuncture. We conducted a 4-week toxicity test of repeated intramuscular injections of WCF in Sprague-Dawley rats. Methods: Forty male and 40 female rats were divided into 4 groups of 10 male and 10 female SD rats: The control group received 0.5 mL/animal/day of normal saline whereas the three experimental groups received WCF at doses of 0.125, 0.25, and 0.5 mL/animal/day, respectively. For 4 weeks, the solutions were injected into the femoral muscle of the rats alternating from side to side. Clinical signs, body weights, and food consumption were observed; opthalmological examinations and urinalyses were performed. On day 29, blood samples were taken for hematological and clinical chemistry analyses. Then, necropsy was conducted in all animals to observe weights and external and histopathological changes in the bodily organs. All data were tested using a statistical analysis system (SAS). Results: No deaths were observed. Temporary irregular respiration was observed in male rats of the experimental group for the first 10 days. Body weights, food consumptions, opthalmological examinations, urinalyses, clinical chemistry analyses, organ weights and necropsy produced no findings with toxicological meaning. In the hematological analysis, delay of prothrombin time (PT) was observed in male rats of the 0.25- and the 0.5-mL/animal/day groups. In the histopathological test, a dose-dependent inflammatory cell infiltration into the fascia and panniculitis in perimuscular tissues was observed in all animals of the experimental groups. However, those symptoms were limited to local injection points. No toxicological meanings, except localized changes, were noted. Conclusion: WCF solution has no significant toxicological meaning, but does produce localized symptoms. No observed adverse effect level (NOAEL) of WCF in male and female rats is expected for doses over 0.5 mL/animal/day.

Protective Effects of Prunella Vulgaris on Glomerular Injury in Streptozotocin-Induced Diabetic Rats (하고초 추출물의 streptozotocin 유발 당뇨 랫트 사구체 손상 개선 효과)

  • Prunella vulgaris, well-known traditional medicinal plant, is used for the cure of abscess, scrofula, hypertension and urinary diseases. Diabetic nephropathy is the most common cause of end-stage renal disease. The pathological characteristics of diabetic nephropathy are glomerular and tubular basement membrane thickening. The aim of the present study was to evaluate the effect of Prunella vulgaris, on diabetic glomerular injury in streptozotocin-induced diabetes rats. Diabetes mellitus was induced by a single intraperitoneal injection of streptozotocin (STZ; 45 mg/kg) and confirmed by random glucose level higher than ${\leq}300mg/dL$. The experimental rats were divided into five groups: control group (Male SD rats), STZ group (Male SD rats injected STZ), Aminoguanidine group (Male SD rats injected STZ + AG 100 mg/kg/day), Low dose group (Male SD rats injected STZ + APV 100 mg/kg/day), High dose group (Male SD rats injected STZ + APV 300 mg/kg/day). AG or APVs were administered once a day for 8 weeks. Body weight and food/water intake were measured every four weeks. At the end of study, the kidneys were collected and cut into pieces for immunohistochemistry and western blot analysis. Our study showed that body weight and water/food intake were no significant differences between untreated STZ-induced diabetic rat and APV treated-STZ rat. However, phosphorylation of receptor-regulated Smads (Smad3) was significantly decreased in APV treated-STZ rat as compared with the diabetic group. In addition, APV was improved nephrin level in kidney tissue. Therefore, we suggest that APV has a protective effect against STZ-induced diabetic glomerular injury.

The ProtectiveEffect of Oryeongsan, Geumgwe-sinkihwan, and Jwagwieum on Renal Injury in Rats with Unilateral Ureteral Obstruction (편측성 요관폐색으로 유발된 신장 질환 백서 모델에서 오령산, 금궤신기환, 좌귀음의 보호효과)

  • Ureteral obstruction can be causes of renal dysfunction and renal injury at late period of kidney pathology. The purpose of this study was to determine the protective effects of Oryeongsan (ORS), Geumgwe-sinkihwan (GSH), and Jwagwieum (JGE) in rats with unilateral ureteral obstruction (UUO). The animal models were divided into five groups randomly at the age of 5 weeks; Control group: SD male rats (n=10), UUO group: SD male rats with UUO surgery (n=10), ORS group: SD male rats with UUO surgery + ORS 200 mg/kg/day (n=10), GSH group: SD male rats with UUO surgery + GSH 200 mg/kg/day (n=10), JGE group: SD male rats with UUO surgery + JGE 200 mg/kg/day (n=10). Treatment with ORS, GSH, and JGE significantly ameliorate creatinine clearance(Ccr). The present results also showed that ORS, GSH, and JGE improved the morphological aspects of renal tissues. These prescriptions also reduced the expression levels of cytokines such as TNF-α, IL-1β, and IL-6. In Kidney, UUO increased the expression levels of inflamasome markers such as NLRP3, ASC, and Caspase-1. However, ORS, GSH, and JGE suppressed these levele. Treatment with these prescriptions reduced kidney inflammation markers such as Neutrophil Gelatinase Associated Lipocalin (NGAL) and kidney injury molecule -1 (KIM-1). Therefore, these findings suggest that ORS, GSH, and JGE has a protective effect on renal injury by alleviating renal inflammation and improving renal function in rats with UUO.

The Effect of Dietary Caffeine Supplementation on Plasma and Liver Lipid Concentrations in Male Rats (카페인 첨가 식이가 성장기 수컷 흰쥐의 혈중 및 간 지질에 미치는 영향)

  • The purpose of this study was to investigate the effect of dietary caffeine supplementation(0.033%/diet) on the plasma and liver lipid concentrations in rats. Twenty male Sprague-Dawley rats(body weight $200{\pm}10\;g$) were divided into two groups: the control group and the caffeine-supplemented group(0.033% caffeine-supplemented diet). All rats were fed an experimental diet, and deionized water was avaliable ad libitum for 6 weeks. Plasma lipid concentrations were measured with a commercial kit based on an enzymatic method, and liver lipid concentrations were measured using the Folch method. There were no significant differences in body weight gain, food intake or food efficiency ratio between the control and caffeine-supplemented group. The total cholesterol and LDL-cholesterol levels as well as the AI(athrogenic index) were significantly higher in rats fed the caffeine diet(0.033%) than in those fed the control diet. There were no significant differences in the liver total cholesterol and triglyceride levels between the control and caffeine-supplemented groups. Therefore, it seems possible that caffeine(0.033 g/100 g diet) may increase the plasma lipid concentration level in growing rats.
