Purpose: The aim of this study was to present new a model that allows the study of the bone healing process, with an emphasis on the biological behavior of different graft-to-host interfaces. A standardized "over-inlay" surgical technique combined with a differential histomorphometric analysis is presented in order to optimize the use of critical-size calvarial defects in pre-clinical testing. Methods: Critical-size defects were created into the parietal bone of 8 male Wistar rats. Deproteinized bovine bone (DBBM) blocks were inserted into the defects, so that part of the block was included within the calvarial thickness and part exceeded the calvarial height (an "over-inlay" graft). All animals were sacrificed at 1 or 3 months. Histomorphometric and immunohistochemical evaluation was carried out within distinct regions of interest (ROIs): the areas adjacent to the native bone (BA), the periosteal area (PA) and the central area (CA). Results: The animals healed without complications. Differential morphometry allowed the examination of the tissue composition within distinct regions: the BA presented consistent amounts of new bone formation (NB), which increased over time ($24.53%{\pm}1.26%$ at 1 month; $37.73%{\pm}0.39%$ at 3 months), thus suggesting that this area makes a substantial contribution toward NB. The PA was mainly composed of fibrous tissue ($71.16%{\pm}8.06%$ and $78.30%{\pm}2.67%$, respectively), while the CA showed high amounts of DBBM at both time points ($78.30%{\pm}2.67%$ and $74.68%{\pm}1.07%$, respectively), demonstrating a slow remodeling process. Blood vessels revealed a progressive migration from the interface with native bone toward the central area of the graft. Osterix-positive cells observed at 1 month within the PA suggested that the periosteum was a source of osteoprogenitor elements. Alkaline phosphatase data on matrix deposition confirmed this observation. Conclusions: The present model allowed for a standardized investigation of distinct graft-to-host interfaces both at vertically augmented and inlay-augmented sites, thus possibly limiting the number of animals required for pre-clinical investigations.
Although garlic has been reported to have impressive effects in lowering serum lipids, there have been controversial evaluations on these effects. To find the potential fator causing the inconsistency in the previous studies, we examined the effects of two types of garlic according to the producing-area (hangihyung garlic, nangihyung garlic) on serum lipid profiles and leptin level. Thirthy six of 4 wk old Sprague Dawley male rats fed high fat diet (40% of calories as fat) for 6 wks to induce obesity, and subsequently fed 5% garlic powder supplemented (HF+H: hangihyung garlic powder, HF+N: nangihyung garlic powder) high fat diets (w/w) for further 5 wk. For the comparison, normal control group fed AIN-76A diet (11.7% of calories as fat). Supplementation with hangihyung and nangihyung garlic resulted in a significant reduction of high fat induced body weight gain, white fat (i.e., epididymal, visceral and peritoneal fat) development, adipocyte hypertrophy and the development of hyperinsulinemia and hyperliptinemia. Serum triglyceride and total cholesterol level was greatly reduced by hangihyung garlic supplementation (p<0.05). The HDL-cholesterol level was increased by dietary hangihyung and nangihyung garlic. There were slight non-significant decreases in triglyceride and total cholesterol of HF+N group as compared to those of HF group. Leptin level of HF+H group was found to be significantly lower than HF group (p<0.05). There was no significant difference among N group and HF+N group. These results suggest that hangihyung garlic may lead to the higher activity in improving lipid profiles than nangihyung garlic. Whether the hypolipidemic effect of garlic increases in a species-dependent has yet to be determined and awaits further research.

The purpose of this study was to assess the effect on mandibular growth of botulinum toxin type A (BTXA) injection into the unilateral massester muscle of growing rats at three different growing stages. Thirty six male Sprague-Dawley rats were divided into three groups according to the age (group 1: 4 week-old, group 2: 5week-old, group 3: 6week-old). Then each group was randomly divided into 3 subgroups (control group, unilateral injection group, bilateral injection group). Experimental animals were sacrificed after 4 weeks. Then the jaw measurements were evaluated. The results were as follows: 1. In the group 1, mandibular body length, condylar height and coronoid process height of the unilateral group(BTXA side) and the bilateral group were shorter than those of the control group (p<0.05). 2. In the group 2, anterior region height, condylar height, coronoid process height of the unilateral group(BTXA side) and the bilateral group were shorter than those of the control group (p<0.05). 3. In the group 3, mandibular body length, condylar height, coronoid process height of the unilateral group(BTXA side) and the bilateral group were shorter than those of the control group (p<0.05). 4. There was no significant difference in mandibular measurements between the control side and the injection side in the unilateral group in all age groups (p>0.05).
Fumonisin B$_1$ is hepatotoxic in all species, but liver carcinogenic and nephrotoxic in rat. Our objective was to investigate the effects of multiple iv dose of FB$_1$. Male Sprague-Dawley rats were injected intravenously (iv) with FB$_1$ at 1 mg/kg singly (T1), or daily for 2 (T2) or 3 (T3). T1 rats did not show any cytotoxicity in both liver and kidney. However, the most dramatic change occurred in this group was mitotic figures in liver, which increased 5.5-fold to that of control. Hepatotoxic effects were shown in T3, based on histopathology and serum chemistry. A few scattered single cell deaths occurred primarily in the centrilobular area of the liver in T2. Similar but more lesions in liver and a small number of degenerating cells with hypereosinophilic cytoplasm in outer stripe of medulla of kideny were found in T3 rats. Serum chemical profiles included liver enzymes increased, in which cholesterol was very sensitive. This study suggests that multiple exposure of low dose FB$_1$ cause cytotoxic in the liver earlier time point than kideny. FB$_1$$ also stimulates mitosis in liver that may be associated with carcinogenesis.

