Buffer-Optimized High Gradient Magnetic Separation: Target Cell Capture Efficiency is Predicted by Linear Bead-Capture Theory

  • High gradient magnetic separation (HGMS) is the most commonly used magnetic cell separation technique in biomedical science. However, parameters determining target cell capture efficiencies in HGMS are still not well understood. This limitation leads to loss of information and resources. The present study develops a bead-capture theory to predict capture efficiencies in HGMS. The theory is tested with CD3- and CD14-positive cells in combination with paramagnetic beads of different sizes and a generic immunomagnetic separation system. Data depict a linear relationship between normalized capture efficiency and the bead concentration. In addition, it is shown that key biological functions of target cells are not affected for all bead sizes and concentrations used. In summary, linear bead-capture theory predicts capture efficiency ($E_t$) in a highly significant manner.

Single Magnetic Bead Detection in a Microfluidic Chip Using Planar Hall Effect Sensor

  • In this study, we fabricate an integrated microfluidic chip with a planar Hall effect (PHE) sensor for single magnetic bead detection. The PHE sensor was constructed with a junction size of $10{\mu}m{\times}10{\mu}m$ using a trilayer structure of Ta(3 nm)/NiFe(10 nm)/Cu(1.2 nm)/IrMn(10 nm)/Ta(3 nm). The sensitivity of the PHE sensor was 19.86 ${\mu}V/Oe$. A diameter of 8.18 ${\mu}m$ magnetic beads was used, of which the saturation magnetization was ~2.1 emu/g. The magnetic susceptibility ${\chi}$ of these magnetic beads was calculated to be ~0.14. The diluted magnetic beads solution was introduced to the microfluidic channel attributing a single bead flow and simultaneously the PHE sensor voltage was measured to be 0.35 ${\mu}V$. The integrated microchip was able to detect a magnetic moment of $1.98{\times}10^{-10}$ emu.

  • In a past, the method in which it used the fluorescent material or it analyzes a gene was used in order to detect the Alzheimer's disease. However, in this paper, the magnetic bead is used in order to detect the Alzheimer's disease. The 'magnetic bead used in this paper is able to make the amyloid-beta and the selective binding known as cause of the Alzheimer's disease.

  • 스핀밸브 GMR(giant magnetoresistance) 구조를 갖는 Ta/NiFe/CoFe/Cu/NiFe/IrMn/Ta재료의 자유층에 의한 PHR (planar hall resistance) 특성을 이용한 자기 바이오센서를 제작하였다. PHR 소자는 사진식각 및 건식에칭 공정을 통하여 마이크로 사이즈로 제작되었다. 직경이 $2.8\;{\mu}m$인 단일 자기비드가 있는 경우와 자기비드가 없는 경우 자기장의 세기에 다른 PHR 신호를 측정하였으며, 직경이 $2.8\;{\mu}m$인 단일 자기비드 측정에 성공하였다. 따라서 본 연구에서 제작한 PHR센서는 자기비드 입자의 유무에 따른 출력 특성의 차이를 이용하여 단일 자기비드 측정이 가능한 고분해능 자기 바이오센서에 응용될 수 있다.

Immobilization of potassium copper hexacyanoferrate in doubly crosslinked magnetic polymer bead for highly effective Cs+ removal and facile recovery

  • A potassium copper hexacyanoferrate (KCuHCF) embedded magnetic hydrogel bead (HCF-Mbead) was synthesized via a facile double crosslinking methods of $Fe^{3+}$ ionic binding and freeze-thaw for effective $Cs^+$ removal. The HCF-Mbead had a hierarchical porous structure facilitating fast access of $Cs^+$ ions to embedded active sites. The adsorbent showed enhanced $Cs^+$ removal properties in terms of capacity (69.2 mg/g), selectivity ($K_d=4{\times}10^4mL/g$, 1 ppm $Cs^+$ in seawater) and stability (>99.5% removal in pH 3~11) with rapid magnetic separation. This study further opens the possibility to develop an efficient material that links the integration of adsorption and recovery.