Excessive intake of trans fats is known to be a risk factor for cardiovascular diseases. Previously, we have shown that green tea extract (GTE) lowers the intestinal absorption of lipids and lipid-soluble compounds in rats. This study was conducted to investigate a possible role of GTE on the lymphatic absorption of elaidic acid, a major trans fat in the diet. Adult male Sprague-Dawley rats with lymph duct cannula were infused via an intraduodenal catheter at 3.0 mL/hr for 8 hr with a lipid emulsion containing $180.0{\mu}mol$ elaidic acid, $400.0{\mu}mol$ triolein, $20.7{\mu}mol$ cholesterol, $3.1{\mu}mol$${\alpha}$-tocopherol, and $396.0{\mu}mol$ sodium-taurocholate with or without (control) GTE in a 24 mL PBS buffer (pH, 6.4). Simultaneously, lymph was collected hourly for 8 hr via the lymph duct cannula. There was a significant difference in lymph flow by GTE. Also, the lymphatic absorption of elaidic acid for 8 hr was significantly lower in rats infused with GTE than in those not infused with GTE. Similarly, GTE infusion decreased the lymphatic outputs of cholesterol, oleic acid, and phospholipids, compared with the controls. These findings provide clear evidence that GTE has an inhibitory effect on the intestinal absorption of elaidic acid and other lipids. Our work here provides the foundation for further studies to examine and evaluate dietary strategies to ameliorate dietary trans fats from the diet.

Capsaicin has been shown to affect lipid metabolism. However, it is currently not known whether capsaicin would lower the intestinal absorption of cholesterol. Thus, this study was conducted to investigate the effect of intraduodenally infused capsaicin on the lymphatic absorption of cholesterol and lipids in rats. Male Sprague-Dawley rats weighing 300-350 g were starved for 16 hr and the mesenteric lymph duct was cannulated. Each rat was infused at 3.0 mL/hr for 8 hr via the duodenal catheter with a lipid emulsion, which contained 33.3 kBq [$^{14}C$]-cholesterol, 20.7 μmol cholesterol, 452 μmol triolein, 3.1 μmol $\alpha$-tocopherol, and 396.0 μmol Na-taurocholate without (control) or with 5.0 mg capsaicin in 24 mL PBS buffer (pH 6.4). Simultaneously, lymph was collected hourly for 8 hr. There was no significant difference in lymph flow between the groups. However, the lymphatic absorption of 14C-cholesterol for 8 hr was significantly lower in rats infused with capsaicin than in those infused with no capsaicin. Also, the output of oleic acid for 8 hr was significantly decreased by capsaicin. However, the intestinal absorption of $\alpha$-tocopherol did not differ between the groups. The results indicate that the luminal infusion of capsaicin inhibits the intestinal absorption of cholesterol and lipids in rats.
There is considerable interests in the effect of regular, vigorous exercise, in particular weight training as a possible measure in improving myocardial function. The present investigation aimed to examine possible effect of a long-term weight training program on the heart in aging rats. Male rats aged 3-,10-, and 20-month-old were divided at random into a control (sedentary) and the exercise group. The training group was exercised for 5 days a week by 1 RM of ten times with weight training apparatus. This investigation examined the changes of the heart muscle relative to histological, ultrastructural, cytochemical, and stereological studies in rats. Quantity of lipofuscin pigments was clearly increased in the weight training group of 15-month-old rats, and mitochondrial degeneration, vacuolization, and interstitial proliferation were observed as well. In the weight training group of 25-month-old rats the frequency as separated intercalated discs, fraying myofibrils and hypercontraction band increased in number compared with the same 15-mon1h-old group. From the experimental result of glucose-6-phosphatase activity, the enzyme activities decreased in the weight training group of 15-month-old rats, and more decreased in the same 25-month-old group. In stereological study, both 15- and 25-month-old training groups, mitochondrial and myofibrilar volume densities significantly decreased, whereas interstitial volume density significantly increased. From the experimental results obtained in the present study, it is suggested that long-term weight training exercise do not cause any significantly qualitative and quantitative ultrastructural change of the heart muscle in the young. On the contrary, long-term weight training exercise stress may actually induce degenerative changes in the heart muscle in the old age.

Magnolia obovata exhibit several beneficial effects including anti-oxidant effects. Magnolia obovata is used as a therapeutic agent to stop hemorrhages and a tonic to promoted health in Korean and Chinese medicine. The pharmacokinetic profiles of the main Magnolia obovata is still not accurately investigated. In present study, I examined the effects of water extracts of Magnolia obovata on atherosclerosis induced high cholesterol diet in rats in serum and abdominal aorta. A total of 3-week old 9 male rats of Sprague-Dawley were divided into 3 groups and fed with the basal diet (normal group), high cholesterol diet (atherosclerosis induced group) for 8 weeks, high cholesterol diet supplemented with water extracts of Magnolia obovata (Magnolia obovata group) for 2 weeks. And rats were sacrificed, serum lipid level, abodominal aortic anti-oxidant activities and lipid peroxide were measured. These results indicated that serum total cholesterol, LDL-cholesterol, triglycerides concentration significantly lower in Magnolia obovata group than high cholesterol diet group. But HDL-cholesterol concentration had significantly higher in Magnolia obovata group than high cholesterol diet group. In conclusion, Magnolia obovata controls of lipid level of hyperlipidemia and hypercholesterolemia, thus suppresses the proliferation of intima in endothelial cells, which is risk factors of atherosclerosis induced high cholesterol diet in rats. I consider as Magnolia obovata is worth to using as atherosclerosis treatment.
The purpose of the present investigation was to evaluate the effects of swimming training on response of lipid peroxide (MDA) and superoxide dismutase (SOD) enzyme activity of hyperlipidemic rats. Twenty-five male SD rats (6 weeks old) were randomly divided into a control group and 4 swimming groups after hyperlipidemia induction for 4 weeks through a 1% cholesterol diet. Swimming groups were then divided into unloaded swimming group, low-loaded swimming group, moderate-loaded swimming group and high-loaded swimming group by swimming intensity, and made to swim for 6 weeks (6 days/week). The loaded swimming group rats among the swimming groups swam a lead weight equivalent to 0%, 3%, 5% and 7% of body weight attached to the base of the tail. All data were expressed as mean and standard deviation by using an SPSS/$PC^+$ program, and to evaluate the differences between groups, data were analyzed by one-way analysis of variance and Duncan multiple range test (${\alpha}$=0.05) was performed to test the significant levels of differences between groups. The conclusions obtained from this study were as follows: 1) all swimming groups had significantly lower levels of MDA than the control group (p<0.001). Among the swimming groups, the moderate-loaded group had a significantly lower level than the unloaded group, low-loaded group and high-loaded group (p<0.001). 2) all swimming groups had significantly higher levels of SOD than the control group (p<0.01). Among swimming groups, the unloaded group, moderate-loaded group and high-loaded group had significantly higher levels than the low-loaded group (p<0.01).

This study aimed to identify the effects of extracts of fermented Angelica gigas Nakai (A. gigas) with a Monascus purpureus strain on anti-obesity in a high-fat diet (HFD)-induced obesity rat model. Male Sprague-Dawley rats were randomly divided into seven dietary groups (n=8 per group), as follows: a normal diet group (N) and six HFD groups (C: control, HFD and no treatment; AG: HFD +10% A. gigas extracts; FAG2.5: HFD +2.5% fermented A. gigas extracts; FAG5: HFD +5% fermented A. gigas extracts; FAG10: HFD +10% fermented A. gigas extracts; GC: HFD + Garcinia cambogia extracts). FAG-fed rats exhibited effectively lowered rates of increasing body weight and visceral fat accumulation in the HFD-induced obesity model. The activities of several hepatic marker enzymes, including aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase (LDH), and alkaline phosphatase (ALP), were increased with HFD-induced obesity, but levels of these marker enzymes were significantly decreased in FAG-fed rats. The consumption of FAG reduced triglyceride concentrations in serum to normal levels. FAG-fed rats showed effectively increased leptin concentrations in the HFD-induced obesity model. HFD ingestion induced a significant increase in the thiobarbituric acid reactive substances (TBARS) levels, which was decreased in FAG-fed rats. Hematoxylin and eosin staining and Oil Red O staining of the liver showed that the lipid deposits were decreased via FAG feeding. Moreover, hematoxylin and eosin staining of epididymal adipose adipose tissue showed that the adipocyte were decreased by feeding FAG.