Detection of Fish Virus by Using Immunomagnetic Separation and Polymerase Chain Reaction (IMS-PCR)

  • Immunomagnetic separation of virus coupled with .reverse transcription-polymerase chain reaction (IMS-PCR) was performed with infectious hematopoietic necrosis virus (IHNV). A DNA fragment of expected size was synthesized in the RT-PCR with total RNA extracted from IHNV inoculated CHSE-214. In a SDS-PAGE analysis, a protein band of over 70kDa was detected from non-infected cells and cells inoculated with IHNV and infectious pancreatic necrosis virus (IPNV). This protein was detected in the Western blot analysis probably because of non-specific reaction to monoclonal antibody against IHNV nucleocapsid protein. In the immunomagnetic separation, magnetic beads coated with monoclonal antibody against the IHNV nucleocapsid protein was incubated with supernatant from IHNV inoculated CHSE-214 cells. During this process, the non-specifically reacting protein could be removed by washing the magnetic bead with PBS in the presence of an external magnetic field, and viral proteins were detected from the remaining, cleaned magnetic beads. It was necessary to extract viral RNA from the captured virus particles before RT-PCR, and no DNA product was detected when the captured virus was only heated 5 min at $95^{\circ}C$. A PCR-product of expected size was synthesized from IMS-PCR with magnetic beads double coated either by goat anti-mouse IgG antibody -monoclonal antibody or streptavidin - biotin conjugated monoclonal antibody.

Simple Screening Method for Double-strand DNA Binders Using Hairpin DNA-modified Magnetic Beads

  • We designed an effective screening method for double strand DNA (dsDNA) binders using DNA-modified magnetic particles. Hairpin DNA was immobilized on the surface of magnetic particle for a simple screening of dsDNA binding materials in a solution containing various compounds. Through several magnetic separation and incubation processes, four DNA-binding materials, DAPI, 9AA, AQ2A, and DNR, were successfully screened from among five candidates. Efficiency of screening was demonstrated by HPLC analysis using a C2/18 reverse-phase column. In addition, their relative binding strengths were verified by measuring the melting temperature ($T_m$). If hairpin DNA sequence is modified for other uses, this magnetic bead-based approach can be applied as a high-throughput screening method for various functional materials such as anti-cancer drugs.

Magnetic Bead-Based Immunoassay on a Microfluidic Lab-on-a-Chip

  • This paper presents a basic concept of lab-on-a-chip systems and their advantages in chemical and biological analyses. In addition, magnetic bead-based immunoassay on a microfluidic system is also presented as a typical example of lab-on-chip systems. Rapid and low volume immunoassays have been successfully achieved on the demonstrated lab-on-a-chip using magnetic beads, which are used as both immobilization surfaces and bio-molecule carriers. Total time required for an immunoassay was less than 20 minutes including sample incubation time, and sample volume wasted was less than $50{\mu}l$ during five repeated assays. Lab-on-a-chip is becoming a revolutionary tool for many different applications in chemical and biological analysis due to its fascinating advantages (fast and low cost) over conventional chemical or biological laboratories. Furthermore, simplicity of lab-on-a-chip systems will enable self-testing capability for patients or health consumers overcoming space limitation.

Planar Hall Sensor Used for Microbead Detection and Biochip Application

  • The Planar Hall effect in a spin valve structure has been applied as a biosensor being capable of detecting $Dynabeads^{(R)}$ M-280. The sensor performance was tested under the application of a DC magnetic field where the output signals were obtained from a nanovoltmeter. The sensor with the pattern size of $50{\times}100{\mu}m^2$ has produced high sensitivity; especially, the real-time profiles by using that sensor revealed significant performance at external applied magnetic field of around 7.0 Oe with the resolution of 0.04 beads per $\mu m^2$. Finally, a successful array including 24 patterns with the single sensor size of $3{\times}3{\mu}m^2$ has shown the uniform and stable signals for single magnetic bead detection. The comparison of this sensor signal with the others has proved feasibility for biosensor application. This, connecting with the advantages of more stable and high signal to noise of PHR sensor's behaviors, can be used to detect the biomolecules and provide a vehicle for detection and study of other molecular interaction.
